Dabigatran with IUPAC name 3-({2-[(4-Carbamimidoyl-phenylamino)-methyl]-1-methyl-1H-benzoimidazole-5-carbonyl}-pyridin-2-yl-amino)-propionic acid, which can be used to prevent strokes in those with atrial fibrillation due to causes other than heart valve disease, and at least one additional risk factor for stroke (congestive heart failure, hypertension, age, diabetes, and prior stroke), with molecular formula C34H41N7H5 was studied in detail for functional group analysis with FTIR, characteristic absorbance by UV-NIR, thermal behavior by TGA-DTA-DSC, particle size and stability of the molecule with Nano particle size analyzer. The structure was confirmed by LC-MS/MS with ESI probe and mass was found in aggregation with the reported standard values. The purity of drug was determined by Prep. HPLC analysis. FTIR analysis showed the characteristic peak of carboxylic acid, UV-NIR analysis showed that the λmax was 224 nm with methanol as a diluent, the compound was found stable in the thermal analysis, the average particle size was found to be 25.2 nm, Z-average as 0.2 nm and zeta potential as -67.6 mv hence showed excellent stability in the zeta potential analysis as per ASTM standards D4187-82, American Society of Testing and Materials, 1985.

Opis:
Multiple sclerosis (MS) is a demyelinating, inflammatory, autoimmune disease of the central nervous system which affects most commonly young adults. It has wide spectrum of clinical and radiological presentations with relapses or steady progression. Recent years have brought new reports on the pathogenesis of MS. This systematized the current MS classification and created new parameters describing the course of the disease, such as activity and progression. Attention has been paid to the need for new drugs that focus on the treatment of progressive MS. Until now, the primary and secondary progressive MS have been somewhat forgotten, and most of modifying-disease drugs have been registered in the treatment of relapsing-remitting subtype. In recent years, not only new drug has been registered for the treatment of progressive MS (ocrelizumab) and another one is planned to be approved soon (siponimod), but also indications of old medicines (interferon-beta1b, cladribine, mitoxantrone, cyclophosphamide, azathioprine) have been extended. Despite intensive development, there is still a great need to seek new drugs that will stop the progression of disability in MS patients.

The aim of the dissertation was to determine (select) the best growth medium for a basket willow (Salix viminalis L.) which contains optimal concentration of kinetin. The plant material taken from three varieties of willows: Bjor, Jorr and Tora, which grew in the experimental field, was sterilized and placed on a complete MS (Murashige and Skoog, 1962) growth medium. The propagated plants were used in an experiment, where MS substrate was enriched by growth regulator – kinetin, in various concentrations (0.5 mg∙dm-3, 1 mg∙dm-3, 2 mg∙dm-3 and 3 mg∙dm-3). The substrate which contained 0.5 KIN was considered to be the best for willow’s propagation, because plants cultivated with usage of this medium have shown the largest mass, the highest number of shoots and leaves, and also the highest quantitative and elongation growth of the roots. Among examined varieties of Salix, the best one was Bjor variety, where evaluated parameters were distinctive in comparison to two remaining varieties.

The aim of the study was to determine an optimal composition of growth media for Drosera rotundifolia L. cultured in vitro. Adventitious shoots of Drosera rotundifolia originating from an established in vitro culture were placed on complete Murashige and Skoog (MS) (1962) medium, MS medium with nutrient content reduced to ¾ MS, ½ MS, ¼ MS, 1/6 MS and MS medium with selected macronutrient (KNO3, KNO3 + NH4NO3, KH2PO4 or CaCl2) content reduced by ½. The medium containing ¼ of standard MS nutrients was found the most effective, as plants in this variant had the greatest weight and developed the greatest number of adventitious shoots (37.1). It was also conducive to root growth and development. Flowering was only observed on complete MS, ¾ MS and ½ MS media and MS with reduced content of KNO3 or KNO3 + NH4NO3. Reducing mineral content to ¼ MS or more resulted in red coloration of glandular tentacles.

Opis:
Some new cyanopyridine derivatives have been synthesized and characterization of the synthesized compounds was done by IR, NMR and mass spectral data. The thermal properties of these compounds were studied by thermogravimetry. The thermal stability, melting temperature and some kinetic parameters such as energy of activation, frequency factor, order of reaction and entropy of activation were evaluated from thermograms. The degradation is single step process for some compounds whereas for others, it is multi step process.

