Opis:
Among the numerous protocols that describe the extraction of DNA, those relating to the isolation of DNA from infected plants, are rare. This study describes a rapid and reliable method of extracting a high quality and quantity of DNA from rhododendron leaves artificially infected with Phytophthora cactorum, P. cambivora, P. cinnamomi, P. citrophthora, and P. plurivora. The use of the modified Doyle and Doyle protocol (1987) allowed us to obtain high quantity and quality DNA (18.26 μg from 100 mg of the fresh weight of infected leaves at the ratios of A260/280 and A260/230 – 1.83 and 1.72, respectively), suitable for conventional polymerase chain reaction (PCR) and real-time PCR amplifications.

Opis:
This work has derived the general formula, sufficiently precise for engineering calculations of base frequency of axisymmetric free vibrations of uniform, circular diaphragm type plates clamped at the edge with linearly variable thickness. To solve the boundary problem, the Cauchy’s function method and characteristic series have been applied. The above formula has been derived on the basis of Dunkerley’s formula which is based on the first major term of the characteristic series and results in the simplest, lower bound estimator. The analysis of the formula shows that the base frequency coefficient for diaphragm plates clamped at the edge depends to only a small extent on the Poisson’s ratio, and therefore it may be averaged in the case of construction materials. Comparison of the calculations results of the simplest lower bound estimators for the base frequency obtained by using proposed method, with the results known from the literature as precise solutions, including Conway method, confirmed the high accuracy of the proposed method.

In this article, a well-known technique, the variational iterative method with the Laplace transform, is used to solve nonlinear evolution problems of a simple pendulum and mass spring oscillator, which represents the duffing equation. In the variational iteration method (VIM), finding the Lagrange multiplier is an important step, and the variational theory is often used for this purpose. This paper shows how the Laplace transform can be used to find the multiplier in a simpler way. This method gives an easy approach for scientists and engineers who deal with a wide range of nonlinear problems. Duffing equation is solved by different analytic methods, but we tackle this for the first time to solve the duffing equation and the nonlinear oscillator by using the Laplace-based VIM. In the majority of cases, Laplace variational iteration method (LVIM) just needs one iteration to attain high accuracy of the answer for linearization anddiscretization, or intensive computational work is needed. The convergence criteria of this method are efficient as compared with the VIM. Comparing the analytical VIM by Laplace transform with MATLAB’s built-in command Simulink that confirms the method’s suitability for solving nonlinear evolution problems will be helpful. In future, we will be able to find the solution of highly nonlinear oscillators.

Opis:
In this study, a finite volume method (FVM) is suitably used for solving the problem of a fully coupled fluid flow in a rectangular domain with slip boundary conditions. Numerical solutions for the flow variables, viz. velocity, and pressure have been computed. The FVM, with an upwind scheme, has been implemented to discretize the governing equations of the present problem. The well known SIMPLE algorithm is employed for pressure-velocity coupling. This was executed with the aid of a computer program developed and run in a C-compiler. Computations have been performed for unknown variables with Reynolds numbers (Re) = 50, 100, 250, 500, 750 and 1000. The behavior of steady-state solutions of velocity and pressure of the fluid along horizontal and vertical through geometric center of the rectangular domain have been illustrated. We observed that, with the increase of the Reynolds number, the absolute value of velocity components decreases whereas the pressure value increases.
