Opis:
Service in a special unit of the Police is extremely tedious, both in physical and mental terms, with very high expectations for their policemen. It cannot arise only from official duties, but – above all – specific lifestyle, which should be the primary stimulus for taking them. It is confirmed by research conducted among policemen-antiterrorists, relating to their motivation in taking service in anti-terrorist subunit of the Police. Studies have also shown that such service is not fully in line with the expectations of the officers, and their priorities change over its course. A lot of factors have impact on it, negatively affecting the attitude of policemen, which were revealed in the study. The results can be used to create such conditions of service, which will reduce or eliminate loss of idealistic motives for taking service in the special unit of the Police.

Opis:
Chemical deglycosylation (dg) of sturgeon Acipenser gueldenstaedti Br. (αGTH) resulted in the loss of 83% of its initial carbohydrate content. It altered also recombinant dgαGTH + βGTH dimer molecule, reducing its immunoreactivity by 30%, and fully blocking the hormonal function. CD spectroscopy showed that deglycosylation led to changes in the secondary structure of dgαGTH and in the α-β recombinant. The sugar moiety of sturgeon αGTH is suggested to play an important role in maintaining the biological function of the hormone dimer molecule.

Opis:
The present study was undertaken to elucidate whether an increased, but physiological, amount of progesterone (P4) supplied to the porcine corpus luteum affects luteal secretion of activin A and inhibin a-subunit (Inha) in freely moving gilts. On day 9 of the estrous cycle (EC), both ovarian arteries and both ovarian veins of gilts (n=5) were cannulated. Progesterone was infused into the right ovarian arteries in gilts on days 10, 11 and 12 of the EC at a rate adequate to its physiological retrograde transfer found during the middle luteal phase of the EC. The P4 infusion rate was 0.62 μg/min (day 10), 2x0.62 μg/min (day 11) and 3x0.62 μg/min (day 12). The left ovarian arteries were infused with saline (control). Blood samples were collected from both ovarian veins on days 10-12 of the EC before and after P4 or saline infusion. The mean plasma activin A level in the ovarian vein ipsilateral to the P4-infused ovary was higher (PcO.OOOl) on days 10-12 of the EC than this found in the contralateral ovarian vein. The level of activin A in the ovarian vein ipsilataral to the infusion of P4 was higher on days 11 (PcO.Ol) and 12 (P<0.0001) and tended to be higher (P<0.07) on day 10 of the EC than this in contralateral ovarian vein. The level of Inha in the ovarian vein ipsilateral to the P4-infused ovary on days 10-12 of the EC was not significantly different (P>0.05) than this found in the contralateral ovarian vein. The results of the present study indicate that a local elevation of P4 concentration in blood supplying the ovary during the middle luteal phase of the porcine EC affects ovarian secretion of activin A. The effect of P4 on the secretion of activin A suggested the existence of a short regulatory loop of a positive feedback between P4 being retrogradely transferred into the ovary and the secretion of this peptide.

We studied the shell and penis morphology, and cytochrome oxidase subunit I (COI) gene sequences, in minute, valvatiform hydrobiid gastropods from Slovensky Kras, Slovakia. The morphology confirmed the assignment of the studied snails to the genus Hauffenia, while in the molecular tree they were placed within the Hydrobiidae, and not close to Hauffenia. The results indicate that the Slovak valvatiform hydrobiids are two taxa which presumably represent two genera: Hauffenia Pollonera, 1898 and Lobaumia Haase, 1993. More molecular data on the Austrian and Hungarian taxa are needed.

Within the last decades the slug Arion lusitanicus has expanded its range over wide areas of Europe, in most of them it is now a serious pest. Poland has been invaded relatively recently (since the late 1980s). Considering the ecological importance of the slug, very little is known yet about the mechanism of invasion, establishment of new populations and influence on the native fauna and flora. The analysis of nucleotide sequences of mitochondrial cytochrome oxidase subunit 1 gene (cox1) revealed a great inter- and intrapopulation variation in the Polish populations of A. lusitanicus. The differentiation of all the studied Polish populations of A. lusitanicus is 0.2–2.2%, while two analysed Belgian populations are monomorphic and moderately genetically diverse at 0.8%. This indicates a heterogeneous origin of the Polish populations, probably resulting from multiple independent introduction events. The genotype found in the first four Polish populations (S. Poland) suggests that their origin is different from the remaining populations.

