Opis:: Bone marrow transplantation dates back to the ‘50s, but greatest progress in this therapeutic modality applied successfully in the treatment of several conditions took place mostly during the past 25 years. Among all organ transplantations, bone marrow transplants are second only to kidney transplants. In Poland this therapeutic technique also undergoes a rapid development. During the past 10 years, absolute numbers of various forms of transplantation increased 100-fold, starting from 6 procedures in 1989 to over 700 at present. Essentially, the term bone marrow transplantation (BMT), present in names of international associations and registers, is in fact a colloquialism (e.g. European Group for Blood and Marrow Transplantation). A much more appropriate term would be hematopoietic cell transplantation (HCT). This is because progenitor cells for transplantation may be obtained not only from bone marrow, but also from peripheral blood and umbilical cord blood. The term hematopoietic cell transplantation has a much broader meaning and includes: classic transplantation of bone marrow obtained at surgery, transplantation of peripheral blood stem cells (PBSCT) and umbilical cord blood-derived stem cells. All hematopoietic stem cells express on their surface the CD34+ antigen and glycosylated transmembrane protein, a member of the family of adhesion molecules. In healthy persons, expression of this antigen in bone marrow cells is at the level of 1-3%, while in peripheral blood – 0.01-0.1%, and in umbilical cord blood – 0.1-0.4%. The first source (transplantation material) of hematopoietic progenitor cells was bone marrow, while transplantation of stem cells obtained from peripheral blood and umbilical cord blood started much later (during the ‘80s of the past century).

Opis:: This article is about issues of application which initiate proceedings on the subject of consent of the guardianship court of draw marrow or haematopoietic cells peripheral blood from juvenile. First of all the author described general remarks about proceedings on the subject of draw marrow or haematopoietic cells peripheral blood from juvenile. Afterwards the author described formal requirements of application with reference to formal requirements of procedural document in general and formal requirements of summons. Further considerations affect return and withdraw of application.
W artykule została omówiona problematyka wniosku wszczynającego postępowanie w przedmiocie wyrażenia zgody przez sąd opiekuńczy na pobranie szpiku lub komórek krwiotwórczych krwi obwodowej od małoletniego. W pierwszej kolejności zostały przedstawione uwagi ogólne dotyczące postępowania w przedmiocie pobrania od małoletniego szpiku lub komórek krwi. Następnie przybliżono zagadnienia dotyczące warunków formalnych tego wniosku w odniesieniu do warunków formalnych pism procesowych w ogólności oraz warunków formalnych pozwu. Dalsze rozważania dotyczą zwrotu wniosku oraz jego cofnięcia.

Opis:: Nowadays, increasing attention has been focused on relation between increased D-dimer levels and cancer among patients without detectable thrombosis. The aim of the study was to measure plasma D-dimer levels in portal and peripheral blood in pancreatic cancer patients with absence of venous thromboembolism. Material and methods. Fifteen consecutive patients hospitalized in the Department of General and Transplant Surgery of Medical University in Łódź, from January to March 2012 who underwent surgery due to a pancreatic cancer were enrolled. At laparotomy, portal and peripheral blood were sampled concurrently. D-dimer and fibrinogen levels were measured. Moreover, to investigate overall coagulation function prothrombin time (PT), prothrombin index (PI), international normalized ratio (INR), thrombin time (TT), activated partial thromboplastin time (APTT), TT and APTT index were evaluated. Results. Peripheral plasma D-dimmer levels above normal range were found in 10/15 patients (66,67%), whereas D-dimer above normal values were confirmed in all portal blood samples. Mean D-dimer values were higher in portal than in peripheral blood (3279.37 vs 824.64, by 297%, p=0,025). These discrepancies were accompanied by normal limits of portal and peripheral levels of fibrinogen and comparable coagulation function indexes. Conclusion. Our preliminary study showed the close relation between activation of hemostasis, reflected by elevated D-dimers in portal blood and presence of pancreatic cancer. These data suggest that measurement of portal blood D-dimer levels may be a potentially useful technique for screening the pancreatic cancer.

