Zinc is a vital micronutrient for all life forms, and Zn-solubilizing bacteria (ZSB) present in the soil convert inorganic zinc into forms available for plants. This study assessed ZSB isolated from cow dung for their plant growth-promoting (PGP) characteristics and potential to enhance tomato plant growth. The experiment assayed a total of 30 bacteria from cow dung for Zn-solubilization using insoluble ZnO and ZnCO3. Atomic absorption spectroscopy quantitatively evaluated Zn-solubilization, and the isolates were further studied for Zn-solubilization and plant growth in Solanum lycopersicum. The CDS7 and CDS27 isolates were the most significant Zn solubilizing strains. CDS7 exhibited increased ZnO solubility (32.1 mg/l) compared to CDS21 (23.7 mg/l). PGP trait quantitative results revealed that the CDS7 and CDS21 bacterial strains solubilized insoluble phosphate (287.2 and 217.7 μg/ml, respectively) and produced indole acetic acid (22.1 and 14.8 μg/ml, respectively). Based on 16S rRNA gene sequencing, CDS7 and CDS21 were identified as Pseudomonas kilonensis and Pseudomonas chlororaphis, and 16S rDNA sequences were submitted to the GenBank database. Furthermore, ZSB strains were administered to tomato seeds under a pot study. The treatments with CDS7 inoculant and a consortium of both isolates were reported with maximum plant development (stem length 63.16 and 59.89 cm, respectively) and zinc content (3.13 and 2.36 mg/100 g, respectively) in tomato fruit compared to the control. In conclusion, microorganisms isolated from cow dung with PGP activity can improve Zn bioavailability and plant growth sustainably. They can be used as biofertilizers in agricultural fields to improve plant growth and production.

Background: We present a case of combined Pseudomonas aeruginosa, Acanthamoeba and Candida keratitis and clinical course of treatment in two adolescents girls who did not follow the rules of proper use of soft contact lenses. Case reports: Two teenage contact lens users sought medical attention, reporting visual impairment, redness and pain of the eye. Slit lamp examination showed unilateral corneal ulcer. Confocal microscopy revealed images compatible with amoebic cysts and yeasts cells, whereas the results of conjunctival swabs confirmed the presence of Pseudomonas aeruginosa. After topical and general treatment in the first patient, the corneal defect was healed, and visual acuity improved. The girl remains under the constant supervision of an ophthalmological clinic. The medications were gradually reduced, but with time she developed crystalline keratopathy. Regrettably, the second patient, despite the applied treatment had recurrent inflammation and progressive corneal thinning, which required amniotic membrane transplantation to prevent perforation. Conclusion: The early identification of the pathogen as well as adequate treatment is crucial, as contact lens-associated keratitis are potentially sight threatening. Patient and caregiver education, adequate contact lens hygiene and consistent follow-up are important to prevent serious complications.

Urinary tract infections (UTI’s) are a bacterial infection that affects any part of the urinary tract. Urinary tract infection is one of the major diseases that affect people of all age groups and sexes and can be separated into asymptomatic and symptomatic. The normal urinary tract is sterile but gets infected with the normal flora and opportunistic pathogens. Urinary tract infections (UTIs) are a serious health problem affecting millions of people each year. This is the second most common type of infection in the body. In the present study 20 morphologically different bacterial isolates were recovered from 50 different urine samples from UTI’s patient. The prevalence of the isolates was varied with E. coli (40%), followed by Staphylococcus spp., Pseudomonas spp. and Klebsiella spp. each of (20%) respectively. All the isolates of different bacteria were tested for antibiotic sensitivity patterns against common antibiotics viz. Ampicillin, Tetracycline, Chloramphenicol, Vancomycin, Nitrofurontione and Ciprofloxacin with potencies of 30 μg each. The sensitive pattern of the isolates towards antibiotic Nitrofurontoine was (75%) followed by Tetracycline (55%) and Ciprofloxacin (45%). Intermediate activity was reported against Ampicillin (45%), Chloramphenicol and vancomycin (40%). Resistant pattern shown against vancomycin (60%), Chloramphenicol (45%), while both tetracycline and Ciprofloxacin shown (30%) against the UTI’s isolates. Therefore, drugs like Nitrofurontoine, Tetracycline and Ciprofloxacin are the most effective choices against the common UTI’s isolates.

