Opis:
Objectives: Growing evidence supports the reproductive and developmental toxicity of polycyclic aromatic hydrocarbons (PAHs) from prenatal and postnatal exposure, but the results of epidemiological studies regarding harmful effects of PAHs exposure on male reproductive system still remain limited and inconclusive. The aim of the present study was to investigate the relationship between 1-hydroxypyrene, a biomarker of polycyclic aromatic hydrocarbons exposure and semen quality. Materials and Methods: The study population consisted of 277 men attending an infertility clinic for diagnostic purposes and having normal semen concentration of 20-300 mln/ml or slight oligozoospermia (semen concentration: 15-20 mln/ml) (WHO 1999). All the men were healthy and under 45 years of age. All participants were interviewed and provided a semen sample. The interview included questions concerning demographics, socio-economic status, medical history related to past diseases which may have an impact on semen quality, lifestyle factors and occupational information. Concentrations of 1-hydroxypyrene (1-OHP) in the urine samples were analyzed using high performance liquid chromatography (HPLC). Results: A positive association was found between the level of 1-OHP in urine and sperm neck abnormalities as well as the percentage of static sperm cells (p = 0.001, p = 0.018, respectively). Additionally, exposure to PAHs measured by 1-OHP in urine decreased semen volume and the percentage of motile sperm cells (p = 0.014, p = 0.0001, respectively). Conclusions: Presented findings indicate that the environmental level of PAHs exposure adversely affects male semen quality. The future large-scale studies should incorporate different biomarkers to generate a more accurate and full assessment of the effects of PAHs exposure on male fertility.

Objectives The objective of this study was to assess sperm quality and deoxyribonucleic acid (DNA) integrity of coke oven workers exposed to polycyclic aromatic hydrocarbons (PAHs) as compared to control subjects. Material and methods The coke oven workers (N = 52) and administrative staff (N = 35) of a steel plant served as the exposed and control groups, respectively. Exposure to PAHs was assessed by measuring 1-hydroxypyren. Analysis of sperm quality (concentration, motility, vitality, and morphology) was performed simultaneously with sperm DNA integrity analysis, including DNA fragmentation, denaturation, bulky DNA adducts, and 8-oxo-7,8-dihydro-2’-deoxyguanosine (8-oxo-dGuo). A questionnaire was conducted to collect demographic and potential confounding data. Results The coke oven workers had lower percentages of sperm motility, vitality and normal morphology than the control group, but the difference was not significant. For DNA integrity, the coke oven workers had significantly higher concentrations of bulky DNA adducts and 8-oxo-dGuo than the control subjects (p = 0.009 and p = 0.048, respectively). However, DNA fragmentation percentages did not significantly increase as compared to those in the subjects from the control group (p = 0.232). There was no correlation between sperm quality parameters and DNA integrity indicators. Conclusions Occupational exposure of the coke oven workers to PAHs was associated with decreased sperm DNA integrity. Int J Occup Med Environ Health 2016;29(6):915–926

Introduction and objective. The integrity, stability and composition of sperm chromatin are of great importance in the fertilizing potential of male gametes and their capacity to support normal embryonic development. In this study, the author presents the current state of knowledge about the sperm epigenetic profile and risk of cancer. Abbreviated description of the state of knowledge. The obtaining of pregnancy and the state of health of the baby depends on the quality of the genetic material of both the female and the male. Health behaviours and environmental factors directly affect the quality of sperm, as well as the human egg cell and, consequently, on the reproductive capabilities, the course of pregnancy and the state of the newborn. There exist two thoroughly investigated epigenetic modifications: DNA methylation and histone modifications. The process of DNA methylation can be also a fundamental factor contributing to the development of cancer, where epigenotype undergoes significant modifications. When considering numerous DNA aberrations in the male gamete, the most commonly encountered is DNA fragmentation, particularly in infertile subjects. Surprisingly, an intracytoplasmatic sperm injection study of mice oocytes, using spermatozoa with a high DNA Fragmentation Index (DFI), revealed that a considerable percentage of adults born as a result of this method, showed a significant increase in the incidence of abnormal behavioural tests, malformations, cancer and signs of premature aging. Summary. The issue of assisted procreation raises the need to look for an appropriate treatment for males with sperm chromatin abnormalities. As a result, the fight against smoking addiction becomes the obvious necessity. Moreover, the reasonable solution nowadays seems to be supplementation with micronutrients and folic acid. It has been proved that the process of DNA fragmentation is a phenomenon that intensifies over time. Therefore, there should be a pursuance for, as close as possible, to the moment of ejaculation, application of semen to reproductive techniques. Finally, epigenetic changes are suspected of being one of the factors responsible for the deterioration of male sperm parameters observed in recent decades.

Phthalate are found in the environmental samples due to their wide use in the industry as plasticizers. Di-n-butyl phthalate (DBP) is mainly used in nitrocellulose and polyvinyl acetate products as well as in personal-care products. This study was performed to investigate the influence of exposure to DBP on the quantity and quality (motility, morphology) and DNA damage (induction of micronuclei and DNA strand breaks) of male mice gametes. The estimation of DBP residues was also done. Eight weeks exposure to DBP (500 mg/kg bw and 2000 mg/kg bw) did not significantly affect testes and epididymes weights as well as sperm count. DBP clearly diminished sperm motility, enhanced frequency of abnormal sperm heads and not significantly increased DNA strand breaks in germ cells as well as frequency of micronuclei in spermatids. There were no bioacumulation of DBP in mice. Results suggest that DBP may affect the male mice germ cells.
Ftalany są powszechnie wykorzystywane w przemyśle jako plastyfikatory, dlatego też często występują w próbach środowiskowych. Ftalan di-n-butylu (DBP) jest składnikiem produktów nitrocelulozowych i poliwinylowych, jak również produktów do higieny osobistej. Celem pracy było zbadanie wpływu podawania DBP na ilość i jakość gamet męskich myszy (ruchliwość, morfologia) oraz na występowanie uszkodzeń DNA (indukcja mikrojąder oraz pęknięć nici DNA). Zbadano także stężenie pozostałości DBP w gametach. Ośmiotygodniowe narażanie na DBP (500 mg/kg mc i 2000 mg/kg mc) nie powodowało istotnych zmian w ciężarze jąder i najądrzy ani w liczebności plemników. Narażenie na DBP, zwłaszcza w dawce 2000 mg/kg mc wpływało wyraźnie na zmniejszenie ruchliwości plemników i zwiększenie odsetka plemników o nieprawidłowej budowie morfologicznej. Powodowało też nieznaczne zwiększenie pęknięć nici DNA w gametach oraz zwiększenie częstości występowania mikrojąder w spermatydach. Nie wykazano bioakumulacji DBP w gonadach samców myszy. Uzyskane wyniki sugerują, że DBP może oddziaływać niekorzystnie na męskie komórki rozrodcze myszy.
