Opis:
In this paper, herbal bitters are widely used due to their numerous acclaimed health benefits in many Nigerian homes; however, many have not been subjected to scientific scrutiny. The aim of this study was to determine the phytochemical composition and antioxidant capacity of a non-alcoholic polyherbal formulation, Yoyo bitters, towards validating its broad pharmacological claims. The phytochemical components of Yoyo bitters were ascertained by phytochemical screening assays and gas chromatography-mass spectrometry (GC-MS). The antioxidant activity was investigated in vitro using 2,2-diphenyl-1-picryhydrazyl (DPPH) radical, hydrogen peroxide (H2O2) scavenging activity, total antioxidant capacity (TAC) and ferric reducing antioxidant power (FRAP) assays. Qualitative phytochemical analysis of Yoyo bitters showed the presence of saponins, tannins, flavonoids, terpenoids, cardiac glycosides and anthocyanins. The total phenols, flavonoids, flavanols, tannins and carotenoids content were 14.741 ±0.64 mg GAE/ml, 0.152 ±0.01 mg RE/ml, 0.437 ±0.02 mg RE/ml, 0.368 ±0.04 mg TAE/ml and 0.016 ±0.00 mg CAE/ml respectively. GC-MS chromatogram revealed the presence of forty-three (43) phytochemical compounds with D-allose (41.81%), 1,6-anhydro-beta-D-glucofuranose (24.15%), 5-hydroxymethylfurfural (8.02%) and Z-6-pentadecen-1-ol acetate (3.50%) as the most abundant constituents. Yoyo bitters demonstrated effective antioxidant activity against DPPH and H2O2 with IC50 values of 0.492 mg/ml and 0.629 mg/ml respectively compared to ascorbic acid of 0.161 mg/ml and 0.130 mg/ml respectively. Total antioxidant capacity and ferric reducing antioxidant power of Yoyo bitters were 0.432 mg AAE/ml and 2.236 mg AAE/ml respectively. This study validates the antioxidant capacity of Yoyo bitters and provides chemical basis for its acclaimed pharmacological actions.

Rumex thyrsiflorus Fingerh. is mentioned as a European folk medicinal plant. This species has also been traditionally used as an edible plant in Eastern Europe because of its nutritional value. During the study, qualitative and quantitative sex-related differences of phenolic constituents in methanolic leaf extracts of R. thyrsiflorus were evaluated. The presence of the same substances (nine phenolic acids before, and six phenolic acids after acid hydrolysis, nine flavonoids, and a catechin) was estimated in both female and male specimens, using the HPLC-DAD method. A statistically significant higher content of eleven constituents in female plant extracts (acids: chlorogenic, p-coumaric, cryptochlorogenic, gallic, protocatechuic, neochlorogenic, vanillic; flavonoids: quercitrin, rhamnetin, rutoside; and a catechin) was shown. This is the first report concerning the relation between the sex and the content of biologically active phenolic secondary metabolites in leaf extracts of R. thyrsiflorus. Female plants of R. thyrsiflorus could be useful for pharmaceutical purposes as a preferential source of bioactive phenolic acids, flavonoids and especially catechin.

