This study evaluated the effect of supplementing the freezing extender with exogenous antioxidants on apoptotic-like changes in post-thaw boar spermatozoa. A total of 36 ejaculates were resuspended in standard lactose-egg yolk-glycerol extender supplemented with antioxidant to final concentrations of 0 (as control), 2.5mM GSH (group I), 5.0 mM GSH (group II), 150 IU/mL SOD (group III), 300 IU/mL SOD (group IV), 200 IU/mL CAT (group V), 400 IU/mL CAT (group VI), 150 IU/mL SOD+200 IU/mL CAT (group VII), 300 IU/mL SOD+400 IU/mL CAT (group VIII). Sperm motility and apoptotic-like changes were determined before and after freeze-thawing. The various markers of apoptotic-like changes were measured: plasma membrane permeability by YO-PRO-1/PI assay, phosphatidylserine (PS) translocation across the plasma membrane using fluorescein-labeled Annexin-V, mitochondrial transmembrane potential detected by JC-1, and DNA fragmentation evaluated by TUNEL assay. The highest percentage of progressive motile sperm was noticed in group II (PM% 64.2±15.4) compared with control (PM% 36.8±5.5). The supplementation of 400 IU/mL CAT (group VI) revealed significant (P<0.01) reduction of apoptotic-like changes (YO-PRO-1⁺/PI⁻: 13.1±7.5%, AnV⁺/PI⁻: 9.9±4.1%) in frozen-thawed spermatozoa compared with extendersupplemented with200 IU/mL CAT (groupV). Irrespectiveof the concentrationused, SOD and CAT in combination (group VII and group VIII) significantly (P<0.01) improved post-thaw sperm survival compared with the control. Evaluation by TUNEL assay revealed that cryopreservation and thawing did not induce DNA fragmentation in boar spermatozoa.

A total of 100 Malassezia pachydermatis strains recovered from skin and mucosal membranes of dogs were evaluated for their adhesive properties. Two types of growth, related to colony morphology on Sabouraud agar, were observed (type I and II). The mean number of fungal cells attaching to canine buccal epithelial cells was found to be 17. The number of adhered cells was greater (statistically significant at the level of p < 0.01) in strains belonging to the type I.

The objective of this study was to determine the effect of different levels of dietary methionine on selected immune parameters of young turkeys. A total of 357 one-day-old female Hybrid Converter turkeys were randomly divided into three groups with low, medium and high level of dietary methionine (LM, MM, HM) of seven replicates each. Methionine was added to the basal diet (LM) at 0.16% (MM) and 0.32% (HM). At 17 days of age, seven birds per group were vaccinated against Ornithobacterium rhinotraheale (ORT) infection, with ORNITIN (ABIC) vaccine. At 28 days of age, blood was sampled and the birds were euthanized. The serum concentrations and activity of selected biochemical parameters, total IgY and IgM, and vaccine-induced antibody titers (IgY) against ORT were determined. The percentages of CD4⁺, CD8⁺, CD4⁺/CD8⁺ T cell subpopulations and IgM⁺ B cell subpopulation were determined in blood and organs by flow cytometry. Different supplementary levels of methionine had no significant effect on vaccine-induced antibody titers against ORT or total serum IgM and IgY levels, as well as on the percentages of peripheral blood T and B cell subsets. Increasing dietary methionine rates decreased the percentage of CD4⁺ T cell subpopulation, however it has increased the percentage of IgM⁺ B cell subpopulation in the spleen. Vaccination against ORT resulted in a significant decrease in the percentage of CD4⁺ T cell subset and an increase in the percentage of CD8⁺ T cell subset in the spleen. It could be concluded that MM turkeys have developed the most desirable values of immune parameters.

The aim of this study was to compare progesterone concentrations in bitches during ovulation with a different course of follicular collapse as determined by ultrasonography. The study was conducted on 36 animals of different breeds, from 2 to 7 years old, and from 2 to 44 kg of body weight. Ultrasound examinations of ovaries were conducted in the peri-ovulatory period until ovulation was detected. Based on USG scans the animals were divided into two groups: group A with fast follicular collapse within 1 day, and group B with gradual follicular collapse within 2-3 days. Progesterone measurements were performed in both groups on the day when the beginning of ovulation was diagnosed by USG. Follicular collapse in group A was observed in 27.8% of animals (n=10) and in group B in 72.2% (n=26). There were no statistical differences (p<0.05) in progesterone concentrations between groups A and B (6.68 ± 0.92 ng/ml and 6.81 ± 0.57 ng/ml respectively). From 31 bred bitches (natural mating or insemination) pregnancy was confirmed in 29 females (93.5%). These results provide information about the sufficient stability of progesterone concentration during ovulation regardless of the follicular collapse speed. Ultrasonography during the peri-ovulatory period combined with progesterone measurements allows progress in ovulation management.

