Opis:
Photodynamic therapy (PDT) is a promising treatment method for non-oncological and oncological diseases. PDT requires the use of photosensitizer, light, and cell oxygen. A selective, cytotoxic effects can be achieved in cancer cells, thanks to formation of radical oxygen species (ROS). Recent data indicate, that PDT with aminolevulinic acid (ALA) is successful in treatment of colorectal cancer. However, anti-cancer effects PDT in vivo, can vary in PDT in vitro, due to low level of oxygenation of tumor cells. Experimental research confirms the role of cholesterol in oncogenesis and indicates, that in colorectal cancer the cholesterol synthesis is disrupted; among others lovastatin belong to inhibitors 3-hydroxy-3-methyl-glutharyl-coenzyme A (HMG-CoA) enzyme of cholesterol synthesis pathway. Anti-tumor potential of statins: antiproliferative and pro-apoptotic, were demonstrated in various tumors in vitro and in vivo. Colorectal cancer, which often has genes mutations that are related to programmed death cells, shows resistance to chemotherapy. The objective of the research was to investigate the effect of antiproliferative and cytotoxic lovastatin and ALA-PDT (in low dose) working separately and in combination method (lovastatin and PDT) − for apoptosis induction, in metastatic colon cell line SW620. Our finding showed, that lovastatin inhibits the growth of cancer cells, depending on the dose and incubation time. ALA-PDT in low dose works on cell cytotoxic, yet do not initiate apoptosis. Whereas, preincubation of cells with lovastatin (IC50) increases the cytotoxic effect after PDT and induces apoptosis in cancer cells SW620, as demonstrated using an assay of Annexin V for flow cytometry. Our research demonstrates for the first time, the effectiveness of ALA-PDT connection with an inhibitor of cholesterol synthesis pathway – lovastatin, in increasing the cytotoxicity and induction of apoptosis in colon cancer cells, that are resistant to chemotherapy.
Terapia fotodynamiczna (ang. photodynamic therapy, PDT) jest obiecującą metodą w leczeniu nowotworowych i nienowotworowych schorzeń. Działanie PDT wymaga użycia fotouczulacza, światła i tlenu komórkowego, dla uzyskania wybiórczego cytotoksycznego efektu w komórkach patologicznych. Efekt ten jest wynikiem powstania wolnych rodników (ROS). Badania pokazują, że PDT z kwasem aminolewulinowym (ALA) jest skuteczna w leczeniu raka jelita grubego. Jednak efekty PDT in vivo, ze względu na niski poziom oksygenacji guza, mogą odbiegać od działania PDT in vitro. Dane eksperymentalne potwierdzają rolę cholesterolu w procesie onkogenezy i wskazują na zaburzony szlak syntezy cholesterolu, m.in. w raku jelita grubego. Statyny, np. lowastatyna, należą do inhibitorów 3-hydroksy-3-metylo-glutarylo-koenzymu A (HMG-CoA) − enzymu szlaku syntezy cholesterolu. Potencjał antynowotworowy statyn, antyproliferacyjny i proapoptotyczny, wykazano w nowotworach in vitro i in vivo. Rak jelita grubego, cechujący się mutacjami genów związanych z apoptozą, wykazuje oporność na standardowe chemioterapeutyki. Celem pracy było zbadanie efektu antyproliferacyjnego i cytotoksycznego lowastatyny i ALA-PDT (w niskiej dawce) działających osobno, oraz w metodzie połączonej (lowastatyna i PDT) – na indukcję apoptozy w przerzutowej linii raka jelita grubego SW620. Wyniki pokazały, że lowastatyna hamuje wzrost komórek w związku zależnym od dawki i czasu inkubacji. ALA-PDT w niskiej dawce działa cytotoksycznie, ale nie inicjuje apoptozy. Natomiast preinkubacja komórek raka z lowastatyną (IC50) zwiększa efekt cytotoksyczny po PDT i indukuje apoptozę w komórkach, co wykazano w teście z aneksyną V na cytometrze przepływowym. Nasze wyniki po raz pierwszy demonstrują skuteczność metody łączonej: ALA-PDT i inhibitora syntezy szlaku cholesterolu – lowastatyny, w zwiększeniu cytotoksyczności i indukcji apoptozy w komórkach raka jelita grubego, opornego na chemioterapię.

Opis:
Simple methods for detection and isolation of protein-porphyrin complexes were elaborated in our laboratory. They are based on the separation of protein-porphyrin complexes in native polyacrylamide gel and measurement of their fluorescence, with the use of two detection systems: the commercially available Gel DocTM 2000 system, and a system specially designed for the purpose of these investigations, concerning protein-porphyrin interactions. The fluorescent complexes can be electro-transferred from the gel onto PVDF membrane, eluted and analyzed in order to identify the protein interacting with porphyrins.

