На основании анализа трех полиформных систем белков ( Hb ,Tf и CA), рассматривая частоту аллелей, коэффициент гомозиготности, коэффициент „чистопородности", коэффициент сродственности и генетическую дистанцию, можно говорить о существенных генетических различиях между сравниваемыми четырьмя популяциями овец: мериносом, низинной овцой и овцой разновидности Желязна.
The analysis of three polymorphic arrangements of proteins (Hb, Tf and CA) and consideration of the frequency of alleles, heterozygocity coefficient, "breed purity" coefficient, relation coefficient and genetic distance proved 1tie existence of significant genetic differences between the four sheep populations compared, viz.: Merino, Lowland, Wielkopolska and Żelazna sheep.

Зависимость между полиморфизмом трансферрина и гемоглобина с одной и оплодотворимостыо с другой стороны исследовали в двух стадах польской низинной овцы. В стадах не установлено стетистически существенных различий в частоте рождаемости близнецов в результате спаривания овец с разными генотипами гемоглобине и происходящих от баранов с различными гемотипами.
The dependence between the polymorphism of transferrin and hemoglobin on the one hand and fecundity on the other was investigated in two flocks of Polish lowLand sheep. No statistically significant differences in the frequency of birth Df twins after ewes and rams of different transferrin and hemoglobin genotypes aere found in any of these flocks. No significant differences were found, either, In the freuency of twin births from those of different hemoglobin genotypes and Driginating from rams of different hemotypes.

Opis:
α-Amylase isozymes from rye endosperm were analysed by means of isoelectric focusing, polyacrylamide gel electrophoresis, immunoelectrophoresis and colorimetric assay. Α-AMY 1 (high pI) group was separated into 13 IEF bands, whereas in group α-AMY2 (low pI) 2 intensive and 6-8 faint bands were found. Two linked (2±1 cM) polymorphic loci and a single locus with two alleles encoding for α-AMY 1 and α-AMY 2 groups, respectively, were identified after genetic analysis of the IEF patterns. All α-amylase isozymes developed on PAGE, were shown to belong to α-AMY 1 group. It was demonstrated that a single PAGE isozyme corresponds to 2-4 separate IEF bands and that most of the IEF bands can be attributed to more than one PAGE isozyme. The activity of α-amylases from PAGE zone I was 2.3 times higher than the activity of zone II isozymes. A strong correlation between the activity and protein amount of particular α-AMY 1 isozymes (r=0.94) was found.

The bovine kappa-casein (CASK) gene is known as a potential quantitative trait locus in dairy cattle breeding. However, the molecular basis of the effect of the CASK allele B on different milk properties remains unclear. In this report, a 214 bp fragment of the 5' untranslated region of the CASK gene containing 5 potential consensus sequenses for different transcription factors was PCR-amplified to find RFLPs. A Dde I RFLP was identified. In a population of 112 Bos taurus (86 cows and 26 bulls of Polish Black and White crossbred Holstein-Friesian) and 7 Bison bonasus individuals, 7 had no recognition sites for Dde I, 23 were heterozygous and 89 were cut completely into two fragments.

C-banding identified centromeric heterochromatin polymorphism, most often located in pair no. 16, was a basis for selecting animals for the experiment. The aim of the experiment was to assess the impact of centromeric heterochromatin polymorphism on pig fertility, expressed by litter size. The research included the first two litters obtained from 30 pairs of parents with different genotypes as regards the parameter under study. A statistical analysis of the number of offspring from different mating types showed no significant differences between the experimental groups of animals. The results obtained did not confirm suggested correlation between polymorphism of the centromeric heterochromatin regions and pig fertility, estimated by mean litter size. Thus the polymorphism cannot be regarded as a selection criterion.

Opis:
Genetic variability of a selected population of Dreissena polymorpha, an invasive species playing a significant role in aquatic ecosystems, was studied. Starch gel electrophoresis was used to analyse 8 enzymatic loci in 200 individuals collected from 20 sites in a lake. The population was found to have 75.0% of polymorphic loci, 2.8 alleles per locus, 3.5 alleles per polymorphic locus, 0.393 coefficient of expected heterozygosity, and 149 genotypes. Zebra mussel clumps were strongly polymorphic; almost every individual had a different genotype. The high polymorphism observed in the D. polymorpha clumps had most likely resulted from external cross-fertilisation and the presence of free-swimming veliger larvae as well as from a considerable heterozygosity of individual bivalves. Genetic variability of the population studied was found to be similar to that of populations inhabiting other Western Pomeranian lakes, including both highly polluted ones and those formed as recently as about 40 years ago. This provides evidence for a mass colonisation of freshwater reservoirs effected by very polymorphic parent populations of D. polymorpha. The literature data on North American zebra mussel populations which invaded that continent about 10 years ago show them to be polymorphic, too, but not as much as European ones.

The usefulness of mutagenic treatment to enlarge isozymic variability of barley and the use of induced mutants for genetic analysis were evaluated. N-methyl-N-nitroso urea, sodium azide and gamma rays were employed as mutagenic agents. Electrophoretic assays of 3848 M₂ seedlings obtained by chemical mutagenic treatment of the spring barley cultivars Dema, Aramir, Bielik and 3100 M₂ seedlings obtained by physical mutagenic treatment of the cv. Dema revealed 70 isozymic mutants, which represent 30 separate mutants in 25 M₁ plants. Most of mutations (27) were induced by chemical mutagen at polymorphic esterase loci. The occurrence of induced mutants at monomorphic loci, Got2 and Lap2, made it possible to perform genetic analysis of those loci in barley including mapping respective genes within chromosomes.

