Ceratopteris thalictroides is a water fern very sensitive to both dehydration and low temperature. This study focuses on the cryopreservation of this species by encapsulation-dehydration technique, in particular on the effects of pre-culture step, alginate bead size and the physical conditions of culture on the cryopreservation efficiency. Encapsulated and non-pre-cultured gametophytes did not survive cooling with liquid nitrogen. When cryopreservation was preceded by a 2-week period of pre-culture, regrowth reached 42.1%. Reduction in the size of the alginate bead, and culture in total darkness resulted in improved gametophyte regrowth capacity (75.5% or 81.7%, respectively). The best results (91.3%) were obtained when all factors tested occurred simultaneously. The gametophytes recovered very quickly and sporophytes were formed within 4 weeks after rewarming. These simple improvements can be used, not only for the cryopreservation of gametophytes in cryptogams but also for some recalcitrant species of seed plants.

Shoot tips from in vitro plants of four rose species were cryopreserved by the droplet vitrification method. Optimized conditions involved exposure to loading solution for 20 min, then treatment with plant vitrification solution (PVS2) for 20 min (Rosa agrestis, R. canina and R. dumalis) or 30 min (R. rubiginosa) followed by freezing in liquid nitrogen. Survival rate ranged from 78.3 to 95.1%, depending on the species. Regrowth rate of shoot tips was 50.5% for R. agrestis, 63.2% for R. rubiginosa, 71.4% for R. dumalis and 78% for R. canina. The preculture of donor plants in a medium with 0.25 цЫ sucrose facilitated the isolation of shoot tips and increased regrowth rate after cryopreservation. Plant regeneration was carried out in Murashige and Skoog medium with 1 цЫ 6-benzylaminopurine, 1.5 цЫ gibberellic acid and 0.087 M sucrose. Plants regenerated from cryopreserved shoot tips did not display morphological alterations in comparison with non-cryopreserved shoot tip - derived plants.

Opis:
The purpose of the research is to prepare a mathematical and numerical model for the phenomenon of heat transfer during cryopreservation. In the paper, two popular methods, slow freezing and vitrification, are compared. Furthermore, the basic model of thermal processes is supplemented by the phenomenon of phase transitions. To determine the temperature distribution during cryopreservation processes, one uses the heat transfer equation proposed by Pennes. An integral part of the energy equation is the substitute thermal capacity (STC) performed according to the concept named one domain method (fixed domain method), The numerical model is developed using the finite difference method (FDM) connected with directed interval arithmetic. The final part of the article contains the results of numerical simulations.

The aim of this study was to compare the cryoprotective effects of egg yolk from different avian species (hen, goose and quail) on post-thaw quality of dog semen. Total motility (TMOT) and progressive motility (PMOT) of frozen-thawed spermatozoa were not significantly differed among the extenders, but were higher in the quail-egg yolk based extender compared with extender containing hen or goose egg yolk. It was found that post-thaw sperm motion parameters, velocity VCL and ALH, were significantly higher in the quail-egg yolk based extender. No marked differences in post-thaw sperm plasma membrane integrity (PMI) and mitochondrial membrane potential (MMP) were observed among the extenders. In conclusion, the results of the present study suggest that goose or quail egg yolk is a suitable alternative to hen egg yolk for the cryopreservation of dog semen.

Embryogenic cultures of plants are exposed to various stress factors both in vitro and during cryostorage. In order to safely include the plant material obtained by somatic embryogenesis in combination with cryopreservation for breeding programs, it is necessary to monitor its genetic stability. The aim of the present study was the assessment of somaclonal variation in plant material obtained from embryogenic cultures of Picea abies (L.) Karst. and P. omorika (Pančić) Purk. maintained in vitro or stored in liquid nitrogen by the pregrowth-dehydration method. The analysis of genetic conformity with using microsatellite markers was performed on cotyledonary somatic embryos (CSE), germinating somatic embryos (GSE) and somatic seedlings (SS), obtained from tissues maintained in vitro or from recovered embryogenic tissues (ETc) and CSE obtained after cryopreservation. The analysis revealed changes in the DNA of somatic embryogenesis-derived plant material of both Picea spp. They were found in plant material from 8 out of 10 tested embryogenic lines of P. abies and in 10 out of 19 embryogenic lines of P. omorika after in vitro culture. Changes were also detected in plant material obtained after cryopreservation. Somaclonal variation was observed in ETc and CSE of P. omorika and at ETv stage of P. abies. However, most of the changes were induced at the stage of somatic embryogenesis initiation. These results confirm the need for monitoring the genetic stability of plants obtained by somatic embryogenesis and after cryopreservation for both spruce species.

