Opis:
Despite numerous theories, the etiology and pathogenesis of primary varicose veins remain unclear. The etiology of chronic venous diseases (CVDs) known as chronic venous insufficiency (CVI) is related to leukocyte trapping. Leukocyte trapping involves trapping of white cells in vessel walls followed by their activation and translocation outside the vessel. Release of reactive oxygen species (ROS) from trapped white cells has been documented. Superoxide dismutase (SOD) directly inhibits the generation of free radicals and compounds that are produced during oxidation by ROS, such as malonyldialdehyde (MDA). The aim of this study was to determine the involvement of free radicals in the etiology of venous changes. The following material was used for the study: fragments of sufficient or insufficient venous system and varices from 31 patients diagnosed with chronic venous disease in the 2nd or 3rd degree, according to clinical state, etiology, anatomy and pathophysiology (CEAP), which were qualified for surgical procedure. The levels of oxidative stress markers strongly correlated with lesions observed by USG in insufficient and varicose veins. In both a higher concentration of MDA was observed, which is a sign of lipid peroxidation. Antioxidative mechanisms, SOD activity and total antioxidative power expressed as FRAP were inversely proportional to MDA concentration. In insufficient and varicose veins both FRAP and SOD activities were significantly lower than in normal veins. The severity of clinical changes was inversely dependent on the efficiency of scavenging of ROS, which additionally proves the participation of free radicals in pathogenesis of CVDs.

Plants are constantly exposed to the influence of biotic and abiotic stress factors that significantly affect the induction of resistance responses. Biotic factors include bacteria, fungi and insects such as the common cockchafer (Melolontha melolontha) that harm many tree species, including pine and larch. The adult beetles damage needles, while the larvae (grubs) feed on roots. The aim of the experiment was to determine how plants cope with the damage caused by these insects. The activity of antioxidant enzymes such as peroxidase (POD) and superoxide dismutase (SOD) were determined three and six weeks after the insect-induced damage and the results show that the peroxidase activity, as measured with guaiacol (GPOD), was higher in the damaged roots of larch. The POD activity measured with syringaldazine (SPOD) was slightly higher in damaged pine needles as well as pine roots. SOD activity was higher in the damaged roots of both species as compared to the needles. It is believed that changes in the activity of POD and SOD as well as the presence of another form of SOD in the roots of larch, indicate direct involvement of these enzymes in the plants’ response to damage caused by M. melolontha.

Mutants of Saccharomyces cerevisiae devoid of Cu,Zn-superoxide dismutase are hypersensitive to a range of oxidants, hyperbaric oxygen and hyperosmotic media, show lysine and methionine auxotrophy when grown under the atmosphere of air and have a shortened replicative life span when compared to the wild-type strain. Ascorbate and other antioxidants can ameliorate these defects, which may be a basis of simple tests sensing the presence of antioxidants. In particular, tests of growth on solid medium (colony formation) in the absence of methionine and/or lysine, or in the presence of 0.8 M NaCl can be useful for detection and semiquantitative estimation of compounds of antioxidant properties. Hypoxic atmosphere was found to increase the sensitivity of detection of antioxidants. The test of abolishment of lysine auxotrophy showed a concentration dependence of the antioxidant effects of cysteine and N-acetylcysteine which, however, lost their protective action at high concentration, in contrast to glutathione which was effective also at higher concentrations.

The aim of the study was an evaluation of changes in protein level and activity of SOD isoenzymes, and the participation of AP-1 and NF-κB in subsequent stages of colorectal cancer development. Studies were conducted on 65 colorectal cancers. Controls were unchanged colon regions. Activity of SOD isoenzymes, lipid peroxidation level (TBARS), and protein level of SOD1, SOD2, AP-1 and NF-κB were determined. We found that the protein level and activity of SOD isoenzymes and protein level of AP-1 and NF-κB change in subsequent stages of clinical advancement of colorectal cancer, according to UICC (I-IV), and in grades of tumor cells differentiation (G1-G3). These results indicate adaptation of colorectal cancer cells to oxidative stress, and show that the observed changes of SOD activity and protein level depend on gradual progression of colorectal cancer, and suggest an impairment of processes regulated by AP-1 and NF-κB which are critical for tumor progression (proliferation, differentiation and apoptosis).

