Opis:
Proteomics is the large scale of study of proteins, particularly their function and structure. Proteomics is an excellent approach for studying changes in metabolism in response to different stress conditions. In the present review focused on different types of techniques for the analysis of expressed proteins. The techniques includes 2-D gel electrophoresis, MALDI-TOF/MS etc., play a vital role for the analysis of novel proteins and their role in disease maintenance and treatment. The review also concentrated on applicative perspective of proteomics in the fields of biomedical, agriculture and food.

Opis:
VPS33B, a human Vacuolar Protein Sorting (VPS) protein which mediates the phagolysosomal fusion in macrophage of the eukaryotic organisms. This protein has a great role during the mycobacterial infections, which binds with the Mycobacterium protein tyrosine phosphatase A (PtpA). A single functional domain of PtpA has been identified using SMART domain databases, followed by finding the antigenicity of PtpA using CLC main workbench tool. The protein-protein interaction network predicts the interface of biological functions of proteins, built by using Cytoscape 2.8.3 version tool for manual literature survey of protein sets. According to the literature the specific interactivity of PtpA with VPS33B of human lead to pathogenesis, and provided a good platform to find the structure of VPS33B as it lacks the 3 dimensional structure in PDB. Homology Modelling of VPS33B provides a significant properties to design a specific drug through screening the drug databases (eDrug3D). The modelled protein has been validated through SAVES server maintained by NIH and UCLA with the standard Ramachandran plot with accuracy of 90.7 %. From our findings the interface residues are very crucial points which has been found through docking the modelled protein and Mycobacterium protein and interface residues were selected manually using PyMol software.

Introduction: Neurogenic bladder (NB) most often is caused by meningomyelocele (MMC) and manifests with various lower urinary tract dysfunctions. Condition of NB is worsened by inflammatory process or oxidative status imbalance. Purpose: To estimate of urinary uric acid (UA), hs-CRP, thiol status in association with NB function in MMC patients.Materials/Methods: 33 MMC children and 20 healthy individuals were included into the study. The first daytime urine samples were collected from all examined participants and urinary thiol status, hs CRP and UA were measured.Results: MMC children presented higher urinary UA level. The median hs-CRP level were also higher in MMC patients compared to the reference. Thiol status were lower in MMC individuals compared to reference group. We found positive correlation between serum creatinine, serum UA and urine creatinine and negative between serum creatinine and GFR. Correlations between urinary UA and physical development parameters, renal function, hsCRP, thiol status and urodynamic findings in MMC and reference groups were found.Conclusions: UA is a marker potentially having direct effect on the bladder function. Disturbed oxidative status and increased markers of inflammation may be a potentially modifiable factors affecting function of lower urinary tract in MMC children.

Opis:
The effect of the Panax ginseng powder extract on reproduction, growth, renal and liver functions and some blood biochemical indices of Japanese quails were investigated. Panax ginseng powder extract at 2 mg and 4 mg/bird daily increased egg number per hen, average egg weight and hatchability. It slightly improved the efficiency of feed utilization and increased the levels of serum total protein, alkaline phosphatase and aspartate amino transferase (AST). The extract decreased significantly the levels of albumin, total lipids, triacylglycerols, cholesterol and glucose, and had no effect on body weight and the levels of serum bilirubin, urea and creatinine. This suggests that long term studies should be done on many different biochemical and physiological parameters and laboratory animals.

Opis:
The in vitro protein biosynthesis has the potentials to become a powerful technology for biochemical research. Beside the determination of structure and function the in vitro evolution of proteins is also of great interest. The system described was used to produce bovine heart fatty acid binding protein (FABP) and bacterial chloramphenicol acetyltransferase (CAT) with and without fusion of the Strep-tag II affinity peptide. The proteins were purified after and during protein biosynthesis by using a StrepTactin Sepharose matrix. No significant influence of the Strep-tag and the conditions during the affinity chromatography on maturation or activity of the protein was observed. The in vitro evolution of proteins is feasible by means of ribosome display. The selection of a specific mRNA coding for a shortened FABP with a N-terminal His-tag via the accompanying protein property was shown. Goal of the selection was to bind the FABP via the His-tag on Ni(II)-IDA-agarose. After nine cycles of transcription, translation, affinity selection and RT-PCR the protein with the His-tag could be enriched 108-fold. In order to correlate a possible relationship between changes in protein population and biological function studies were initiated in which 2-dimensional protein patterns of the total in vitro system were compared after 0 and 2 h reaction time. The very interesting findings are that a number of proteins disappear, while others are newly formed during protein synthesis.

Opis:
The muscles and internal organs of marine animals contain many proteases. These enzymes fulfill different metabolic functions in the living organisms and affect the sensory quality and functional properties of the catch. Their activity depends on the species, life cycle, and the feeding status of the organism. Many proteases of marine origin differ from their counterparts in terrestrial animals in being more active at lower temperature and less resistant to thermal denaturation. Of practical importance in the industry is their role in ripening of salted fish, fish sauces and marinades, modyfying fish protein concentrates, ensilaging of seafood offal, and deskinning of fishery products. The negative effects comprise mainly the early post morten quality degradation of the catch and deterioration of the rheological properties of fish gels. Proteolytic preparations form different marine sources may be used successfully in food processing and as industrial enzymes, e. g. tanning agents.
