Opis:
Grapevine seedlings Vitis vinifera were grown in a greenhouse under optimum conditions (soil moisture ca 70%) and under drought stress (soil moisture ca 35%). In addition, some of the plants subjected to drought underwent subsequent regeneration under optimum conditions. Drought stress caused accumulation of total phenolic compounds in grapevine roots, which may indicate that these compounds play an important role in the adaptation of roots to growth under stress conditions. Phenolic acids found in the roots occurred in the ester-bound form only. p-coumaric acid was present in the highest concentrations (6.2 to 10.5 µg/g fresh matter). The content of ferulic acid was lower, ranging from 2.4 to 4.6 µg/g fresh matter. The lowest concentration in grapevine roots was achieved by caffeic acid (2.4 to 2.9 µg/g fresh matter). The levels of p-coumaric and ferulic acids in roots rose significantly under the drought stress, while the concentration of caffeic acid increased during the post-drought recovery period. This may suggest that some of the phenolic acids protect plants under stress conditions. All the extracts from grapevine roots had antioxidative properties, but the antiradical activity of the extracts obtained from roots subjected to drought stress was inferior to the control. The same extracts were also characterised by depressed reducing power. The results imply that tolerance of grapevine to soil drought may be associated with the value of antioxidative potential in root tissues of these plants.

Opis:
Roots of radish (Raphanus sativus L. var. radicula Pers.) of Rowa cv. seedlings grown in water cultures (pH 4.3) have been tested. Aluminum as AlCl₃·6H₂O has been applied at 0, 10, 20 and 40 mg·dm⁻³ concentrations. Inhibition of the growth of root elongation and the following changes of root morphology have been observed after aluminum treatment: browning and thickening, root cap elongation or falling off, bending of the apex root and cracks on its surface. Roots have been characterized with the change of hair length and lateral zones. First lateral roots occurred closer to the basal root apex than first hair. Reduction of hair length was noticed too. Atrophy of outer root tissues has been accompanied by great enlargement of cortex cells size as a result of their hypertrophy.
Badano korzenie siewek rzodkiewki ‘Rowa’ (Raphanus sativus L. Var. radicula Pers.) hodowanych w kulturach wodnych o pH 4,3. Glin zastosowano w stężeniach: 0, 10, 20 i 40 mg·dm⁻³ w formie AlCl₃·6H₂O. W obecności glinu obserwowano inhibicję wzrostu elongacyjnego korzeni oraz zmiany w ich morfologii: brązowienie, wydłużanie się lub odpadanie czapeczki, zaginanie się szczytowego odcinka korzenia oraz spękania na jego powierzchni. Korzenie o znacznie powiększonej średnicy odznaczały się zmianą długości i układu stref korzeniowych: włośnikowej i korzeni bocznych oraz redukcją długości włośników. Obumieraniu tkanek obwodowej strefy korzenia towarzyszyło znaczne powiększenie rozmiarów komórek kory w wyniku ich hipertrofii.

Background: Breaking an endodontic instrument is a complication that really hinders or even makes impossible complete disinfection and tight obturation of the root canal system. Case presentation: A 74-year-old patient suffered a fracture of the clinical crown of the second incisor in the maxilla. The examination revealed necrotic pulp and the presence of obliteration significantly limiting the lumen of the tooth canal. It was decided to perform root canal treatment and then to do a reconstruction reinforced by crown-root inlay. An endodontic instrument was broken in the lumen of the root canal at the beginning of the endodontic treatment. A broken piece tightly filled nearly 80% of the total length of the canal. It was decided to use endodontic forceps to remove the fragment.Conclusion: Choosing the right method is a key step in the complication described above. The management technique should entail the least risk of iatrogenic complications because they can greatly affect the long-term maintenance of the tooth. The article discuss on the basis of current literature standards of conduction in the case of tool breakage in the root canal system and presents the problem on the basis of own experience.

