Badania przeprowadzono na 2 grupach indorów typu Big-6 podczas 16-tygodniowego odchowu. Czynnikiem doświadczalnym był dodatek miedzi w formie chelatu z lizyną w ilości 10 mg Cu-dm-3H2O. Celem badań było określenie wpływu miedzi jako dodatku do wody pitnej na masę ciała oraz kumulację składników mineralnych w wybranych tkankach indyków. Wykazano, że dodatek chelatu spowodował zwiększenie końcowej masy ciała indorów, natomiast nie wpłynął w znaczący sposób na masę analizowanych tkanek. Stwierdzono, że zastosowanie Cu wpłynęło na wzrost zawartości Cu, Zn i Fe we wszystkich badanych tkankach. Największą retencję Cu wykazano w przypadku wątroby, natomiast najmniejsze ilości tego pierwiastka kumulowały się w mięśniach piersiowych ptaków. Długotrwała aplikacja miedzi nie spowodowała nadmiernej kumulacji tego składnika w nerkach i mięśniach indyków, co nie stanowiło zagrożenia dla zdrowia konsumentów, gdyż jej zawartość nie przekraczała dopuszczalnych norm (5 mg-kg-1; wg PN-A-86524:1994).
Investigations were performed using 2 groups of turkeys Big-6 type turkeys during 16 - week rearing. Addition of copper in the form of chelate with lysine in the amount of 10 mg Cu-dm-3H2O was an experimental factor. The aim of the present study was to define the effect of copper, as a supplement to drinking water on body mass and on accumulation of mineral components in selected turkey tissues. Analysis of the obtained results has shown that a supplement of chelate caused an increase of the final body mass of the turkeys, but it did not considerably influence the mass of the analyzed tissues. It was found that the copper affected the increase of Cu, Zn and Fe contents, in all tissues studied. Livers were characterized by the highest Cu retention, whereas the least amounts of this element were accumulated in breast muscles of the birds. A long application of copper did not effect an excessive accumulation of this mineral component in kidneys and muscles of turkeys and did not pose a threat for the consumers' health, as the copper content in these tissues did not exceed the permissible norms (5 mg-kg-1; according to PN-A-86524:1994).

