    Opis:
    A proof of concept for using paper test as a suitable method in the production of monoclonal antibodies (MAbs) is reported. The paper test which detects antibodies against porcine circovirus type 2 (PCV2) using colloidal gold-labelled capsid protein as the antigen probe was applied exclusively in the screening of anti-PCV2 MAbs. It allowed the detection of 118 single cell clones within 30 min using naked eyes. MAbs with specific binding to authentic epitopes on the virus were selected using a blocking strategy in which the antibody was pre-incubated with PCV2 viral sample before applying to the test paper. Five hybridomas secreting MAbs against the capsid protein were obtained, with only three of them capable of binding to PCV2. The results were validated and confirmed using enzyme-linked immunosorbent assay and immunofluorescence assay. The paper test is simple, rapid, and independent on professional technicians and proves to be an excellent approach for the screening of MAbs against specific targets.

    The wastewater generated from pulp and paper production still contains pollutants and contaminants, so appropriate and economical materials are needed to reduce pollutants by using adsorbents from bottom ash. This study examined the characteristics of bottom ash and pulp and paper wastewater, the effect of stirring time, stirring speed, and adsorbent activation temperature on decreasing the environmental parameters of wastewater. The synthesis of bottom ash as an adsorbent was carried out by heating at 100 and 200 °C for 1 hour. The wastewater treatment process with maximum yield occurs at a stirring speed of 100 rpm for 50 min. The use of bottom ash without heating as an adsorbent in the pulp and paper wastewater treatment process has reduced TSS 77.5%, COD 85.72%, chloride 26.9%, TDS 1143 ppm, and EC 2180 s/cm, which have met the environmental quality standards.

    The study focuses on the process of assessing (a micro-skill) goal attainment in EFL pronunciation course via measures which can foster different students’ attitudes and self-perceptions. Standard (pen-and-paper) tests offer immediate evidence of success but they put heavy demands on students’ cognitive, performance and stress-controlling skills. CALL-related techniques can be used as supplementary ones, even if technically assessing different sub-skills, Kahoot or Moodle quizzes can complement and re-orientate the assessment as well as the learning processes. To investigate the impact of the diverse assessment measures a small-scale research was conducted among Year 1 students of the English Department at the Pedagogical University in Kraków. They are participants in a 90-hours-a-year pronunciation course, where one of the components involves mastering transcribing skills. The specific element of the course evaluated by standard and CALL-related measures in the study were the phonetic variants of the -es and -ed endings in English. Through analysis of test scores, coupled with the ideas obtained via semi-structured interviews, the study hoped to verify the claim that matters such as student comfort, instant individual feedback and personal safety are most efficiently handled by the Moodle quizzes. Apart from providing well-balanced scores, they offer the least-threatening, stress-free environments for learning and assessment, thus developing students’ self-monitoring their progress.

    W artykule przedstawiono zagadnienia dotyczące identyfikacji własności dynamicznych konstrukcji wsporczej części prasowej maszyny papierniczej poddanej w warunkach eksploatacyjnych działaniu wielu poliharmonicznych wymuszeń dynamicznych. W pierwszej części artykułu omówiono okoliczności wskazujące na możliwość występowania wzmocnień rezonansowych odpowiedzi drganiowych na działające wymuszenia dynamiczne oraz przeanalizowano potrzeby i ograniczenia związane z realizacją identyfikacji własności dynamicznych konstrukcji. Ponadto wykazano, że wiarygodna estymacja własności dynamicznych omawianej konstrukcji wsporczej wymaga zastosowania testu dynamicznego z wykorzystaniem wymuszeń działających podczas normalnej eksploatacji maszyny papierniczej. Wyniki rozważań przeprowadzonych w pierwszej części artykułu były podstawą realizacji badań własności dynamicznych konstrukcji wsporczej części prasowej maszyny papierniczej w warunkach eksploatacyjnych. Analiza wyników badań własności dynamicznych tej konstrukcji została przedstawiona w drugiej części artykułu. Dla uzyskanych przebiegów odpowiedzi drganiowych wyznaczono charakterystyki teoretyczne i dopasowano je do tych przebiegów uzyskując w ten sposób parametry dynamiczne badanej konstrukcji. Wyniki badań własności dynamicznych porównano następnie z wynikami obliczeń dynamicznych charakterystyk amplitudowych w oparciu o metodę elementów skończonych i na tej podstawie podjęto decyzję o realizacji modernizacji konstrukcji.
    The paper presents identification of dynamic properties of a paper machine press part support structure subjected to many polyharmonic excitations during operation. First part of the article contains description of circumstances pointing at possibility of resonant amplification of vibration responses to the dynamic excitations and analysis of needs and limitations connected with identification of the structure dynamic properties. Moreover it was pointed out that in order to realize reliable estimation of the support structure dynamic properties one should apply dynamic test using excitations acting during standard operation of the paper machine. Results of considerations presented in first part of the article were the base to conduct the research of paper machine press part support structure dynamic properties in operation conditions. Analysis of research results of dynamic properties of this structure is presented in second part of the article. Theoretic characteristics were estimated for courses of vibration responses and matched to these courses in order to estimate dynamic parameters of the considered structure. Next, research results of structure dynamic properties and results of dynamic calculations realized on the base of finite element method were compared and decision concerning realization of the support structure modernization was made.

    Paper chromatography is a method that allows rapid assessment of the condition and identification of fuel, water and soot contaminants in engine oil. The paper contains an analysis of the suitability of rapid paper chromatography for assessing the operating quality of engine oil. It takes into account the comparison of physicochemical parameters by classical methods with the quality of commercial paper chromatography. The presented research allowed verifying the suitability of paper chromatography for laboratory evaluation of engine oil quality. It was found that a more extensive analysis of the oil condition is possible provided that the chromatographic separation time of oil dispensed onto filter paper is increased from 1 minute to 40 minutes. Linear relationships were observed between the surface area of the stain core after 40-minute separation and selected viscosity parameters (e.g. kinematic viscosity measured at 40°C). Based on rapid paper chromatography, it was found that reasonable decisions can be made about the quality of engine oil, and thus about replacement or further use of the oil.
