Opis:
The application of biochemical and molecular techniques in parasitological studies has provided increasing evidences of genetic polymorphism among parasite populations. This review presents possible origins of genetic variation within populations of various protozoan species. Since the mode of reproduction has an important influence on genetic polymorphism within parasite populations these considerations refer mainly to some protozoan parasites which have various life cycles, e.g. Giardia, Trypanosoma, Cryptosporidium, Toxoplasma. Also other factors associated with parasites (such as: transmission and passage history in laboratory conditions; occurrence in different hosts or geographic regions; selective pressure of drugs; competitive interactions between populations) that affect parasite genetic diversity are discussed. However, the number of examined isolates of parasites and genetic markers, assortment of methods, probes, primers and reagents used is also of significance. The significance of genetic variability in parasite populations is still the subject of much interest and controversy. A simple interpretation of such variation is impossible because of the complexity of host-parasite interactions. The knowledge of parasite diversity at the nucleic acids level has continually increased, but a corect interpretation of this phenomenon requires at least the same knowledge of genetic variability in host populations. Nevertheless, genetic variability in protozoan parasites has many important implications, e.g. for taxonomy, epidemiology, control and evolution. Genetic differences within parasite populations might also be associated with phenotypic variability, e.g. virulence, antigenicity, infectivity, drug sensitivity, hostpreference etc.

After a long period of using basic microscopic, immunological and biochemical methods for diagnosis, rapid development of nucleic acids investigation enabled introduction of specific and sensitive methods of detection of pathogenic agents on the molecuiar level. Among others, polymerase chain reaction (PCR), discovered in mid of 80'ies and then automatized, offered an attractive alternative to conventional testing systems. In this paper we describe reliable diagnostic tests widely used in the world, including Poland, and capable of detecting different disease agents as parasites and fungi in clinical specimens and pathogens of emerging zoonotic diseases in ticks. The possibilities of using molecular methods for determination of Plasmodium falciparum drug resistance is also discussed. Moreover, the report offers information concerning kinds of molecular tests and institutions in which there are executed.

Peroxidase-antiperoxidase (PAP) method was used to detect Toxoplasma gondii in the central nervous system of experimentally infected mice. Due to its capability for accurate localization of a parasite antigen among the populations of host cells, the method proved very useful in the diagnosis as well as in observations on the development of pathological changes.

Opis:
Immunocapture assays ISAGA PLUS IgA/IgM (bioMerieux) and IgE ISAGA were used to determine their usefulness in the diagnosis of acquired and congenital toxoplasmosis. Specific IgM, IgA and IgE antibodies were tested in 134 patients, namely pregnant women who seroconverted during gestation (n= 20), children with congenital toxoplasmosis (n= 5), patients with toxoplasmic lymphadenitis (n= 56) and immunocompetent individuals with chronic Toxoplasma gondii infection (n= 53). Altogether 172 sera were examined. Specific IgM antibodies were detected in all sera from pregnant women (100%) with recent T. gondii infection (1- 8 weeks after seroconversion), in all patients with toxoplasmic lymphadenopathy (1-3 months after onset of symptoms) and in their control examinations after 2 and 5 months (100%) and also in 35 (66%) out of 53 patients with chronic infection. In infants with congenital toxoplasmosis IgM were found only in one new-born; equivocal results were obtained in 3 children during the asymptomatic serological reactivation in the second year of life. Specific IgA antibodies were present in sera from 15 (75%) out of 20 women seroconverted during pregnancy; in 3 cases the results were equivocal. IgA antibodies were detected in sera from 30 (81.1%) out of 37 patients with toxoplasmic lymphadenitis examined once; in 19 patients examined 3 times IgA antibodies were present in all the cases in the first serological examination performed when clinical symptoms were first observed (100%), in 17 patients after 2 months (89.5%) and in 11 patients after 5 months (57.9%). IgA antibodies were also detected in 21 sera (39.6%) from patients with chronic T. gondii infection. In children with congenital toxoplasmosis IgA antibodies were found in 3 cases during serological reactivation after discontinuation of pyrimethamine-sulfadiazine therapy; in these cases equivocal results of IgM antibodies were present, and positive result of IgE antibodies in one case. Specific IgE antibodies were detected in sera from 17 (85%) out of 20 women with seroconversion and in 18 patients with lymphadenopathy (32.1%); in the last group IgE antibodies were not present in the follow-up examination after 5 months. IgE antibodies were detected only in 5 cases (9.4%) with chronic infection. IgA and IgE antibodies in ISAGA begin to appear about a week later than IgM antibodies; in sera collected between the 2nd and 3rd week after invasion the positive results were obtained in all cases (100%). Therefore, ISAGA PLUS IgA/IgM (bioMerieux) is useful for the diagnosis of recent T. gondii infection especially in women with suspected seroconversion during pregnancy. ISAGA PLUS IgA/IgM is more sensitive than any conventional method routinely used and so far is a specially eflicient technique for newborns and infants suspected for congenital infection and/or in diagnosing congenital toxoplasmosis during immunological recrudescence. This test has a limited value in toxoplasmosis with lymphadenopathy by reason of possibility of a long persistence of IgM and IgA antibodies detected by ISAGA. Detection of specific IgE antibodies using ISAGA technique may be useful for differential diagnosis of acute and chronic phase of T. gondii infection and also in some cases of serological reactivation of congenital toxoplasmosis.