Opis:
Chloramine-T is a widely used disinfectant for the treatment of gill diseases of fish in freshwater, and more recently attention has turned to its use in seawater. However, despite the wide use of chloramine-T, few studies have examined its toxicity to fish. Therefore, the aim of the current study was to examine the effects of disinfection by Chloramine-T on the muscle tissue of rainbow trout (Oncorhynchus mykiss Walbaum) using oxidative stress biomarkers (levels of 2-thiobarbituric acid reactive substances and derivatives of oxidatively modified proteins) and biochemical enzymes’ activity (alanine- and aspartate aminotransferases (ALT and AST), lactate dehydrogenase (LDH)) to observe the its toxic effects. The endpoints obtained from this study will be useful to monitor the effects of disinfectant bathing with Chloramine-T for this species of fish. In the disinfectant group, rainbow trout (n = 11) were exposed to Chloramine-T in final concentration of 9 mg per L. Control group of trout (n = 11) was handled with water from basin in the same way as Chloramine-T exposed group. Fish were bathed with Chloramine-T for 20 min and repeated three times every 3 days. Two days after the last bathing fish were sampled to study. Our results showed that Chloramine-T bathing caused the decrease of the lipid peroxidation as well as ALT and AST activity and significant decrease of LDH activity (by 339%, p = 0.017) compared to controls. Chloramine-T markedly affects on lactate and pyruvate metabolism and resulted to decrease of LDH activity. Correlative analysis revealed that the lipid peroxidation level is correlated with ALT and AST activity in the muscle tissue of unhandled control group. In the muscle tissue of trout disinfected by Chloramine-T, LDH activity is correlated positively with ALT and AST activity. Thus, the skeletal muscles of fish play an important role in the processing of lactate through the gluconeogenic and glycogenic pathways including a greater potential for biosynthesis. Our studies indicated that Chloramine-T in dose of 9 mg per L could at least partly attenuate oxidative stress and can be used for prophylactic disinfecting treatment of rainbow trout. Oxidative stress and biochemical alterations could be effectively used as potential biomarkers of Chloramine-T toxicity to the fish in the warning signal for pharmaceutical exposure to aquatic organisms. However, more detailed studies on using of these specific biomarkers to monitor the disinfectant treatment in aquaculture are needed.
Chloramina-T jest szeroko stosowanym środkiem dezynfekcyjnym i terapeutycznym do leczenia chorób skrzeli ryb w wodach słodkich i morskich. Jednak pomimo szerokiego stosowania tego środka, tylko w niewielu badaniach analizowano jego toksyczność dla ryb (Powell i Harris 2004). W związku z tym celem pracy było zbadanie wpływu dezynfektanta chloraminy-T na tkankę mięśniową pstrąga tęczowego (Oncorhynchus mykiss Walbaum) z wykorzystaniem biomarkerów stresu oksydacyjnego (poziom produktów reagujących z kwasem 2-tiobarbiturowym, aldehydowe i ketonowe pochodne oksydacyjnej modyfikacji białek) oraz przemian metabolicznych (aktywność aminotransferaz alaninowej i asparaginianowej, dehydrogenazy mleczanowej, stężenie mleczanu i pirogronianu). Uzyskane wyniki końcowe będą przydatne do monitorowania skutków dezynfekujących kąpieli z chloraminą-T dla tego gatunku ryb. Nasze wyniki wskazują, że chloramina-T znacznie obniża peroksydację lipidów na tle zmniejszenia aktywności aminotransferaz alaninowej i asparaginianowej oraz dehydrogenazy mleczanowej. Ponadto obniżona aktywność dehydrogenazy mleczanowej spowodowała zmniejszenie aktywności aminotransferaz. Zatem mięśnie szkieletowe ryb odgrywają ważną rolę w obróbce mleczanu przez glukoneogenezę i glikogenezę. Chloramina-T w dawce 9 mg na litr może przynajmniej częściowo złagodzić stres oksydacyjny w tkance mięśniowej pstrąga tęczowego i może być stosowana do dezynfekcji tego gatunku ryb. Konieczne są jednak bardziej szczegółowe badania dotyczące korzystania z tych specyficznych biomarkerów do monitorowania dezynfekcji w akwakulturze.

With the numerous improvements in cereal tissue and wheat anther culture, it is necessary to determine which of the improvements should be combined for optimal response. This study was conducted using one highly responsive cultivar of wheat (Triticum aestivum L. cv. Pavon 76) to test the effectiveness of pre-culture cold treatment (0 or 3-7 days at 5°C) of anthers, five initiation basal media, and various changes in Murashige-Skoog regeneration media. A cold pre-culture treatment was inhibitory for all initiation media for embryoid initiation. Of the initiation media, P1, 85D12, and N6 were similar for embryoid initiation (0.80 to 0.90 embryoids/anther) without a cold pre-culture treatment. Plant regeneration was improved by the addition of amino acids or glucose, increased sucrose concentration, filter sterilizing the medium, and altering plant growth regulator concentrations. P1 medium which is normally used for embryoid initiation was also beneficial for improving plant regeneration. Ethylene inhibitors were generally not beneficial.

