Opis:
Today, occurrence of nanotechnology is making life easier, increasing the individual happiness and attracts attention to the benefits to be provided by it. As with any technology, nanotechnology also has risks, which are discussed here. In this study, toxic effects of ZnOTiO₂ nanocomposite on various growth parameters of Drosophila melanogaster and removal of these effects by Cimin grape ( Vitis vinifera L. ssp., Cimin) were investigated. For this purpose, assessment of the number of offspring and the survival rate were performed by feeding flies with ZnOTiO₂ (1 mg/ml) and Cimin grape seed and fruit extracts (5 and 10 mg/ml). In the first stage, larvae of the the same age were put into the medium of each application group and the adult individuals were recorded. In the second stage, in order to determine the number of offspring in all treatment groups, in a set of experiments the grape extracts and ZnOTiO₂ were only applied to the female members, while in another set of experiments, the grape extracts and ZnOTiO₂ were applied only to male members, belonging to the same application group. The total number of flies developed from the eggs laid during ten days of pair mating was counted. As a result, survival rate and the offspring number in the application groups, that were only exposed to ZnOTiO₂ were found to show a decline, compared with the other groups. At the same time, the values obtained from the groups, exposed to the ZnOTiO₂ and grape extracts together, were close to control group values. This result supports the healing effects of Cimin grape, which is abundant in Erzincan.

OmpF plays very important roles in the influx of antibiotics and bacterial survival in the presence of antibiotics. However, high-grade mutant OmpF and its function in decreasing bacterial survival rate have not been reported to date. In the present study, we cloned a high-grade mutant OmpF (mOmpF) and sequence analysis suggested that over 45 percent of the DNA sequence was significantly mutated, leading to dramatic changes in over 55 percent of the amino acid sequence. mOmpF protein was successfully expressed. When grown in the presence of antibiotic, the bacterial survival rate decreased and the antibiotic inhibition zone became larger with the increase of the mOmpF. It was concluded that concentration of high-grade mutant mOmpF dramatically influenced the bacterial survival rate. The study presented here may provide insights into better understanding of the relationships between structure and function of OmpF.

Opis:
HER-2/neu overexpression is considered an important prognostic factor, which may predict an aggressive clinical course of several tumours, including cervical cancer. Material and method: HER-2/neu expression was assessed by immunohistochemical technique in 298 patients with stage IB and IIA cervical cancer, of either planoepithelial or adenomatous type. HER-2/neu expression was compared in groups of patients selected based on generally accepted prognostic factors, e.g. clinical stage, histological type, presence of metastases to regional lymph nodes, invasion of vascular space and degree of radiation-induced destruction of tumor. Using the Cox model, a multivariate analysis of effect of selected prognostic factors was performed, both in relation to overall survival and to symptom-free survival. Results: Positive reaction to HER-2/neu was obtained in 135 cases (45.3%); the reaction was pronounced (3+) in 23 cases (7.8%) and moderate (2+) in 41 cases (13.7%). No positive reaction was obtained in 163 cases (54.7%). Analysis of survival yielded the following results: probability of 5-year overall survival for the entire patient population was 90.9% and that of 10 years – 85.2%. Overall survival curve for the entire study population is presented. Recurrence-free 5- and 10-year survival rates were 83.3% and 82.3%, respectively. Symptom-free 5- and 10-year survival rates were significantly influenced by: lymph node involvement and their number, invasion of vascular space, histological type and HER-2/neu expression. Cox analysis demonstrated a significant correlation between risk of recurrence or death and the following factors: lymph node metastases and their number, invasion of vascular space, histological type of tumour and HER-2/neu expression. The risk of recurrence or death with a single nodal metastasis and several nodal metastases is 2.4- and 5.1-fold greater than when no nodal metastases are present. The fact of vascular space involvement results in a 3-fold increase of risk of recurrence or death. In the case of planoepithelial cancer, the risk of recurrence or death is 0.4-fold that for adenomatous cancer. Finding of a strong HER-2/neu expression (2+, 3+) increases 2.7-fold the risk of therapeutic failure. Conclusions: No statistically significant differences were found in the HER-2/neu expression in groups of patients defined based on clinical-pathological parameters (i.e. clinical stage, histological type, regional lymph node metastases, radiation-induced cervix sterilization). The following prognostic factors had a significant impact on symptom-free survival: histological type, presence and numbers of lymph node metastases, invasion of vascular space, as well as TP53, BCL2 and HER-2/neu expression. Among all factors analysed, most significant prognostic impact, both in terms of overall survival and symptom-free survival, was associated with involvement of pelvic lymph nodes.
Prawdopodobieństwo przeżycia bez nawrotu choroby 5 i 10 lat wyniosło odpowiednio 83,3% i 82,3%. Stwierdzenie silnej ekspresji HER-2/neu (2+, 3+) zwiększa 2,7 razy ryzyko niepowodzenia leczenia. Wnioski: Nie stwierdzono istotnych statystycznie różnic w ekspresji HER-2/neu w grupach chorych wyróżnionych na podstawie parametrów kliniczno-patologicznych (tj.: stopnia klinicznego zaawansowania, typu histologicznego, przerzutów do regionalnych węzłów chłonnych i popromiennej sterylizacji szyjki macicy). Istotny wpływ statystyczny na przeżycia bezobjawowe miały następujące czynniki prognostyczne: typ histologiczny, przerzuty do węzłów chłonnych i ich liczba, inwazja przestrzeni naczyniowej oraz ekspresja TP53, BCL2 i HER-2/neu. Spośród wszystkich analizowanych czynników największy wpływ prognostyczny zarówno na przeżycia całkowite, jak i bezobjawowe miała obecność przerzutów do węzłów chłonnych miednicy.