Endophytes are an important constituent of sustainable agriculture because of their ability to produce a large number of agriculturally important metabolites. A salt-tolerant fluorescent green pigment-producing endophytic bacterium was isolated on 2.5% NaCl-supplemented nutrient agar from the leaf samples of Zanthoxylum alatum Roxb. The isolate Z1B4 was identified as Pseudomonas fluorescens based on morphological features, fatty acid methyl ester analysis, biochemical tests, and 16S rRNA gene sequencing. P. fluorescens Z1B4 showed positive results for tricalcium phosphate solubilization; 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity; and production of auxins, siderophores, hydrogen cyanide, and ammonia. P. fluorescens Z1B4 also showed strong antagonistic activity against Curvularia lunata (MTCC 283), Fusarium verticillioides (MTCC 3322), and Alternaria alternata (MTCC 1362) and exhibited stress tolerance to a wide range of temperature and pH and concentrations of NaCl and calcium salts. Under natural conditions, following inoculation with the isolate Z1B4, a significant increase in the growth of pea and maize test plants in pots was observed compared to that of uninoculated control plants. The rifampicin-resistant mutant Z1B4Rif was recovered from the roots, shoots, and leaves of the test plants, indicating that the isolated endophytic bacterium can grow well within different plant tissues. The present study indicated that the endophytic bacterium P. fluorescens Z1B4 can be used as a bacterial inoculant in stressed environments for sustainable agriculture.

Improperly treated heavy metal wastewater discharged into water sources could cause a serious issue for the environment. The aim of this study was to bioaccumulate iron (Fe) using native bacteria isolated from the laboratory drainage water containing a high concentration of iron. The experiment was conducted in 250 mL conical flasks containing 150 mL Fe solution in concentrations of 25, 100, and 250 mg/L, respectively. Approximately 10% of bacteria inoculum was cultivated in each Fe concentration for 24 and 48 hours. The results showed that Pseudomonas hibiscicola was identified as an effective iron-accumulating species of bacteria. The species could remove Fe up to 82% (25 mg/L), 77.8% (100 mg/L) and 32% (250 mg/L). This promising result indicates that the native bacteria isolated from the environment pose a great potential for the remediation of wastewater containing iron.

Opis:
Phenol is one of the main pollutants that have a serious impact on the environment and can even be very critical to human health. The biodegradation of phenol can be considered an increasingly important pollution control process. In this study, the degradation of phenol by Pseudomonas fredriksbergsis was investigated for the first time under different growth conditions. Six different initial concentrations of phenol were used as the primary substrate. Culture conditions had an important effect on these cells' ability to biodegrade phenol. The best growth of this organism and its highest biodegradation level of phenol were noticed at pH 7, temperature 28 °C, and periods of 36 and 96 h, respectively. The highest biodegradation rate was perceived at 700 mg/L initial phenol concentration. Approximately 90% of the phenol (700 mg/L) was removed in less than 96 hours of incubation time. It was found that the Haldane model best fitted the relationship between the specific growth rate and the initial phenol concentration, whereas the phenol biodegradation profiles time could be adequately described by the modified Gompertz model. The parameters of the Haldane equation are: $0.062 h^{−1}$, 11 ppm, and 121 ppm for Haldane’s maximum specific growth rate, the half-saturation coefficient, and the Haldane’s growth kinetics inhibition coefficient, respectively. The Haldane equation fitted the experimental data by minimizing the sum of squared error (SSR) to $1.36×10^{-3}$.

