In mutation breeding of chrysanthemum the regeneration in vitro of adventitious shoots from ligulate florets can lead to the separation of chimera components and, as a result, to producing a new original cultivar. The success of that method considerably depends on the result being the number of the shoots formed. The more is produced, the greater the chances for an effective separation of chimera components and creating a new stable cultivar. The present research defines the effect of such factors as the inflorescence development stage, the type of the explant as well as the position of its inoculation on the increase in the efficiency of adventitious shoots regeneration. The ligulate florets of Chrysanthemum × grandiflorum (Ramat.) Kitam. ‘Cool Time’ were inoculated on the Murashige and Skoog [1962] medium supplemented with 2 mg·dm-3 BAP and 0.5 mg·dm-3 NAA. There was shown no significant effect of the inflorescence development stage (incompletely open with a partially visible disk or with the entire visible disk in which tubular florets do not produce pollen or completely open in which two or half of the whorls of tubular florets produce pollen) on the shoot regeneration efficiency. Most shoots regenerate on transversely- or lengthwise-cut into half or on the entire pierced ligulate florets – horizontally inoculated, with the abaxial side on the medium.
W hodowli mutacyjnej chryzantem regeneracja in vitro pędów przybyszowych z kwiatów języczkowatych doprowadzić może do rozdzielenia komponentów chimer i w rezultacie do uzyskania nowych, oryginalnych odmian. W tej metodzie o sukcesie w dużej mierze decyduje wynik w postaci liczby uzyskanych pędów. Im większa jest ich liczba, tym większe są szanse na skuteczną separację komponentów chimer i uzyskanie nowej stabilnej odmiany. W badaniach określono wpływ takich czynników jak stadium rozwoju kwiatostanu, rodzaj eksplantatu oraz pozycja jego inokulacji na zwiększenie wydajności kaulogenezy przybyszowej. Kwiaty języczkowate Chrysanthemum × grandiflorum (Ramat.) Kitam. ‘Cool Time’ wykładano na pożywka Murashige i Skooga [1962] uzupełnioną 2 mg·dm-3 BAP i 0.5 mg·dm-3 NAA. Nie udowodniono wpływu stadium rozwoju kwiatostanu (niecałkowicie otwarty z widocznym częściowo oczkiem lub z widocznym całym oczkiem, w którym nie pylą kwiaty rurkowate albo całkowicie otwarty, w którym dwa lub połowa okółków kwiatów rurkowatych pyli) na wydajność regeneracji pędów. Najwięcej pędów powstaje na przeciętych poprzecznie lub podłużnie na pół albo na całych nakłuwanych kwiatach języczkowatych inokulowanych horyzontalnie stroną zewnętrzną.

Opis:
The effects of TDZ and paclobutrazol on the primary regeneration on tulip flower stalk explants of six cultivars and subsequent shoot multiplication were examined. Explants, flower stalk slices, were excised from cooled and subsequently forced bulbs. The explants were incubated for two months in darkness on medium containing NAA and cytokinins, 2iP and BAP, as control, or TDZ (0.5-4 mg l-1) and paclobutrazol (0.05-0.4 mg l-1). Then, the regenerating explants were subcultured on medium with TDZ and NAA applied at low concentrations. Different regeneration capabilities were found depending on cultivar and growth regulators. The percentage of explants forming leaf-like structures ranged, on the control medium, from 80% in 'Blue Parrot' and 'Prominence' to below 30% in 'Apeldoorn' and 'Mirjoran'. TDZ, applied at optimum for each cultivar concentration, greatly increased the regeneration potential up to 70-100%. Paclobutrazol, added to the TDZ-containing medium, significantly enhanced the response of explants, resulting in high numbers of leaf-like structures formed per explant (13.7-22.8). The structures developed gradually into characteristic forms: the growing up cotyledonary leaf, the probable root primordium formed at its base, the growing downwards stolon and the shoot meristem developed finely on its tip. It is suggested that such primary regeneration may have a nature of somatic embryogenesis. Then, the adventitious shoots developed and formed clusters, which were divided into 2-3 smaller ones every two months. The growth regulators, used at initial stage, markedly influenced subsequent shoot multiplication. Thus, the most intensive shoot formation was noted with TDZ at concentrations of 0.5-2 mg l-1 and paclobutrazol of 0.05-0.1 mg l-1.

