The SeqA protein of Escherichia coli is not only the main negative regulator of DNA replication initiation but also a specific transcription factor. It binds to hemimethylated GATC sequences and, with somewhat different specificity, to fully methylated GATC regions. Recently, a microarray analysis was reported, in which transcriptomes of wild-type and ΔseqA strains were compared. Although in the seqA mutant the levels of some transcripts were significantly decreased while certain transcripts were evidently more abundant relative to wild-type bacteria, no correlation between the presence of GATC motifs in promoter sequences and transcription activity was found. However, here we show that when larger DNA fragments, encompassing positions from -250 to +250 relative to the transcription start site, are analyzed, some common features of GATC distribution near the promoters activated by SeqA can be demonstrated. Nevertheless, it seems that the GATC pattern is not the only determinant of SeqA-dependence of promoter activity.

The work tends to classify the stylistic characteristics which can be found in those par- ts of Serbian folk poetry that deal with the motif of the “end of time” vision. The “end of time” is a Biblical topie about the curse and the finał judgement that will lead to the de- struction of the Earth due to the sins and lawlessness of the people living on it. The “end of time “vision consists of a longer or a shorter listing of the human lawlessness and the events that foretell the coming of the end. In this world destruction the four basie life sym- bols will disappear: the sheep and the wheat, the bee and the flower.  In order to express the apocalyptic vision of the world disaster, a specific “scenario” is madę; the “scenario” in which the apocryphal picture of paradise and heli is given. That picture enabled the folk poet to produce specific poetic images with a lot of stylistic figu- res in a finely sharpened poetry.

W pracy przedstawiono wybrane metody analizy powierzchni (ISO 25178) wskazując na rodzaj informacji, jaką można za pomocą danej metody uzyskać. Opisywane algorytmy i procedury zostały zaimplementowane w opracowanym w Wydziałowym Zakładzie Metrologii Mikro- i Nanostruktur Politechniki Wrocławskiej programie TOPOGRAF. W pracy zaprezentowano przykładowe wyniki działania zaimplementowanych algorytmów i procedur.
Progress in the scanning probe microscopy makes it become a much more common tool. In the paper the principle of operation of the scanning probe microscope is presented. It is emphasized that the measurement result is an image of the investigated surface. Two groups of parameters connected with lateral properties of the tested surface image are introduced. The first group of parameters is connected with the image autocorrelation function and two dimensional Fourier transform. The texture aspect ratio and surface autocorrelation length (Fig. 1) are given as examples of the autocorrelation parameters. The second group of parameters is related to the so called 3D motif analysis. At the first stage of this analysis the image is segmented by means of the watershed segmentations algorithm. At the second stage the statistics connected with the shape and dimensions of the segments are calculated. The segmentation algorithm as well as the parameters describing shape and dimensions of segments (Fig 2) are presented in the paper. Due to the quantum nature of the micro- and nano-world, the uncertainty of products of the micro and nanotechnology will always be greater than that of products of the conventional technology. For this reason there is strong demand for flexible techniques capable of handling this increased uncertainty. The 3D motif analysis is very well suited to meet this challenge, since it enables the extension of the basic set of the parameters describing segments. Therefore the segment parameters might be adapted to the function of the investigated nanostructures. The images presented in the paper were obtained by means of the TOPOGRAF software that is developed at the Division of Micro- and Nanostructures Metrology.

The calcium-activated neutral proteases, μ- and m-calpain, along with their inhibitor, calpastatin, have been demonstrated to mediate a variety of Ca2+-dependent processes including signal transduction, cell proliferation, cell cycle progression, differentiation, apoptosis, membrane fusion, platelet activation and skeletal muscle protein degradation. The cDNA coding for yak calpastatin was amplified and cloned by RT-PCR to investigate and characterize the nucleotide/amino-acid sequence and to predict structure and function of the calpastatin. The present study suggests that the yak calpastatin gene encodes a protein of 786 amino acids that shares 99 % sequence identity with the amino-acid sequence of cattle calpastatin, and that the yak protein is composed of an N-terminal region (domains L and XL) and four repetitive homologous C-terminal domains (d1–d4), in which several prosite motifs are present including short peptide L54–64 (EVKPKEHTEPK in domain L) and GXXE/ DXTIPPXYR (in subdomain B), where X is a variable amino acid. Our results suggest the existence of other functional sites including potential phosphorylation sites for protein kinase C, cAMP- and cGMP-dependent protein kinase, casein kinase II, as well as N-myristoylation and amidation sites that play an important role in molecular regulation of the calpain/calpastatin system. The regulation of the calpain/calpastatin system is determined by the interaction between dIV and dVI in calpains and subdomains A, B, and C in calpastatin.