The aim of this study was to compare the cryoprotective effects of egg yolk from different avian species (hen, goose and quail) on post-thaw quality of dog semen. Total motility (TMOT) and progressive motility (PMOT) of frozen-thawed spermatozoa were not significantly differed among the extenders, but were higher in the quail-egg yolk based extender compared with extender containing hen or goose egg yolk. It was found that post-thaw sperm motion parameters, velocity VCL and ALH, were significantly higher in the quail-egg yolk based extender. No marked differences in post-thaw sperm plasma membrane integrity (PMI) and mitochondrial membrane potential (MMP) were observed among the extenders. In conclusion, the results of the present study suggest that goose or quail egg yolk is a suitable alternative to hen egg yolk for the cryopreservation of dog semen.

Opis:
Both epidermal growth factor (EGF) and transforming growth factor (TGF) play an important physiological role in the processes of proliferation and differentiation of several different cell types. However, the expression profiles of these factors in domestic bitches endometrium are still poorly recognized. The aim of the present study was to identify and analyze the differential expression of these factors in various stages of the estrus cycle. Endometrial tissue from proestrus (n=17), estrus (n=10), day 10 diestrus (n=15), day 35 diestrus (n=18) and anestrus (n=25) was collected soon after ovariohysterectomy. Total RNA was isolated from the endometrium by means of Chomczyński and Sacchi method, treated by DNase I, and reverse- transcribed into cDNA. Quantitative analysis of EGF, TGFβ1, TGFβ2, and TGFβ3 cDNA was performed by real-time quantitative polymerase chain reaction (RT-PCR). EGF expression in canine endometrium was increased in the estrus stage as compared to proestrus (P<0.05), day 10 diestrus (P<0.05), day 35 diestrus (P<0.01) and anestrus (P<0.001). We also found the differences in EGF expression between day 10 and day 35 of estrus as well as between day 35 of estrus with anestrus (P<0.05, P<0.01, respectively). The TGFf1 transcript contents were also higher in estrus as compared to other stages (P<0.01). The TGFβ2 and TGFβ3 in the estrus stage was increased compared to proestrus, day 10 diestrus, day 35 diestrus and anestrus (P<0.05). We proved that expression of EGF and TGFβ transcript isoforms is related to the phase of estrus in bitches and therefore may be regulated by specific hormone concentrations during these periods. Our results confirm the hypothesis that these growth factors play a role in the regulation of biochemical changes in the endometrial tissues during the estrus cycle.

The objective of this study was to determine the effect of different dietary levels of sodium in diets with and without sodium chloride on mineral metabolism, including blood electrolyte levels and tibia mineralization parameters, in young turkeys (to six weeks of age). The influence of diets with a low (L), medium (M) and high (H) sodium content, at 0.34, 1.34 and 2.82 g/kg respectively, was compared. The content of chloride and potassium in turkey diets (1.7 - 5.9 and 11 g/kg, respectively) was above the recommended levels. The sodium-deficient diet L decreased the serum concentrations of sodium, chloride and phosphorus, and it increased the serum levels of calcium and magnesium in turkeys, compared with groups M and H. Relative to group L, different dietary sodium levels in groups M and H contributed to a similar increase in the body weights of birds (1.06 vs. 1.46 and 1.44 kg, p<0.001) and in the absolute (4.60 vs. 6.83 and 6.62 g, p<0.001) and relative tibia weight (0.42 vs. 0.46 and 0.46% body weight, p=0.031). No significant differences were found between groups with respect to the content of ash, calcium and phosphorus in tibia dry matter. Supplemental sodium increased the bone density index (from 50.6 to 68.4 and 66.3 mg/mm in groups L, M and H, respectively, p<0.001), the maximum bending moment (from 5.27 to 7.40 and 7.33 N/mm, p=0.002) and the minimum breaking strength of tibia (from 0.136 to 0.191 and 0.189, p=0.002). In conclusion, our study indicates that the applied dietary treatment with a moderate sodium level (1.34 g/kg) resulted in a rate of bird growth and tibia mineralization similar to those observed with the treatment with much higher Na content (2.82 g/kg).

The aim of the study was to determine the effect of age of cows, i.e. subsequent lactation and stage of lactation, on bioactive whey protein content in milk of dairy cows of six primary breeds kept in Poland. In all cases the significant correlations between lactoferrin concentration in milk and stage of lactation was stated. Its content gradually increased with the course of lactation but the changes in the content of this protein were highly dependent on breed of cows, what was indicated also by high interactions between breed and stage of lactation.

