This review summarizes and discusses several analytical methods involving the preparation and use of new solid phase extractant. A literature survey of the last ten years offering a critical review of these new sorbents available for use in trace analyte enrichment is provided.

This study describes optimization of headspace solid phase microextraction followed by gas chromatography equipped with a flame ionization detector for toluene at trace level in spiked urine. The parameters affecting the extraction and gas chromatographic determination of analytes were studied: extraction time and temperature; desorption time and temperature; addition of NaCl; and pH, volume and agitation of the sample. Optimized headspace extraction was carried out at 30 °C for 6 min in the presence of 0.2 g⋅m–1of NaCl in the sample solution. Also, sample volume and sample pH were optimized at 5 ml and 7 (neutral pH), respectively. Desorption of the analytes was carried out at 250 °C for 60 s. The optimized procedure was validated with 3 different pools of spiked urine; it showed good reproducibility over 6 consecutive days and 6 within-day experiments. The study also determined the accuracy, linearity and detection limits of this method.

Opis:
Ceftiofur sodium (CFT), a third-generation cephalosporin for parenteral use, is commonly used in veterinary medicine against aerobic Gram-positive and Gram-negative bacteria as well as certain anaerobes. Its broad spectrum of activity and resistance to beta-lactamases result from the presence of methoxyimino and aminothiazole moieties at C-7 in the cephalosporin structure. The aim of this study was a comprehensive evaluation of the stability of CFT in the solid phase and in aqueous solutions. A fast and sensitive HPLC isocratic method was used for the determination of CFT degradation in the solid phase and in aqueous solutions. CFT degradation occurred according to a first-order reaction depending on the substrate concentration. The kinetic and thermodynamic parameters of CFT degradation in the solid phase were calculated. General acid-base hydrolysis of CFT was not observed in the solutions of hydrochloric acid, sodium hydroxide, phosphate (pH 5.84 – 7.25), acetate (pH 3.65 – 5.48) and borate (pH 7.49 – 10.07) buffers. CFT was the most stable in the pH range 2 – 6. The susceptibility of CFT to degradation under the influence of stress factors (pH, temperature, buffer components concentration, relative air humidity) should be considered in terms of storage conditions and the preparation of the product for administration.

Simple and rapid reverse phase high-performance liquid chromatography (RP-HPLC) method was developed and validated using Phenomenex Gemini c18 (4.6 x 250 mm, 5 μ) reverse phase column for the determination of LAF in human plasma, Solid Phase Extraction (SPE) technique was used for the extraction of analyte, detection was carried out by Photo Diode Array detector at 216 nm. Chromatographic resolution of the LAF was achieved within 4.6 min by using mobile phase Methanol and 5 mM Di-Potassium Hydrogen Phosphate Buffer (pH 9.5) (80:20, v/v), flow rate was 1.0 mL/min. Calibration curve was linear with correlation coefficient of 0.9996 in the range of 50-1000 ng/mL, Limit of Detection (LOD) and Limit of Quantitation (LOQ) were 10 ng/mL and 30 ng/mL respectively, intra and inter-day deviations were lower than 3.92 % and 3.98 % respectively. The overall mean recovery of LAF was 94.57 %. No any endogenous constituents were found to interfere at retention time of the analyte. This new RP-HPLC method was successfully validated and may be applied to conduct bioavailability & bioequivalence studies of LAF.

The paper presents the results of DSC analysis of steel B500SP produced in the process of continuous casting, which is intended for the production reinforcement rods with high ductility. Studies were carried out in the temperature range below 1000°C in a protective atmosphere of helium during samples heating program. The main objective of the study was to determine the temperature range of austenite structure formation during heating. As a result of performed experiments: Ac1s, Ac1f – temperatures of the beginning and finish of the eutectoid transformation, Ac2 – Curie temperature of the ferrite magnetic transformation and the temperature Ac3 of transformation of proeutectoid ferrite into austenite were elaborated. Experimental determination of phase transformations temperatures of steel B500SP has great importance for production technology of reinforcement rods, because good mechanical properties of rods are formed by the special thermal treatment in Tempcore process.