Tropospheric ozone is a harmful air pollutant which may cause oxidative stress in plant cells, leading to biochemical and physiological changes and yield reduction. The aim of the study was to examine the cumulative effect of long-term ozone stress on the activity of antioxidative enzymes in leaves of ozone sensitive and resistant tobacco cultivar growing at the sites of various ozone concentrations. The one-month experiment was conducted in the 2009, 2010 and 2011 growing seasons with different meteorological conditions and ozone concentrations. The activity of SOD, APX and GuPX was measured four times at weekly intervals. The highest tropospheric ozone concentration was recorded in 2010 along with high level of solar radiation and temperature. The enhanced ozone level caused the increase in the activity of all examined enzymes. However, at more elevated ozone level the activity of all enzymes was higher in sensitive (Bel W3) than resistant (Bel B) tobacco cultivar. On the other hand, at lower ozone level the activity of examined enzymes was rather similar in both cultivars or even higher in ozone-resistant one, which was especially valid for SOD. A positive correlation between the activity of all enzymes and ozone concentration was shown in both cultivars. The presented results show that meteorological conditions modify tropospheric ozone concentration and real plant response to this environmental pollutant. It is highly important to interpret week-by-week plant response and environmental conditions bearing in mind the cumulative ozone effect resulting from previous weeks conditions.
Zwiększone stężenie ozon troposferycznego w powietrzu jest szkodliwe dla roślin, gdyż przyczynia się do powstania stresu oksydacyjnego prowadzącego do zmian biochemiczno-fizjologicznych oraz redukcji plonów. Celem pracy było zbadanie wpływu długotrwałego stresu ozonowego na aktywność enzymów antyoksydacyjnych w liściach wrażliwej oraz odpornej na ozon odmiany tytoniu rosnących na stanowiskach ekspozycyjnych różniących się stężeniem tego zanieczyszczenia. Przeprowadzono trzy trwające jeden miesiąc doświadczenia w latach 2009, 2010 i 2011 charakteryzujących się różnymi warunkami meteorologicznymi oraz różnym stężeniem ozonu. Aktywność SOD, APX oraz GuPX oznaczano czterokrotnie (w odstępach tygodniowych) w czasie ekspozycji roślin. Najwyższe stężenie ozonu stwierdzono w roku 2010, kiedy odnotowano również wysoki poziom promieniowania słonecznego oraz temperatury powietrza. Zwiększone stężenie ozonu przyczyniło się do wzrostu aktywności badanych enzymów. Większą aktywność enzymów w warunkach podwyższonego stężenia ozonu stwierdzono u wrażliwej (Bel W3) niż odpornej (Bel B) odmiany tytoniu. Przy niższym stężeniu ozonu, aktywność badanych enzymów była zbliżona u obu odmian, lub nawet wyższa u odmiany odpornej, co było szczególnie widoczne w przypadku SOD. U obu odmian stwierdzono pozytywną korelacje pomiędzy aktywnością badanych enzymów oraz stężeniem ozonu. Uzyskane wyniki wskazują, że warunki atmosferyczne wpływają na stężenie ozonu troposferycznego, co z kolei wpływa na rzeczywistą odpowiedź rośliny na to gazowe zanieczyszczenie powietrza. Niezwykle ważne jest, aby uzyskane dane dotyczące odpowiedzi roślin na zanieczyszczenie powietrza ozonem interpretować z uwzględnieniem czasowych zależności, w szczególności efektu kumulowania się wpływu ozonu w poprzedzającym przedziale czasowym.

