Abstract A novel floating controlled release drug delivery system of Sitagliptin phosphate was formulated in an effort to increase the gastric retention time of the dosage form and thereby increased drug bioavailability. The tablets were prepared by wet granulation method using Psyllium Husk and tragacanth gum as a release retarding polymers and sodium bicarbonate as a gas generating agent. All the designed nine batches of floating tablets were evaluated for physical characteristics viz. weight variation, thickness, content uniformity, hardness, floating capacity, swelling studies. All formulations had floating lag time of less than 1 min and constantly floated for 12 h except F1, F4 and F7 (that dissolved completely in 8 h or less than 8 h). In vitro drug release studies were carried out for 8 h and release mechanism was further evaluated by linear regression analysis, F9 composed of 30% Psyllium Husk, 10% tragacanth gum and 18% sodium bicarbonate sustained the drug release for longer period. The formulations followed first order kinetics, Higuchi drug release kinetics with diffusion as the dominant mechanism of drug release and the release exponent ranged (0.452-0.635) indicating that the drug release from all formulations was by non-Fickian diffusion mechanism. The prepared floating tablets of STP (F9) might be a promising drug delivery system with sustained release action and improved bioavailability.

Opis:
Six warm-blooded horses in the age of 6 till 16, were researched. The horses were divided into two groups: experimental and testing (each per 3 horses). In the first stage the trial was taken up to estimate the grade (rank) of sand accumulation in the alimentary canal of all horses with using the floatation test. In the second stage the experimental group with the given foragegot the supplement of Plantago Psyllium seeds in the amount of 210 g per horse per day during 7 days. The researches revealed that preventive supplement of Plantago Psyllium seeds three times increases the amount of sand excreted from the alimentary canal. So using the supplement of Plantago Psyllium seeds can prevent the excessive of sand concentration in horses alimentary canal and this way decreases the risk of occurring the intestinal colic diseases – in the matter.
Badaniami objęto 6 koni gorącokrwistych w wieku od 6 do 16 lat. Konie podzielono na grupę doświadczalną i kontrolną (liczące po trzy konie). Badania przeprowadzono w dwóch etapach. W pierwszym etapie podjęto próbę określenia stopnia zapiaszczenia jelit u wszystkich koni z wykorzystaniem testu flotacji. W drugim etapie grupa doświadczalna wraz z zadawaną paszą przyjmowała dodatek nasion babki płesznik w ilości 210 g na konia na dzień przez 7 kolejnych dni. Badania wykazały, że profilaktyczne podawanie koniom nasion babki płesznik trzykrotnie zwiększa ilość piasku wydalanego z przewodu pokarmowego. Stosowanie dodatku nasion Plantago psyllium może więc zapobiegać nadmiernemu gromadzeniu się piasku w przewodzie pokarmowym koni, a tym samym zmniejsza ryzyko wystąpienia schorzeń kolkowych na tym tle.

This study demonstrates the comparison in the method of fabrication and thus evaluates the potential of psyllium husk powder and gelatin-based composite microporous scaffolds for tissue engineering applications. The scaffold is being prepared in three different ratios of 50:50, 75:25 and 100 (w/w of psyllium husk powder and gelatin, respectively) by employing a suitable cross-linking agent, EDC-NHS, followed drying. We have demonstrated the use and outcomes of two different methods of scaffold drying, i.e., vacuum desiccation along with liquid nitrogen dip and lyophilization. It was concluded from the SEM micrographs that the scaffolds dried under vacuum accompanied with liquid nitrogen exposure exhibited less porous architecture when compared to those prepared using a lyophilizer, that resulted in pores in the range of 60-110 μm. Scaffolds fabricated using the former technique lost porosity and sponge-like characteristics of a scaffold. In spite of the above fact, water retaining capacity and stability in the cell culture of such scaffolds is significant, nearly 40-50% of its initial dry weight. Cell culture experiments support the potential of the scaffolds prepared from different methods of fabrication for its cytocompatibility and suitability for cell growth and proliferation for a substantial duration. Erosion in the porous design of the scaffolds was observed after 14 days via SEM micrographs. It was inferred that freeze-drying is a better technique than vacuum desiccation for scaffold preparation. The present investigation has been conducted keeping in mind the importance of drying a scaffold. Scaffold drying is a necessary step to increase its shelf-life, makes it easy to transport and much importantly, controlling the pore size of the scaffold.
