Plants were regenerated from immature embryo cultures of 35 winter wheat genotypes. General responses of regenerated plants were investigated and a total of 7142 R₂ spike lines from 1593 R₁ plants were assessed in the field for somaclonal variants in 1985/86, 1986/87 and 1987/88. Selected variants were studied for their possible genetic inheritance. From regenerated plantlets, 81% survived and 63% produced fertile plants. Forms with reduced plant height, length of spike and other morphological abnormalities were found in this progeny. Populations of R₁ plants were highly variable due mainly to the physiological disturbances resulting from the in vitro process. Overall somaclonal variation frequencies were 14.2% per plant basis and 5.3% per R₂ spike basis. The variants were similar in the three different R₂ generations with predominant variants being negative in plant height, maturity, awns, spike type and plant type. Both uniform R₂ variant families and spike lines were found in addition to the segregating variants which constituted the majority. On average, in a variant family or line, 18% and 14% of their component lines and plants were variants, respectively. Inheritability was demonstrated for the uniform variant families and spike lines as well as segregated variants. Of those 134 selections, about 70% were classified as inheritable. Both recessive and dominant gene mutations at one, two or three loci were evident in some variants as suggested by the segregating data.

There are limited published data on in vitro reproduction of Fagus sylvatica L. (European beech). This study was aimed to determine the efficiency of induction of somatic embryogenesis or organogenesis of beech from different types of explants in various culture conditions. Explants derived from immature, fresh seeds (collected in 2011 and 2013) and from mature seeds, stored at –10ºC and some stratified at 3ºC, were placed on induction media with various combinations of plant growth regulators: zeatin, 2,4-dichlorophenoxyacetic acid (2,4-D) and/or benzyladenine (BA). Initial cultures were kept in darkness or weak light (white fluorescent or blue-red LED). Limited success has been achieved in initiation of somatic embryogenesis. We obtained friable, yellow-white callus with characteristic PEM-like structures (cPEM-ls, from embryonic axes or fragments of immature embryos with embryonic axes), which may be an early developmental stage of embryogenic callus of Fagus sylvatica. This type of callus regenerated from explants incubated in darkness, mainly on WPM medium with addition of 6.8 μM zeatin or WPM and MSG media with 9.1 μM 2,4-D and 2.2 μM BA. The highest frequency of regeneration of callus with cPEM-ls was 5%. Instead, we succeeded to induce organogenesis from both immature and mature zygotic embryos and from embryonic axes. The best results were obtained for mature zygotic embryos incubated on ½WPM medium (half-strength Woody Plant Medium) with 9.1 μM 2,4-D and 2.2 μM BA. Adventitious buds were regenerated on up to 15% of the explants. The induced buds developed into shoots, enabling us to establish tissue cultures of beech. Induction of organogenesis from the tested explants was more efficient than induction of somatic embryogenesis.

Recent advances at Institute of Grassland and Environmental Research (Aberystwyth, U.K.) in cytogenetics of the Lolium/Festuca complex places us in the advantageous position of being able to map genes of agronomic importance onto chromosome arms using fluorescence in situ hybridization (FISH). The ability to physically map genes leads to the capability for "dissecting" quantitative traits into their different components and will lead to better understanding of the complex physiological processes involved and the identification of their genetic control. By tagging genes of interest, using molecular and morphological markers, it will be possible to select and combine suites of desirable genes in a single genotype and thus produce novel cultivars by conventional breeding procedures. Programmes for introgression depend on the relationships between species and on levels of chromosome pairing. Phylogenetic relationships within the Lolium/Festuca complex are being determined using both genomic in situ hybridization (GISH) and FISH. With recent advances in genetic manipulation within the Lolium/Festuca complex, opportunities now arise for gene transfer from Lolium and Festuca species into other important agricultural crops.

Under a scanning microscope, leaves from tissue cultures (after 8 weeks in vitro culturing), from 1 year old regenerants and from the 100 years old donor tree were studied. More abnormalities were observed in the tissue culture compared to regenerant and donor. During tissue culture transfer rapid, leaf dehydration occurs due to defective regulation mechanisms making stomata of tissue culture stay open. Epidermis as well as stomata cells were observed already on the leaves in in vitro conditions, but they become physiologically functioning only after transfer into conditions of natural environment.
Przy zastosowaniu mikroskopu scaningowego porównywano cechy budowy komórek liści pochodzących z 100 letniej rośliny matecznej, 8 tygodniowych kultur tkankowych oraz 1 rocznych regeneratów. W regenerantach z kultur tkankowych obserwowano więcej nienormalności w budowie komórek epidermy, komórek szparkowych i komórek przyszparkowych w porównaniu z komórkami pochodzącymi z rośliny matecznej. Podczas przenoszenia tkanek w kulturze in vitro obserwowano odwodnienie tkanek liści a mimo to komórki szparkowe pozostawały otwarte. Komórki epidermy oraz komórki szparkowe były obecne na liściach hodowli in vitro jednak stawały się one fizjologicznie funkcjonalne tylko wtedy gdy zostały przeniesione do warunków naturalnych.