Opis:
Toxoplasma gondii infection is one of the most common human zoonosis. Humans acquire toxoplasmosis by consuming raw or undercooked meat, from environments and from via vertical transmission to the foetus through the placenta during pregnancy. In the present study, a total of 784 persons (forestry workers), from north−eastern region of Poland (pomorskie and warmińsko−mazurskie provinces), were tested by using commercially available ELISA test (DiaSorin) to determine the frequency of Toxoplasma gondii infection. The study was based on detection of IgG and IgM specific antibodies in collected serum samples. The T. gondii−specific IgG were found in 490 samples (62.5%), while the IgM antibodies were noted only in 18 persons (3.7%). High prevalence of seropositive results in forestry workers may suggest an occupational exposure of this group to T. gondii.

Opis:
Dot-immunobinding technique was used to detect circulating Toxoplasma antigens in the sera of men and animals. Serum samples were collected from mice and rabbits infected experimentally with parasites of the RH strain of T. gondii while human sera were obtained from persons suspected of having active toxoplasmosis. Circulating Toxoplasma antigens were detected in the samples of mice sera that had been collected since the 2-nd day of iniection and in the sera of rabbits during 2 and 3 week of iniection. From amongst 146 human sera, 35 (24%) gave positive reaction in the test while 14 (9,5%) produced doubtful results. The experiments proved the method to be sensitive, reproducible and easy to perform. It obviates the need of special equipment while most of the necessary reagents are commercially available. High percentage of doubtful results is the main drawback of the method.

Opis:
Detection of Toxoplasma gondii in human placenta by PCR and placental histologic findings. Since isolation of Toxoplasma gondii from human placenta strongly correlates with fetal infection, the aims of the study were: to detect fragments of T. gondii B1 gene in human placentae by PCR and to evaluate their pathology. 36 placentae included in three groups were obtained: group I (n = 7) from pregnancies with prenatal diagnosis of fetal toxoplasmosis; II (n = 17) from women with serologic features of primary infection during pregnancy; III (n&13) from pregnancies with fetal T. gondii infection based on clinical signs. T. gondii DNA was found in 2/4 samples from the I group and in 1/14 from the II group. Villitis was identified in 3/15 other placentae from the II group. In the III group we did not recognize neither T. gondii DNA nor villitis. We consider PCR and pathologic evaluations of placentae as the two complementary methods. PCR can be especially helpful in pregnancies not screened against T. gondii as positive result in placenta can confirm mother's primary infection.