Photodynamic therapy (PDT) is based on photosensitizers activated by light of appropriate wavelength. Their activation leads to generation of singlet oxygen and free radicals responsible for the cytotoxic effect. The aim of this project was to compare the bactericidal effect of PDT using different porphyrin photosensitizers against a methicillin-resistant Staphylococcus aureus strain. Exogenous sensitizers (protoporphyrin IX and newly synthesized derivative, protoporphyrin diarginate) induced a 3 log10-unit reduction in bacterial viable counts. With the use of endogenous, ALA-induced porphyrins, a 1.6 log10-unit reduction was obtained. The sensitizers tested executed their antibacterial activity with no essential change in the antibiotic resistance pattern of the studied strain.

Terapia fotodynamiczna postrzegana jest obecnie jako metoda przydatna w leczeniu wielu typów nowotworów w różnym stadium zaawansowania oraz chorób innego pochodzenia. Aktywacja fotouczulacza wybiórczo zgromadzonego w nieprawidłowych komórkach przez światło laserowe o odpowiedniej długości fali, nie dość, że pozwala na oszczędzenie zdrowych, niezaatakowanych procesem chorobowym obszarów, to jest także wysoce skuteczna i dobrze znoszona przez pacjentów. Jednakże często opisywanym przez nich dyskomfortem towarzyszącym leczeniu jest ból. Reakcja bólowa w PDT zależy w dużym stopniu od rodzaju, umiejscowienia i rozmiaru leczonej zmiany chorobowej. Nie bez znaczenia są także inne czynniki charakterystyczne dla PDT jak typ oraz stężenie fotouczulacza, fluencja, gęstość strumienia cząstek, promieniowanie, źródło światła, długość fali, frakcjonowanie naświetlania, wzrost temperatury, wiek i płeć pacjenta oraz fototyp i rodzaj skóry. Współczesna medycyna dysponuje skutecznymi narzędziami w walce z bólem pojawiającym się także w wyniku postępowania terapeutycznego, jak np. PDT. W niniejszej pracy dokonano przeglądu stosowanych dotychczas metod niwelowania i leczenia dolegliwości bólowych towarzyszących fototerapii.
Photodynamic therapy is perceived nowadays as useful method of treating many types of tumors of diverse severity, as well as other diseases. Firstly, the activation of photosensitizing agents selectively accumulated in tumor cells by laser light, suitable for the wavelength of the substance allows to spare healthy, not undergoing disease process regions. Secondly, this procedure is very effective and well tolerated by patients. However, the main side effect described by patients is pain. The pain reaction caused by PDT depends largely on the type, localization and extent of treated lesion. The other typical for PDT factors such as type and concentration of photosensitizing agent, fluence and irradiance, light source and wavelength, fractionated light, temperature increase, patient age and sex, skin phototypes are not insignificant. There are effective methods of controlling the pain occuring during treatment procedures, such as PDT. This article also provides a review of commonly used relieving procedures of pain caused by phototherapy.

Due to its fluorescent and phototoxic properties, rose bengal (RB) is used in photodynamic therapy. To improve the delivery of RB to its site of action, the application of nanocarrier systems has been proposed. The most promising approach includes the use of pH-responsive nanoparticles. To evaluate the pattern of drug release in different buffers, equilibrium dialysis is commonly used. Here, we used water and two buffers to determine the impact of solvent composition on the aggregation and dialysis rate of RB through a cellulose membrane. The results show that buffer composition does not influence the fluorescent properties of RB. However, the presence of additional ions causes a change in diffusion rate that is most probably linked to the size of RB aggregates.

Photodynamic therapy is a treatment that uses a combination of light-absorbing photosensitizers and dissolved oxygen to kill cancer. One specific limitation of photodynamic therapy is that the visible light used for photosensitizer excitation has a short tissue penetration depth of several millimeters. This limits the application of photodynamic therapy to surface cancers in the absence of a technique to illuminate deeper tissue. Efforts to extend tissue depth to which photodynamic therapy can be applied have been attempted with use of up-conversion and persistent-luminescent nanoparticles that absorb near infrared light and emit visible light for photosensitizer excitation, yet an initial excitation with an external light source is still required. More recently, systems employing chemiluminescence as an excitation energy source designed to bypass the use of external light have been developed and investigated as potential agents that could overcome the problem of achieving photodynamic therapy in deep tissue. We wish to provide an overview of several systems that have been recently reported that employ both radiative and non-radiative chemiluminescent energy transfer for photosensitizer excitation that have been developed in the hope of achieving “dark” photodynamic therapy. This article reviews several of these important new developments in the design of photodynamic therapeutic systems that utilize chemiluminescence.

