Opis:
Nitric monoxide (NO) exerts a great variety of physiological functions. L-Arginine supplies amino groups which are transformed to NO in various NO-synthase-active isoenzyme complexes. NO-synthesis is stimulated under various conditions increasing the tissue of stable NO-metabolites. The major oxidation product found is nitrite. Elevated nitrite levels were reported to exist in a variety of diseases including HIV, reperfusion injury and hypovolemic shock. Denitrifying bacteria such as Paracoccus denitrificans have a membrane bound set of cytochromes (cyt cd1, cyt bc) which were shown to be involved in nitrite reduction activities. Mammalian mitochondria have similar cytochromes which form part of the respiratory chain. Like in bacteria quinols are used as reductants of these types of cytochromes. The observation of one-e- divergence from this redox-couple to external dioxygen made us to study whether this site of the respiratory chain may also recycle nitrite back to its bioactive form NO. Thus, the aim of the present study was therefore to confirm the existence of a reductive pathway which reestablishes the existence of the bioregulator NO from its main metabolite NO2-. Our results show that respiring mitochondria readily reduce added nitrite to NO which was made visible by nitrosylation of deoxyhemoglobin. The adduct gives characteristic triplet-ESR-signals. Using inhibitors of the respiratory chain for chemical sequestration of respiratory segments we were able to identify the site where nitrite is reduced. The results confirm the ubiquinone/cyt bc1 couple as the reductant site where nitrite is recycled. The high affinity of NO to the heme-iron of cytochrome oxidase will result in an impairment of mitochondrial energy-production. "Nitrite tolerance" of angina pectoris patients using NO-donors may be explained in that way.

Opis:
Previously, we showed that anaerobic induction of respiratory nitrate reductase (NR) activity in Bradyrhizobium sp. (Lupinus) USDA 3045 is strongly enhanced by nitrate or nitrite through de novo synthesis. Here, multiple NR-active soluble forms, ranging from 75 kDa to 190 kDa, were observed under anaerobic conditions. Electrophoretic activity band patterns differed depending on the level and the type of the N oxyanion added. The intensity of the membrane-bound NR activity band of 230 kDa changed with time along with consumption of 2 mM nitrate. It was associated with a parallel 5-fold increase and then 2-fold reduction in the amount of membrane-bound NR protein. In contrast, on 4 mM nitrate, the level of NR protein was much more stable, apparently due to slower nitrate depletion. Moreover, in cells anaerobically grown without nitrate addition, a 42-kDa derivative of NR degradation was immunodetected, which was not observed if nitrate was present in the medium. These findings suggest that the amount of the respiratory NR protein could be negatively regulated by endogenous proteases in relation to the level of nitrate available. It seems, therefore, that multiple native forms might be not different isoenzymes but immature complexes or derivatives of the enzyme protein turnover. This report adds to a modest list of bacterial enzymes apparently regulated by proteolysis, such as GS, MurAA, EnvA, GdhA, and MetA.

Opis:
Sprawdzono oddziaływanie jonów magnezowych na aktywność reduktazy azotanowej kapusty głowiastej białej, a następnie zbadano ich wpływ na zmiany poziomu azotanów i azotynów w niektórych kiszonkach warzywnych. Uzyskane wyniki badań wykazały, że jony magnezowe powodowały istotny wzrost aktywności reduktazy azotanowej, a dodatek w odpowiednich ilościach chlorku magnezu do surowca w czasie przygotowywania do kiszenia spowodował istotny spadek zarówno poziomu azotanów jak i azotynów.
The aim of this study was to examine the effect of magnesium ions on some changes in the nitrate reductase activity in the white headed cabbage as well as a practical application of the results obtained in vegetable souring. As experimental material were used: white and red headed cabbage, red beets and the same vegetable in the soured form. Vegetable souring was performed using a traditional method as well as a modified one, considiering a supplement of magnesium chloride at various levels during the process of souring. The activity of nitrate reductase in the white headed cabbage was determined by the Jaworski method. In raw and soured vegetables the level of nitrates was determined by brucine method and that of nitrites by the Griess colorimetric method. A supplement of magnesium ions to the incubation mixture containing raw white cabbage resulted in an increased activity of the studied enzyme by 17,0-34,8% and was significant in all magnesium ions concetrations used. The introduction of adequate amounts of magnesium chloride to soured vegetable production resulted also in a significant decrease in the nitrate and nitrite levels. An organoleptic evaluation of soured vegetables made by a traditional method and by using a supplement high quality, although the smell of white headed cabbage and "beet acid" has slightly however, significantly deteriorated.
