Opis:
During 2015, samples from 22 apple trees showing proliferation symptoms were collected in southwest Bulgaria and Central and South Poland and tested for phytoplasma presence. ‘Candidatus Phytoplasma mali’ was identified in 18 samples based on results of restriction fragment length polymorphism (RFLP) analysis of the 16S rRNA gene amplified in nested PCR using primer pair P1/P7 followed by R16F2n/R16R2 and F1/B6 primer pairs. The nitroreductase and rhodonase-like genes and ribosomal protein genes rpl22 and rps3 were then analyzed using PCR-RFLP technique to study the genetic variability of the phytoplasma strains. Two restriction profiles, P-I or P-II, were obtained from fragments of 16S rDNA plus 16S-23S spacer region digested with HpaII enzyme. Restriction fragment length polymorphism analysis of nitroreductase and rhodonase-like genes using digestion with HincII endonuclease revealed that all ‘Ca. P. mali’ strains belonged to the subtype AP-15. Analysis of rpl22 and rps3 ribosomal protein genes digested with AluI enzyme resulted in classification of detected phytoplasma strains to rpX-A subgroup.

Parabronema skrjabini is a spirurid nematode of the family Habronematidae that lives in the abomasum of ruminants such as sheep and goats. The purpose of this study was to investigate the molecular aspects of Parabronema skrjabini in sheep. The worms were collected from sheep in Sanandaj (west of Iran). The first internal transcribed spacer (ITS) of ribosomal DNA (rDNA) nucleotide fragments of Parabronema skrjabini were amplified by polymerase chain reaction (PCR) using two pairs of specific primers (Para-Ir-R and Para-Ir-F). ITS1 homology in the sequence of this study was 69% compared with the sequence data in GenBank. To our knowledge, this is the first study in the world exploring the genetic diversity of P. skrjabini in sheep based on ITS1.

We report the first case of chafer beetle [Anisoplia austriaca (Herbst 1783)] mortality caused by Actinomucor elegans var. elegans in wheat fields of the Kurdistan province, Iran. For three years, dead larvae of Anisoplia austriaca were collected from wheat fields of the Kurdistan province. Similar isolates of a fast-growing fungus were recovered from all samples. The fungal isolates were identified as A. elegans var. elegans based on morphological and cultural characteristics. The identity of the species was further confirmed using sequence data of the ITS (Internal Transcribed Spacer) region of ribosomal DNA. Koch’s postulates were fulfilled by the inoculation of the larvae of A. austriaca and Galleria mellonella (Linnaeus, 1758) (as the model insect) using the spore suspension of A. elegans var. elegans. The viability of sporangiospores was evaluated using a spore dilution technique on germination medium. The results on the pathogenicity (100% mortality in A. austriaca larvae) and viability tests (germination: 95.45%) demonstrated that A. elegans var. elegans can be considered as a potential biocontrol agent against the chafer beetle. Field experiments are still required to evaluate the capacity of A. elegans as a biological control agent.

We characterized 17 insect-pathogenic entomophthoralean fungal isolates (Entomophthoromycotina: Entomophthorales) using morphological and molecular techniques. We identified four species from various insect hosts: (i) Entomophthora planchoniana, six specimens from aphids; (ii) Pandora neoaphidis, three specimens from aphids; (iii) Zoophthora phalloides from an aphid; and (iv) Z. radicans, seven specimens from insects in the orders Diptera, Hemiptera, and Lepidoptera. Analysis of ITS1 data from E. planchoniana showed clustering in accordance to aphid host species. Entomophthora planchoniana from Macrosiphum euphorbiae clustered together, separate from the isolate from Myzus persicae. The P. neoaphidis specimens clustered with sequences from other aphid-pathogenic Pandora species in GenBank. In this study, Z. phalloides from Brevicoryne brassicae and Z. radicans from an unidentified species of Chironomidae (Diptera) in Argentina were characterized for the first time. The present study was initiated to elucidate the taxonomy of the entomophthoralean fungi in Argentina according to their morphological and molecular characters. The presented results emphasize the significance of the combination of molecular data and information on morphology, ecology, and host range for accurate identification of entomophthoralean and allied genera.