Telomerase, undetectable in normal somatic cells, plays a critical role in carcinogenesis of the majority of human tumors including lung carcinoma. The aim of our study was to determine human telomerase reverse transcriptase (hTERT) mRNA expression in patients with non-small cell lung cancer (NSCLC) in order to estimate its usefulness as diagnostic and/or prognostic factor. hTERT expression was analyzed in a group of 12 females and 28 males with NSCLC using Quantitative Real-Time Polymerase Chain Reaction (QRT-PCR method) in cancerous and non-cancerous lung tissues. Results were analyzed according to clinical data and one-, two-, and five-year survival rates. hTERT expression in the cancerous tissue was significantly higher than in the lung parenchyma free from neoplasm infiltration (p<0.05). There was no significant association between hTERT expression in the tumor tissue and age, gender, grading or clinical stage. A significant difference in hTERT expression between two types of histopathological patterns (adenocarcinoma and squamous cell carcinoma) was detected (p=0.01). No association between hTERT expression in NSCLC specimens and survival rates was found. hTERT mRNA detection by QRT-PCR in tumor and corresponding cancer-free tissues can be used as a diagnostic marker in patients with NSCLC, but seems not to be a prognostic factor.

The natural occurrence of the shrimp (Penaeus monodon Fabricius, 1798) and seagrass (Cymodocea rotundata Asch. & Schweinf.) is found in the area of Indian Ocean, Indonesia, for central Pacific. Vibriosis can cause death in larvae, juvenile and adult shrimp almost close to 100%. Antibacterial substances from secondary metabolites are produced by plants and animals to reduce the use of synthetic antibiotics. The purpose of this research was to determine the ability of secondary metabolites contained in the bacterial extract of the association of sea grass Cymodocea rotundata (isolate code BA.1) to treat vibriosis in tiger shrimp (Penaeus monodon). This research was conducted at the Laboratory of Integrated Biotechnology and Laboratory of Aquaculture, Fisheries and Marine Sciences Faculty, Universitas Padjadajaran, and at the Brackish and Southern Ocean Aquaculture Development Center Pangandaran, from June to September 2018. The research method used was experimental, with completely randomized design (CRD) consisting of five treatments with three replications including; control treatment (concentration of 0 mg·L–1), 150 mg.L–1, 300 mg·L–1, 450 mg·L–1 and 600 mg·L–1. Observations made include clinical symptoms (morphology, behavior) and survival rate. Clinical symptoms of infected tiger shrimp during the in vivo test were red spots on the abdomen and necrosis of some leg and tail segments. The behavior of tiger shrimp mostly shows less responsive movements to fish-feed on the first day, and showing responsive and active behavior on the fifth day. In vivo test results for 14 days showed that the highest survival rate of 51.67% was in case of the addition of antibacterial extracts of BA.1 with a concentration of 300 mg·L–1.