Rye plants were grown in a nutrient solution prepa- red according to Hoagland for 2 weeks at pH 7, next for 14 days at pH 4.5 (without Cu+2) and in the presence of 20, 50, or 100 mg dm-3 copper ions. The control plants were grown continuously at pH 7. The physicochemical surface properties of the roots were exa- mined using two adsorbates – polar (water vapour) and non-polar (nitrogen). The surface properties of the roots grown at pH 4.5 without Cu+2 were apparently the same as those of controls. The roots of rye which grew in the presence of Cu+2 were characte- rized by lower (relative to controls) specific surface area values. Statistically significant differences in the size of the apparent surface area (determined by water vapour) were reported for roots in- cubated with copper ions at a concentration of 20 and 50 mg dm-3. The average water vapour adsorption energy of the root surface decreased under the stress conditions. There were no statistically significant differences for the free surface area and characteristic energy of nitrogen adsorption.

At the plants’ exposal to abiotic stress, organic acids, including citric acid, are exuded through their roots. Previous studies have suggested that the exogenous application of citric acid increases antioxidant activity within the plant. Thus, we postulated that organic acids released into the surroundings during times of environmental stress may function as signaling molecules to increase antioxidant enzyme activity. To gain further insight into this phenomenon, we identified individual organic acids exuded from the roots of leafy vegetables under drought stress. We then analyzed enzyme activity and the root/shoot lengths of seedlings after treatment with the types of organic acids found to be exuded from the studied leafy vegetables, including acetic, citric, lactic, and tartaric acids. There was a significant increase in catalase and ascorbate peroxidase enzyme activity in Napa cabbage (Brassica rapa var. pekinensis) after exogenous citric acid application. Root lengths of cabbage (Brassica oleracea var. capitata) and Napa cabbage seedlings were significantly longer in citric and lactic acids pretreated seedlings compared to those of the control. The above results support the conclusion that exogenous application of citric acid alleviates drought stress. However, there is insufficient evidence to prove that organic acids act as signaling molecules to prime neighboring plants for upcoming stress.

The study was performed to evaluate range of the genotypie variation in drought susceptibility at juvenile growth stages among 23 old and modern winter cultivars of Triticum aestivum and one old cv. of T. spelta. Response of germinated seeds to mannitol-simulated drought stress as well as that in seedling shoot and root growth, water use and leaf transpiration under varied soil moisture were measured. Considerable genotypie variation for all morpho-physiological traits was found between wheat cultivars. There were significant differences between the older and modern cultivars. The former showed usually an enhanced rooting ability and higher water requirements, while the newer indicated more efficient water use. The older cultivars were more resistant to both iso-osmotic stress and low soil moisture at the juvenile growth stage. However, no clear differences were found between the two groups of cultivars in the variation range of the drought susceptibility indices. Complex patterns of the response to drought were observed. Cultivars, which distinguished by both a lower shoot : root ratio and a stronger drought-induced stimulative root growth, tended to be more resistant. Although only moderate the relationships noticed, a consideration of the root plasticity screening in local wheat breeding appears to be justified.