Opis:: Human peripheral blood cells stimulated by phytohemagglutinin (which serve as a model of cancerous cells) and resting cells were incubated in dimethyl sulfoxide solutions of various phthalocyanines. In order to diminish the influence of atmospheric oxygen the cells were embedded in a polymer (polyvinyl alcohol) film. Fluorescence spectra of the samples were measured over two regions of excitation wavelengths: at 405 nm (predominant absorption of the cell material) and in the regions of strong absorption of phthalocyanines (at about 605 nm and 337 nm). The intrinsic emission of cell material became changed as a result both of cells' stimulation and of incubation of cells in dye solution. In most cases the stimulated cells when stained by dye exhibited higher long wavelength fluorescence intensity than resting cells. This suggests higher efficiency of dye incorporation into cancerous cells than into healthy cells. The absorption spectra of samples were also measured. The spectra of various phthalocyanines in incubation solvent, in polymer and in the cells embedded in polymer, were compared. The comparison of properties of the cells stimulated for different time periods enabled to establish the conditions of stimulation creating a population of cells incorporating a large number of sensitizing molecules.

Opis:: Sildenafil is a selective type 5 phosphodiesterase (PDE5) inhibitor, commonly used in the treatment of erectile dysfunction (ED) and pulmonary arterial hypertension (PAH). The results of recent studies suggest that this drug modulates function of the immune system. However, only few studies were performed in humans. Therefore, the aim of this study was to investigate the ability of sildenafil to regulate the production of pro-inflammatory cytokines including tumor necrosis factor alpha (TNF-α), interleukin (IL)-1β and IL-6 in peripheral blood mononuclear cells (PBMCs) from healthy men. Phorbol myristate acetate (PMA) - stimulated and unstimulated PBMCs from 16 participants were cultured in the absence or presence of sildenafil (400 ng/ml and 4 µg/ml). TNF-α, IL-1β and IL-6 concentrations in culture supernatants were measured by enzyme-linked immunosorbent assay (ELISA). Cell viability was assessed by trypan blue staining. Sildenafil at a concentration of 400 ng/ml significantly increased TNF-α production in stimulated PBMCs (p<0.05) but had no effects on IL-1β and IL-6. The drug did not affect PBMCs viability. This is the first report describing such effects of sildenafil in humans.

Opis:: Exposure to phenol causes ultrastructural disorders in supporting and receptor cells in the peripheral segment of the olfactory organ of yellowfin (Cottocomephorus grewingkii) and in cells in peripheral blood of stone sculpins (Paracottus knerii), yellowfin and perch (Perca fluviatilis). Identical responses to phenol exposure in the blood cells were observed in endemic Lake Baikal fish and in nonendemic fish. Differences were found only in the rate and intensity of the recovery response. Fast activation of immune processes were found in the coastal nonendemic perch, but they were slow in the stone sculpin, a coastal endemic species.

Opis:: This article concerns the ruling rendered in one of the civil proceedings which are regulated in Act of 1 July 2005 on the cell, tissue and organ recovery, storage and transplantation, i.e. in the proceeding on the subject of giving consent of the guardianship court to collect bone marrow or peripheral blood haematopoietic cells from the juvenile. First of all, the author describes the issues pertaining to the content of the ruling, its time and place, as well as its form and legal nature. Next, the author presents the characteristics and results of the ruling, including: legitimacy, inalterability, res iudicata, effectiveness and enforceability. Moreover, the author touches upon a very important issue of justification of ruling.
Artykuł dotyczy problematyki orzeczenia wydanego w jednym z postępowań cywilnych uregulowanych w ustawie z dnia 1 lipca 2005 r. o pobieraniu, przechowywaniu i przeszczepianiu komórek, tkanek i narządów, a mianowicie w postępowaniu w przedmiocie wyrażenia zgody przez sąd opiekuńczy na pobranie szpiku lub komórek krwiotwórczych krwi obwodowej od małoletniego. W pierwszej kolejności zostały omówione zagadnienia dotyczące treści orzeczenia, czasu i miejsca orzekania, postaci orzeczenia oraz jego charakteru prawnego. Dalsze rozważania dotyczą cech i skutków orzeczenia, w tym: prawomocności, niezmienności, powagi rzeczy osądzonej, skuteczności i wykonalności. Autorka porusza ponadto istotną problematykę uzasadnienia postanowienia i doręczenia tego uzasadnienia.