The effect of two types of modified atmosphere (MA1: 69% N2, 25% CO2, 5%O2, 1% CO; MA2: 70% N2, 30% CO2) on changes in physical and chemical parameters (pH, aw - water activity, TVBN - total volatile basic nitrogen, TMA - trimethylamine, FFA - free fatty acids, PV - peroxide value, TBA - thiobarbituric acid) in muscle tissues of the silver carp was monitored in the study. The samples were stored at temperatures +2 ± 2oC for 18 days. Changes in gas volumes (CO2 and O2) in MAs were also monitored. CO2 levels increased in MA1 but decreased in MA2. At the end of 18 days of storage, a significantly (P < 0.01) lower water activity (aw) levels were found in samples packaged under MA1, in contrast to samples packaged under MA2 where water activity values showed considerable fluctuation. Variations in pH values in the two types of MA showed similar trends. Sample pH gradually decreased until day 9 of storage. On day 11, muscle tissue pH increased markedly and then began to decrease again. The overall decrease in pH values was more profound in samples packaged under MA1. TVBN and TMA levels in samples packaged under the two types of MAs remained almost identical until day 9 of the experiment. Later, however, significantly (P < 0.01) higher levels of both parameters were found in muscle tissues packaged under MA1. FFA concentrations in silver carp samples in MA1 were significantly lower (P < 0.01) throughout the experiment. The PV increased significantly in both muscle samples tested. Greater fluctuations in this parameter’s values throughout the experiment were observed in samples packaged under MA2. Faster rates of oxidation (P < 0.01) were found in samples packaged under MA1 starting on day 9. Maximum TBA values in MA1 and MA2 were observed on days 14 and 18 of the experiment, respectively. From the course of proteolytic and oxidative changes point of view, the more appropriate combination of gases for silver carp storage seems to be the mixture of 70% N2 and 30% CO2 (MA2), which allows for muscle storage of up to 9 days. We recommend TVBN as a suitable indicator of freshness, and TBA assay as a suitable indicator of the extent of oxidative processes.

In the literature available hitherto there are many reports on enzymatic changes in tissues and a correlated rise in enzymatic activity in blood serum during experimental and human trichinellosis. In this study we were characterised proteinase activity in crude extracts from muscles of mice infected with Trichinella spiralis and T. pseudospiralis and the dynamics of their changes in different stages of disease. The activity of proteinase in muscle of mice infected with T. spiralis showed an increase in the 1 st- 5th week post infection, and then a slight decrease. The biggest proteinase activity was observed in 5th- 6th week post infection. In the muscle of mice infected with T. pseudospiralis the increase of proteinase activity was observed in 1st-4th week post infection. In the 4th week the activity reached its maximum and in the 5th- 10th week post infection there was a decrease of the activity in comparison with the control. As we could see, the dynamics of the changes of proteinase activity in mice is similar in the case of the disease with other biochemical and immunological indices observed in trichinellosis and with the increase of regeneration and transformation processes observed in histopathological studies.

A variety of Acanthamoeba spp. pathogenic strains were isolated from water pools in the area of Poznań. In many cases amoebae invaded lugs, kidney heart, liver and diaphragm causing significant changes in these Organs. The presence of amoeba outside of CNS was found mainly in the early periods of infection. Amoeba isolated from different organs of dead animals did not exhibit any specificity in relation to the organs in following infections. Infection with both low-and high-virulent strains resulted in pathomorphological changes of similar character. However, changes induced by low-virulent parasites developed later and were less extensive.

Opis:
Apple leaves of the cvs. Pionier and Romus were observed to have cytological changes under the influence of A. schlechtendali feeding. Cells of lower epidermis and spongy parenchyma were subjected to destruction. No changes, however, were found in the cell layers of palisade parenchyma and upper epidermis.
W liściach jabłoni odmian Pionier i Romus obserwowano zmiany cytologiczne zachodzące pod wpływem żerowania A. schlechtendali. Zniszczeniu ulegały komórki epidermy dolnej oraz komórki miękiszu gąbczastego. Nie stwierdzono natomiast żadnych zmian w warstwie komórek miękiszu palisadowego i epidermy górnej. Uszkodzenia komórek epidermy polegały na rozerwaniu ścian i nekrozie. W warstwie miękiszu gąbczastego następował proces degradacji pojedynczych lub partii komórek, ściany komórkowe ulegały pogrubieniu, a w przestworach międzykomórkowych pojawiały się złogi.
