Verticillium wilt caused by Verticillium dahliae, is one of the most devastating diseases in pistachio orchards in the world including Iran. In search for an eff ective non-chemical strategy for the management of this disease, we evaluated the biocontrol potential of Trichoderma harzianum isolates obtained from the rhizosphere of healthy pistachio trees in diff erent locations of the Kerman province of Iran against V. dahliae under laboratory and greenhouse conditions. Dual culture tests in the laboratory were conducted in a completely randomized design using 72 T. harzianum isolates. Twenty isolates showed the highest in vitro antagonistic activity. Th e results indicated that all 20 isolates were capable of inhibiting the mycelial growth of V. dahliae signifi cantly. Among them, isolates Tr8 and Tr19 were the most eff ective by 88.89% and 85.12% inhibition, respectively. Extracted cell free metabolites of all eff ective isolates also inhibited the growth of V. dahliae in the culture medium signifi cantly. According to the results, isolates Tr4 and Tr6 inhibited fungal pathogen growth by 94.94% and 88.15% respectively, through production of non-volatile metabolites. In the evaluation of volatile metabolites, isolates Tr5 and Tr4 were the most eff ective by 26.27% and 24.49% growth inhibition, respectively. Based on the results of the in vitro experiments, the fi ve most eff ective isolates were selected for evaluation under greenhouse conditions for their biocontrol potential in controlling Verticillium wilt of pistachio. Results of the greenhouse, (in vivo) experiments were positive and indicated that the occurrence of wilt disease in plants treated with the antagonists alone or in combination with pathogenic fungus was lower than in plants inoculated with pathogen alone. Th e overall results of this study suggest that Trichoderma fungal antagonist may be an eff ective biocontrol agent for the control of Verticillium wilt of pistachio.

The article presents factors which can influence the occurrence the pine wilt disease, i.e. the causative agent of this disease – pine wood nematode, Bursaphelenchus xylophilus, the transmission mechanism of pine wood nematode, the insect vectors – cerambycid beetles of the genus Monochamus, and environmental conditions which favour development of this disease. Furthermore, the methods of prevention of pine wilt disease are discussed.

Tomato spotted wilt wirus (TSWV), powodujący brązową plamistość pomidora, występuje na całym świecie, atakując 1090 gatunków roślin. Najbardziej efektywną metodą ograniczenia strat ekonomicznych powodowanych przez tę chorobę jest wyhodowanie odmian odpornych. W IUNG-PIB w Puławach uzyskano linię hodowlaną tytoniu, charakteryzująca się wysoką odpornością na TSWV. Celem badań było określenie sposobu dziedziczenia czynnika odporności, występującego u linii L-9. W wyniku obustronnych krzyżowań linii L-9 z podatną odmianą Wiślica otrzymano pokolenie F1, z którego powstały krzyżówki wsteczne z rodzicem odpornym i podatnym oraz pokolenie F2. Uzyskane populacje przetestowano pod względem odporności na TSWV w warunkach szklarniowych, metodą mechanicznej inokulacji izolatem wirusa pochodzącym z plantacji produkcyjnych tytoniu. Obecność TSWV sprawdzano metodą DAS-ELISA. Stwierdzono dominujący charakter odporności i brak wpływu czynnika matecznego na ujawnianie się tej cechy. Segregacja fenotypów odpornych i podatnych w krzyżówkach testowych potwierdziła hipotezę o jednogenowym sposobie dziedziczenia odporności na TSWV pochodzącej z linii L-9.
Tomato spotted wilt virus (TSWV) has been reported worldwide in a 1090 different host species. The most effective way to minimize the damage of this disease is to develop resistance of the field crops. The tobacco breeding line developed in IUNG-PIB in Puławy exhibits a high level of TSWV resistance. The objective of this study was to investigate the inheritance of the resistance present in the L-9 line. The resistant parental L-9 line and susceptible cultivar Wiślica were crossed in both directions to create F1 hybrids, from which backcrosses with resistant and susceptible parent were made and F2 generation were obtained. All populations were evaluated for response to artificial inoculation with one TSWV isolate of local origin, in greenhouse conditions. The presence of the virus was verified using DAS-ELISA. A dominant character and no material effect were stated for the resistance trait. Segregation ratios of resistant and susceptible phenotypes in the testing crosses were in agreement with the hypothesis that inheritance of TSWV resistance of the L-9 line is dominant and monogenic.