The desert is a favourite setting for adventures in the Roman d’Alexandre composed by Alexandre de Paris, around 1188. The space of anxiety, of fear, of transgression, of initiation into the mysteries of life and death, it all belongs to one of the most important parts of the medieval imagination, fed by myths and tales of exotic travels, inherited from oriental and occidental Antiquity. There are many things going on in this area, such as quests, conquests, victories, defeats, friendships, love, hardships, death and treason.The motif of a desert becomes a dream carrier, expressing the protagonists will of surpassing themselves, the will which could be shared with the public to whom the text was addressed. Instead of being a simple element of oriental decoration, it expresses a certain vision of the world, as it transforms itself from a simple geographic and climatic space into a mythic Le désert dans le Roman d’Alexandre d’Alexandre de Paris (XIIe sičcle) space. Its representation, connected with the omnipresence of marvels, echoes the images of the Orient, as they were perceived by the medieval Occident. The marvels of the desert, seen as projections of desires, dreams, and occidental traumatisms and nightmares, create a complex figure of a mysterious Elsewhere – dangerous, disturbing, fascinating and irresistibly attractive.

The article explores the textual intricacy and the epistemological uncertainty projected by the 1999 novel of Peter Straub titled Mr. X. Already the title of the novel hints at secrecy, hidden identities, and cryptic messages. Indeed, the novel seems to be conceived as a cryptogram and a kind of literary “Russian doll”. This applies as much to the person of the narrator as to the construction of the narrative, its layers upon layers of secrecy and deception. This cryptic character of the novel is reinforced by the lack of closure and the ambiguity of the ending: “the fog” that shrouds everything as well as “undecipherable signs” that Ned meets along the way serve as a trope that projects the implied reader’s loss of certainty. The sense of being lost in the textual maze is compounded by the abundant instances of doubling and mirroring in the text. This is first observed in the composition of the novel as the dual-level narrative, consisting of the part narrated by Ned, and of the diary of Mr. X, the mysterious figure seen by Ned in his dreams, later revealed to be his father. The two, Ned and Mr. X, in their own peculiar ways act out the same pattern: Ned’s quest to find out the identity of his father is paralleled by his father’s (futile) attempts to confirm his own unearthly origins. However, duality is most powerfully expressed by the motif of doppelganger: Ned, the narrator, in time realizes the existence of his not-entirelyhuman brother, Robert, his “shadow self”, his “dark half”. Finally, even the novel, Mr. X, finds itself reflected in the work of fiction written by Mr. X, a story titled Blue Fire. Blue Fire, introduced at length into the narrative, serves as its specular text, mise en abyme, encapsulating its two central themes, as verbalized by the narrator: “the obsession with the ancestral house” and “the flight from and the pursuit of the Other”.

This article proposes some considerations on a particular variation of the elegiac mood represented by and manifested in lyrical farewells of departing mothers. A review of the variants commences with an analysis of a particularly important work by Tadeusz Różewicz written at the beginning of this century — Matka odchodzi. The book was, at the time, a particular reference point for the following poetic volumes in which the theme of the death of the poet’s mother was paramount and significant. The article also focuses on volumes of poems, written by poets that belonged to different generations but shared the same date of publication, i.e. the year 2009. Both clear similarities and marked differences in the actual commitment in carrying out the theme and in creating the profiles of mothers that have passed away are to be found in the works of Piotr Sommer (Dni i noce), Jan Polkowski (Cantus) and Eugeniusz Tkaczyszyn-Dycki (Piosenka o zależnościach i uzależnieniach).

