The influence of an antiestrogen, indole-3-carbinol (I3C) on the expression of CYP1A1, CYP1B1 and AhR genes was investigated in an attempt to establish whether I3C could increase the expression of genes involved in estrone metabolism. Another purpose was to examine the proliferation of an estrogen-dependent breast cancer cell (MCF-7 line) under the influence of I3C and both I3C and DDT. In MCF-7 cells incubated with I3C or I3C and DDT combined, quantitative RT-PCR analysis revealed a significant increase in the level of CYP1A1, AhR, and CYP1B1 transcripts. The proliferation rate of MCF-7 cells was increased by treatment with DDT or estradiol (E2), whereas I3C did not affect the proliferation of MCF-7 cells but greatly reduced the stimulatory effect of DDT, and abolished the effect of E2. The level of p21 transcript, encoding p21 protein involved in the cell cycle, was increased several-fold by I3C comparing to its level in cells incubated with estradiol or DDT. The results suggest that the proliferation of MCF-7 cells is accompanied not only by expression of genes encoding cytochromes involved in estrogen metabolism, but also by changes in the expression of other genes including that encoding p21 protein involved in the cell cycle.

The aim of this study was to analyze the molecular mechanism of inositol hexaphosphate (InsP6) action through which it may inhibit proliferation of colon cancer cells and cell cycle progression. A kinetic study of p53 and p21WAF1 mRNA increase was performed on human colon cancer HT-29 cells after treatment with 1, 5 and 10 mM InsP6 for 6, 12, 24 and 48 h. Real-time-QPCR based on TaqMan methodology was applied to analyze quantitatively the transcript levels of these genes. The transcription of β-actin and GAPDH genes was assessed in parallel to select the control gene with least variability. The 2-ΔΔCt method was used to analyze the relative changes in gene transcription. InsP6 stimulated p53 and p21WAF1 expression at the mRNA level, with the highest increase in p21WAF1 mRNA occurring at 24 h, i.e., following the highest increase in p53 mRNA observed at 12 h. Based on these studies it may be concluded that the ability of InsP6 to arrest the cell cycle may be mediated by the transcriptional up-regulation of the p53-responsive p21WAF1 gene.

Introduction and objective. E2F transcription factor 2 (E2F2) protein is the transcription factor that plays an important role in tumorigenesis. E2F2 effects the cell cycle, tumor suppressor proteins, and can also be transformed by proteins of small DNA tumor viruses. The objective of the study is to provide a summary of the current knowledge on the neoplastic pathways that involve E2F2. State of knowledge. Numerous studies have demonstrated a role for E2F2 in various signaling pathways. Certain components of these pathways may serve as potential targets for oncological therapy. E2F2 has been shown to be associated with neoplasms of various locations and histological types (breast, colon, gastric, laryngeal, liver, lung, ovarian, pancreatic, and prostate cancers). Conclusions. Further investigations of E2F2 pathways are warranted for a clearer understanding of neoplastic processes and to identify novel pharmacological treatments.

Opis:
Theory of optical properties of interacting electrons and holes in quasi-2D quantum dots in strong magnetic fields is discussed. In two dimensions and the lowest Landau level, hidden symmetries control the interaction of the interacting system with light. By confining electrons and holes into quantum dots hidden symmetries can be removed and the excitation spectrum of electrons and excitons can be observed. We discuss a theory electronic and of excitonic quantum Hall droplets at a filling factorν=2. For an excitonic quantum Hall droplet the characteristic emission spectra are predicted to be related to the total spin of electron and hole configurations. For the electronic droplet the excitation spectrum of the droplet can be mapped out by measuring the emission for increasing number of electrons.

Carrier recombination dynamics in polar and nonpolar GaN epilayers and GaN/AlGaN multiple quantum wells grown over sapphire substrates with various crystallographic orientation were studied under high photoexcitation by 20 ps laser pulses. The transient of luminescence featured a significant enhancement in nonradiative recombination of free carriers for nonpolar a-plane GaN epilayers compared to conventional c-plane samples. The epitaxial lateral overgrowth technique was demonstrated to significantly improve the quality of nonpolar a-plane films. This was proved by more than 40-fold increase in luminescence decay time (430 ps compared to ≤10 ps in the ordinary a-plane epilayer). Under high-excitation regime, a complete screening of built-in electric field by free carriers in multiple quantum wells grown on c-plane and r-plane sapphire substrates was achieved. Under such high excitation, luminescence efficiency and carrier lifetime of multiple quantum wells was shown to be determined by the substrate quality.