Opis:
Isatis concstricta Davis (an endemic plant of Turkey) suffers from low propagation rates under natural conditions and is threatened due to fast unplanned urbanisation. The study compared the effects of variants of BA + NAA on shoot regeneration on leaf and petiole explants excised from one week old in vitro regenerated seedlings. MS medium containing 1 mg L–1 BA + 1 mg L–1 NAA induced maximum proliferation on petiole and leaf explants with 13.33 and 12.75 shoots per explant repectively. However, leaf explant induced shoots were sturdy and healthier compared to petiole explant induced shoots. These shoots were rooted on MS medium containing 0.5 mg L–1 IBA and the plants were acclimatized in peat moss and sand (v/v). They grew to flowering under ex vitro conditions. This system of regeneration is advantageous for conventional propagation and the results will help in establishment of a powerful and meaningful micropropagation system for I. constricta.

The experiments were carried out on Lotus corniculatus (L.) seedling root explants of the cultivar varieties Skrzeszowicka, Caroll A10 and strain 175. Callus formation and shoot regeneration were the major explant response depended mainly on of the studied genotype and used plant growth regulators (PGRs). Primary cortex of proximal and distal end of explant was the most active tissue for callus proliferation. For shoot primordia differentiation deeper zones of cortex took a part. The process of meristematic centre initiation was not uniform and various level of shoot differentiation events were observed not earlier than 3 weeks of culture. Usually, the shoot primordia regeneration began on proximal rather than distal end of the explant. BAP rather than urea derivatives stimulated shoot proliferation in extended cultures. Increasing of BAP and TDZ concentrations brought about the explant polarity and expansion of the meristematic zones. The explant position in root did not have significant influence on the number of regenerated shoots. The cultures only had better bud formation by TDZ when compared to BAP. BAP stimulated bud formation and development of the shoots from them. Short term of TDZ treatment of explants stimulated meristem formation which developed into buds and shoots. CPPU stimulated callus proliferation and bud formation when explants pretreatment was prolonged from 12 to 36 hrs.

Various explants from 30-day-old seedlings of Centaurium erythraea Rafn were evaluated for their morphogenetic capacity under in vitro culture conditions. Shoot formation from shoot tip explants was achieved mainly through adventitious bud differentiation. The highest number of shoots (up to 43.3 ± 2.2 from a single shoot tip) was obtained on Murashige and Skoog medium (MS) supplemented with indole-3-acetic acid (IAA) (0.57 μM) and 6-benzylaminopurine (BAP) (4.4 μM). Adventitious shoot regeneration was also achieved through organogenesis from calluses obtained from hypocotyls, cotyledons, roots and leaves on MS medium containing IAA (2.85 μM) and BAP (0.88 μM). Significant differences were noted between explant types in their effects on shoot regeneration. In the primary culture, the best response was obtained either from calluses derived from roots or leaves (44.4 ± 4.5 and 40.2 ± 6.0 shoots per callus, respectively). The number of subcultures of inoculated calluses affected both the multiplication rate (the number of shoots/explant) and shoot morphology (the frequency of shoot hyperhydricity). Shoots rooted with the frequency of 94-100% after culture on MS medium without growth regulators. Plantlets were successfully acclimatized (97%) under high relative humidity and then moved to the greenhouse.

Opis:
A vanishing species in Poland - Plantago maritima L. was regenerated in vitro from tips of shoots (obtained in vitro) and from different explants of 4-week-old seedlings: seedling tips, hypocotyls, cotyledons, roots. Murashige and Skoog basal medium, supplemented with 0.6 pM indole-3-acetic acid in combination with cytokinins 6-benzyladenine, zeatin or kinetin, was used. The plants obtained in the result of micropropagation were normal in appearence. It was proved that Plantago maritima species was amenable to propagation from different kinds of explants. The method may be of significance for protection of sea plantain.

Opis:
Observations of Japanese pagoda trees indicate that they undergo a full cycle of vegetative and generative development without self-renovation. The aim of this study was to obtain a successive media protocol propagation of Japanese pagoda tree by in vitro cultures. The effects of growth regulators were studied with reference to primary regeneration, shoot multiplication and rooting. As explants source were used the part of shoot of 90-year-oldtree. Explants were placed on MS (Murashige and Skoog 1962) basal medium with the addition of 6-benzylaminopurine – BAP (1.0–2.0 mg dm-3), thidiazuron – TDZ (0.1–0.3 mg dm-3) and indole-3-acetic acid – IAA (0.5–1.0 mg dm-3). BAP was the growth regulator which significantly increased shoot regeneration on initial explants. TDZ in turn, inhibited the formation of adventitious shoots and caused the explants which had been placed on the medium to die. The multiplication of the Japanese pagoda tree by in vitro cultures should be conducted on MS media with the addition of 0.5 mg dm-3 BAP, and they should be rooted on media with the addition of 0.3 mg dm-3 indole-3-butyric acid – IBA. It seems, that devising an efficient method of Japanese pagoda tree micropropagation is realistic.