A simple and effective cooling device based on a thermoelectric cooler was applied to cool the SPME fiber. The device was used for quantitative extraction of aromatic hydrocarbons in the air. Several factors such as coating temperature, extraction temperature and relative humidity in the laboratory setting were optimized. Comparison of the results between the cold fiber SPME (CF-SPME) and NIOSH 1501 method on standard test atmosphere indicated a satisfactory agreement. The CF-SPME and SPME method were also compared. The results revealed that CF-SPME has the most appropriate outcome for the extraction of aromatic hydrocarbons from the ambient air. The cold fiber SPME technique showed good results for several validation parameters. Under the optimized conditions, the limits of detection (LOD) and the limits of quantification (LOQ) ranged from 0.00019 to 0.00033 and 0.0006 to 0.001 ng ml−1 , respectively. The intra-day relative standard deviation (RSD) showed ranging from 4.8 to 10.5%.

Mackerel of Scomberomorus commerson (Lacepède, 1800) is a fish that has thick meat of a distinctive taste and is a favorite of Indonesian people. It is made into various types of processed seafood. The amount of edible flesh of the fish is 65%, meaning that the waste from the fish is 35%, including the head and bones. The purpose of this study was to determine the type and class of volatile compound components that can be drawn from mackerel head and fish bone, by making these into a broth. This study uses an experimental method by testing the composition of volatile flavor compounds and then undertaking proximate testing. The method of extracting volatile flavor compounds that was applied is Solid Phase Microextraction (SPME), while Gas Chromatography / Mass Spectrometry (GC / MS) was harnessed to identify the volatile flavor. Accordingly, 150 compounds were detected in the sample of mackerel head broth, as compared with 133 compounds in mackerel bone broth. The compounds that were detected are hydrocarbons, aldehydes, alcohols, ketones, organic compounds and others. That which has the largest proportion is pentadecane from the hydrocarbon group, with a value of 18,545%. The proximate analysis results showed that mackerel head broth samples had a 96.08% water content, 1.55% ash content, 0.28% fat content and 2.78% protein content, while samples of mackerel bone broth had a 96.69% water content, 1.54% ash content, 0.44% fat content and 1.84% protein content.

Steroid compounds are a kind of pollutants which are increasingly more frequently found in environmental samples. Due to the ever more increasing level of such pollutants and their biological activity, they are of interest to analysts. Our study involved the development of a simple and fast method for the assay of dutasteride in treated industrial waste water using high-performance liquid chromatography. As the steroid concentration in water is much lower than the limit of detection of the UV detector used, the compound assayed in the test water was pre-concentrated using solid-phase extraction. Our procedure has high accuracy and a precision of 2.6%, and the recovery of the analyte from water is 78.6%. The procedure enables the assay of dutasteride at a level of 2 microg/L in water.

The adsorption of pentafl uorobenzene on nine ionic liquid-based silicas was investigated using solid phase extraction. The effects of several variables such as the type of ionic liquid groups, adsorption time, temperatures and water ratio in the solution system were experimentally evaluated. The imidazole-chloride ionic liquid group based silica exhibited the highest adsorption efficiency under the optimized conditions of 5 min adsorption at 30°C in water/methanol (30:70, vol%) solution. In addition, the effects of pH, as well as type and concentrations of chloride salts were investigated. At pH values other than neutral and high salt concentration, the adsorption efficiency was reduced. Finally, the relative standard deviation of less than 5.8% over a 5-day period showed a high precision for the nine tested sorbents.

The applicability of silica gels, modified with ketoimine groups, in solid-phase extraction was tested. The surface characteristics of the modified silica was determined by elemental analysis, NMR spectra for the solid phases (29Si CP MAS NMR), analysis of pore size distribution of the silica support, and nitrogen adsorptiondesorption. The newly proposed sorbents with ketoimine groups were used for extraction of endocrine-disruptor compounds, extracted from water and powder milk. The properties of the sorbents were compared with commercial C18 ones.