Iris belongs to the Iridaceae family. It is one of the most important pharmaceutical and ornamental plants in the world. To assess the potyvirus incidence in natural resources of iris plants in Iran, Antigen Coated-Plate ELISA (ACP-ELISA) was performed on 490 symptomatic rhizomatous iris leaf samples, which detected the potyvirus in 36.7% of the samples. Genomic 3’ end of one mechanically non-transmitted potyvirus isolate, comprising a 3’ untranslated region (390 bp) and C-terminus of the coat protein (CP) gene (459 bp), was amplified by reverse transcription polymerase chain reaction (RT-PCR), which was ligated into pTG19-T vector. The nucleotide sequence of amplicons was compared with related sequences, using Blastn software available at NCBI GenBank, and showed the highest similarity with Iris severe mosaic virus (ISMV) isolates. The nucleotide and deduced amino acid sequence of the CP C-terminus region was more than 83% identical with other ISMV isolates, therefore this isolate was designated as ISMV-Ir. This new ISMV isolate is closely related to the Chinese ISMV-PHz in phylogenetic analysis, based on the partial nucleotide and deduced amino acid sequence of the CP region. This is the first report of ISMV occurrence on Iris spp. in Iran.

The Erwinia species are well-known pathogens of economic importance in Malaysia causing serious damage to high-value fruit crops that include pineapple [Ananas comosus (L.) Merr.] and papaya (Carica papaya L.).The 16S rRNA sequence using eubacteria fD1 and rP2 primers, identified two bacteria species; Dickeya zeae from pineapple heart rot, and Erwinia mallotivora from papaya dieback. Phylogenetic analysis based on the neighbor-joining method indicated that all the bacterial isolates clustered in their own taxa and formed monophyletic clades. From the pathogenicity test, all isolates of D. zeae and E. mallotivora showed pathogenic reactions on their respective host plants. Genetic variability of these isolates was assessed using repetitive sequence-based PCR (rep-PCR) fingerprinting. The results indicated interspecies, and intraspecies variation in both species’ isolates. There were more polymorphic bands shown by rep-PCR fingerprints than enterobacterial repetitive intergenic consensus (ERIC) and BOX- PCRs, however both species’ isolates produced distinguishable banding patterns. Unweighted pair-group method with arithmetic averages (UPGMA) cluster analysis indicated that all Dickeya and Erwinia isolates from the same species were grouped in the same main cluster. Similarity among the isolates ranged from 77 to 99%. Sequencing of 16S rRNA using eubacteria fD1 and rP2 primers, and rep-PCR fingerprinting revealed diversity among Dickeya and Erwinia isolates. But this method appears to be reliable for discriminating isolates from pineapple heart rot and papaya dieback.

This study presents the first molecular and serological evaluation of Echinococcus granulosus infections in wild boars in Iran. Twenty five wild boars were collected in south-western Iran, during authorized hunting program, from March to October 2013, necropsied and examined for E. granulosus infection. Furthermore, seroprevalence of cystic echinococcosis in hunted boars was evaluated by an ELISA system. A fertile hydatid cyst due to E. granulosus was detected in the lung of one of the animals. Genotype analysis of the isolate was determined by analyzing a mitochondrial gene, cytochrome C oxidase subunit 1 (co1). DNA was extracted from the cyst sample and polymerase chain reaction amplification and DNA sequencing of the specific region of the co1 gene was performed. Molecular evaluation confirmed the presence of a sheep strain, the G1 genotype, in the wild boar in south-western Iran. This is the first report of the presence of G1 genotype of E. granulosus in wild boar in Iran. Serological evaluation of hydatid cyst by antigen-B ELISA revealed E. granulosus antibodies in 5 (20%) of 25 wild boars. A statistically significant difference was observed between the prevalence of E. granulosus antibodies and gender while the difference between the seroprevalence of E. granulosus and age was insignificant. Findings of this study might have important implications for the prevention and control of cystic echinococcosis.