Chitosan and its derivatives are renewable biopolymers characterized by high biocompatibility; therefore, they are harmless to humans and allow immune tolerance and improved hydrophilicity. Moreover, chitosan has been the most studied of all polysaccharides used in biomedical applications during the last decade. Combinations of chitosan and porphyrinoid compounds in hybrid materials have revealed many potential applications for biomedical sciences. The main advantage of such materials is an increase in the solubility of porphyrinoids in body fluids and therefore greater release of singlet oxygen to the treated tissue. Chitosan-based drug delivery systems can improve the targeting of porphyrinoids and their release at predetermined locations and finally achieve desired therapeutic effects with minimal side effects. Hence, porphyrinoid-chitosan materials can be applied in drug delivery systems, cancer theranostics and magnetic resonance imaging. The combination of chitosan and porphyrinoids also appears useful in the healing and repairing of damaged organs, tissue engineering, regenerative medicine, as well as dressing materials. Huge benefits are related to the treatment of wounds, which has been presented for self-healing hydrogels based on chitosan and porphyrinoids. Furthermore, the chitosan/porphyrinoid combinations have revealed enormous benefits for antimicrobial photodynamic therapy.

Colon cancer is one of the most frequent causes of death in the world. Despite the easily accessible diagnostic tools, the disease is still diagnosed at its advanced stage, when radical treatment is no longer possible and only palliative therapy can be provided. Cancer recurrence, which worsens the patient’s prognosis, is a separate problem. Currently, scientists are seeking modern, multidirectional treatment methods, which would enable the inhibition of neoplasia, elongation of patients’ lives and improvement in their quality. Immunotherapy and photodynamic therapy seem to be promising as they provide good results, also at advanced stages of cancer. Due to its cytotoxic and immunomodulating effect, photodynamic therapy exhibits an antineoplastic effect. Using monoclonal antibodies in the targeted therapy of colon cancer is a new, promising concept, which requires further studies.
Rak jelita grubego jest jedną z najczęstszych przyczyn zgonów na świecie. Pomimo łatwo dostępnych narzędzi diagnostycznych nadal często rozpoznawany jest w stadium zaawansowanym, kiedy niejednokrotnie nie ma już możliwości leczenia radykalnego i pozostaje jedynie terapia paliatywna. Odrębnym problemem jest wznowa nowotworu, pogarszająca rokowanie pacjenta. Współcześnie poszukuje się nowoczesnych, wielokierunkowych metod leczniczych, które pozwoliłyby zahamować proces nowotworzenia, wydłużyć życie chorych i poprawić jego jakość. Obiecującym kierunkiem badań okazują się immunoterapia oraz terapia fotodynamiczna, które dają pomyślne wyniki również w zaawansowanych stadiach raka. Terapia fotodynamiczna wykazuje działanie przeciwnowotworowe poprzez efekt cytotoksyczny oraz immunomodulujący. Zastosowanie przeciwciał monoklonalnych w celowanej terapii raka jelita grubego jest nową, dobrze rokującą koncepcją wymagającą dalszych badań.

Nevoid basal cell carcinoma syndrome (NBCCS) is a rare genetic disease that is manifested in a number of disorders concerning the skin, skeleton, cardiovascular and nervous systems. Various defects which can be observed at first contact with a patient visiting a dermatologist or dentist may help to diagnose this syndrome. Frequent problems with odontogenic cysts and metastatic basal-cell carcinomas result in patients being under the constant care of a specialist. This short study presents the case of 31-year-old patient with Gorlin-Goltz syndrome treated with combination therapy using CO2 laser and photodynamic therapy.

Opis:
Human prostate cancer cells were evaluated for growth after photodynamic therapy, radiotherapy, and combined treatment. Indocyanine green was tested as a photosensitizer and radiosensitizer. Two human cell lines were used: PC-3 derived from prostate carcinoma, and EPN derived from normal prostate tissue. The light source used for the photoactivation experiments was a diode laser peaked at 805 nm. The light dose incident on cells was 108 J/cm2. Ionizing radiation was produced by a linear accelerator, and the dose was 2, 4 and 6 Gy. Cytotoxicity was evaluated by measuring the colony forming ability of cells. Our results show that indocyanine green induces cell death by photoactivation, but it does not act as a radiosensitizer if used with ionizing radiation. The combined treatment of photodynamic therapy and radiotherapy produces an additive effect which does not depend on the sequence of the two treatments. Combined treatments could be more useful since they allow the reduction of the ionizing radiation dose to obtain the same effect as one obtainable by radiotherapy alone.