In vitro flowering and micropropagation are useful for plant breeding programs and commercial production of important ornamental plants. In vitro conditions including media components, kind, concentration and ratio of plant growth regulators and culture conditions significantly affect in vitro flowering and micropropagation. There is no any report dealing with the in vitro flowering of Lisianthus (Eustoma grandiflorum). Here, a protocol was developed for flowering and high frequency in vitro micropropagation of E. grandiflorum, an ornamental plant. Micropropagation is an effective tools for propagation of ornamental plants in large scale. The aim of the present study was to evaluate the effect of different concentrations of NAA and BA on micropropagation and flowering of Lisianthus, in vitro. Used culture medium was MS enriched with 0, 0.1, 0.2 and 2 mg L-1 of NAA and BA. In establishment process of explants, the most shoot length (2.07 cm per plant) was obtained on medium supplemented with 0.1 mg L-1 BA (without NAA). Maximum shoot number (5.80 per plant) was produced in medium containing 0.1 mg L-1 BA along with 0.2 mg L-1 NAA. Bud explants in culture media containing 0.2 mg L-1 NAA (without BA) and 0.1 mg L-1 NAA along with 2 mg L-1 BA produced maximum node number (3.20 per plant). The largest number of root (14.53 per plant) and root length (3.87 cm per plant) were produced on 0.2 mg L-1 NAA without BA, also 0.2 mg L-1 BA plus 0.2 mg L-1 NAA and 0.2 mg L-1 BA without NAA. Explants produced flower on medium containing 0.1 mg L-1 BA along with 0.1 mg L-1 NAA without transition of callus formation. Flower was produced from callus in medium containing 0.1 mg L-1 BA along with 2 mg L-1 NAA. Regenerated plants showed 98% survival in greenhouse during acclimatization. Acclimatized plants were morphologically similar to the mother plants.
Kwitnienie i mikropropagacja in vitro są użyteczne w programach hodowli roślin oraz produkcji komercyjnej ważnych roślin ozdobnych. Warunki in vitro, łącznie ze składnikami pożywek, rodzajem, stężeniem oraz proporcją regulatorów wzrostu roślin, a także warunkami hodowli, w sposób istotny wpływają na kwitnienie i mikropropagację in vitro. Nie istnieje żadne badanie dotyczące kwitnienia in vitro lisanthiusa (Eustoma grandiflorum). W niniejszym badaniu opracowano kwitnienie i wysoką częstotliwość mikropropagacji in vitro dla E. grandiflorum, który jest rośliną ozdobną. Mikropropagacja jest skutecznym narzędziem rozmnażania roślin ozdobnych na dużą skalę. Celem niniejszego badania była ocena wpływu różnych stężeĔ NAA i BA na mikropropagację i kwitnienie lisianthiusa in vitro. Używana pożywka hodowlana została wzbogacona za pomocą 0; 0,1; 0,2 i 2 mg L-1 NAA i BA. Przy powstawaniu eksplantów największa długość łodygi (2,07 cm na roślinę) była uzyskana na pożywce uzupełnionej o 0,1 mg L-1 BA (bez NAA). Maksymalna liczba łodyg (5,80 na roślinę) została wytworzona na pożywce zawierającej 0,1 mg L-1 BA wraz z 0,2 mg L-1 NAA. Eksplanty pączków na pożywce hodowlanej zawierającej 0,2 mg L-1 NAA (bez BA) oraz 0,1 mg L-1 NAA wraz z 2 mg L-1 BA wytworzyły maksymalną liczbę węzłów (3,20 na roślinę). Największą liczbę korzeni (14,53 na roślinę) oraz największą długość korzenia (3,87 na roślinę) zaobserwowano na 0,2 mg L-1 NAA bez BA jak również 0,2 mg L-1 BA plus 0,2 mg L-1 NAA oraz 0,2 mg L-1 BA bez NAA. Eksplanty tworzyły kwiat na pożywce zawierającej 0,1 mg L-1 BA wraz z 0,1 mg L-1 NAA bez przeniesienia kalusa. Kwiat był tworzony z kalusa na pożywce zawierającej 0,1 mg L-1 BA wraz z 2 mg L-1 NAA. Zregenerowane rośliny wykazały 98% przeżycie w szklarni podczas aklimatyzacji. Zaaklimatyzowane rośliny były morfologicznie podobne to swych roślin macierzystych.