Factors affecting solid phase extraction (SPE) of trans,trans-muconic acid (ttMA), as a benzene biomarker, including sample pH, sample concentration, sample volume, sample flow rate, washing solvent, elution solvent, and type of sorbent were evaluated. Extracted samples were determined by HPLC-UV (high performance liquid chromatography-ultraviolet). The analytical column was C18, UV wave length was 259 nm, and the mobile phase was H2O/methanol/acetic acid run at flow rate of 1 ml/min. A strong anion exchange silica cartridge was found successful in simplifying SPE. There was a significant difference between recoveries of ttMA when different factors were used (p < .001). An optimum recovery was obtained when sample pH was adjusted at 7. There was no significant difference when different sample concentrations were used (p > .05). The optimized method was then validated with 3 different pools of samples showing good reproducibility over 6 consecutive days and 6 within-day experiments.

Carcinoembryonic antigen (CEA) is an oncofoetal cell surface glycoprotein that serves as an important tumour marker for colorectal and some other carcinomas. Its immunoglobulin-like structure places CEA within the immunoglobulin superfamily. CEA functions in several biological roles including homotypic and heterotypic (with other CEA family members) cell adhesion. Cell-cell interaction can be modulated by different factors, e.g., post-translational modifications such as glycosylation. The purpose of this study was to examine whether changes in carbohydrate composition of CEA oligosaccharides can influence homotypic (CEA-CEA) interactions. In order to modulate glycosylation of CEA we used two different glycosylation mutants of Chinese hamster ovary (CHO) cells, Lec2 and Lec8. Lec2 cells should produce CEA with nonsialylated N-glycans, while Lec8 cells should yield more truncated sugar structures than Lec2. Parental CHO (Pro5) cells and the glycosylation deficient mutants were stably transfected with CEA cDNA. All three CEA glycoforms, tested in a solid-phase cell adhesion assay, showed an ability to mediate CEA-dependent cell adhesion, and no qualitative differences in the adhesion between the glycoforms were observed. Thus, it may be assumed that carbohydrates do not play a role in homotypic adhesion, and the interactions between CEA molecules depend solely on the polypeptide structure.

A devised methodology presented here allows the determination of di-n-butyl phthalate in environmental samples (water and landfill leachate) using solid-phase extraction (SPE) and gas chromatography. It is developed based on the use of a gas chromatograph with an FID detector. Preliminary testing has also provided extraction parameters and conditions for chromatographic determination, with calibration applied by reference to an internal standard. The linearity of the calibration curve has been tested in DBP concentrations ranging from 0 to 7.5 mg/L, with the data obtained showing that, throughout this range, the detector readings as a function of the DBP concentrations remain linear (R2 coefficient >0.99). The average levels of recovery of DBP from aqueous solutions of phthalates are in the range of 97-109%, while the corresponding figures for leachates are 85-101%. The values of the coefficients of variation associated with the results obtained do not exceed 5%. The results, therefore, indicate that the applied extraction method is effective as regards DBP extraction from both water and landfill leachate, while numerous other substances present in the leachate from landfill sites apparently do not affect the correct determination of di-n-butyl phthalate by the method developed.

A majority of biochemical reactions are often catalysed by different types of enzymes. Adsorption of the enzyme is an imperative phenomenon, which protects it from physical or chemical degradation resulting in enzyme reserve in soil. This article summarizes some of the key results from previous studies and provides information about how enzymes are adsorbed on the surface of the soil solid phase and how different factors affect enzymatic activity in soil. Many studies have been done separately on the soil enzymatic activity and adsorption of enzymes on solid surfaces. However, only a few studies discuss enzyme adsorption on soil perspective; hence, we attempted to facilitate the process of enzyme adsorption specifically on soil surfaces. This review is remarkably unmatched, as we have thoroughly reviewed the relevant publications related to protein adsorption and enzymatic activity. Also, the article focuses on two important aspects, adsorption of enzymes and factors limiting the activity of adsorbed enzyme, together in one paper. The first part of this review comprehensively lays emphasis on different interactions between enzymes and the soil solid phase and the kinetics of enzyme adsorption. In the second part, we encircle various factors affecting the enzymatic activity of the adsorbed enzyme in soil.