Recombinant subunit vaccines based on hemagglutinin proteins produced in bacteria (bacterial HAs) are promising candidates for enhancing the supply of vaccines against influenza, especially for a pandemic. Over 20 years after the failure to obtain the antigen with native HA characteristics in the early 1980’s, there are increasing data on successful production of HA proteins in bacteria. The vast majority of bacterial HAs have been based on the HA1 subunit of HA expressed separately or as a component of conjugate vaccines, but those based on the ectodomain and the HA2 subunit have also been reported. The most of HAs have been efficiently expressed as insoluble aggregates called inclusion bodies. Refolded and purified proteins were extensively studied for structure, the ability to bind to sialic acid-containing receptors, antigenicity, immunogenicity and efficacy. The results from these studies contradict the view that glycosylation determines the correct structure of the hemagglutinin, as they proved that bacterial HAs can be valuable vaccine antigens when appropriate folding and purification methods are applied to rationally designed proteins. The best evidence for success in bacterial production of protective HA is that vaccines based on proprietary Toll-like Receptor (VaxInnate) and bacteriophage Qβ-VLPs (Cytos Biotechnology) technologies have been advanced to clinical studies.

The effect of light irradiance on the amount of ATP synthase α-subunit in mesophyll (M) and bundle sheath (BS) chloroplasts of C4 species such as maize (Zea mays L., type NADP-ME), millet (Panicum miliaceum, type NAD-ME) and guinea grass (Panicum maximum, type PEP-CK) was investigated in plants grown under high, moderate and low light intensities equal to 800, 350 and 50 µmol photons m-2 s-1, respectively. The results demonstrate that α-subunit of ATP synthase in both M and BS chloroplasts is altered by light intensity, but differently in the investigated species. Moreover, we identified two isoforms of the CF1 α-subunit, called α and ά. The CF1 α-subunit was the major isoform and was present in all light conditions, whereas ά was the minor isoform in low light. A strong increase in the level of the ά-subunit in maize mesophyll and bundle sheath thylakoids was observed after 50 h of high light treatment. The α and ά-subunits from investigated C4 species displayed apparent molecular masses of 64 and 67 kDa, respectively, on SDS/PAGE. The presence of the ά-subunit of ATPase was confirmed in isolated CF1 complex, where it was recognized by antisera to the α-subunit. The N-terminal sequence of ά-subunit is nearly identical to that of α. Our results indicate that both isoforms coexist in M and BS chloroplasts during plant growth at all irradiances. We suggest the existence in M and BS chloroplasts of C4 plants of a mechanism(s) regulating the ATPase composition in response to light irradiance. Accumulation of the ά isoform may have a protective role under high light stress against over protonation of the thylakoid lumen and photooxidative damage of PSII.

New stands of Paramecium tredecaurelia, a rare species of the P. aurelia spp. complex, were identified in Thailand and Madagascar on the basis of mating reactions and molecular markers (rDNA and mtDNA). Analysis of DNA fragments showed that all P. tredecaurelia strains, the recently recorded ones and the ones known previously from France, Mexico, and Israel, form a monophyletic and well-defined clade in the P. aurelia species trees. All of these strains, collected from different localities around the world, represent identical or nearly identical haplotypes in terms of all the studied DNA fragments. Given the huge distances between particular collection sites, such a low level of variability of the studied sequences may result from a slow rate of evolution in P. tredecaurelia.

Paraphysomonas vestita is a unicellular, colorless, silica-scaled chrysophyte that plays an important ecological role in freshwater microbial communities as a consumer of prokaryotic and eukaryotic prey. There is little biogeographical information for this minute protist despite its significant role in aquatic food webs. In addition, the phylogenetic relationship of P. vestita to other taxa is unclear as P. vestita may be polyphyletic or a cryptic species complex. In this study, a clonal isolate from a freshwater sample of Laguna de Bay, Philippines was subjected to morphological study by electron microscopy and its small subunit ribosomal RNA (SSU rRNA) gene was sequenced. Morphological studies showed that the isolate possesses two unequal flagella emerging from the anterior part of the cell. Negative staining revealed the structure of the scales which consist of a baseplate with slightly thickened rim. The narrowing spine arises from the center of the baseplate. These results agree with previously studied isolates of P. vestita. The 18S rRNA gene sequence of the isolate had a very high similarity (99%) to P. vestita strain PV10. Phylogenetic analysis also showed that the isolate clustered with other Paraphysomonas sequences with high bootstrap support. Phylogenetic studies confirmed that P. vestita may be polyphyletic. No studies on the ultrastructure and phylogeny of a silica-scaled chrysophyte isolated in the Philippines have been reported so far. Results from this study may contribute to further ultrastructural and phylogenetic studies on aquatic flagellates and specifically to a revision of this potentially polyphyletic species.

To downregulate expression of the β1 integrin subunit in endothelial cells, plasmid encoding the ribozyme cassette containing hammerhead ribozyme flanked at the 5' terminus by tRNAVal and at the 3' terminus by constitutive transport element sequences was constructed. When used to transfect immortalized human endothelial cell line EA.hy 926, it selectively blocked the synthesis of the β1 integrin subunit and thus inhibited migration and proliferation of the cells. Thus, this construct may be a valuable tool to control the proangiogenic phenotype of stimulated endothelial cells.
