Recently we have shown that in vitro binding of the proximal part of the human tyrosine hydroxylase gene to the nuclear matrix is correlated with its transcriptional activity. The strongest binding potential was predicted by computing for the first intron sequence (Lenartowski & Goc, 2002, Neurosci Lett.; 330 : 151-154). In this study a 16 kb fragment of the bovine genomic DNA containing the tyrosine hydroxylase gene was investigated for its affinity to the nuclear matrix. Only a 950 bp fragment encoding the distal part of the first intron, second exon and a few nucleotides of the second intron bound to the nuclear matrix. The binding was independent of the tissue-specific tyrosine hydroxylase gene activation. The fragment was subcloned and sequenced. Computer search pointed to one potential intronic matrix attachment region with two AP1-like sites embedded in the sequence. We conclude that even if the position of the matrix binding region is conserved among the tyrosine hydroxylase genes in mammals, its tissue specificity and/or function is not preserved or is achieved by different mechanisms.

Opis:
Nucleoside transporters (NT) facilitate the movement of nucleosides and nucleobases across cell membranes. NT-mediated transport is vital for the synthesis of nucleic acids in cells that lack de novo purine synthesis. Some nucleosides display biological activity and act as signalling molecules. For example, adenosine exerts a potent action on many physiological processes including vasodilatation, hormone and neurotransmitter release, platelet aggregation, and lipolysis. Therefore, carrier-mediated transport of this nucleoside plays an important role in modulating cell function, because the efficiency of the transport processes determines adenosine availability to its receptors or to metabolizing enzymes. Nucleoside transporters are also key elements in anticancer and antiviral therapy with the use of nucleoside analogues. Mammalian cells possess two major nucleoside transporter families: equilibrative (ENT) and concentrative (CNT) Na+-dependent ones. This review characterizes gene loci, substrate specificity, tissue distribution, membrane topology and structure of ENT and CNT proteins. Regulation of nucleoside transporters by various factors is also presented.

Opis:
Tissue engineering is a very rapidly developing domain of science. There is a huge interest in the biology of stem cells, especially in their proliferation, differentiation and spreading in the response to different factors. Biomaterials with properties similar to the natural environment of human body (accelerating wounds healing and not causing immunological response) as well as these that can control cells behavior are in constant need. This review presents the most popular polymeric biomaterials for tissue engineering and regenerative medicine [3–5, 8–20], explains interactions between cells and biomaterials [3, 13, 22–33], describes the most important surface properties that can have an influence on cells i.e. topography [36, 43–47, 53–72], roughness [1, 5, 31, 51, 66, 73–84], stiffness [54, 84–102], hydrophobicity, chemistry [4, 11, 16, 20, 27, 31, 32, 36–38, 42, 58, 103–112] and surface charge [18, 37, 113–116].
