Opis:
The symptoms of oxidative stress and antioxidative response were investigated on Norway spruce seedlings subjected to freezing desiccation conditions. Three-year-old seedlings were exposed to freezing desiccation at -3oC and -10oC for 45 days in two acclimation stages: autumn (October) and winter (January). The stress enhanced the production of reactive oxygen species (ROS): superoxide radical anion (O2.-), and hydrogen peroxide (H2O2). Concentrations of low molecular antioxidants: glutathione (GSH), ascorbic acid (AsA) and a-tocopherol declined at both low temperatures and acclimation stages. The activity of superoxide dismutase (SOD) increased with ROS production, while guaiacol peroxidase (POX) activity decreased. The freeze-induced desiccation of needles was significantly correlated with the cold hardiness (LT50), the level of low-molecular antioxidants, and POX activity, but not with SOD activity. Under extreme freezing desiccation conditions, these reactions continued, leading to the degradation of membrane phospholipids and a strong decrease in cold hardiness. The results show that membranes are the primary site of injury induced by ROS, produced under the influence of low temperature combined with dehydration. The acclimation response of Norway spruce needles to the oxidative stress generated by long-term cold and/or freezing desiccation is discussed.

Opis:
The unique goniatite−rich pyritic level, 1.6 m thick, exposed at Kostomłoty (Holy Cross Mountains, central Poland) represents a distinct, local biotic event in the Early Frasnian interval corresponding to the inception of a major geochemical (carbon cycling) perturbation in the stagnant deep−water and oxygen−deficient Kostomłoty basin. The taxonomic and palaeoecologic characteristics of molluscan fossil associations from the Goniatite Level are presented. Most of the goniatitids and orthoconic nautiloids from the studied fossil assemblages are juvenile conchs or protoconchs, or incomplete phragmocones and represent the genera Acanthoclymenia and Linguatornoceras, the former being predominant, and a single adult specimen probably of the genus Koenenites. In contrast, gastropods and bivalves are generally well preserved and identifiable to species level. Two new species are described: a gastropod Palaeozygopleura (Bohemozyga) pyritica sp. nov. and a bivalve Glyptohallicardia multicostata sp. nov. These studied molluscs and also amphiporoids are allochthonous elements which must have been transported into the deeper settings during sea−level rise and flooding of fringing reefs (Timan Event) and/or storm events, and there were mixed with pelagic cephalopods. Probably, a sea−level rise even led to flooding of nearby areas, and thus to introduction of pelagic material (juvenile cephalopods) into reefal settings, and then back to the deeper water again by the quasi−estuarine circulation of water masses.

Opis:
The method of ploidy level determination depends on the equipment, expertise of the lab or farm staff, and on the material studied (fish species, specimens' age and size, etc.). The combination of some simple techniques can be very helpful for fast, reliable and inexpensive assessment of effects of genome manipulations. Among indirect methods the nucleoli quantification seems to be the most convenient one, except that it is limited to species possessing one locus of nucleolar organizer regions (NORs) (rainbow trout as an example). Species with multilocus NORs (like brook trout) need another method, for instance sizing of erythrocytes nuclei. These techniques can be useful for identification of products resulting from new approaches in biotechnology applied to fish culture, as polyploidisation or hybridisation, for characteristics of transformation levels of organisms found in the wild or those produced due to experimental treatments.

Opis:
The aim of the research described in this paper is to classify groundwater level patterns in pine stands of forest fresh sites. The approach has been employed with the view to facilitate explaining interactions between physiographic characteristics and groundwater dynamics in prospective research. The methods which have been employed so far for explanation of groundwater level dynamics interactions with local physiographic conditions do not let to draw firm conclusions. The classification of groundwater dynamics patterns requires employing more sophisticated methods, because of a relatively extensive range of groundwater dynamics site-to-site variability expressed, e.g., by amplitude and cycle period. The methods of groundwater patterns classification proposed in the literature focused either on hydrogeological criteria or, if related to forest ecosystems, focused on water balance elements in forest habitats. The area selected for investigation represented typical features for the Northern European Lowland forests defined by soil and form of terrain pattern shaped by the last glacial period (Vistulian glaciation) and dominant share of Scots Pine (Pinus sylvestris L.) in stand species composition. The research period covered the 2002-2007 hydrological years. The measurement used in analysis covered 35 sites equipped with measurement wells.

