Opis:
Both epidermal growth factor (EGF) and transforming growth factor (TGF) play an important physiological role in the processes of proliferation and differentiation of several different cell types. However, the expression profiles of these factors in domestic bitches endometrium are still poorly recognized. The aim of the present study was to identify and analyze the differential expression of these factors in various stages of the estrus cycle. Endometrial tissue from proestrus (n=17), estrus (n=10), day 10 diestrus (n=15), day 35 diestrus (n=18) and anestrus (n=25) was collected soon after ovariohysterectomy. Total RNA was isolated from the endometrium by means of Chomczyński and Sacchi method, treated by DNase I, and reverse- transcribed into cDNA. Quantitative analysis of EGF, TGFβ1, TGFβ2, and TGFβ3 cDNA was performed by real-time quantitative polymerase chain reaction (RT-PCR). EGF expression in canine endometrium was increased in the estrus stage as compared to proestrus (P<0.05), day 10 diestrus (P<0.05), day 35 diestrus (P<0.01) and anestrus (P<0.001). We also found the differences in EGF expression between day 10 and day 35 of estrus as well as between day 35 of estrus with anestrus (P<0.05, P<0.01, respectively). The TGFf1 transcript contents were also higher in estrus as compared to other stages (P<0.01). The TGFβ2 and TGFβ3 in the estrus stage was increased compared to proestrus, day 10 diestrus, day 35 diestrus and anestrus (P<0.05). We proved that expression of EGF and TGFβ transcript isoforms is related to the phase of estrus in bitches and therefore may be regulated by specific hormone concentrations during these periods. Our results confirm the hypothesis that these growth factors play a role in the regulation of biochemical changes in the endometrial tissues during the estrus cycle.

The aim of the present study was to define the effect of TGF-β1 on C2C12 myoblasts myogenesis. TGF-β1 together with its receptor is a negative auto-paracrine regulator of myogenesis, which influences the proliferation, differentiation, and functions of muscle cells. TGF-β1 exerts highly significant inhibitory effect on differentiation of C2C12 mouse myoblasts manifested by the impairment of cell fusion and very low expression of myosin heavy chain. The study of differentiating C2C12 mouse myoblasts treated with TGF-β1 revealed 502 genes (436 down-regulated and 66 up-regulated) with statistically different expression. TGF-β1-regulated genes were identified to be involved in 29 biological processes, 29 molecular functions groups and 59 pathways. The strongest inhibiting effect of TGF-β1 was observed in the cadherin and Wnt pathways. The key-genes that could play the role of TGF-β1 targets during myoblasts differentiation was identified such as: Max, Creb1, Ccna2, Bax, MdfI, Tef, Tubg1, Cxcl5, Rho, Calca and Lgals4.

WC1+ cells in cattle exhibit both regulatory and effector activities. However, it has not been elucidated whether they are so plastic that both activities co-exist in one cell or there are separate subpopulations of effector and regulatory cells. Since the production of IFN-γ and IL-10 seems to be related to WC1+ cells’ effector and regulatory function, respectively, the main aim of this study was to determine whether those cytokines are produced by separate subpopulations of WC1+, or are co-produced by the same cells. Due to increasingly frequent emphasised role of consumption of IL-2 in the mechanism of suppressor action of mouse CD25+CD4+ T regulatory cells, expression of the receptor’s α chain for interleukin 2 (CD25) on WC1+ lymphocytes has been evaluated. An average of 5.21% of WC1+ cells obtained from PBMCs of 12-month-old heifers show constitutive expression of the CD25 molecule, with CD25highWC1+ and CD25lowWC1+ cells accounting for 1.05% and 4.10% of WC1+ lymphocytes, respectively. For detection of intracellular cytokine production, PBMCs were stimulated with concanavalin A. Both IFN-γ- and IL-10-producing cells within the CD25-WC1+ and CD25+WC1+ subpopulations were mainly separate subpopulations. The average percentage of IFN-γ+IL-10-, IFN-γ-IL-10+ and IFN-γ+IL-10+ cells among CD25-WC1+ lymphocytes was 4.03%, 2.67% and 0.51%, respectively. A positive correlation was observed between the presence of the CD25 molecule on WC1+ lymphocytes and production of IL-10 and TGF-β, because the average percentage of IFN-γ-IL-10+ and IFN-γ+IL-10+ among CD25+WC1+ lymphocytes was 3 and 4.5 times higher as compared to the corresponding cells in the CD25-WC1+ subpopulation, whereas the percentage of IFN-γ+IL-10- cells in both the subpopulations was not significantly different. The percentage of TGF-β+ cells within the CD25+WC1+ subpopulation was 2.72 times as high as that of CD25-WC1+ lymphocytes. Therefore, with respect to the production of IFN-γ, IL-10 and TGF-β, CD25+WC1+ lymphocytes turn out to have a more suppressor profile than CD25-WC1+.