Background . Melanin finds enormous applications in different industries for its unique photoprotective and anti-oxidant properties. Due to its emerging demand, scientific researchers are putting efforts to unravel more microorganisms with a potential of producing melanin on large scale. Hence, the present study was aimed at the isolation of extracellular melanin producing microorganisms from lime quarries of Karnataka, India. Besides this, the tyrosinase gene governing melanin synthesis in different organisms were compared in silico to understand its evolutionary aspects. Material and methods . Melanin producing microorganisms were screened on tyrosine gelatin beef extract agar medium. Potential isolate was explored for submerged production of melanin in broth containing L-tyrosine. Melanin was characterized by UV-Vis spectroscopy, thin layer and high performance liquid chromatographic techniques. Antibacterial activity of melanin was performed by agar well assay. Comparative tyrosinase gene sequence analysis was performed by using Geneious 2021.1 trial version software. Results . Pseudomonas otitidis DDB2 was found to be potential for melanin production. No antibacterial activity was exerted by the melanin against tested pathogens. The in silico studies showed that the common central domain of tyrosinase protein sequence of selected Pseudomonas sps. exhibited 100% identity with the common central domain of Homo sapiens tyrosinase (NP_000363.1). Conclusions . Our study shows the production of melanin in good quantities by the isolate Pseudomonas otitidis DDB2 which can be explored for scale-up process. Since the melanin formed is of eumelanin type and the tyrosinase gene sequence of several Pseudomonas sp. showed relatedness to humans, this molecule may be further developed for sunscreen formulations.

Consumption of water contaminated with pathogens and heavy metals is responsible for the demise of millions of people around the globe. To find a low cost, facile and benign route for developing a biosorbent capable of handling the challenges associated with unsafe drinking water, the study report the preparation, activation, adsorption and removal of cadmium and pathogenic organisms from water samples. Modified plantain peel biochar (MPPB) was characterized using Braunauer-Emmett-Teller (BET) surface area and X-Ray diffraction (XRD). Surface area and pore size of the MPPB determined from BET surface area plot are 8.79 cc/g and 16.69 m2/g respectively with XRD crystallite size of 14.56 nm. The maximum adsorption capacity was observed to be 366.300 mmolkg-1. Equilibrium isotherm indicated the coefficient of determination to increase from Jovanovich →Elovic→Temkin→Freundlich→Langmuir model. Surface interaction and mass transfer kinetics shows a mixture of intra-particle diffusion and mass transfer process. The adsorption kinetics and surface interaction models coefficient of determination increased from Bangham pore diffusion →mass transfer model→Liquid film diffusion→pseudo first order→Dunwald-Wagner→Elovic→Pseudo-second order→Weber and Moris model. The adsorption indicated a chemisorption process with rate limitimg step likely to involve electron pair donor-acceptor interactions, hydrogen bonding and dipole dipole interactions. Antimicrobial evaluation of MPPB showed that it is a potential antimicrobial agent against common micro-organisms.

White button mushrooms, Agaricus bisporus, is susceptible to several pathogenic microorganisms, including Pseudomonas tolaasii, which causes brown blotch disease. The disease is devastating and results in severe loss of cultivated mushrooms. For this reason, the study was undertaken of the effect of two biological basic substances (EC 1107/2009) – hydrogen peroxide and wine vinegar, on the reduction of brown bacterial blotch. The preparations were evaluated in laboratory and cultivation conditions. High bactericidal and bacteriostatic activity of the tested basic substances was demonstrated in laboratory conditions. In a crop experiment, the basic substances showed varying efficacy in reducing brown blotch depending on the inoculum concentration and the substances concentrations. The limiting effect of hydrogen peroxide and wine vinegar on the development of brown bacterial blotch in the second flush of fruiting bodies was shown. It can be concluded that the use of hydrogen peroxide and wine vinegar to reduce bacterial diseases of mushrooms can be an alternative to other chemical protection methods.