The aim of this study was to evaluate the influence of copper supplements on superoxide dismutase (SOD) activities in the kidneys and liver of rats exposed to moderate doses of cadmium chloride. SOD activities were measured weekly for a period of 28 days. The examinations involved male Wistar rats divided into three groups. Rats in group I were considered the controls whereas rats in group II and III were administered intragastrically for 7 d, 14 d, 21 d, and 28 d a water solution of cadmium chloride corresponding to 10 mg Cd/kg diet. All rats fed a standard laboratory chow LSM (Agros Motycz, Poland) containing 5 mg Cu/kg except for the rats in group III in which animals were fed an LSM diet supplemented up to 50 mg Cu/kg. The experimental procedure included evaluation of water and feed intake, body weight gains, liver, kidneys, testicles, heart and spleen to body ratio, and hepatic and renal SOD activities measured after 7 d, 14 d, 21 d, and 28 d exposure to cadmium or cadmium plus copper. Results indicated that SOD activity in cadmium or cadmium plus copper groups changed in comparison with the control values. Supplements of copper increased significantly hepatic and renal SOD activities after 21 and 28 d of exposure when compared to those found in the controls and cadmium exposed rats. Moreover, rats fed the copper supplement diet improved body weight gain in comparison with that in the cadmium exposed rats. Results suggest that copper may stimulate a protective activity against toxic action of cadmium given at doses resembling its concentrations in the areas contaminated with this metal.
Celem pracy było określenie wpływu diety wzbogaconej w miedź na aktywność dysmutazy ponadtlenkowej (SOD) w wątrobie i nerkach szczurów szczepu Wistar eksponowanych na umiarkowane dawki chlorku kadmu. W badaniach uwzględniono 3 grupy zwierząt: grupę I stanowiły zwierzęta kontrolne, grupę II zwierzęta eksponowane na kadm podawany dożołądkowo w ilościach odpowiadających 10 mg Cd/ kg paszy oraz grupę III, w której kadm podawano tak jak w grupie II, a ponadto zwierzęta eksponowane były na chlorek miedzi podawany w paszy LSM (Agros Motycz, Polska) w stężeniu 50 mg Cu/kg. Stężenie miedzi w standardowej paszy LSM podawanej zwierzętom w grupach I i II wynosiło 5 mg Cu/kg. W doświadczeniu określano spożycie paszy i wody, przyrosty masy ciała, masy względne wątroby, nerek, serca, śledziony i jąder oraz aktywność SOD po 7, 14, 21 i 28 d podawania kadmu. Wyniki badań wskazują, że zatruwanie szczurów chlorkiem kadmu zmieniało aktywność SOD w wątrobie i nerkach w porównaniu z danymi w grupie kontrolnej. Dodatek miedzi do paszy zwiększył statystycznie istotnie aktywność SOD po 21 i 28 dniach ekspozycji na kadm w porównaniu z danymi w grupie kontrolnej i grupie eksponowanej tylko na kadm. Warto także podkreślić, że dodatek miedzi do paszy szczurów zatruwanych kadmem powodował zwiększone przyrosty masy ciała w porównaniu z danymi uzyskanymi od zwierząt karmionych standardową paszą i eksponowanych na kadm. Uzyskane wyniki sugerują, że miedź może mieć ochronne działanie u zwierząt eksponowanych na kadm drogą pokarmową w ilościach, które mogą być obecne w paszy na terenach skażonych tym metalem.

The activity of Cu,Zn superoxide dismutase in the fluid obtained from eye lens capsules after cataract surgery was investigated in samples obtained from patients with senile cataract and with senile cataract combined with diabetes mellitus. Two parameters were measured and compared: the frequency of occurrence of detected superoxide dismutase activity and the relative activity of the enzyme in samples derived from senile cataract patients versus those from the patiens affected additionally by diabetes mellitus. It was confirmed that the decrease of superoxide dismutase activity during cataract was additionally promoted by diabetes mellitus.

Superoxide dismutase 1 (SOD1) is a metalloenzyme that catalyzes the disproportionation of superoxide into molecular oxygen and hydrogen peroxide. In this study, the human SOD1 (hSOD1) gene was cloned, expressed and purified. The hSOD1 gene was amplified from a pool of Bxpc3 cell cDNAs by PCR and cloned into expression vector pET-28a (+). The recombinant soluble hSOD1 was expressed in E. coli BL21 (DE3) at 37°C and purified using nickel column affinity chromatography. Soluble hSOD1 was produced with a yield of 5.9 μg/mL medium. As metal ions can have a certain influence on protein structure and activity, we researched the influences of different concentrations of Cu2+ and Zn2+ on hSOD1 activity at induction and the time of activity detection. The results implied that Cu2+ and Zn2+ do not enhance SOD1 expression and solubility; they can, however, improve the catalytic activity at induction. Meanwhile, Cu2+ and Zn2+ also enhanced the enzyme activity at the time of detection. Furthermore, most other bivalent cations had the potential to replace Zn2+ and Cu2+, and also improved enzyme activity at the time of detection.