A rapid, accurate and precise LC-MS method is described for the quantitative determination of pramipexole in human plasma matrix using ropinirole as internal standard. Pramipexole and ropinirole were extracted from plasma by liquid-liquid extraction technique. The method was validated over the concentration range of 100-2514 pg/mL. The method was found to have acceptable accuracy, precision, linearity and selectivity. The mean extraction recovery from spiked plasma samples was in the range of 79.415-87.00 %. The intra-day accuracy of the assay ranged from 98.924 to 112.236 % and intra-day precision ranged from 3.489 to 6.756 %. Inter-day accuracy and precision results for quality control samples ranged between 100.340 and 107.443% of nominal and precision is observed to be 3.970-5.714 %. The pramipexole was found to be stable after several stability studies. The proposed method yielded a quick, simple and reliable protocol for estimating pramipexole concentrations in human plasma.

Opis:
Successful in vitro propagation through ex vitro rooting mechanism has been achieved in Piper longum by nodal shoot segment cultures. Shoot tip explants were less proliferative compared to the nodal meristems. The explants were sterilized using 0.1% HgCl2 and cultured on Murashige and Skoog (MS) medium with various concentrations of growth regulators. MS medium supplemented with 1.0 mg/L 6-benzylaminopurine (BAP) was found suitable for bud breaking within four weeks. Kinetin (Kin) was not reported impressive compared to BAP in culture induction response. The shoots were multiplied and elongated on MS medium supplemented with 0.5 mg/L each of BAP and Kin and 0.1 mg/L indole-3 acetic acid (IAA), where the shoots were elongated up to 5.8 cm length. Rooting and acclimatization was achieved by ex vitro rooting methods using 300 mg/L indole-3 butyric acid (IBA) for 5 min, and 5.3 roots with 3.2 cm average length were observed. The rooted shoots were transferred to the greenhouse for acclimatization. The hardened plantlets were transferred to earthen pots and maintained in the greenhouse. Normal flowering was observed in micropropagated plants of P. longum.

In vitro propagation methods using nodal segments of young tissues of Hibiscus subdariffa plant have been established. The nodal shoot segments responded better than apical shoot tip explants. Murashige and Skoog (MS) semi-solid medium supplemented with 1.0 mg/L 6-benzylaminopurine (BAP) induced 4.2 ±0.24 shoots per node with 2.40 ±0.18 cm in length after four weeks of inoculation. About 92% explants responded on this medium combination. The shoots were further multiplied (8.2 ±1.37 shoots with 5.9 ±1.05 cm length) by repeated subculture of mother explants and freshly induced shoot clumps on MS medium with the reduced concentrations of BAP and Kinetin (0.5 mg/L each) with additives within 4 weeks. Half strength MS medium augmented with 2.0 mg/L indole-3 butyric acid (IBA) was found most favorable for root induction with 98% response. On this medium 7.5 ±0.61 roots with 4.8 ±0.23 cm length of roots were recorded. The regenerated plantlets were successfully hardened and field transferred.

Talipariti elatum (Sw.) Malvaceae, also known as Blue Mahoe or Mountain Mahoe, commonly named as Majagua azul or Majagua, with red or crimson flowers, is a medicinal tree traditionally used against cough, asthma, catarrh, and expectorant. Its flowers contain a lot different chemical compounds, mainly flavonoids. From red petals of the flowers a flavonol glucoside was isolated and characterized by TLC, UV, IR, HPLC-MS/MS and NMR spectroscopy. Structure analyses of that chemical component revealed that It have the identical glucoside moiety attached to a flavonol skeleton like gossypitrin (gossypetin-7-O--glucoside) but in different position for which the structure of gossypetin-3’-O-glucoside was deduced from HSQC, HMBC, COSY and NOESY correlations.

Gas Chromatography coupled with mass spectrometry was applied to study the fragmentation of selected flavonoids from the ethanolic extract of Smilax domingensis Willd., Smilacaceae, known as zarzaparrilla, after Soxhlet extraction during 20 hours. Compounds belonging to one of the major subgroups found in common plants, i.e. flavanols were studied. Following solvent extraction and derivatization using BSTFA, possibly two different metabolites from the same chemical group were characterized in one analytical run: (+)-catechin and (-)-epicatechin. For the very first time, qualitative data on these analytes in the drug were determined after detailed validation of a sensitive, cheap and reliable GC-MS method.

The essential oil from flowering Marrubium (Marrubium vulgare L.) an aromatic member of the Lamiaceae family, from Algeria, obtained by hydrodistillation, was analysed by GC/FID and GC/MS. The constituents were identified by their mass spectra and Kovats’ indices. Fivety (50) components in the oil of M. vulgare were identified. The results demonstrated that the major components of the essential oil were: 4,8,12,16-Tetramethyl heptadecan-4-olid (16.97 %), Germacrene D-4-ol (9.61 %), α- pinéne (9.37 %), Phytol (4.87 %), Dehydro-sabina ketone (4.12 %), Piperitone (3.27 %), δ-Cadinene (3.13 %), 1-Octen-3-ol (2.35 %) and Benzaldehyde (2.31 %). Essential oil of M vulgare was evaluated for its antibacterial activities against Gram-positive and Gram negative pathogenic bacteria: Bacillus subtilis, Micrococcus luteus, Escherichia coli and Klebsiella pneumoniae.