Opis:
Allium aflatunense and Allium karataviense are valuable bulbous plants grown in a garden. Moreover, Allium aflatunense is one of the most decorative species for cut flowers industry. These plants characterize with a low multiplication rate. Immature inflorescence stem sections and leaf bases isolated from ‘Allium aflatunense ‘Purple Sensation’ and Allium karataviense ‘Ivory Queen’ bulbs in October were used as source of explants for shoot regeneration. The use of a two – step sterilization procedure (2% Cl- – 30 min and 1% Cl- –15 min) gave much better results than a one – step (2% Cl- – 30 min). Inflorescence explants in inverted orientation were cultured on a Murashige and Skoog [1962] medium containing BA in concentration of 2 mg·dm⁻³ and NAA in concentration of 1 mg·dm⁻³. Leaf bases placed in normal and inverted orientation were incubated on MS medium supplemented with: BA 2 mg·dm⁻³ plus NAA 1 mg·dm⁻³, BA 5 mg·dm⁻³ plus NAA 1 mg·dm⁻³ and TDZ 1 mg·dm⁻³ plus NAA 1 mg·dm⁻³. In subsequent experiments clumps consisting of 2 shoots were cultured on MS medium containing BA 0.5, 1, 2 mg·dm⁻³ with NAA 0.1 mg·dm⁻³ and BA 2 mg·dm⁻³ with IAA 0.1 mg·dm⁻³ for 6 weeks. A multiplication rate during four subcultures on medium with addition of BA 0.5 mg·dm⁻³ and NAA 0.1 mg·dm⁻³ was also examined. Explants from young inflorescences and from immature leaf bases were found effective to form shoots. The orientation of leaf explants did not have a significant effect on the number of regenerating shoots. 5.7 to 7.5 shoots were achieved for Allium aflatunense ‘Purple Sensation’ on studied media during 6 weeks of multiplication and 3.5 to 5.1 shoots for Allium karataviense ‘Ivory Queen’. Multiplication rate of 31.3 shoots for Allium aflatunense ‘Purple Sensation’ and 26.6 shoots for Allium karataviense ‘Ivory Queen’ was possible to obtain on medium supplemented with BA 0.5 mg·dm⁻³ and NAA 0.1 mg·dm⁻³ after 32 weeks of culture.
Allium aflatunense i Allium karataviense to cenne rośliny cebulowe do uprawy w ogrodach. Ponadto Allium aflatunense jest jednym z najbardziej dekoracyjnych gatunków uprawianym na kwiat cięty. Rośliny te charakteryzują się niskim współczynnikiem rozmnażania. Źródłem materiału do zapoczątkowania kultur były fragmenty młodych pędów kwiatostanowych i nasady liści izolowane w paĨdzierniku z Allium aflatunense ‘Purple Sensation’ i Allium karataviense ‘Ivory Queen’. Zastosowanie sterylizacji dwukrotnej (2% Cl- – 30 min i 1% Cl- –15 min) dało lepsze rezultaty niż sterylizacja pojedyncza. Eksplantaty z pędów kwiatostanowych wykładano w pozycji odwrotnej do naturalnej i kultywowano na pożywce Murashige i Skooga [1962] zawierającej BA 2 mg·dm⁻³ i NAA 1 mg·dm⁻³. Podstawy liści układano w pozycji naturalnej i odwróconej i inkubowano na pożywce MS z dodatkiem: BA 2 mg·dm⁻³ + NAA 1 mg·dm⁻³, BA 5 mg·dm⁻³ + NAA 1 mg·dm⁻³ i TDZ 1 mg·dm⁻³ + NAA 1 mg·dm⁻³. W kolejnym doświadczeniu zespoły pędów zawierające 2 pędy namnażano na pożywkach MS uzupełnionych: BA 0,5; 1; 2 mg·dm⁻³ oraz NAA 0,1 mg·dm⁻³ i BA 2 mg·dm⁻³ oraz IAA 0,1 mg·dm⁻³ podczas 6 tygodni. Oceniono również współczynnik namnażania pędów podczas 4 pasaży na pożywce z dodatkiem BA 0,5 mg·dm⁻³ i NAA 0,1 mg·dm⁻³. Fragmenty młodych pędów kwiatostanowych i podstawa liści wykazywały duże zdolności regeneracyjne pędów. Sposób ułożenia eksplantatów nie wywierał istotnego wpływu na liczbę regenerujących pędów. Na badanych pożywkach uzyskano od 5,7 do 7,5 pędów Allium aflatunense ‘Purple Sensation’ i od 3,5 do 5,1 pędów Allium karataviense ‘Ivory Queen’ podczas 6 tygodni namnażania. Podczas 32 tygodni namnażania pędów na pożywce zawierającej BA 0,5 mg·dm⁻³ i NAA 0,1 mg·dm⁻³ uzyskano 31,3 pędów Allium aflatunense ‘Purple Sensation’ i 26,6 pędów of Allium karataviense ‘Ivory Queen’.