Multiresidue matrix solid-phase dispersion (MSPD) technique with capillary gas chromatographic detection for determination and quantification of ten commonly used organophosphorus pesticides (chlorpyrifos, chlorpyrifos-methyl, diazinon, dimethoate, fenitrothion, formothion, malathion, parathion, phosalone, pirimiphos-methyl) in fruits and vegetables is described. The method gave high recoveries for all spiked samples in 80-120% range. The limits of detection were from 0.01 to 0.1 mg/kg for the compounds studied and were much lower than maximum residue levels (MRLs) allowed. The procedure was applied to the determination of ten organophosphorus pesticides in samples collected during the routine crop monitoring.
Celem badań było sprawdzenie możliwości zastosowania metody rozproszenia matrycy w fazie stałej (MSPD) do oznaczania pozostałości najczęściej stosowanych związków fosforoorganicznych w owocach i warzywach. Porównano wyniki oznaczeń dziesięciu najczęściej stosowanych insektycydów z tej grupy związków uzyskane dwiema metodami: MSPD i klasyczną z wykorzystaniem ekstrakcji rozpuszczalnikiem organicznym. Obie metody okazały się równie skuteczne, a ich parametry (wartości odzysku, odchylenie standardowe, granice oznaczalności) porównywalne. Metoda MSPD jest jednak prostsza, mniej czasochłonna, a przede wszystkim bardziej ekonomiczna od metody klasycznej. Ograniczenie zużycia rozpuszczalników organicznych jest ważne ze względu na ich negatywny wpływ na zdrowie osób pracujących w laboratorium i środowisko. Metodę MSPD zastosowano w analizie pozostałości związków fosforoorganicznych w monitoringowych badaniach próbek owoców i warzyw.

Due to the difficulty of detecting traces of organic acid mixture in an aqueous sample and the complexity of resolving UV-Vis spectra effectively, a combinatory method based on a self-made radical electric focusing solid phase extraction (REFSPE) device, UV-Vis detection and partial least squares (PLS) calculation is proposed here. In this study, REFSPE was used to enhance the extraction process of analytes between the aqueous phase and the membrane phase to enrich the trace of mixed organic acid efficiently. Then, the analytes, which were eluted from the adsorption film by ethanol with the assistance of an ultrasonic cleaning machine, were detected with UV-Vis spectrophotometry. After that, the PLS method was introduced to solve the problem of overlapping peaks in UV-Vis spectra of mixed substances and to quantify each compound. The linearly dependent coefficients between the predicted value of the model and the actual concentration of the sample were all higher than 0.99. The limit values of detection for benzoic acid, phthalic acid and p-toluene sulfonic acid were found at 9.9 μg/L, 12.2 μg/L and 13.8 μg/L with the relative recovery values between 84.8% and 117.9%. The RSD (n = 20) values of each component are 1.17%, 1.11% and 0.86%, respectively. Therefore, the proposed combined method can determine traces of complex materials in an aqueous sample efficiently and has wonderful potential applications.