An effective protocol has been developed for the adventitious root culture of Cannon Ball tree (Couroupita guianensis Aubl.) from the leaves of in vitro germinated 30 days’ old seedlings. Seeds germinated on cytokinins and auxins supplemented with Murashige and Skoog (MS) medium. Half strength MS medium supplemented with 2.0 mg/L indole-3-butyric acid (IBA) was found the most suitable for the induction of adventitious roots (92%) from the midrib region of the leaves than the other auxins. The roots were proliferated on half strength MS liquid medium supplemented with IBA 0.5 mg/L. After eight weeks of inoculation the roots in the medium yielded 5.933 ±0.135 gm fresh and 2.731 ±0.010 gm dry biomass of adventitious roots. The growth rate was increased by 8-9 folds in this investigation.

Aims of this study were to investigation the presence of phytochemical compounds in the methanolic extract of Cressa cretica L. leaves by using GC-MS method and report the functional groups by using FT-IR spectroscopy. The identification of phytochemical compounds based on the peak area, retention time, molecular weight, molecular formula, MS fragment ions. Thirty four phytochemical compounds were identification in the methanolic extract of Cressa cretica leaves. The GC-MS analysis provide the existence of 5-Methyl-6-phenyltetrahydro-1,3-oxazine-2-thione, Lactose, 3-Deoxy-L-ribose-2,5-dibenzoate, Sarreroside, Pterin-6-carboxylic acid, Octadecadiynoic acid, methyl ester, d-Mannose, Dodecanoic acid, 3-hydroxy-, Geranyl isovalerate, Tetradecanoic acid , 6-epi-shyobunol, Paromomycin, Cis-9-Hexadecenoic acid, and others. The FT-IR analysis revealed the presence of Alkenes, aliphatic amines, nitro compounds, alkanes. These are chemical compounds are may be useful for various herbal formulation as antifungal, antibacterial, anti- inflammatory, anti-oxidant and others.

Few studies have shown that Cnestis ferruginea possess some therapeutic properties. The present study was aimed screening the extracts of the fruits and leaves of Cnestis ferruginea for phytochemicals using crude screening and gas chromatography-mass spectrophotometer (GC-MS) methods, and also antimicrobial activity against clinical isolates. Preparation of the extracts (aqueous and ethanolic), identification of isolates, and antimicrobial sensitivity were done using previously reported standard methodologies. Replicate readings for the antimicrobial sensitivity were analysed using analysis of variance at 95% level of significance. The result of the crude qualitative screening revealed the presence of phenol, terpenes, alkaloids, saponins, tannin and polyphenol in various amounts in both the leaves and fruits. Quantitative analysis using GC-MS revealed a total of 14 similar phytochemicals each in the leaves and fruits of the study plant in varying amounts. In the fruits, phenol had the highest concentration of 15.01%, followed by terpenes (10.64%), alkaloid (5.43%), and tannins (5.16%). Others were anthocyanins, phytate, phytosterol, steroids, saponins, cardiac glycosides, oxalate, flavonoids, cyanogenic glycosides and coumarin with concentrations that ranged from to 0.01- 4.21%. On the other hand, in the leaves, alkaloids had the highest concentration of 31.62%, followed by phenol (20.59%), phytate (15.18%), and tannin (12.34%). Others include were flavonoid, terpenes, phytosterol, cardiac glycosides, saponins, cyanogenic glycosides, oxalate, anthocyanins, steroids and coumarin with concentrations that ranged from 0.16-8.45%. The isolates were identified as Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa and Proteus spp. The results of the antimicrobial sensitivity revealed varying zones of inhibitions; however, it increased as the concentration of the extract increased. The observed highest zones of inhibitions were 33.86 mm for the fruit ethanolic extract and 30.56 mm for the aqueous extract of the leaves against Proteus and Staphylococcus aureus, respectively.

Garcina kola leaves were collected, washed, dried and powdered. The ethanol extracts were first prepared by the simple method of cold extraction. The extracts were then characterized by the Phytochemical analysis, Fourier transform infrared spectroscopy (FTIR) and Gas chromatography mass spectroscopy (GC-MS) to identify the fatty acids and heterocyclic compounds present in the leaf extracts. The FTIR analysis shows a variation of the peaks that indicate there is synergy among the functional groups of the extract. Moreover, the phytochemical analysis revealed the presence of alkaloids, flavonoids, phenol, saponins, tannins and steroids. The study reveals that the chemical constituents of Garcina kola leaf may be behind their medicinal values in phytomedicine.