Opis:
The effect of transferrin genotypes and genes and globulin antigens on sheep resistance/susceptibility to mastitis was analysed. The udder health condition was diagnosed on the basis of the somatic cell count in 1 ml of milk and results of bacteriological tests. It was found that sheep with the transferrin genotype AC, AD and CC were characterised by a better health of the mammary gland than the remaining ewes. Among 9 transferrin alleles observed in the examined sheep, only the presence of transferrin I allele in the ewes genotype was connected with the somatic cell count below the mean value of this trait. The presence of transferrin A allele in the sheep's genotype was connected with a small infection of milk with mastitis pathogens. The serum globulin antigens analysed did not significantly affect the somatic cell count in the milk of the examined ewes. However, the antigens A2, NS1 (antigens of ß-globulin) and GB2 (antigen of class IgG immunoglobulins) as well as the globulin antigen A6 and GA1 antigen of class IgG immunoglobulins showed to have a significant effect on the level of sheep milk infection.

We have analysed allele distribution at the highly polymorphic variable number of tandem repeats (VNTR) locus D1S80 (pMCT118) in the Polish population using the, polymerase chain reaction (PCR) technique. Characteristics of the D1S80 locus makes it a very useful marker for population genetic research, genetic linkage studies and forensic identification of individuals. During our routine application of the D1S80 marker to paternity testing in several cases of homozygosity detected by polyacrylamide gel electrophoresis, heteroduplex formation for alleles 18 and 24 was also observed. Direct sequencing of PCR products revealed that alleles 18 and 24 of locus D1S80 actually represent a mixture composed of different sequences. Our observations indicate that identification of some 18 and 24 VNTR alleles based only on size estimated in electrophoretic analyses could lead to errors in paternity testing and DNA profiling.

Malignant hyperthermia (MH) is a clinical syndrome in which genetically susceptible individuals respond to the administration of potent inhalation anaesthetics and depolarization skeletal muscle relaxants with skeletal rigidity, unstable blood pressure, tachycardia, arrhythmias, hyperventilation, hypoxia, lactic and respiratory acidosis and high fever. In studies of the genetic basis of MH, a mutation was identified in the porcine (C1843T) and human (C1840T) skeletal muscle ryanodine receptor (RYR1) gene. This gene is mapped on human chromosome 19q13.1. The RYR1 gene contains 106 exons, of which two arc alternatively spliced.

The present study describes a rapid, simple method of bovine IVF embryo sexing by use of PCR technique. A pair of primers corresponding to the bovine amelogenin sequence has been used. The Rapid Cycler (Idaho Technology, USA) used in the current experiment enabled the PCR programme consisting of 55 cycles to be completed in less than 40 minutes. Therefore the total sexing procedure could be performed in less than 90 minutes. The described method succeded in case of 85% analysed embryos.

Dreissena polymorpha inhabits about 46.5% of the bottom area in Lake Dabie. It is most abundant in the northern part of the lake and sporadically occurs in its southern part. The average density of the zebra mussel is 891 individuals/m², while its wet weight is 1374 g/ m². In the settled areas the density of the zebra mussel reaches 1734 individuals/ m². There were presently established 7 age-groups from 0 to 6+. The highest number of specimens occurred in the age-groups between 1+ and 3+, while the lowest number - in the groups 0, 5 and 6+. Strong eutrophication and pollution of Lake Dabie on its southern side, combined with sedimentation of the extensive amounts of seston carried in by the Płonia and Regalica rivers, do not create favourable conditions for development of Dreissena. D. polymorpha plays a very positive role in the lake ecosystem through its biofiltrating action, contributing thus to an increase of water clarity and to limitation of phytoplankton development through cumulating nutrients in its biomass. Electrophoretic analysis of 9 enzymatic loci revealed strong polymorphism of the studied population of the zebra mussel: percent of polymorphic loci - 100, mean number of alleles in locus – 3.4, coefficient of expected heterozygosity per locus in the population, HS - 0.335, percentage of separate genotypes - 69%, in this number unique genotypes - 58%. A very strong scattering of the alleles was stated within the entire population. In each aggregation, on average, 90% of the specimens have a separate genotype. Each of the five defined groups of D. polymorpha, representing profiles I-V, respectively, had similar genetic composition. The values of the genetic similarity among the studied groups of the zebra mussel ranged from 0.96 to 0.99.

The RYR1 gene encoding the Ca²⁺ channel of sarcoplasmic reticulum of human skeletal muscle has been cloned and its nucleotide sequence has been determined earlier. We have used the polymerase chain reaction single strand conformation polymorphism (PCR-SSCP), and sequencing analysis for human, porcine (Sus scrofa), and zebrine (Equus grevyi) ryanodine receptor (ryrl) gene. The fragment of exon 17 of the ryr1 gene was characterized by a high homology between all the analysed species (substitution of a nucleotide is underlined): porcine ryr1 ¹⁸³⁴GTG GCC GTG CGC TCC AAC CAA GAT CT¹⁸⁵⁹ human RYR1 ¹⁸³¹GTG GCC GTG CGC TCC AAC CAA GAT CT¹⁸⁵⁶ zebrine ryr1 GTG GCC GTG CGC TCC AAC CAA GAC CT.

The goal of the study was to evaluate intragenic polymorphic sites in COL1A1 and COL1A2 loci. For COL1A1 the following intragenic markers were used: PCR-RFLP (COL1A1), G/A polymorphism in exon 45 of COL1A1 and C/T polymorphism in +88 position of COL1A1 non-translatable 3’ end. For COL1A2 PCR-VNTR was analyzed. 17 families were examined (6 of the "simplex" type and 11 of the "multiple" type). In 8 out of 11 "multiplex" families the segregation of the markers revealed correlation with OI, whereas the other 3 were non-informative. The method was not useful in "simplex" families.