Fungal antagonist, Talaromyces flavus, is one of the most important biological agents of soil-borne fungal diseases including Verticillium and Fusarium wilt. In this study, to increase the effectiveness of T. flavus isolates obtained from greenhouse cucumbers and field grown tomatoes five chemical stabilizers were evaluated. Based on the results of previous studies, the most effective substrate for the growth, sporulation and stability of T. flavus isolates related to the above-mentioned plants was a mix of rice bran and peat-moss. Different chemical stabilizers were mixed with the above-mentioned substrate containing spore suspensions of various T. flavus isolates. For each plant, a completely randomized experiment was conducted under greenhouse conditions with seven treatments and three replications. The results of this study indicated that treatments containing sodium nitrate and D-cycloserine were more effective than those containing other stabilizers. The overall results of this study suggest that the use of some chemical stabilizers may enhance the biocontrol potential of fungal antagonists in controlling different plant diseases including Verticillium and Fusarium wilt.

In this study, about 112 isolates of Streptomyces were isolated from chickpea rhizospheric soils. Among the isolated strains, five showed strong inhibitory effects against chickpea Fusarium wilt caused by Fusarium oxysporum f. sp. ciceris in vitro using plate assay and selected for further studies. The selected strains were identified as Streptomyces spp. based on morphological and biochemical characterization as well as 16S rDNA sequences analysis. Our results assigned them to strains related to genus of Streptomyces. In vitro, antagonistic effects of Streptomyces strains against the disease were evaluated through the dual-culture method, volatile and non-volatile metabolites, siderophore, protease and chitinase production. All bacterial strains inhibited mycelial growth of the pathogen ranging from 26 to 44.2% in dual culture assay. The non-volatile extract of five of the Streptomyces strains inhibited more than 50% growth of the pathogen, whereas volatile compounds were less effective on mycelial growth inhibition (20.2 to 33.4%). The ability of the biocontrol agents to produce siderophore and protease were varied, whereas, production of chitinase was detected for all strains. Results of the greenhouse assay indicated that all biocontrol agents reduced disease severity (ranging from 38.7 to 54.8%). Accordingly, strain KS62 showed higher control efficacy (54.8%). In addition, the biomass of chickpea plants (plant height and dry weight) significantly increased in plants treated with Streptomyces strains compared to non-bacterized control. The results of this study showed that it may be possible to manage chickpea Fusarium wilt disease effectively by using Streptomyces species, as biocontrol agents. Therefore, evaluating their efficiency under field conditions is needed.

Potato is one of the most important food crops with a capacity to abate hunger among farmers in the region. However, its productivity has stagnated and remained low due to poor agronomic practices applied by smallholder farmers. To avert this situation, dissemination through training of already existing better technologies and practices should be prioritized. A lot of research has been done with better recommendations- use of disease-free tuber seeds from high yielding and tolerant varieties, adequate application of balanced nutrients, timely scouting and integrated management of pests and diseases and good harvesting practices are available for immediate adoption. The adoption and dissemination could be facilitated by currently existing policies that promote cross-border movement and diffusion of research technologies and new varieties.

Fusarium species are causal agents of fungal diseases occurring frequently in numerous agriculturally important plants, including potato, garlic and are one of the common pathogens of tomato, causing root rot in the west part of Iran. Therefore, the objectives of this study were to isolate and identify disease-causing Fusarium species from infected tomatoes based on the morphological and molecular characteristics. Twenty-five isolates of Fusarium were obtained from infected root of tomato plants collected from the fields in different regions of western Iran. Based on morphological features, the strains were classified into four following Fusarium species: F. oxysporum, F. redolens, F. proliferatum and F. verticillioides. The phylogenetic trees based on tef1 and tub2 dataset clearly distinguished closely related species. All of the isolates were evaluated for their pathogenicity on healthy tomato seedlings in the greenhouse. This is the first report on molecular identification of Fusarium species isolated from tomato plants cultivated in Iran.

Pine wilt disease caused by pine wood nematodes is endemic to North America. Pine wood nematodes have already spread to East Asia, including China, Taiwan and Korea in addition to Japan. The pine wood nematode, Bursaphelenchus xylophilus, was discovered in Portuguese pine forests in 1999. If it were to become established in the pine forests in Europe, it could become one of the most serious threat to coniferous forests worldwide.

Tobacco (Nicotiana tabacum L.) is an important industrial crop. Among the diseases causing losses in tobacco cultivation, viral ones are of particular importance due to the very low effectiveness of chemical protection allowing only for vector control. The greatest threat to tobacco cultivation in Poland and in many countries of the world is posed by potato virus Y (PVY), tomato spotted wilt virus (TSWV) and, in recent years, the increasingly frequent tobacco mosaic virus (TMV). The genetic resources studies carried out in the genus Nicotiana have shown a wide variation in species resistance, assessed biologically, serologically and molecularly, depending on the virus used and, in the case of PVY, also on the specific isolate. The results of the assessment of resistance in Nicotiana accessions presented in this paper, gathered from literature data as well as from our own research, allow us to broaden and systematise our knowledge on the sources of resistance to viral diseases. This is the only such an extensive study in this field. It provides an excellent information base for the appropriate selection of accessions for use in resistance breeding.