GaN epilayers and AlGaN/GaN multiple quantum wells grown by metalorganic chemical vapor deposition on different crystal planes (c, a, and r) of the sapphire substrate were studied by excitation intensity dependent and time-resolved photoluminescence. In polar multiple quantum wells grown on a- and c-planes, a blueshift of the luminescence band with increasing the excitation energy was observed, indicating that screening of built-in field by free carriers takes place, whereas in nonpolar r-plane grown multiple quantum wells, the luminescence band maintained an almost constant peak position. Full screening of built-in field was achieved at the excitation densities higher than 0.3 mJ/cm$\text{}^{2}$. Under conditions of screened built-in electric field the structures were characterized by carrier lifetime. It was shown that nonpolar multiple quantum wells suffer from high density of nonradiative traps that can be due to substrate related threading dislocations.

Optical properties of CdTe/ZnTe quantum dots are studied as a function of a capping layer thickness by means of time-integrated and time-resolved microphotoluminescence. The samples are grown by MBE and covered with 10 nm and 100 nm capping layer. Despite that the proximity of the surface may result in an enhanced rate of non-radiative processes limiting the quantum dots optical performance, the set of results indicates that reduction of the capping layer thickness down to 10 nm has no effect on the quantum dot emission intensity and decay rate, contrary to the previously reported case of InAs/GaAs quantum dots.

We report on photoluminescence characterization of InGaN based laser structures grown by homoepitaxial radio frequency plasma-assisted molecular beam epitaxy. Owing to Si doped barriers, the structures show a negligible impact of the built-in electric field, which was proved by excitation intensity dependent and quantum well width dependent luminescence experiments. Relatively low variation in band potential due to inhomogeneous distribution of In was quantitatively estimated from the photoluminescence temperature behavior using Monte Carlo simulation of in-plane carrier hopping and optically detected cyclotron resonance experiments. Efficient stimulated emission with a low threshold for optically pumped laser structures was observed.

Time-resolved correlation measurements were performed to examine the statistics of photon emission from a single quantum dot at high excitation levels. The range of strong powers was determined by the saturation of intensity of excitonic lines. The significant contribution of pulsed background luminescence forced us to sophisticated analysis of the photoluminescence spectra. For quantum dots grown in the Stranski-Krastanov mode, one-photon emission was found to a good approximation. Fluctuation type quantum dots exhibit a significant level of multiphoton emission.

We report on optical orientation of excitons and trions (singly charged exciton) in individual charge-tunable self-assembled InAs/GaAs quantum dots. When the number of electrons varies from 0 to 2, the trion photoluminescence under quasi-resonant excitation gets progressively polarized from zero to ≈100%. We discuss this behavior as the efficient quenching of exciton spin quantum beats in anisotropic quantum dots due to the trion formation. This result indicates a long hole-spin relaxation time larger than the radiative lifetime, confirmed by time-resolved photoluminescence measurements carried out on a quantum dots ensemble.

We describe the design, construction, and use of fiber-coupled terahertz antennas for performing ultrafast coherent THz spectroscopy directly in the cryogenic bore of high-field magnets. With an aim towards measuring the high-frequency (100 GHz to 2000 GHz) complex conductivity of correlated electron materials in the regime of low temperatures and high magnetic fields, these miniature THz emitters and receivers are demonstrated to work down to 1.5 K and up to 18 T, for eventual use in higher-field magnets. Results from a variety of semiconducting and superconducting samples are presented. This paper also describes a separate effort towards achieving coupling between colloidal semiconductor nanocrystal quantum dots, wherein we realize and study inter-dot communication via resonant (Förster) energy transfer. We present studies of the dynamics of resonant energy transfer in monodisperse and energy gradient (layered) assemblies of CdSe nanocrystal quantum dots. Time- and spectrally-resolved photoluminescence data directly reveal the energy-dependent transfer rate of excitons from smaller to larger dots. Results from layered nanocrystal quantum dot assemblies demonstrate unidirectional energy flows, a first step towards artificial light-harvesting structures. Lastly, time-resolved studies at millikelvin temperatures elucidate the nature of ground-state"dark" excitons in these quantum dots.