Sewage sludge constitutes a source of valuable biogenic raw materials, but it is a carrier of many pathogenic microorganisms and viruses. Subjected to an effective sanitization by means of the process of composting, it is suitable to use in agriculture as fertilizers. The aim of this study was to observe the survival rate of Suid Herpesvirus under the influence of the temperature alone (water bath) as well as in sewage sludge subjected to the process of composting (pile). The samples were taken at different time intervals, and the virus titres were determined. The viruses survived considerably longer under laboratory conditions: at 30°C as long as 21 days, at 40°C - 93 hours, and at 50°C - less than an hour. In the compost pile, in spite of the lack of the thermophylic phase, the total survival time of the viruses ranged from 34 to 44.5 hours, which indicates the vast importance of other physicochemical factors, apart from the temperature, contributing to virus inactivation.

Modelling lifetime data with simple mathematical representations and an ease in obtaining the parameter estimate of survival models are crucial quests pursued by survival researchers. In this paper, we derived and introduced a one-parameter distribution called the Agu-Eghwerido (AGUE) distribution with its simple mathematical representation. The regression model of the AGUE distribution was also presented. Several basic properties of the new distribution, such as reliability measures, mean residual function, median, moment generating function, skewness, kurtosis, coefficient of variation, and index of dispersion, were derived. The estimation of the proposed distribution parameter was based on the maximum likelihood estimation method. The real-life applications of the distribution were illustrated using two real lifetime negatively and positively skewed data sets. The new distribution provides a better fit than the Pranav, exponential, and Lindley distributions for the data sets. The simulation results showed that the increase in parameter values decreases the mean squared error value. Similarly, the mean estimate tends towards the true parameter value as the sample sizes increase.

White lupin is an important source of proteins, the content of which amounts to 36–40% in its seeds. Breeding of new varieties usually takes several years. The present study aimed to shorten the breeding cycle by applying the single-seed descent technique together with in vitro culture of embryos dissected from immature seeds. The effects of temperature on embryo development were studied in indeterminate vs. determinate varieties in in vitro culture using the following temperature regimes:(1) 8 °C; (2) 16 °C in the first week, then 8 °C; (3) 20 °C in the first week, then 20/18 °C; and (4) 16 °C throughout the culture period. The embryos were cultured on the standard Murashige and Skoog medium. The observed growth rate of plantlets as well as the survival of plants ex vitro showed that at lower temperatures (8–16 °C) the plants developed slowly, but their ex vitro survival rate was higher compared to those grown at a higher temperature (20 °C). In addition, the results indicated that the in vitro embryo development rate of determinate varieties is significantly lower than that of indeterminate varieties.

Opis:
This research was conducted from October 2018 until April 2019 in the Faculty of Fisheries and Marine Sciences, Universitas Padjadjaran. This study aims to determine the optimum potassium diformate dosage which was added to feed to increase the immune status of Pangasianodon hypophthalmus (Sauvage, 1878). The research method used is the experimental Complete Random Design (CRD) with 4 treatments and 3 replications. The treatments are A (control), B (0.3% KDF), C (0.5% KDF), and D (0.7% KDF). Observed parameters are survival rate, gross clinical signs, intestine pH levels, total leukocyte count (white blood cells), total erythrocyte count (red blood cells), and water quality. The results showed that the 0.5% KDF addition in feed is the most effective to improve immune status. This treatment also is the most effective to improve Pangasius immune status after being challenged with A. hydrophila with survival rate of 93.33%, leukocyte count 2.98×106 cells/mm3 and erythrocyte count 1.02×106 cells/mm3. Increased leukocyte count was 65.26% and erythrocyte count - 58.23%.