The objective of this study was to verify the hypothesis that increasing levels of dietary methionine can stimulate the mechanisms of cell-mediated and humoral immunity in young turkeys. The blood and organs involved in cell-mediated and humoral immune responses were analyzed in 8-week-old turkeys that had been vaccinated against Ornithobacterium rhinotraheale (ORT) infection (on days 17 and 48). The birds were fed diets with a low (LM), medium (MM) and high (HM) methionine content (0.45 and 0.40%, 0.60 and 0.51%, 0.71 and 0.57% in weeks 1 – 4 and 5 – 8, respectively). Dietary methionine supplementation led to a significant increase in body weights of turkeys at 56 days of age, from 3532 g in group LM to 3720 g in group MM and 3760 g in group HM (p=0.001). A significant increase in vaccine-induced antibody titers against ORT was noted in group HM relative to group LM (p=0.006). Increasing levels of methionine had no significant effect on total serum IgG nor IgM levels and most serum biochemical parameters, TP, ALB, GLOB, GLU, AST, ALP, P and Ca. In comparison with group LM, group HM turkeys were characterized by a lower percentage of IgM⁺ B cell subpopulation in the blood and bursa of Fabricius. The percentages of CD4⁺ and CD8⁺ T cell subpopulations in the bursa of Fabricius in group HM were significantly different from those found in groups LM and MM. The highest percentages of CD4⁺ T cells and CD8⁺ T cells in the spleen were observed in groups LM (p<0.001) and HM (p=0.04), respectively. The differences were statistically significant relative to the remaining groups. Turkeys of group LM were characterized by a lower CD4⁺ T cell percentage in the thymus (p<0.001) and a lower CD8⁺ T cell percentage in the cecal tonsils (Cts) (p<0.01). Vaccination against ORT resulted in a significant increase in the percentage of CD4⁺/CD8⁺ T cell subpopulation and a decrease in the percentage of CD8⁺ T cell subset in the spleen.

Depending on the intensity, duration and type of physical exercise, equine metabolism has to adapt to nervous, cardiovascular, endocrine and respiratory system requirements. In horses, exercise and training are known to have considerable effects on the mechanisms of hemostatic system involving platelet activity. The aim of the present study was to evaluate the effect of different training schedules on platelet aggregation in 15 Italian Saddle jumping horses. Animals were divided into three equal groups: Group A was subjected to a high intensity-training program; group B to a light training program, group C included sedentary horses. From each animal, blood samples were collected by jugular venipuncture at rest on the 1st, 3rd and 5th days, and afterwards, once a week, for a total of 5 weeks data recording, in order to assess the maximum degree of platelet aggregation and the initial velocity of aggregation (slope) platelet aggregation. Two-way analysis of variance (ANOVA) showed a significant effect of the different training schedules on studied parameters. The results revealed a different degree of platelet aggregation and a different initial velocity of platelet aggregation that changes during the different training schedules in horses that could represent a different protective endothelial mechanism. These findings could have an important role for a clearer knowledge of the physiological reference values of platelet aggregation and for a better interpretation of these variations during the training.

This study aimed to compare post-thaw quality of boar semen following freezing in an ostrich egg yolk lipoprotein (LPFo) extender supplemented with 0%, 0.25% and 0.50% Orvus Es Paste (OEP). Sperm assessments included total motility (TMOT), mitochondrial function (MF), plasma membrane integrity (PMI) and acrosome integrity (normal apical ridge, NAR). Considerable variations among boars and OEP treatments had a significant effect (P < 0.001) on post-thaw sperm characteristics. It was observed that post-thaw sperm characteristics were significantly compromised in semen samples frozen in the absence of OEP. By contrast, lactose-LPFo-glycerol extender supplemented with either 0.25% OEP or 0.50% OEP markedly enhanced post-thaw sperm characteristics. In all boars, there were no marked differences in post-thaw sperm TMOT between the freezing extenders supplemented with 0.25% and 0.50% OEP. However, a decline in the percentage of post-thaw motile spermatozoa was more pronounced in the extender supplemented with 0.50% OEP following a 120-min incubation period. Furthermore, the proportions of frozen-thawed spermatozoa with MF, PMI and NAR acrosomes varied significantly among the boars in the OEP-supplemented extenders. The findings of this study indicate that different OEP concentrations, in the presence of ostrich egg yolk lipoproteins, could have varying effects on post-thaw sperm survival.

To compare the genotypes of Campylobacter jejuni, isolates of cattle origin were collected from 9 Polish farms and genotyped by ERIC-PCR. We identified 28 genotypes among the 43 C. jejuni isolates, and demonstrated high genomic diversity. The highest level of diversity was observed in strains isolated from stanchion-barn animals in opposition to those from the loose-housing system.