Ovarian cancer is characterized by an extremely poor prognosis in women with an advanced stage of this carcinoma. In United States every year 20,000 women are diagnosed with ovarian cancer. It is the fifth most frequent cause of death from cancer in females and the most common cause of death for 15 years after diagnosis in women with stage III-IV tumors. Among ovarian neoplasms the most frequent are surface epithelial-stromal tumors. Moreover, non-epithelial ovarian tumors are distinguished. This group matters 10% of patients with diagnosed ovarian tumors. Due to a huge variety of ovarian tumors, the following work focuses mainly on the current clinical treatment of the epithelial ovarian cancer. The following paper summarizes current clinical treatment of epithelial ovarian cancer, including surgical procedure, chemo- and immunotherapy including modern therapeutic approaches.

Opis:
Malignant melanoma is a highly invasive cancer which derives from neuroectodermic melanocytic cells. It is characterized by the highest frequency of cases in both sexes and all age groups. High metastatic potential of melanoma and its multidrug resistance might be a significant problem in the treatment. Permanent and complete remissions are rare, while the 5-year survival rate remains relatively low. In the last years there has been a breakthrough in the systemic therapy of malignant melanoma. Modern drugs have revolutionized previous therapeutic management of melanoma. As part of therapeutic programs, new medicines are available: dabrafenib, ipilimumab, pembrolizumab, trametinib and vemurafenib. The following paper presents current clinical management standards for melanoma diagnoses, based on the widely accepted scientific data and expert experience - with emphasis on modern methods of treatment of this cancer.

Mitochondria are cell energetic centers where ATP is produced. They also play a very important role in the PDT as intracellular sites of photosensitizer localization. Photosensitizers gathering in mitochondria (like porphyrin derivatives used in this work) are more effective in tumor cell destruction. Moreover, it was assumed that di-amino acid substituents attached to porphyrin ring will strengthen the effectivity of interaction with membrane receptors of examined cells. MTT assay was performed to investigate the influence of PP(Arg)2 and PP(Ala)2(Arg)2-based PDT on breast cancer cell viability for 24 h, 48 h and 120 h after cell irradiation. Then the influence of PP(Ala)2(Arg)2- and PP(Arg)2-mediated PDT on early mitochondrial apoptosis induction via measurements of the transmembrane mitochondrial potential changes was examined. Results showed that lower energy doses and maximal nontoxic photosensitizer doses of PP(Ala)2(Arg)2 and PP(Arg)2 applied in PDT can imply apoptotic cell death. It was confirmed that modification of the protoporphyrin IX by attaching two alanine substituents raised the efficiency of photodynamic therapy.

Opis:
Photodynamic therapy (PDT) is a clinically approved and rapidly developing cancer treatment regimen. It is a minimally invasive two-stage procedure that requires administration of a photosensitizing agent followed by illumination of the tumor with visible light usually generated by laser sources. A third component of PDT is molecular oxygen which is required for the most effective antitumor effects. In the presence of the latter, light of an appropriate wavelength excites the photosensitizer thereby producing cytotoxic intermediates that damage cellular structures. PDT has been approved in many countries for the treatment of lung, esophageal, bladder, skin and head and neck cancers. The antitumor effects of this treatment result from the combination of direct tumor cell photodamage, destruction of tumor vasculature and activation of an immune response. The mechanisms of the direct photodamage of tumor cells, the signaling pathways that lead to apoptosis or survival of sublethaly damaged cells, and potential novel strategies of improving the antitumor efficacy of PDT are discussed.

Introduction and aim. Photodynamic therapy is a complex process involving the introduction of photosensitizers into the patient’s body and irradiation of them in order to destroy the lesion, and activate the immune system. An important role in photodynamic therapy is played by photobiochemical and physical mechanisms that affect the tumor vessels and lead to the death of the damaged cell. The aim of the study is to determine the effect of photodynamic therapy with the use of Hypericin (Hyp) on the secretion of selected cytokines by colorectal cancer cells. Material and methods. Two colorectal cancer cell lines SW480 and SW620 were used in the study. Cells treated Hypericin were exposed to visible light. Then cell viability was determined by the MTT assay with 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium. Assays were performed for control samples without hypericin and light exposure, with Hyp without light exposure, without Hyp and irradiated with light, and test samples with Hyp and light exposure. Results. In the experiment we reveal, that Hyp- photodynamic activity does not influence the secretion of cytokines. Conclusion. The obtaining results confirming the destructive effect of Hyp- PDT on the colon cancer cells, show a possibility of extending the indication for photodynamic therapy using Hyp, qualification of precancerous changes.