The effect of aluminum was investigated on the cell ultrastructure of roots of fourteen-day-old `Trapez` red pepper seedlings grown in water culture. Disorders in the cell structure were observed using transmission electron microscopy (TEM). An analysis of longitudinal sections of the apical region of the control plants’ roots showed that cells of the cap and cells of the meristematic region had an arrangement and shape typical for these root regions, and cell organelles were properly developed. Changes in the cell structure under the infl uence of aluminum involved a reduction in the number of starch grains in the leucoplasts of the cap, the formation of lobate nuclei and a reduction in the number of cisternae in the dictyosomes as well as the damage of the cell membranes. Moreover, the swelling of mitochondria was observed with a simultaneous reduction in the number of mitochondrial cristae or the bursting of the membrane of these structures. In the vacuoles of the investigated root section, the presence of numerous electron-opaque large-sized formations was found, being most probably aluminum deposits. The cell wall, often thickened, was wavy or the formation of two walls in close distance was observed. The obtained results prove the high sensitivity of the studied red pepper cultivar to aluminum stress.
Badano wpływ glinu na ultrastrukturę komórek korzenia czternastodniowych siewek papryki rocznej `Trapez` uprawianych w kulturach wodnych. Zaburzenia struktury komórek obserwowano w transmisyjnym mikroskopie elektronowym (TEM). Analiza podłużnych przekrojów wierzchołkowej strefy korzeni roślin kontrolnych wykazała, że komórki czapeczki i komórki strefy merystematycznej miały układ i kształt typowy dla tych stref korzenia, a organella komórkowe były prawidłowo wykształcone. Zmiany w strukturze komórek pod wpływem glinu polegały na redukcji liczby ziaren skrobi w leukoplastach czapeczki, tworzeniu płatowatych jąder oraz redukcji liczby cystern w diktiosomach, a także uszkodzeniu membran komórkowych. Zaobserwowano ponadto pęcznienie mitochondriów przy jednoczesnym zmniejszaniu się liczby grzebieni mitochondrialnych lub pękaniu otoczki tych struktur. W wakuolach badanego odcinka korzenia stwierdzono obecność licznych nieprzejrzystych elektronowo tworów o pokaźnych rozmiarach, będących najprawdopodobniej depozytami glinu. Ściana komórkowa była często zgrubiała, pofałdowana lub obserwowano tworzenie się dwu ścian w bliskim sąsiedztwie. Uzyskane wyniki świadczą o dużej wrażliwości badanej odmiany papryki na stres glinowy.

The research has been performed on roots of Vitis vinifera, cv. Himrod, obtained from seedlings grown under chill stress conditions (+10oC in the day and +7oC at night), under optimum conditions (+25oC in the day and +18oC at night) and from seedling which underwent a recover period after the chill stress treatment. The purpose of the study has been to determine quantitative and qualitative changes in phenolic compounds as well as to demonstrate changes in antiradical properties of extracts from grapevine roots, which appeared as a result of chill stress and during recovery under the optimum conditions following the stress. Phenolic compounds from grapevine roots were extracted using 80% acetone. The total content of phenolics was determined by colorimetry. The content of tannins was tested by precipitation with bovine serum albumin. The reducing power as well as DPPH• free radical and ABTS+• cation radical scavenging activity of the extracts were also tested. In order to identify phenolic compounds present in the extracts the RP-HPLC technique was employed. The tested material was found to contain tannins and three identified phenolic acids: ferulic, caffeic and p-coumaric ones. The latter occurred in the highest concentrations (from 4.46 to 6.28 µg/g fresh matter). Ferulic acid appeared in smaller amounts (from 1.68 to 2.65 µg/g fresh matter), followed by caffeic acid (from 0.87 to 1.55 µg/g fresh matter). Significantly less total phenolic compounds occurred in roots of seedlings subjected to chill stress. However, the total content of these compounds increased significantly in roots of plants which underwent recovery after chill stress. Concentration of tannins was determined by two methods. The content of condensed tannins was depressed in roots as a result of low temperature stress, whereas the content of condensed and hydrolysing tannins (determined via the BSA method) rose under chill stress conditions. A significant increase in tannins in root extracts (determined with both methods) was found during the recovery process after the stress. The three identified phenolic acids appeared in grapevine roots as ester-bound compounds. It has been demonstrated that the content of phenolic acids significantly fell as a result of low temperatures, but increased during recovery after chill stress. The weakest ability to scavenge DPPH• and ABTS+• free radicals as well as the reducing power were shown by the extract obtained from grapevine roots from the seedlings subjected to chill stress. Both free radical scavenging activity and reducing power were observed to increase considerably during recovery after stress. This seems to prove that during the recovery process following chill stress the synthesis of antioxidative compounds in grapevine roots is much more intensive.
