This paper is a short review of historical development of the tissue culture method in Poland. Similarly to the rest of the world, in vitro technology in Poland has progressed in many directions simultaneously. Its main fields are closely interconnected by natural sequences of biological processes and integrade one into another. The best results, driven by the prospects of practical applications, have been achieved within the areas of meristem culture and micropropagation of valuable genotypes, somaclonal variation, production of haploids and DH plants, somatic embryogenesis, in vitro culture of embryos and ovules, improvement of plant disease resistance, somatic hybridization and transformation of plants. Due to the fact that in vitro domain is a very broad science the authors are aware that this review might not fully cover all the scientists working with tissue culture and their achievements.

We investigated direct and indirect formation of somatic embryogenesis in Brassica oleracea var. botrytis (cauliflower), a very important vegetable crop worldwide. Direct somatic embryogenesis, which is rather rare, was achieved in culture of 2-week-old hypocotyl explants of Brassica oleracea var. botrytis on MS medium supplemented with 1.0 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D) and 0.5; 1.0; and 1.5 mg/l kinetin. Initial induction of embryogenic callus was achieved on MS supplemented with very low concentrations of 2,4-D (0.05 mg/l and 0.1 mg/l). Indirect somatic embryogenesis from leaf sections was obtained on MS supplemented with 0.05 or 0.1 mg/l 2,4-D. We examined various stages of somatic embryos (globular, heart, torpedo, cotyledonary). More embryos per explant were produced through the indirect pathway (23–25) than through the direct pathway (14–19). The number of embryos produced was high. There is a potential for recurrent, repeated or secondary somatic embryogenesis, possibly an unlimited source for mass propagation and ideal for synthetic seed production in this species. Plant regeneration was achieved on half-strength MS medium without any hormones.

The aim of the experiment was to determine suitable substrate type and optimal plant size for transfer of plantlets from in vitro to ex vitro under experimental outdoor conditions. Tests focused on the effect of substrate type (muddy and sandy) and starting size of plantlets gained through in vitro seed germination (0-3, 3.1-5,5.1-6, 6.1-10 cm) on plant growth. Three parameters (fresh weight, length, and the number of leaves) were compared to evaluate growth. Basic water parameters in experimental water tanks were regularly measured (pH, temperature, electrical conductivity, shadow intensity) and controlled to reach similar conditions to those in the natural habitat of this species. Overwintering was studied in a cellar with newly defined size categories (<6, 6.1-8, 8.1-10, 10.1-12, 12.1-15 cm). Both substrate type and starting size of plantlets significantly impacted growth. Plantlets grew better in the muddy substrate while a 100% success rate of rooting was gained with a starting size of 6.1-10 cm in both substrates. The biggest increase in fresh weight was observed with a starting size of 3.1-5 cm and 5.1-6 cm in both substrates. The greatest increase in fresh weight was observed in plants with a starting size of 3.1-5 cm in the muddy substrate (more than 95% increase). The best overwintering results were gained in the 6.1-8 cm size category.

Opis:
Initiation of tissue culture of many plant species is a very difficult stage due to appearance of many contaminations. The other problem might be a choice of media for regeneration. Initiation of grass species tissue cultures are thought to be very difficult. Therefore, a research was undertaken to evaluate the use of nano-silver particles for plant material disinfection and to estimate a medium Pennisetum alopecuroides. The plant material were buds and nodal explants that were disinfected in 2% NaOCl for 30 min or 0.1% HgCl2 for 1 min. Half of the explants disinfected with NaOCl were soaked in 50, 100 or 250 mg·dm–3 Ag NPs for 1 hour. Explants not soaked in nano-silver were placed on media with Ag NPs at concentrations of 4, 8 or 16 mg·dm–3. An influence of growth regulators on Pennisetum alopecuroides was evaluated in vitro. Regenerated shoots were placed on MS media with: 3 mg·dm–3 BA + 0.3 mg·dm–3 IBA, 3 mg·dm–3 KIN + + 0.3 mg·dm–3 IAA, 1 mg·dm–3 BA + 0.1 mg·dm–3 IBA. It was observed that the use of nano-silver particles lowered the level of contamination. The best results were obtained when Ag NPs was used at concentration of 100–250 mg·dm–3 alone or as a supplementation of the media, at concentration of 4 mg·dm–3 for nodes and 16 mg·dm–3 for adventitious buds. The use of nodal explants allowed to obtain less contamination. Regeneration depended on a media content. The most regenerated shoots were obtained on the MS media supplemented with 1 mg·dm–3 BA and 0.1 mg·dm–3 IBA.

The aim of the work was to evaluate the influence of cytokinins on in vitro propagated dahlia and their consequent effect on acclimatization. Plant material consisted of shoot tips and nodes. Among the three cytokinins, benzyladenine, kinetin and 2-isopentenyl-adenine, only BA effectively stimulated the shoot multiplication from axiliary buds. The highest multiplication rate was obtained from nodes in the presence of 0.25– 0.5 mg·dm–3 BA. Higher concentrations shortened the internodes and decreased the leaf blades and growth of callus. 1 mg·dm–3 of KIN and 2iP positively influenced the shoot growth and size of leaves. Gibberellic acid (GA3) used with BA increased the number of auxillary shoots. The best quality shoots and the highest multiplication rate were obtained when 2 mg·dm–3 BA was used with 5 mg·dm–3 GA3. Cytokinins affected the rooting and acclimatization ex vitro. Dahlia shoots multiplicated in the presence of 1 mg·dm–3 KIN or 2iP rooted faster in the soil and 100% survived in field, while those from 1 mg·dm–3 BA media rooted slowly, had shorter shoots and only 60% of them survived. Plants bloomed after 11–12 weeks in the field. Dahlia plants that had been multiplicated in the presence of KIN had larger diameter and fresh weight in the field. BA and 2iP positively influenced the flower diameter, length of flower stalk and a number of the first-order shoots.

In vitro tissue culture technique, especially micropropagation, is attracting increasing attention in the production of ornamental plants. This technique will probably dominate the horticultural market in the near future. Light is one of the several factors affecting the success of in vitro plant tissue cultures. It directly affects the widely understood morphogenetic response of the explant, i.e., the ability of the explant to grow or regenerate, produce roots, etc. Lighting conditions provided during the in vitro stage may also greatly affect the plant vigor after transferring to nonsterile conditions. Moreover, the necessity of providing artificial light significantly contributes to the total cost of maintaining tissue cultures (related to energy consumption and the need to cool down the heat generated by lamps). Light quantity (intensity) and quality (spectral composition) are the two main parameters that determine its influence on in vitro cultures. This impact depends on the species and other accompanying factors. The aim of this mini-review is to summarize information on the influence of light on the morphogenetic and biochemical response of explants of some selected ornamental plant species grown under in vitro conditions.