The Oomycete pathogen is frequently sampled, isolated to pure cultures, stored, and char-acterized. The article presents methods for collection, storage and prep-aration of P. infestans isolates for inoculation of plant tissues, based on the publication by Zarzycka (2001), later updated and modified.

The aim of this study was to determine the effect of two variants of Tris-citric acid-fructose (TCF) extender containing whole hen egg yolk (TCF-HEY) and lyophilized lipoprotein fractions extracted from ostrich egg yolk (TCF-LPFo) on selected biological properties of cryopreserved sperm cells. Post-thaw percentage of motile sperm (MOT) was significantly higher (P<0.05) for TCF-HEY extender (66.3 ± 3.2%) than for TCF-LPFo extender (52.4 ± 3.4%). Moreover, there was no significant difference in the percentage of sperm with progressive motility (PMOT). Both diluents effectively preserved sperm plasma membrane integrity and mitochondrial function. However, it was observed that cryopreservation impaired the functionality of antioxidant sperm enzymes. The above was manifested by reduced SOD activity, in particular in samples preserved in the TCF-HEY extender, as well as decreased GPx activity. Both diluents inhibited the rate of lipid peroxidation in sperm plasma membrane during freezing-thawing. Our results suggest that LPFo is a satisfactory alternative to hen egg yolk in the extender used for canine sperm cryopreservation.

The aim of this study was to evaluate the influence of sucrose concentration in the preculture medium, as well as the duration of osmotic dehydration on the efficiency of chrysanthemum ‘Richmond’, ‘Lady Orange’ and ‘Lady Salmon’ cryopreservation by encapsulation-dehydration technique. For all cultivars tested, the best regrowth of cryopreserved shot tips was recorded with 0.25 M sucrose concentration and 10 µM ABA during a two-week preculture, followed by a 4- or 7-day osmotic dehydration. The survival rate ranged from 56.8–58.0% (the Lady group) to 63.6% (‘Richmond’). However, the ability to grow was smaller and reached 18.2–50.7%. It was found that higher sucrose concentration during the preculture slowed the growth of chrysanthemum shoot tips and led to an increased formation of multiple shoots (by activating axillary buds) or deformed adventitious shoots (incapable of further growth). The frequency of tissue hyperhydricity also increased, while the rhizogenesis efficiency decreased when higher sucrose concentration in the preculture medium was applied. The influence of osmotic dehydration duration on the explants morphogenetic response was cultivar-dependent.

To optimize the protocol, the shoot tips of Chrysanthemum × grandiflorum (Ramat.) Kitam. radiomutants have been cryopreserved using the encapsulationdehydration technique, following different approaches. In the experiment the influence of abscisic acid – ABA (0; 10; 20 and 30 μM), sucrose concentration (0.09; 0.25 and 0.5 M) and temperature (5ºC or 22ºC) during preculture were tested. Furthermore two dehydration methods, gradual and one-step, were considered. Since the best results were reported with 10 μM ABA + 0.09 M sucrose at room temperature and gradual dehydration, only this combination was selected for the next desiccation-adjustment step. From the time of air desiccation under laminar air-flow applied (0; 1; 2; 3; 4 and 5 hours), 3 hours provided the best results. Due to the growth inhibition on the hormone-free medium, the addition of plant growth regulators during that phase was also tested, confirming the need to apply cytokinins into the medium.
W celu optymalizacji procedury pąki wierzchotkowe radiomutantów chryzantemy wielkokwiatowej pochodzące z różnych warunków kultury in vitro poddane zostały krioprezerwacji techniką kapsułkowania-dehydratacji. W trakcie doświadczenia zbadano wpływ kwasu abscysynowego – ABA (0; 10; 20 i 30 μM), stężenia sacharozy (0,09; 0,25 i 0,5 M) oraz temperatury (5ºC lub 22ºC) w trakcie prekultury. Uwzględnione zostały dwie metody dehydratacji (stopniowa i jednoetapowa). Ponieważ najlepsze wyniki uzyskano na pożywce zawierającej 10 μM ABA + 0,09 M sacharozy w temperaturze pokojowej i po zastosowaniu stopniowej dehydratacji, tylko tę kombinację wykorzystano w kolejnym etapie – optymalizacji czasu desykacji. Spośród zastosowanych okresów suszenia (0, 1, 2, 3, 4 i 5 godzin), suszenie przez 3 godziny zapewniło najlepsze wyniki. W związku z zahamowaniem wzrostu roślin na pożywce pozbawionej regulatorów wzrostu określono wpływ auksyn i cytokinin na ten proces, potwierdzając konieczność stosowania tych ostatnich.