Opis:: The degree of activation of cells involved in cellular immune re-sponse against tumor antigens (cytotoxic lymphocytes Te) as well as efficiency of the mechanisms which promote immunosuppression (Treg — regulatory cells CD4±CD25±Foxp3±) may determine the course of the neoplastic disease. The aim of this study was to assess the function of autologous peripheral blood mononuclear cells (PBMCs) involved in the immunological processes on the basis of expression of Foxp3 and RORgamma t molecules as well as analysis of the relationships with clinical and morphological features of the tumor (pT and pN stage, G feature, degree of invasiveness according to the TFG classification) in laryngeal carcinoma. Materia! and methods: The analysis included a group of 59 patients with verified squamous celt carcinoma of the larynx. In the pathologic evaluation pTNM classification criteria, depth of invasion and degree of histological dif-ferentiation were used. Expression levels of mRNA for Foxp3 and RORgamma t in peripheral blood mononuclear cells by quantitative analysis of the amplified product in real time (real-time RT2-PCR) were evaluated. The level of Foxp3 and RORgamma t protein expression by Western blot analysis was determined. Results: In squamous celt carcinomas of the larynx, with the highest tumor aggressiveness the significantly highest level of mRNA and protein expres-sion for Foxp3 molecule were observed. The severity of Foxp3 expression at both gene and protein level were positively linearly correlated with the degree of local extent of the tumor (pT3-4), depth of invasion (invasion of cartilage) and the degree of histological differentiation (low-differentiated tumors G3). In the study group of laryngeal cancers significantly lower level of RORgamma t expression in carcinomas with less invasive changes (pT1-2, high-differentiated tumors G1, carcinomas with microinvasion without evi-dence of invasion beyond the lamina propria) was also noted. Conclusions: The study results indicate the important role of immune celt activity as indicators of advancement of clinical and morphological changes in squamous celt carcinoma of the larynx.

Opis:: Celem pracy było określenie indukcji mikrojąder w retikulocytach i erytrocytach polichromatycznych myszy pod wpływem subchronicznej ekspozycji na promieniowanie X, nonylofenol (NF) lub na skojarzone działanie obu czynników. Myszy Pzh: SFIS narażane były przez 8 tygodni, 5 razy w tygodniu. Dawki wynosiły 0.05 Gy lub 0,10 Gy promieniowania X, 25 mg/kg mc lub 50 mg/kg mc nonylofenolu, a w przypadku skojarzonego działania 0,05 Gy + 25 mg/kg mc NF lub 0,10 Gy + 50 mg/kg mc NF. Zarówno promieniowania X, jak i NF działając oddzielnie indukowały powstawanie mikrojąder w retikulocytach i erytrocytach polichromatycznych myszy. Skojarzone działanie obu czynników w retikulocytach krwi obwodowej oraz w erytrocytach polichromatycznych szpiku kostnego indukowało zwiększoną częstość występowania mikrojąder w porównaniu do efektów działania każdego z czynników oddzielnie. W retikulocytach szpiku kostnego skojarzone działanie obu czynników w małych dawkach (0,05 Gy + 50 mg/kg mc NF) powodowało zwiększenie efektu mutagennego. Natomiast, w przypadku skojarzenia obu czynników w większych dawkach (0,10 Gy + 50 mg/kg mc NF), nonylofenol może wywierać efekt ochronny w stosunku do DNA retikulocytów powodując zmniejszenie uszkodzeń indukowanych przez promieniowanie X.
The aim of the research was to investigate the level of micronuclei induction in reticulocytes and polychromatic erythrocytes of mice following subchronic exposure to X-rays, nonylphenol (NP) or to a combination of both. Male mice Pzh:SFIS were exposed during 8 weeks, 5 day per week to doses 0.05 Gy and 0.10 Gy of X-rays, 25 mg/kg mc and 50 mg/kg bw of nonylphenol as well as to 0.05 Gy + 25 mg/kg bw NP and 0.10 Gy + 50 mg/kg bw NP for combined exposure. Both X-rays and NP, acting alone induced micronuclei in reticulocytes and in polychromatic erythrocytes of mice. Combined X-rays-NP exposure of peripheral blood reticulocytes and bone marrow polychromatic erythrocytes caused enhanced frequency of micronuclei compared to the effect of each agent alone. In bone marrow reticulocytes, combined exposure in lower doses (0.05 Gy + 25 mg/kg bw NP) induced enhanced mutagenic effect. Contrary, after combined exposure to both agents in higher doses (0.10 Gy + 50 mg/kg bw NP), nonylphenol may protected DNA of reticulocytes against damage induced by X-rays.