Mandevilla sanderi is an important commercial ornamental pot plant. Traditional vegetative propagation is limited due to the low rate, therefore there is a need to develop an alternative, more efficient method. There is an interest in development of micropropagation technology for the species, as it allows to obtain a lot of offsprings in a relatively short time. The aim of the present work was to estimate an influence of explants type and position on regeneration of Mandevilla sanderi in tissue culture. Four different types of explants (leafy shoot tips, decapitated leafy shoot tips, defoliated shoot tips, decapitated and defoliated shoot tips) were used in the experiment, which were placed on the media vertically, while defoliated shoot tips were placed horizontally or vertically upside down. The explants were cultivated on a Murashige and Skoog (MS) medium supplemented with 1 mg·dm–3 benzyladenine (BA) and 0.5 mg·dm–3 indole-3-butyric acid (IBA). It was noted that both explants orientation and positioning, influenced the multiplication rate. Defoliated shoot tips placed horizontally were characterized by higher multiplication rate (6.8) in comparison to upside down vertical positioning (3.2). It was also observed that removal of shoot apex improved axillary branching, while defoliation of shoots placed in a normal position reduced multiplication rate.

Investigations were conducted on the strongly infested by microbes embryos isolated from seed stratified in non-sterile conditions or from tree buds. The results showed, that PPM (Plant Preservative Mixture, produced by Plant Cell Technology, Inc.) was very useful for increasing the percentage of sterile cultures disinfected in a 5% solution of NaOCl. The best way of applying PPM turned out to be adding the substance directly to the growing medium. At the concentration of 2 or 4 cm3 x dm-3 of PPM there was an 30% increase in percentage of sterile of in vitro cultures developed from embryos and 70-80% of cultures developed from buds, as compared with the control without PPM. At the range of concentrations employed in this study, no negative effects of PPM upon the development and growth of callus were observed.

Opis:
In vitro tissue culture technique, especially micropropagation, is attracting increasing attention in the production of ornamental plants. This technique will probably dominate the horticultural market in the near future. Light is one of the several factors affecting the success of in vitro plant tissue cultures. It directly affects the widely understood morphogenetic response of the explant, i.e., the ability of the explant to grow or regenerate, produce roots, etc. Lighting conditions provided during the in vitro stage may also greatly affect the plant vigor after transferring to nonsterile conditions. Moreover, the necessity of providing artificial light significantly contributes to the total cost of maintaining tissue cultures (related to energy consumption and the need to cool down the heat generated by lamps). Light quantity (intensity) and quality (spectral composition) are the two main parameters that determine its influence on in vitro cultures. This impact depends on the species and other accompanying factors. The aim of this mini-review is to summarize information on the influence of light on the morphogenetic and biochemical response of explants of some selected ornamental plant species grown under in vitro conditions.

Opis:
The aim of the work was to evaluate the influence of cytokinins on in vitro propagated dahlia and their consequent effect on acclimatization. Plant material consisted of shoot tips and nodes. Among the three cytokinins, benzyladenine, kinetin and 2-isopentenyl-adenine, only BA effectively stimulated the shoot multiplication from axiliary buds. The highest multiplication rate was obtained from nodes in the presence of 0.25– 0.5 mg·dm–3 BA. Higher concentrations shortened the internodes and decreased the leaf blades and growth of callus. 1 mg·dm–3 of KIN and 2iP positively influenced the shoot growth and size of leaves. Gibberellic acid (GA3) used with BA increased the number of auxillary shoots. The best quality shoots and the highest multiplication rate were obtained when 2 mg·dm–3 BA was used with 5 mg·dm–3 GA3. Cytokinins affected the rooting and acclimatization ex vitro. Dahlia shoots multiplicated in the presence of 1 mg·dm–3 KIN or 2iP rooted faster in the soil and 100% survived in field, while those from 1 mg·dm–3 BA media rooted slowly, had shorter shoots and only 60% of them survived. Plants bloomed after 11–12 weeks in the field. Dahlia plants that had been multiplicated in the presence of KIN had larger diameter and fresh weight in the field. BA and 2iP positively influenced the flower diameter, length of flower stalk and a number of the first-order shoots.
