The mechanical link coupling cytoskeletal and contractile proteins to the sarcolemma of smooth muscle cells is essential for transmitting tension from the cell's interior to exterior. In addition to the well-characterized actin-integrin associations present in adhaerens junctions, our recent work has postulated the existence of a reversible annexin-dependent membrane-cytoskeleton complex, forged in response to a rise in intracellular Ca2+ concentration following smooth muscle cell stimulation (Babiychuk et al., J. Biol Chem. 1999, 274, 35191-35195). Detailed biochemical characterization of the interactions responsible for the formation of this complex revealed that annexins II and VI interact with actomyosin, or detergent-insoluble glycosphingolipid-enriched membrane domains (rafts) purified from smooth muscle, in a concentration- and Ca2+-dependent manner. Annexin II interacted with lipid rafts with high Ca2+-sensitivity, while for annexin VI this interaction required non-physiologically high concentrations of free Ca2+. However, the Ca2+-sensitivity of the latter interaction strongly increased in the presence of purified smooth muscle actomyosin. The detailed biochemical analysis of the interactions occurring between annexin II, annexin VI, actomyosin and rafts suggests that annexins regulate sarcolemmal organization during smooth muscle cell contraction.

We and others have recently obtained data suggesting that cytokine-STAT signaling in many different cell-types is a chaperoned pathway initiated at the level of specialized plasma membrane microdomains called "rafts" (the "raft-STAT signaling hypothesis"). These findings are of broad significance in that all cytokines and growth factors initiate signaling in target cells by interacting with respective cell-surface receptors. The new data suggest that raft microdomains represent the units of function at the cell-surface through which ligand-stimulated STAT signaling is initiated. Moreover, recent evidence shows the involvement of chaperone proteins in regulating the STAT signaling pathway. These chaperones include the human homolog of the tumorous imaginal disc 1 protein (hTid1) which associates with Janus kinase 2 (JAK2) at the level of the plasma membrane, heat shock protein 90 (HSP90) which associates with STAT3 and STAT1 proteins in caveolin-1-containing raft and cytoplasmic complexes, and glucose regulated protein 58 (GRP58/ER-60/ERp57), a thiol dependent protein-disulfide isomerase, found in association with STAT3 "statosome" complexes in the cytosol and in the raft fraction. We suggest a function of the HSP90 chaperone system in preserving IL-6/STAT3 signaling in liver cells in the context of fever. The identification and function of protein partners associated with specific STAT species in rafts and in cytosolic complexes, and in the efficient departure of cytokine-activated STATs from the cytosolic face of rafts towards the cell nucleus are now areas of active investigation.

In the Santonian chalk glacial deposits exposed at Kornica (eastern Poland) an assemblage of echinoderms comprising asteroids, echinoids, ophiuroids, and crinoids (comatulids, roveacrinids, and Bourgueticrinus, Isocrinus, Isselicrinus, Marsupites) is documented. The Santonian age of sediments at Kornica has been determined with foraminifers. Numerous occurrences of the crinoid Marsupites testudinarius, indicate their late Santonian age.

Opis:
Model systems such as black lipid membranes or conventional uni- or multilamellar liposomes are commonly used to study membrane properties and structure. However, the construction and dimensions of these models excluded their direct optical microscopic observation. Since the introduction of the simple method of liposome electroformation in alternating electric field giant unilamellar vesicles (GUVs) have become an important model imitating biological membranes. Due to the average diameter of GUVs reaching up to 100 µm, they can be easily observed under a fluorescent or confocal microscope provided that the appropriate fluorescent probe was incorporated into the lipid phase during vesicle formation. GUVs can be formed from different lipid mixtures and they are stable in a wide range of physical conditions such as pH, pressure or temperature. This mini-review presents information about the methods of GUV production and their usage. Particularly, the use of GUVs in studying lipid phase separation and the appearance and behavior of lipid domains (rafts) in membranes is discussed but also other examples of GUVs use in membrane research are given. The experience of the authors in setting up the GUV-forming equipment and production of GUVs is also presented.

The article That’s How I’ve Remembered Mietek aims to throw light on the origins and the final artistic form of Władysław Ślesickis documentary The Rafts Sail (1962). The movie depicts the story of a young Mazurian boy named Mietek, who crosses the line between childhood and adulthood (when, as a raftsman, he floats timber down the river). This film was probably the first time when the director’s poetic attitude towards filmmaking resulted in such a sublime documentary portrait, devoid of any words. Looking back, The Rafts Sail (awarded the Lion of San Marco at the Venice Film Festival in 1962) can be perceived as a clear symptom of Ślesickis masterpieces, such as Before Leaves Fall [1964] or The Family of Man [1966].

