During the last few years a growing amount of data has accumulated showing phospholipid participation in nuclear signal transduction. Very recent data strongly support the hypothesis that signal transduction in the nucleus is autonomic. Local production of inositol polyphosphates, beginning with the activation of phospholipase C is required for their specific function in the nucleus. Enzymes which modify polyphosphoinositols may control gene expression. Much less information is available about the role of other lipids in nuclear signal transduction. The aim of this minireview is to stress what is currently known about nuclear lipids with respect to nuclear signal transduction.

A body of evidence, linking inositide-specific phospholipase C (PI-PLC) to the nucleus, is quite extensive. The main isoform in the nucleus is PI-PLCβ1, whose activity is up-regulated in response to insulin-like growth factor-1 (IGF-1) or insulin stimulation. Whilst at the plasma membrane this PI-PLC is activated and regulated by Gαq/α11 and Gβg subunits, there is yet no evidence that qα/α11 is present within the nuclear compartment, neither GTP-γ-S nor AlF4 can stimulate PI-PLCβ1 activity in isolated nuclei. Here we review the evidence that upon occupancy of type 1 IGF receptor there is translocation to the nucleus of phosphorylated mitogen-activated protein kinase (MAPK) which phosphorylates nuclear PI-PLCβ1 and triggers its signalling, hinting at a separate pathway of regulation depending on the subcellular location of PI-PLCβ1. The difference in the regulation of the activity of PI-PLCβ1mirrors the evidence that nuclear and cytoplasmatic inositides can differ markedly in their signalling capability. Indeed, we do know that agonists which affect nuclear inositol lipid cycle at the nucleus do not stimulate the one at the plasma membrane.

Degenerative disc disease (DDD) in the lumbosacral spine is one of the most common causes of pain and the significant associated limitations in physical activity and daily functioning, with the vast majority of patients requiring long-term physiotherapy. Hence, the significance of proper diagnostics, locating the cause of the ailment, implementation of appropriate therapy and prevention. The aim of the study was to investigate the efficacy of outpatient physiotherapy on reducing pain and improving the function of the lumbosacral spine. The research group comprised 95 people (50 women and 45 men) with an average age of 53 years, all patients with DDD in the lumbosacral spine. They underwent 3 physical treatments: magnetotherapy, laser therapy, and systemic cryotherapy, as well as gymnastic exercises, aimed at improving physical fitness, and strengthening the muscular corset. The research methods included the Schober test, the Thomayer test (finger-ground test), the Visual Analogue Scale scale, Laitinen's pain questionnaire, and calculation of BMI. Physiotherapeutic treatments significantly reduced the patients' pain symptoms, significantly increased the range of motion in the lumbosacral spine and improved physical fitness. Better results of the therapy were observed in patients with lower BMI.

Previously we have shown that aldolase (ALD; EC 4.1.2.13) is present in cardiomyocyte nuclei. Now, we focused our attention on ALD localization in smooth muscle cells. Immunocytochemical methods were used to study the subcellular localization of ALD. Aldolase was localized in the cytoplasm as well as in the nuclei. Within the nuclei ALD was located in the heterochromatin region. Native polyacrylamide gel electrophoresis followed by aldolase activity staining in gel was used to study the ALD isoenzyme pattern in porcine smooth muscle cells. Two ALD isoenzymes, A and C, were found in these cells but in the nuclei only the muscle isoenzyme was detected. To support the nuclear localization of ALD, measurement of aldolase activity in the smooth muscle cell nuclei isolated from porcine stomach was performed. The ALD activity in the isolated nuclei was detectable only after preincubation of the nuclear fraction with Triton X-100 and high concentration of KCl.

Opis:
The purpose of this article is to outline Portuguese syllable structure. The shape of the syllable in European Portuguese (the standard southern and central dialect) is suggested to be identical in slow and fast speech (Mateus – d’Andrade 2002). This view is challenged in this article by an analysis of Vowel Deletion and Vowel Nasalisation. It is argued that within the skeletal theory of the syllable, it is impossible to maintain the same syllabic structure for slow speech and fast speech.

People are exposed to heavy metals both in an occupational and natural environment. The most pronounced effects of heavy metals result from their interaction with cellular genetic material packed in form of chromatin. Heavy metals influence chromatin, mimicking and substituting natural microelements in various processes taking place in the cell, or interacting chemically with nuclear components: nucleic acids, proteins and lipids. This paper is a review of current knowledge on the effects of heavy metals on chromatin, exerted at the level of various nuclear components.

