Opis:
In the tradition of Model Theoretic Syntax, we propose a logical approach to the description of grammars. We combine in one formalism several tools that are used throughout computer science for their power of abstraction: logic and lambda calculus. We propose then a high-level formalism for describing mildly context sensitive grammars and their semantic interpretation. As we rely on the correspondence between logic and finite state automata, our method combines conciseness with effectivity. We illustrate our approach with a simple linguistic model of several interleaved linguistic phenomena involving extraction. The level of abstraction provided by logic and lambda calculus allows us not only to use this linguistic model for several languages, namely English, German, and Dutch, but also for semantic interpretation.

The majority of proteins are unable to translocate into the cell interior. Hence for peptide- and protein-based therapeutics a direct intracytoplasmic delivery with the aid of transducing agents is an attractive approach. We wanted to deliver to the cell interior a putatively cytotoxic protein VPg. Protein transduction was achieved in vitro with three different commercial products. However, in our hands, delivery of various control proteins without known deleterious effects, as well as of protein VPg, always induced cell death. Finally, we used a novel transducing peptide Wr-T, which was not toxic to cultured cells, even in a quite large range of concentrations. Most importantly, control protein delivered to cells in culture did not display any toxicity while VPg protein exerted a strong cytotoxic effect. These data show that results obtained with cell-penetrating agents should be interpreted with caution.

The G protein-coupled histamine H4 receptor (H4R) is the last member of histamine receptors family discovered so far. Its expression pattern, together with postulated involvement in a wide variety of immunological and inflammatory processes make histamine H4 receptor an interesting target for drug development. Potential H4R ligands may provide an innovative therapies for different immuno-based diseases, including allergy, asthma, pruritus associated with allergy or autoimmune skin conditions, rheumatoid arthritis and pain. However, none of successfully developed selective and potent histamine H4 receptor ligands have been introduced to the market up to date. For that reason there is still a strong demand for pharmacological models to be used in studies on potent H4R ligands. In current work we present the development of novel mammalian cell line, stably expressing human histamine H4 receptor, with use of retroviral transduction approach. Obtained cell line was pharmacologically characterized in radioligand binding studies and its utility for affinity testing of potent receptor ligands was confirmed in comparative studies with the use of relevant insect cells expression model. Obtained results allow for statement that developed cellular model may be successfully employed in search for new compounds active at histamine H4 receptor.

Opis:
Aluminium toxicity is one of the major factors that limit plant growth and development in many acid soils. Root cells plasma membrane, particularly of the root apex, seems to be a major target of Al toxicity. However, strong interaction of Al3+, the main Al toxic form, with oxygen donor ligands (proteins, nucleic acids, polysaccharides) results in the inhibition of cell division, cell extension, and transport. Although the identification of Al tolerance genes is under way, the mechanism of their expression remains obscure.Soil chemical factors that limit root growth in acid soils, diminish crop production, include Al, Mn and various cations, and also deficiency or unavailability of Ca, Mg, P, Mo, and Si. These effects are further complicated by interactions of Al with other ions in different plant genotypes and under stress conditions (Foy, 1992). Cytotoxicity of Al has been well documented in plants (Delhaize & Ryan, 1995; Horst et al., 1999; Kollmeier et al., 2000; Marienfeld et al., 2000). It is generally known that plants grown in acid soils due to Al solubility at low pH have reduced root systems and exhibit a variety of nutrient-deficiency symptoms, with a consequent decrease in yield. In many countries with naturally acid soils, which constitute about 40% of world arable soil (LeNoble et al., 1996), Al toxicity is a major agricultural problem, and is intensively studied in plant systems.The effects of aluminium on plant growth, crop yield, uptake and nutrients distribution in vegetative and reproductive parts are still not fully understood. This review discusses recent information on aluminium toxicity with an emphasis on plant response to Al stress.

Endocrine disruptors interfere with hormonal action through receptors and signaling pathways, but receptor crosstalk involved in the actions of endocrine disruptors has not yet been well documented. This review summarizes what is known about the actions of endocrine disruptors through receptor crosstalk, focusing on three model cases: crosstalk involving the estrogen receptor, the androgen receptor, and the thyroid hormone receptor. Here, crosstalk is categorized as bidirectional or unidirectional, with the latter category further subdivided into ligand-dependent or -independent crosstalk. More research needs to be done to develop a clearer understanding of the involvement of receptor crosstalk in cell signaling that is induced by endocrine disruptors. This understanding will help to develop in vitro and in silico assays that can replace animal tests.

Despite the fact, that the American mink (Neovison vison) is one of the most intensively economically exploited and problematic, from the ecological point of view, fur-bearing animal, it remains one of the least studied livestock species. It can be proven by the fact, that the research on one of the key hormones, which has systemic effects, that is the growth hormone, are rather poorly advanced. The purpose of this review is to present the summary and the critical analysis of the current state of knowledge on the topic of the growth hormone in the American mink, its biological function, production in physiological conditions, regulation of secretion and transduction of the hormone’s signal. The article contains also an up to date information on the American mink growth growth hormone gene and its protein product.
Mimo że norka amerykańska (Neovison vison) jest jednym z najintensywniej eksploatowanych gospodarczo, a także problematycznych, z ekologicznego punktu widzenia, gatunków zwierząt futerkowych, pozostaje jednym z najsłabiej zbadanych gatunków zwierząt gospodarskich. Dowodzi tego, między innymi fakt, że badania nad jednym z kluczowych hormonów o działaniu systemowym, jakim jest hormon wzrostu, są słabo zaawansowane. Celem niniejszego artykułu jest przegląd i krytyczna analiza aktualnego stanu wiedzy na temat hormonu wzrostu u norki amerykańskiej, jego funkcji biologicznej, produkcji w warunkach fizjologicznych, regulacji wydzielania oraz transdukcji sygnału. Artykuł zawiera również aktualne informacje o genie hormonu wzrostu oraz jego produkcie białkowym.

Intact rat peritoneal macrophages (rPM) treated with 3-isobutyl-1-methylxanthine (IBMX), an inhibitor of phosphodiesterases (PDEs), accumulated more cGMP than untreated cells. A PDE activity toward [3H]cGMP was detected in the soluble and particulate fractions of rPM. The hydrolysis of cGMP was Ca2+/calmodulin-independent but increased in the presence of cGMP excess. Similar results were obtained when [3H]cAMP was used as a substrate. The hydrolytic activity towards both nucleotides was inhibited in the presence of IBMX. Therefore, the PDEs of families 2, 5, 10 and 11 are potential candidates for cGMP hydrolysis in the rPM. They may not only regulate the cGMP level in a feedback-controlled way but also link cGMP-dependent pathways with those regulated by cAMP.