Błona komórkowa komórek eukariotycznych jest niejednorodna i charakteryzuje się obecnością domen nazywanych tratwami błonowymi. Są to liczące kilka nanometrów dynamiczne skupiska sfingolipidów, cholesterolu i wybranych białek, zwłaszcza palmitoilowanych, które wyodrębniają się z otaczającego je środowiska glicerofosfolipidów. Labilne nanodomeny zlewają się w większe struktury, platformy sygnałowe, stabilizowane przez interakcję z cytoszkieletem podbłonowym w czasie aktywacji szeregu receptorów zaangażowanych w reakcje wrodzonej i nabytej odporności. Paradoksalnie, niektóre wirusy i bakterie wykorzystują tratwy błonowe jako miejsca inwazji/opuszczania komórek gospodarza. Z drugiej strony, w błonie bakterii wykryto rejony, w których także skupiają się wybrane białka sensorowe i enzymy, co sugeruje udział domen błony bakteryjnej w procesach przekazywania sygnału. Skład lipidowy bakteryjnych domen błonowych jest słabo poznany, a rolę w ich formowaniu przypisuje się białku flotylinie. Heterogenna organizacja błony komórkowej jest zatem zachowanym ewolucyjnie sposobem zapewnienia właściwej przestrzennej organizacji receptorów oraz białek i lipidów biorących udział w ich kaskadach sygnałowych, która stała się szczególnie istotna dla funkcjonowania komórek układu odpornościowego człowieka i zwierząt.
The plasma membrane of eukaryotic cells contains domains named rafts which are nonscale dynamic assemblies of sphingolipids, cholesterol and selected proteins, mainly palmitoylated ones. During stimulation of distinct immune receptors labile rafts merge into larger structures which are stabilized by submembraneous cytoskeleton and serve as signaling platforms of those receptors. Paradoxically, rafts are also utilized by some viruses and bacteria to invade/escape host cells. On the other hand, bacterial plasma membrane contains domains accommodating sensory proteins and several other enzymes which suggests that those domains are sites of signal transduction. Lipid composition of bacterial membrane domains is poorly characterized and a role in their formation is ascribed to proteins named flotillins. Thus, domain organization of the plasma membrane seems to be common to eukaryotic and prokaryotic cells. It facilitates spatial organization of plasma membrane receptors as well as lipids and proteins involved in their signaling pathways. During evolution rafts of the plasma membrane have become important especially for functioning of human and animal immune cells.

Successful rescue action at sea is based on a. o. a correct choice of rescue means and their reliability. Operational characteristics of life-saving appliances determine their performance in a given water area. Therefore they affect duration time of rescue action and decide this way on survival time of shipwrecked persons. This paper presents impact of characteristics of circular inflatable life rafts on their dynamics in a given sea environment. Particular issues dealing with raft motion were analyzed and their solutions were then presented in the form of computer programs built into MATLAB environment. The programs operate on the open choice basis– the user is requested to introduce data of a raft to be investigated. The program clearly informs which units should be used for the data. The obtained results allow to limit survivours searching area and simultaneously form the basis for working out a model of searched object drift motion. In this work the use was made of the results obtained from the KBN research project No. 4T12C03827 in which an attempt was made to analyzing capsize probability of a raft at given sea states. The raft investigated in the project is also used as a reference raft in the computer programs developed in this work.

Opis:
Władysław Jagiełło and Witold Kiejstutowicz were preparing a war with the Teutonic Order. It was decided to build a bridge across the Vistula near Czerwińsk. The wood for building about 50 riverboats (punts) and spans had to dry for several months. The works which started in February 1409 in the Kozienice Forest were coordinated by Master Jarosław and Staroste Dobrogost the Black. Teams of boatbuilders, carpenters, blacksmiths and raftsmen made the elements of the bridge (boats, spans, anchors, iron buckles). The bridge was erected between Śladow and Czerwińsk on June 28-30, 1410. The barriers on the banks and the military forces ensured an uninterrupted march of the Polish army on June 30-July 3, comprising approx. 15,000 mounted knights, several hundred heavy tabor wagons, and several dozen cannons. The combined Polish and Lithuanian-Ruthenian armies departed from Czerwińsk for the war with the Teutonic Order. The bridge was dismantled, floated to Płock and rebuilt on the Vistula near Przypust on September 24-27, 1410 for the returning Polish army.

ObjectivesThe role of the cardiovascular system in the development of seasickness remains uncertain.Material and MethodsOverall, 18 healthy students (10 males and 8 females) aged 18–24 years volunteered in the project, spending 2–7 h on life rafts. The cardiovascular system was examined with impedance cardiography. Susceptibility and symptoms of seasickness were evaluated by the Motion Sickness Susceptibility Questionnaire Shortform (MSSQ-Short) and the Motion Sickness Assessment Questionnaire (MSAQ). The Visual Analogue Scale (VAS), ranging 0–10, was used to assess nausea, dizziness and mood. The parameters were assessed at 2 time points.ResultsDifferences in the heart rate (HR), the thoracic fluid content index (TFCI), the stroke index (SI) and the Heather index (HI) before launching the life rafts and after leaving them were observed (78.6, 20.8, 55.6 and 15.9 vs. 70.1, 19.7, 60.5 and 17.9, with p-values of 0.002, <0.001, 0.003 and 0.004, respectively). Females reacted with changes in SI and HR more vividly, whereas males regulated more HI and TFCI. In addition, HR correlated significantly with the central and peripheral symptoms in MSAQ, stroke volume (SV) with peripheral and sopite-related ones, SI with overall ones, and pulse pressure with overall, gastrointestinal and central ones (Spearman’s rank correlation coefficient [ρ] was –0.478, –0.711, 0.476, 0.472, 0.525, –0.476, –0.579 and –0.584, respectively). As regards MSSQ-Short, it correlated negatively with sopite-related symptoms in MSAQ (ρ= –0.486). Mood in VAS correlated significantly with gastrointestinal symptoms, SI and the cardiac index (CI) (ρ = –0.752, –0.492 and –0.489, respectively).ConclusionsIt was found that HR correlated negatively, and SV/SI correlated positively, with the severity of seasickness symptoms measured with MSAQ. Gender is probably an independent factor influencing reactions to motion. Women react with SI increase whereas men react with increased heart contractility (HI rise). Negative mood in seasickness evaluated with VAS seems to be mostly determined by gastrointestinal symptoms assessed with MSAQ and diminished cardiovascular indices (both CI and SI).