Opis:
The Jacobi and Poincaré shape transitions are very promising way to investigate the shape of the nucleus. The presented here quasi-phenomenological approach allows to estimate the experimental conditions which are necessary to observe these phenomena. The static energy minimum gives the spin ranges and the fissility of atomic nuclei soft for the shape transitions and available experimentally. Dynamical effects taken into account through the solving collective Hamiltonian for zero-point vibration estimation, changes the spin rages for the shape transitions. The static deformation of the nucleus constrained by the minimum of energy for given spin has been enhanced to dynamical nuclear shapes permitted by the zero point energy. The large amplitude vibrations around the static deformation gives the ensemble of nuclear shapes possible to be observed.

We found that in starfish oocytes two different enzymes, phosphatidylserine synthase-1 (PSS1) and -2 (PSS2), which synthesize phosphatidylserine by a base-exchange reaction, are present. We studied phosphatidylserine synthesis in immature oocytes which still contain the nucleus (germinal vesicles) and in mature cells, in which the re-initiation of the meiotic cycle induced by the hormone 1-methyladenine led to structural changes in the endoplasmic reticulum, to the disappearance of the nuclear envelope and to the intermixing of the nucleoplasm with the cytoplasm. It was found that the levels of PSS1 and PSS2 transcripts were higher in immature and mature oocytes, respectively. The level of the expressed PSS2 protein, higher than that of PSS1, was not influenced by the maturation process, whereas the level of PSS1 protein was higher in immature than in mature oocytes. Serine incorporation into phosphatidylserine was enhanced in immature oocytes. The depletion of calcium stores by thapsigargin resulted in 50% lowering of phosphatidylserine synthesis. We suggest that changes in phosphatidylserine synthesis may be affected by the release of calcium stored in the nuclear envelope and in the endoplasmic reticulum, the membranes that undergo disintegration and fragmentation during meiosis. The reason for the greater synthesis of PS may be the higher level of expression of PSS1 in immature oocytes.

The DNA content after Feulgen reaction in the guard cells and epidermis of Omithogalum umbellatum ovary was cytophotometrically measured in different phases of flower development. Only in bud of flowers guard cells DNA content was 2C while in full blown flowers it was higher, between 2C-4C. This observation was supported by autoradiographic studies with 3H-thymidine which was incorporated into guard cell nuclei in the ovary epidermis of newly developed flowers. Thus DNA level in O. umbellatum guard cells was higher than those in other plants described in literature. On the other hand, DNA content in the epidermis cells increased gradually with ovary growth reaching the maximum level of 8C in some cells.

The pyruvate kinase isoenzyme M2 originating from the nucleoplasm and cytoplasm of tumor cells, with its highest affinity to the 2-phosphoenolpyruvate (2-PEP) and sensitivity to L-cysteine, contributes to an increased generation of energy as ATP, necessary for tumor cell proliferation. In the presence of L-cysteine, the isoenzyme M2 PK demonstrates the activity of histone kinase, transferring the phosphoryl group from 2-PEP to the ε-amine residue of the H1 histone lysine. Oligochitosans induce expression of the inducible nitric oxide synthase gene (iNOS), what results in an increased synthesis of nitric oxide, which reacts with L-cysteine and produces L-S-nitrosocysteine. Lack of L-cysteine contributes to inhibition of kinase activity of the H1 histone, an M2 PK isoenzyme. Decreased phosphorylation of the H1 histone contributes to inhibition of EAT cell proliferation. No effect on proliferation of normal cells that include the PK M1 isoenzyme has been observed in the presence of oligochitosans.

W artykule scharakteryzowano sposoby leczenia dysfunkcji i urazów kręgosłupa oraz przedstawiono różne elementy konstrukcyjne stosowane w chirurgii kręgosłupa. Z uwagi na bardziej znaczący aspekt społeczny większą uwagę zwrócono na techniki leczenia inwazyjnego wykorzystujące stabilizatory i implanty krążka międzykręgowego. Przedstawiono różne rozwiązania stabilizatorów płytkowych i prętowych. W zakresie implantów międzytrzonowych omówiono rozwiązania konstrukcyjne dotyczące jądra miażdżystego i całego krążka międzykręgowego. Przegląd konstrukcji stabilizatorów i implantów wskazuje na tendencje rozwojowe idące w kierunku przywrócenia ruchomości implantowanego odcinka kręgosłupa. Rozwiązaniami spełniającymi powyższe oczekiwania są mobilne implanty krążka międzykręgowego.
The methods of treating spinal dysfunctions and injuries were characterized and various components used in spinal surgery were presented in this article. More attention was paid to invasive treatment techniques using intervertebral disk stabilizers and implants due to their greater social significance. Various solutions of plate and rod stabilizers were presented. In the scope of interbody implants, design solutions pertaining to the nucleus pulposus and the entire intervertebral disk were discussed. The survey of stabilizer and implant designs indicates developmental tendencies in the direction of restoring mobility of the implanted segment of the spine. Mobile implants of the intervertebral disk are solutions fulfilling the above expectations.