The aim of the study was to evaluate the ability of Galactomyces geotrichum MK017 for the biosynthesis of 2-phenylethyl alcohol and optimization of the culture medium composition in order to increase the yield of the product. The culture of Galactomyces geotrichum MK017 strain was carried out in the laboratory scale. For isolation of volatiles Solid Phase Microextraction (SPME) has been used. The identification and quantification of aroma compounds produced by examined microflora was determined by gas chromatography and mass spectrometry (GC/MS) system. The results showed that the tested mould Galactomyces geotrichum MK017 shows the potential for the production of 2-phenylethyl alcohol, but also for the synthesis of phenylacetaldehyde, phenylacetic and phenyllactic acids. For the optimization of the 2-phenylethyl alcohol production yield four different medium composition have been tested. The bioprocess of aroma compound production by tested microorganism was the most efficient on the medium composed of sucrose (80 g/l) and L-phenylalanine (21 g/l) and pH value of 5.0. Using this composition in a batch culture of 770 ml volume the highest concentration of 2-phenylethyl alcohol has been obtained – 6 mg/l. At the same time amount of phenylacetaldehyde, phenylacetic acid and phenyllactic acidhas reached 2.2 mg/l, 10.66 mg/l and 32.3 mg/l respectively.
Celem pracy było określenie zdolności pleśni Galactomyces geotrichum MK017 do biosyntezy 2-fenyloetanolu oraz optymalizacja składników podłoża produkcyjnego w celu zwiększenia ilości uzyskanego produktu. Hodowlę pleśni Galactomyces geotrichum MK017 przeprowadzono w skali laboratoryjnej. Do identyfikacji związków lotnych zastosowano metodę mikroekstrakcji do fazy stałej (SPME). Identyfikacji oraz oceny ilościowej uzyskanego związku zapachowego dokonano z wykorzystaniem chromatografii gazowej sprzężonej ze spektrometrią mas (GC/MS). Uzyskane wyniki potwierdziły zdolność badanej pleśni Galactomyces geotrichum MK017 do produkcji 2-fenyloetanolu oraz do syntezy aldehydu fenylooctowego, kwasu fenylooctowego oraz kwasu fenylomlekowego. W celu optymalizacji produkcji 2-fenyloetanolu przetestowano cztery różne podłoża hodowlane. Największą produkcję 2-fenyloetanolu otrzymano na pożywce zawierającej sacharozę (80 g/l) i L-fenyloalaninę (21 g/l) oraz przy pH pożywki na poziomie 5.0. Uwzględniając powyższe zawartości poszczególnych składników pożywki, uzyskano w hodowli okresowej w 770 ml pożywki, najwyższą koncentrację 2-fenyloetanolu – 6 mg/l. Równocześnie w próbie otrzymano aldehyd fenylooctowy, kwas fenylooctowy oraz kwas fenylomlekowy odpowiednio w ilościach: 2.2 mg/l, 10.66 mg/l i 32.3 mg/l.

Odprowadzanie ścieków deszczowych z terenów zurbanizowanych związane jest z koniecznością przeciwdziałania zrzutowi znacznych ładunków zanieczyszczeń, w tym zawiesiny łatwo opadającej i ropopochodnych oraz tłuszczów, do odbiorników ścieków. W tym celu, przed skierowaniem ścieków do kanalizacji deszczowej, stosowane są różnego rodzaju separatory mające za zadanie oddzielić i zatrzymać fazy rozproszone - ciekłą i stałą. Efektywność ich pracy jest zatem miarą ochrony wód odbiorników ścieków deszczowych. Praca niniejsza przedstawia wyniki trójwymiarowych badań modelowych efektywności pracy prostego separatora fazy stałej oczyszczającego ścieki deszczowe. Badania modelowe zostały przeprowadzone w komercyjnym programie CFD Fluent Ansys Inc. Zaprezentowane obliczenia symulacyjne objęły przepływ ścieków deszczowych zawierających trzy frakcje fazy rozproszonej. Zastosowano trzy zróżnicowane prędkości przepływu ścieków deszczowych oraz trzy średnice ziaren fazy rozproszonej charakterystyczne dla frakcji piaskowej, pylastej i ilastej. Czas trwania odwzorowywanego zjawiska przepływu ścieków przez separator oraz zawartość procentową danej frakcji oparto o wartości charakterystyczne dla warunków lubelskich. Wyniki zaprezentowanych obliczeń numerycznych obejmowały rozkład prędkości przepływu wody i poszczególnych badanych frakcji przez separator, a także masę oraz rozkład przestrzenny zatrzymanych osadów w objętości zbiornika. Przeprowadzone badania umożliwiły ocenę skuteczności zatrzymywania poszczególnych frakcji oraz kształt objętości czynnej zbiornika separatora w funkcji prędkości przepływu. Ze względu na brak walidacji modelu otrzymane wyniki należy traktować jako wyniki badań wstępnych.
Storm water management in urbanized areas is directly connected to limiting the significant discharge of pollutants, including TSS, fats and oil derivatives, to the receivers. Thus, the various types of separators allowing to separate and retain the dispersed solid or liquid phase are applied before surface runoff waters enter the storm water system. Therefore, their efficiency is a measure of storm water receiver quality protection. This paper presents the results of three dimensional modeling studies concerning the operation efficiency of simple solid phase separator treating storm water. Our numerical researches were performed in the commercial CFD (computational fluid dynamics) software Fluent Ansys Inc. The presented numerical simulations covered flow of storm water containing there factions of dispersed fraction through the solid phase separator. Three velocities of flow and three particles’ dimensions representative for sand, silt and clay fractions were applied. Time duration of modeled storm water flow as well as percentage distribution of applied solid phases were based on values typical for Lublin region. Results of our numerical calculations covered velocities of flow distribution for storm water and tested solid phases as well as the accumulated masses and spatial development of retained solid phases particles inside the separator’s water body. The performed numerical simulations allowed assessment of retaining efficiency for the applied solid phase fraction and shape of separator’s active volume as a function of storm water velocity of flow. The lacking validation of our modeling attempt influences the fact that our studies should be treated as preliminary.