Opis:
The objective of this study was to determine the abundance and species composition of thrips collected from ears of Triticum durum Desf. in relation to plant protection intensity. The experiment was set up at the Experimental Research Station of Wrocław University of Environmental and Life Sciences, at Pawłowice (51o09’ N; 17o06’ E), Poland, in 2006-2008. It was conducted on durum wheat plants at three different levels of plant protection intensity and on common winter wheat plants. The samples of thrips were collected from the ears of the plants at the milk maturity stage of the grain. During three years of the research, eight species of thrips were identified from Triticum durum. Most often the eudominants were Haplothrips aculeatus, Limothrips cerealium and L. denticornis. We have demonstrated that there was no statistically significant effect of the plant protection intensity on the thrips abundance. The host species, on the other hand, did not affect the species composition of the studied insects.
Celem badań było poznanie liczebności i składu gatunkowego przylżeńców (Thysanoptera) występujących w kłosach pszenicy twardej (Triticum durum Desf.) w zależności od intensywności ochrony tej rośliny. Doświadczenie prowadzono w latach 2006-2008, w Rolniczym Zakładzie Doświadczalnym w Pawłowicach (51o09’ N; 17o06’ E), należącym do Uniwersytetu Przyrodniczego we Wrocławiu. Badaniami objęto pszenicę twardą bez ochrony chemicznej, chronioną środkami chemicznymi oraz porównawczo pszenicę zwyczajną. W celu określenia składu gatunkowego przylżeńców pobierano kłosy, których ziarniaki były w fazie dojrzałości mlecznej. W ciągu trzech lat badań w kłosach pszenicy twardej oznaczono 8 gatunków przylżeńców. Do eudominantów najczęściej należały Haplothrips aculeatus, Limothrips cerealium i L. denticornis. Nie stwierdzono wyraźnego wpływu zastosowanej ochrony roślin na liczebność badanej grupy owadów na pszenicy twardej. W kłosach pszenicy twardej występowały te same gatunki przylżeńców jak w kłosach pszenicy zwyczajnej.

Opis:
The representatives of the genus Paramecium are well-studied ciliates and can be used in water quality assessment and the determinations of saprobic levels. For these applications, a clear and unambiguous identification of ciliate assemblages is essential, which is typically based on morphological characters requiring a sound taxonomic knowledge and experience in species determination including microscopic identification of both living and stained specimens. Therefore, we developed and applied specific PCR primers for the detection of species belonging to the genus Paramecium and the Paramecium aurelia complex. These primers were successfully tested with different Paramecium species including representatives of the P. aurelia complex as well as closely related species like Frontonia sp. and Tetrahymena sp. in both experimental and environmental samples. These primers can be used in a simultaneous approach achieving fast and reliable results with regard to determination of ciliate community and water assessment.

Opis:
The purpose of this study was to investigate the effects of two cropping systems (intensive and integrated) on infection level of winter and spring wheat kernels by Fusarium species. Field experiments were conducted with winter wheat ‘Tonacja’ and spring wheat ‘Bombona’ during two consecutive growing seasons (2007/2008 and 2008/2009 - winter wheat, and 2008 and 2009 - spring wheat). A rainfall level in 2009 from the last decade of May to the first decade of July was 2.5-times higher than that in 2008. After the harvest, kernels were surface disinfected with 1.5% NaOCl solution for 2 min and then analysed for the infection level by different species of Fusarium. Fusaria were isolated on CZID medium and identified on the basis of macro - and micro-morphology on three media (PDA, SNA and a medium containing tannin). Our results demonstrate that the wheat grain infection by Fusarium depended mainly on a rainfall level. The intensive cropping system was more conducive to the grain infection by fusaria in comparison to the integrated one. The most frequent species were F. avenaceum in 2008, and F. graminearum, F. avenaceum and F. poae in 2009.

Opis:
The taxonomie value of phytolith assemblages and their degree of variability within different species of the same genus is still an undervalued issue in the botanical range of phytolith studies. However the understanding of grass phytolith variance and its implications to plant systematics is doubtless. In the present study phytoliths of the lateral shoots (leaves) of nine, globally distributed Poa species (Pooideae - Poaceae) are described. Phytoliths were recovered from Poa specimens by the dry ashing technique. Altogether 6223 disarticulated phytoliths were counted (approximately 500-700 phytoliths per species) in 54 plant samples, which cover six shoots of nine species. Not only the relative frequency of each morphotype was calculated, but measurements were conducted to determine the biogenic silica content of Poa lateral shoots. A phytolith reference collection for the nine selected species of a worldwide importance was also compiled. The description of the most significant phytolith morphotypes and their taxonomic relationships are given here. Results suggest that the biogenic silica content of the Poa lateral shoots was determined to be relatively high within all nine species. Phytolith assemblage data was subjected to multivariate statistical analyses (e.g., CA and PCA) in order to find differences and similarities among the nine Poa species. Results show that the two closely related Poa of the P. pratensis species group, namely the P. pratensis and P. angustifolia, only slightly differ from the other Poa species if we consider their rondel-trapeziform short cells (SC) phytolith frequencies.