In his article Wallenrod’s Garden and the Mystery of Wajdelota the author analyses the role of the motif of garden in Adam Mickiewicz’s epic poem Konrad Wallenrod. The conventionality of the use of the term ‘garden’ is emphasized by pointing to the fact that there were some changes introduced to the order of the valley near the Neman River, which were conducive to his amorous meetings with his Ladylove. The manner in which the picture of ‘the garden’ is outlined in the poem is confronted by the author with the classical and romantic gardening styles. It is stressed that the poet distances himself from the two schools, which results from, among other things, the emphasis on the beauty of the valley, a garden created by Nature herself. The motif of the garden played an important role in Konrad’s last conversation with Aldona, when the main hero of the poem tried in vain to persuade his wife to escape to Lithuania. He tempted her with a mirage of regained happiness, appealed to their mutual memories, in which this old and still existing garden, a witness of their love, occupied the central place. While analyzing the strategy of insistence used by Wallenrod in this situation, the author of the article referred to the conception of memory proposed by a German researcher, Jan Assmann. In the last part of the essay the character of the relationship between Wallenrod and Halban is considered along with the attempt to unravel the mystery hidden in the words “my son!”, which the old Lithuanian bard, the patron of Konrad’s spy mission, used in their last conversation.

Jan Jozef Szczepanski is one of the most uncommon, postwar polish writers and publicist. His wide and rich co-operation with “Tygodnik Powszechny” and the most famous narrations such as: “The Shoes” or novel “The Polish Autumn” are strictly connected with problems like: honor, heroism and dignity of a human being. He is also known as one of the greatest continuator of Joseph Conrad’s tradition and thoughts, especially with understanding the meaning of writer’s responsibility for the world. Szczepanski has created a very special attitude – a pose of contemporaneousness witness, which brings reflections about modern morality and obligates readers to confront different values and ideas. Despite his relationship with catholic “Tygodnik Powszechny”, Szczepanski always declared, that the most important and timeless ethic is not connected with any religion, but is a total amount of all human experiences. That point of view brought the writer nearer the philosophical conception of Jaques Mounier and the idea of humanistic personalization. His publicist debut in 1946 called “The Saint against the Imperium” – a reflection about Mahatma Gandhi – was an introduction to Jan Jozef Szczepanski’s thoughts about modern saintliness without religious labels. His general idea concerned ethic and moral saintliness, without any connections with ontologic position of a person. Mahatma Gandhi’s example shown that every person in modern world could be The Saint, no matter what kind of religion or culture had represented. The Saintliness in Szczepański’s reflection was an original, moral value. That value was also very important in one of his most affecting essays “The Saint”. A history of Maria Maksymilian Kolbe – a catholic priest, who had voluntarily tendered his own life for other Auschwitz’s prisoner freedom – shown that even savaged time of war could bring a shadow of hope. Szczepanski was far from popular hagiography. As he said: “Modern, lay world probably needs saintliness more than any time before. But – I suppose – less than any time before needs hagiography” Szczepanski was trying to find a key to victory of a human morality in modern world through reflections about ethical sources of mankind. The affirmation of summum bonum exists in all religious and ethical systems, but – as the writer admitted – words are sometimes too weak to express essence of modern saintliness.

In the article contemporary natal narrative is studied from the perspective of the influence of mythological and story types of texts on it. Natal narrative being similar to mythological text confirms some norms of world outlook and behavior. Natal narrative, like many texts of the story type, is about extraordinary events in a woman’s life or about an exceptional situation, which has happened to her acquaintances. The heroine of the natal narrative crosses a definite semantic border, reasserting the status of her exceptional nature.

The issue connected with sense of life appears in the works of the Apostolic Fathers not only as a main subject, but also not occasionally. They used the motif of two roads to show the sense of behavior on the earth, according to the God’s commandments and his will. They do not condemn the earthly world but deeds, which lead people to death. Even in the face of his close martyrdom, they saw a clear aim: life on the earth is important, because it leads to God. The Apostolic Fathers tell also the truth connected with eternal life and hope to be used from the dead, which will take place after death. The sense of our life is – it leads a man to the resurrection.

Opis:
The author examines the scope and sources of St Augustine’s knowledge of Manichaeism. He mostly follows the thesis of J.K. Coyle, that as a Christian and anti-Manichaean polemicist, Augustine obtained additional information about the “Religion of Light”. To illustrate Augustine-the-Manichaean and Augustine-the-polemicist’s knowledge of the religion of Mani, the author includes two lengthy excerpts from his writings: from the 7th book of Confessiones and the treatise De natura boni, which features a quotation from the Manichaean work Treasure. The author provides a special comment on this extract.