Fluorimetric method based on oxidation of the fluorogenic 6-methoxy-2-naphthaldehyde was applied to evaluate temporal and population variability of the specific activity of salivary aldehyde dehydrogenase (ALDH) and the degree of its inactivation in healthy human population. Analyzed was also its dependence on drinking and smoking habits, coffee consumption, and its sensitivity to N-acetylcysteine. Both the specific activity of salivary ALDH and the degree of its inactivation were highly variable during the day, with the highest activities recorded in the morning hours. The activities were also highly variable both intra- and interpersonally, and negatively correlated with age, and this correlation was stronger for the subgroup of volunteers declaring abstinence from alcohol and tobacco. Moderately positive correlations of salivary ALDH specific activity with alcohol consumption and tobacco smoking were also recorded (rs ~0.27; p=0.004 and rs =0.30; p=0.001, respectively). Moderate coffee consumption correlated positively with the inactivation of salivary ALDH, particularly in the subgroup of non-drinking and non-smoking volunteers. It was found that mechanical stimulation of the saliva flow increases the specific activity of salivary ALDH. The specific activity of the salivary ALDH was strongly and positively correlated with that of superoxide dismutase, and somewhat less with salivary peroxidase. The antioxidant-containing drug N-acetylcysteine increased activity of salivary ALDH presumably by preventing its inactivation in the oral cavity. Some food-related aldehydes, mainly cinnamic aldehyde and anisaldehyde, were excellent substrates of the salivary ALDH3A1 enzyme, while alkenals, particularly those with short chain, were characterized by lower affinity towards this enzyme but high catalytic constants. The protective role of salivary ALDH against aldehydes in food and those found in the cigarette smoke is discussed, as well as its participation in diminishing the effects of alcohol- and smoking-related oxidative stress.

Cadmium (Cd), similarly to other heavy metals, inhibits plant growth. We have recently showed that Cd2+ either stimulates (1-4 μM) or inhibits (ł 6 μM) growth of soybean (Glycine max L.) cells in suspension culture (Sobkowiak & Deckert, 2003, Plant Physiol Biochem. 41: 767-72). Here, soybean cell suspension cultures were treated with various concentrations of Cd2+ (1-10 μM) and the following enzymes were analyzed by native electrophoresis: superoxide dismutase (SOD), catalase (CAT), peroxidase (POX) and ascorbate peroxidase (APOX). We found a significant correlation between the cadmium-induced changes of soybean cell culture growth and the isoenzyme pattern of the antioxidant enzymes. The results suggest that inhibition of growth and modification of antioxidant defense reactions appear in soybean cells when Cd2+ concentration in culture medium increases only slightly, from 4 to 6 μM.

Opis:
Activities of superoxide dismutase, catalase and glutathione peroxidase in erythrocytes of cystic fibrosis children were studied in order to estimate the severity of their deficiency. Our results point to increased susceptibility of erythrocytes of cystic fibrosis subjects to oxidative injury and indicate that the antioxidant status of patients should be carefully monitored.

Opis:
Lead, similar to other heavy metals and abiotic factors, causes many unfavorable changes at the subcellular and molecular levels in plant cells. An increased level of superoxide anion in Pisum sativum root cells treated with 1 mM Pb(NO3)2 evidenced oxidative stress conditions. We found increased activities of enzymatic components of the antioxidative system (catalase and superoxide dismutase) in the cytosol, mitochondrial and peroxisomal fractions isolated from root cells of Pisum sativum grown in modified Hoagland medium in the presence of lead ions (0.5 or 1 mM). Two isoenzyme forms of superoxide dismutase (Cu,Zn-SOD and Mn-SOD) found in different subcellular compartments of pea roots were more active in Pb-treated plants than in control. Increased amount of alternative oxidase accompanied by an increased activity of this enzyme was found in mitochondria isolated from lead-treated roots. These results show that plants storing excessive amounts of lead in roots defend themselves against the harmful oxidative stress caused by this heavy metal.