Fusarium oxysporum f. sp. pisi (FOP) is a significant and destructive pathogen of field pea in Algeria. In the present study, 50 isolates of F. oxysporum f. sp. pisi, the causal agent of pea (Pisum sativum) wilt, collected from different parts of western Algeria and representing four races of the pathogen, were analyzed for virulence. The wilt incidence ranged from 6.66 to 88.33% on a highly susceptible cultivar (Little Marvel). Twenty-one isolates belonging to four races of FOP and one nonpathogenic F. oxysporum (FO) isolate were analyzed for vegetative compatibility in order to reveal the genetic structure of the population and to check the reliability of the method for the identification of physiological races of FOP. Obtained results showed that the FOP isolates could be classified into four main vegetative compatibility groups (VCGs) that corresponded to races l, 2A, 2B and 5. The race 6 isolate fell into the race 1 VCG. To our knowledge, this is the first such study in Algeria of its kind.

Seven populations of N. alata, 4 populations of N. sanderae and several hundred plants belonging to three generations of the hybrid (N. tabacum cv. TB-566 tetra × N. alata) × N. alata were tested for resistance to TSWV. Tested plants were artificial inoculated with TSWV under greenhouse conditions. Out of 11 accessions of N. alata and N. sanderae 7 were found to be composed entirely of plants hypersensitive to TSWV. In the remaining four accessions there were from 16 to 25% of TSWV susceptible plants. The hybrids (N. tabacum cv. TB-566 tetra × N. alata) × N. alata, regardless of their cytogenetical status, showed hypersensitive response to inoculation with TSWV...

The study aimed at the evaluation of the efficacy of horseradish seedling treatment with chitosan (Biochikol 020 PC) for the protection of the roots against Verticillium wilt and influence on the size and root yield structure. The direct effect of chitosan on the fungi which contribute to the degradation of root health was tested in laboratory conditions. The horseradish root yield and contribution of individual root fractions and the intensity of Verticillium wilt was significantly dependent on the year of cultivation. Soaking the horseradish seedlings in a 2.5% chitosan solution contributed to the mean increase of 1.7 t·ha−1 of the total yield of roots and the seedling fraction with the length of 30-35 cm by 1.6 t·ha−1, as well as to the decrease of the amount of the fraction of roots shorter than 25 cm and waste material. The used chitosan limited the occurrence of horseradish root Verticillium wilt in every year. In the in vitro study, no fungistatic effect of chitosan was reported only in reference to S. sclerotiorium. On the other hand, the growth of the mycelium of the remaining Verticillium wilt causing fungi depended on the chitosan concentration. Normally, the fungistatic activity of chitosan increased with the increase of the dose and its highest level occurred at 2.5%. The mean surface growth inhibition coefficient enabled raking the fungi according to their decreasing sensitivity to chitosan: F. culmorum 64.9%, P. exiqua 58.1%, V. dahliae 44%, R. solani 35.4%, F. oxysporum 34.3%, F. solani 32.4%.
Celem pracy była ocena skuteczności zaprawiania sadzonek chrzanu chitozanem (Biochikol 020 PC) w ochronie korzeni przed czernieniem pierścieniowym oraz wpływu na wielkość i strukturę plonu korzeni. Bezpośrednie oddziaływanie chitozanu na grzyby przyczyniające się do pogorszenia zdrowotności korzeni badano w warunkach laboratoryjnych. Plon korzeni chrzanu oraz udział w nim poszczególnych frakcji oraz nasilenie czernienia pierścieniowego istotnie zależało od lat uprawy. Moczenie sadzonek chrzanu w 2,5% roztworze chitozanu zwiększało średnio o 1,7 t·ha-1 plon całkowity korzeni i frakcji sadzonek o długości 30-35 cm o 1,6 t·ha-1 oraz zmniejszało ilość frakcji korzeni krótszych niż 25 cm i odpadów. Zastosowany chitozan w każdym roku ograniczał występowanie czernienia pierścieniowego korzeni chrzanu. W badaniach in vitro tylko w odniesieniu do S.sclerotiorum nie stwierdzono fungistatycznego oddziaływania. Z kolei wzrost grzybni pozostałych grzybów powodujących czernienie pierścieniowe zależał od stężenia chitozanu. Na ogół aktywność fungistatyczna chitozanu wzrastała wraz ze zwiększeniem dawki i była największa dla 2,5%. Średnie współczynniki zahamowania rozrostu powierzchniowego pozwoliły na uszeregowanie grzybów według ich malejącej wrażliwości na chitozan: F. culmorum 64,9%, P. exiqua 58,1%, V. dahliae 44%, R. solani 35,4%, F. oxysporum 34,3%, F. solani 32,4%.