During the illness are released volatile organic compounds with specific smell which could have in diagnosis of diseases. The first aim of the study was qualitative and quantitative analysis of exhaled breath samples obtained from patients with lung cancer, healthy volunteers and people with other lung diseases by gas chromatography-mass spectrometry. This study showed that twenty compounds propane, ethanol, isobutane, butane, propanal, 1-propanol, 2-propanol, 2-methylfuran, 2-butanone, benzene, 2-pentanone, pentanal, hexanal, cyclohexanone, 4-heptanone, 2,4-dimethylheptane, 2,3,4-trimethylhexane, 2,3,5-trimethylhexane, 4-methyloctane, α-pinene separated two research groups of patients and healthy controls. The second goal was to evaluate the sensitivity and specificity of canine scent detection using 5 station scent lineup. Among lung cancer patients and complementary samples, overall sensitivity of canine scent detection was 85.54%, while specificity was 71.84%.

The oil bearing rose (Rosa damascena) is the most important rose species in terms of fragrances and flavourings. Due to the very short blooming period and excessive amount of flowers, considerable amount of the rose flowers wait for a long time until distillation. There are losses of essential oil yield and quality use of before waited petals. The cold storage and drying applications may be alternative method for evaluate of excessive amount of flowers. Therefore in this study it was aimed to determine the effects of storage on cold (4°C) and room condition (25°C) and convective drying with different temperatures (40, 50 and 60°C) in terms of changes in volatile compositions of oil rose flowers based on direct hexane extraction. Totally 20 volatile compounds were identified in fresh, stored and dried rose petals. The detected compounds varied according to the various storage and drying conditions. It was determined that phenylethyl alcohol, citronellol, geranyl acetate, nonadecane were predominant compounds on all treatments. In the study, storing treatments led to increase on the percentage of oxygenated monoterpenes (OM) while drying treatments led to decrease on OM. It was determined that storing and drying treatments led to increase on the percentage of benzenoid compounds (BC) and aliphatic hydrocarbons (AH).
Róża damasceńska (Rosa damascena) jest najważniejszym gatunkiem róży w kategoriach aromatów i zapachów. Ze względu na krótki okres kwitnienia i dużą liczbę kwiatów, znaczna liczba kwiatów róży czeka przez długi czas na destylację. Oznacza to straty olejków lotnych oraz pogorszenie jakości. Składowanie w chłodni i suszenie może być alternatywną metodą. Niniejsze badanie miało na celu określenie wpływu przechowywania w chłodni (4°C) i w warunkach pokojowych (25°C) oraz suszenia konwekcyjnego w różnych temperaturach (40, 50 i 60°C) na zmiany składu olejków lotnych kwiatów róży damasceńskiej w oparciu o ekstrakcję heksanu. W świeżych, magazynowanych i suszonych płatkach róży zidentyfikowano w sumie 20 składników lotnych. Różniły się one w zależności o warunków przechowywania i suszenia. Stwierdzono, że alkohol fenyloetylowy, cytronelol, octan geranylu oraz nonadekan były głównymi składnikami w przypadku wszystkich zabiegów. W niniejszym badaniu przechowywanie spowodowało wzrost utlenionych monoterpenów (OM), natomiast suszenie prowadziło do spadku OM. Stwierdzono, że zabiegi przechowywania i suszenia prowadzą do wzrostu zawartości składników benzenoidowych (BC) oraz węglowodorów alifatycznych (AH).