Plant growth-promoting rhizobacteria (PGPR) are a collection of microorganisms often used to support and promote plant development and combat plant infectious diseases with various biological control methods. The most significant restricting factors for agricultural productivity worldwide are abiotic constraints. In the present study, seven bacterial isolates from the rice rhizosphere were selected for detailed tests based on results obtained in experiments determining the ACC deaminase synthesis and drought tolerance at !0.30 MPa PEG level. Screening results of the stress tolerance analysis of the seven isolates for elevated temperature (50°C), alkalinity (10% NaCl), and drought (-1.2 MPa) showed that abiotic stress resistance was less prevalent in DRO2, DRO13, and DRO43 isolates than in DRO17, DRO28, DRO35, and DRO51 isolates. During the study, it was observed that DRO17, DRO28, and DRO51 tended to maintain similar cell density at -0.73 MPa PEG level, as observed at -0.30 MPa stress condition. No bacterial growth was observed at higher PEG level (-1.12 MPa) for any bacterial isolate. Four strains of Pseudomonas (DRO17, DRO28, DRO35, and DRO51) exhibited salinity and temperature tolerance. Antifungal screening using the bangle method showed that DRO35 was highly antagonistic towards Rhizoctonia solani 4633, followed by Fusarium moniliforme 4223, with an inhibition of 64.3% and 48%, respectively. The DRO28 isolate exhibited 72.5% growth inhibition for Fusarium moniliforme 4223, while the DRO51 isolate showed 38.2% growth inhibition for Bipolaris hawaiiensis 2445. DRO17 reduced the growth of Rhizoctonia solani 4633, and Curvularia lunata 350 by 36% and 31%, respectively. In conclusion, the screening of bacterial strains with promising stress tolerance and antifungal characteristics could support farmers to achieve the required positive outcomes in the agriculture field.

A new bacteria named Pseudomonas fulva K3 (P. fulva) strain was isolated from the surroundings of weathered crust rare earth tailing with efficient NH4+-N removal ability via heterotrophic nitrification and aerobic denitrification. The nitrogen removal properties could be intensified by the synergistic effect between as-prepared magnesium-modified biochar (MgBC) and P. fulva strain. The results show that P. fulva exhibited a rod-shaped morphology and $NH_4+-N$ can be completely biodegraded under the optimal conditions of pH=7.0~8.0, temperature 30 oC and initial $NH_4+-N$ concentration of 100 ~150 mg/L. The $NH_4+-N$ tolerant concentration for P. fulva was determined to be 300 mg/L. The magnesium-modified biochar (MgBC) worked as an adsorbent of $NH_4+-N$. The kinetics and isotherm model for adsorption could be described by the pseudo-secondorder kinetic and Freundlich model, respectively. The XPS results showed that $NH_4+-N$ was mainly adsorbed on the surface by chemical adsorption. Furthermore, the P. fulva could be immobilized on MgBC due to its large surface area, adjusting the concentration of $NH_4+-N$ to a proper range for the growth of P. fulva by adsorption and desorption equilibrium, and leading to the intensified effect on nitrogen removal. The total nitrogen removal efficiency of the eluted weathered crust rare earth tailing reached 84.7 % in MgBC + P. fulva system.

Opis:
Murraya koenigii L. (family: Rutaceae), commonly referred to as curry leaf, is a highly valued plant due to its aroma and medicinal features. Two dominant AM species Glomus mosseae and Acaulospora laevis were isolated from the rhizospheric soil of M. koenigii. A pot experiment was performed to verify the interactive potential of G. mosseae and A. laevis alone or in combination with Pseudomonas fluorescens on M. koenigii. Various morphological and biochemical parameters were measured after 120 days. Overall results suggest that all co-inoculation treatments showed beneficial effects on all the growth, physiological and oil content. The overall results demonstrate that the co-inoculation of bioinoculants, like P. fluorescens with AM fungi, promotes higher AM colonization and spore number enhancing the nutrient acquisition, especially phosphorus (P), improving the rhizospheric condition of soil.