Opis:
In cyanobacteria, carbonic anhydrase (zinc metalloenzyme) is a major enzyme that converts CO2 to HCO3 maintaining the carbon concentration around the vicinity of RuBisCo, leading to cyanobacterial biomass generation. Anthropogenic activities, disposal of leached micro nutrients effluents from industries into the aquatic environment results in cyanobacterial blooms. The harmful cyanobacteria release cyanotoxins in open-water system which on ingression through oral route causes major health issues like hepatotoxicity and immunotoxicity. A database was prepared consisting of approximately 3k phytochemicals curated from previous literatures, earlier identified by GC-MS analysis. The phytochemicals were subjected to online servers to identify the novel lead molecules which followed ADMET and drug-like candidates. The identified leads were optimized by density functional theory method using B3YLP/G* level of theory. Carbonic anhydrase chosen as target to observe the binding interaction through molecular docking simulations. From the molecules included in the database the highest binding energy exhibited by alpha-tocopherol succinate and mycophenolic acid were found to be -9.23 kcal/mol and -14.41 kcal/mol and displayed interactions with GLY A102, GLN B30, ASP A41, LYS A105 including Zn2+ and their adjacent amino acids CYS 101, HIS 98, CYS 39 in both chain A and chain A-B of carbonic anhydrase. The identified molecular orbitals decipher computed global electrophilicity values (Energy gap, electrophilicity and Softness) of alpha tocopherol succinate and mycophenolic acid were found to be (5.262, 1.948, 0.380) eV and (4.710, 2.805, 0.424) eV demonstrates both molecules are effective and stable. The identified leads may serve as a better anti-carbonic anhydrase agent because they accommodate in the binding site and hampers the catalytic activity of Carbonic anhydrase thus inhibiting the generation of cyanobacterial biomass. This identified lead molecules may serve as a substructure to design novel phytochemicals against carbonic anhydrase present in cyanobacteria. Further in vitro study is necessary to evaluate the efficacy of these molecules.

Opis:
The research study evaluated the phytochemical constituents of Methanol extracts of Moringa oleifera whole leaf by Gas Chromatography-Mass Spectrometry (GC-MS) and Fourier transform infrared spectroscopy (FT/IR) analysis. The leaves was washed, air dried for 2 weeks, then ground into a fine powder and extracted using methanol by maceration for 24 hours using standard procedures. After the contact time elapsed, the solvent filtered to recover the extract. The functional groups and the chemical constituents of the methanol extract of Moringa oleifera leaves investigated using Fourier transform infrared spectroscopy (FT/IR) and Gas Chromatography-Mass Spectrometry (GC-MS) respectively. The FT/IR analysis confirmed the presence of O-H, C=C, C-H, C-O, CH3 and C=C-H bond stretching functional groups, which indicates that the substance is an aliphatic alcohol, ester, aldehyde and carboxylic acid. However, the fingerprint region had a pattern that is specific for every molecule, the presence of -OH function and N-O stretch; suggest that or alcohols and nitrogen or aromatic or aliphatic phenols containing molecules are major components of the Moringa oleifera leaf studied. GC-MS analysis of the extract reveals the identification of twenty compounds, in which two compounds were identified in each peak. N,N'-Pentamethylenebis[s-3-aminopropyl thiosulfuric acid and 2-Myristynoyl pantetheine (100%), 2-Myristynoyl pantetheine and Deoxyspergualin (92.05%), 5-Octadecenal and 9-Hexadecenoic acid (27.94%). N,N'-Pentamethylenebis[s-3-aminopropyl thiosulfuric acid and Pentetic Acid are the major phytoconstituents. Most of the compounds in the list are bioactive and possess medicinal properties, which further justify the application of Moringa oleifera traditional plant in the discovery of novel therapeutics.

Germinating seeds constitute a natural source of substances that can be used to supplement food and increase its functionality. In this work, red mizuna (Brassica rapa L. var. nipposinica) in the form of sprouts was used as research materials. The main goal of this study was to investigate the effect of germination time on the phytochemical profile of health-promoting compounds in red mizuna sprouts. The metabolomic profile of water-methanol mizuna sprout extracts was analyzed by UPLCESI-MS/MS. In this work qualitative analysis of four mizuna sprout extracts collected after 4, 6, 8 and 11 days after sowing was carried out. Moreover, the phytochemical characterization of red mizuna seeds before the sprouting process was undertaken. Thirty-nine compounds were interpreted and the fact of the variability of the qualitative profile over time was confirmed. This is the first report to provide information on the differences in the phytochemical profile between sprouts of red mizuna subjected to the influence of germination time, in order to determine the harvesting and consumption maturity of the tested plant material.