High quality DNA extractions are a prerequisite for genetic studies of a variety of organisms including fish. The current study focused on the applicability of different commercially available solid phase extraction (SPE) methods as the easiest and fastest methods for DNA extraction and their efficiency with different tissue qualities. These were represented by different kinds of pike tissues (fins, muscle, scales) preserved with different methods and stored at different temperatures over different periods of time (0.5 to 10.0 years). All DNA extractions were analysed according to their yield, purity, integrity and functionality in PCR based downstream analysis. Additionally mechanical pre-treatment of poor quality tissues (e.g. old or aged tissues) and efficient ethanol preservation of frozen bulk fin tissue were investigated. All SPE methods yielded functional DNA from very different qualities of pike tissues as shown by PCR analysis of small nuclear (microsatellite) and large mitochondrial (complete D-loop) DNA fragments. DNA from poor quality tissue can be extracted using single column SPE and in some cases mechanical pre-treatment even improved the yield. Good quality tissue as obtained e.g. from commercial fishermen as frozen bulk material is more efficiently preserved by thawing in ethanol at room temperature than at 2-8°C. DNA from these and air dried tissues was very efficiently prepared by applying reverse SPE in 96-well format, allowing for fast processing of a multitude of samples for high throughput analysis.

Ekstrakcja do fazy stałej SPE jest techniką stosowaną powszechnie do selektywnej izolacji analitów z ciekłych matryc. Może być wykorzystana do przygotowania prób analizowanych metodami chromatografii gazowej. W niniejszej pracy przedstawiono zastosowanie techniki SPE w analizie lotnych związków zapachowych ogórka. Zidentyfikowano główne związki lotne, a następnie porównano ich zawartość pomiędzy poszczególnymi próbami owoców ogórka transgenicznego. Otrzymane wyniki były porównywalne z wcześniejszymi wynikami badań własnych uzyskanymi za pomocą innych technik analitycznych. Ekstrakcja SPE może stanowić alternatywę lub uzupełnienie dla dotychczasowych oznaczeń substancji zapachowych.
Solid phase extraction (SPE) is a common technique applied to selective isolation of compounds from liquid matrix. It can be use in sample preparation for gas chromatography. The aim of this study was the application of SPE technique for evaluating the aroma of cucumber. The main volatiles were determined and then the comparison of their concentration between different cucumber samples was done. The results were comparable with those obtained previously by other analytical methods. The SPE technique could be an alternative or complement tool in the analysis of volatiles.

The two-dimensional distribution of gas-solid flow parameters is a great research significance to reflect the actual situation in industry. The commonly used method is the ultrasonic tomography method, in which multiple probes are arranged at various angles, or the measurement device is rotated as that in medicine, but in most industrial situations, it is impossible to install probes at all angles or rotate the measured pipe. The backscattering method, however, uses only one transducer to both transmit and receive signals, and the two-dimensional information is obtained by only rotating the transducer. Ultrasound attenuates greatly in the air, and the attenuation changes with frequency. Therefore, Comsol is used to study the reflection of particles with different radii in the air to ultrasound with various frequencies. It is found that the backscattering equivalent voltage is the largest when the product of ultrasonic frequency and particle radius is about 27.78 Hz·m, and the particle concentration of 30% causes the strongest backscattering. The simulated results are in good agreement with the Faran backscattering model, which can provide references for selecting the appropriate frequency and obtaining the concentration when measuring gas-solid two-phase flow with the ultrasonic backscattering method.
