Opis:
Shiga toxins belonging to a family of structurally and functionally related protein toxins serve as the key virulence factors for pathogenecity of the virulent enteric bacterial strains namely Shigella dysenteriae serotype 1 including Shiga toxin-producing Escherichia coli (STEC). S. dysenteriae type 1 isolates remain major public health concerns due to their widespread outbreaks and the severity of extra-intestinal diseases, including acute renal failure and also affects the central nervous system. Despite practicing improved hygienic conditions and regulating food and drinking water safety, the enteric pathogens are imposing a major threat in the well being of the human society. Shiga toxin on entry into host cells’ endoplasmic reticulum (ER) activates the stress response in ER and inhibits protein synthesis by catalytic inactivation of eukaryotic ribosomes. In many cell types shiga toxins trigger apoptosis. Recent studies have shown that shiga toxins induce autophagy which activates different signaling pathways in toxin-sensitive and toxin-resistant human cells. In this review the molecular basis of Shiga toxins’ effect on host cell leading to manifestation of the infection in affected individuals are discussed with an emphasis on recent findings.

Opis:
Head and neck cancers constitute the sixth most common malignant tumours worldwide, and are one of the majors problems of global public health. In the oral cavity there are several types of oral cancers, but around 90% are squamous cell carcinoma. Many different risk factors play a role in the etiology of head and neck cancer. The aim of this study is presentation from the aspect of molecular, viral and bacterial infection. Knowledge about all factors which influence the development of these malignances is essential for diagnostics and successful treatment.

Opis:
Skin cancers are one of the most common cancers in the Caucasian population. A constantly increasing number of nonmelanoma skin cancers and malignant melanomas is observed. The incidence of skin cancers is associated mainly with exposure to sunlight. Therefore, agricultural workers who work in open spaces are a particularly vulnerable group. Currently, studies on the pathogenesis of skin cancer focus on the molecular basis associated with ultraviolet radiation. This study is an attempt to summarize the current state of knowledge on this issue. There have been demonstrated mutations in different classes of genes associated with carcinogenesis, including protooncogenes, tumour suppressor genes, genes that control apoptosis, genes encoding transcription factors and DNA repair genes in patients with skin cancers. Mutations in the latter result in reducing the effectiveness of DNA repair and fixation of mutations. All changes at the gene level lead to structural changes, quantitative and dysfunction of proteins encoded by these genes. All these factors contribute to the process of carcinogenesis. Due to increasing number of skin cancers, it seems important to increase knowledge of the molecular basis of skin cancers. This knowledge could be crucial for predicting the course of the disease, and for the development of new therapeutic strategies.

Heterocyclic aromatic compounds containing various derivatives of five or six-membered nitrogen-containing rings, viz. triazoles, tetrazoles, furazans, triazines, and tetrazines, and some of their salts have been studied. High nitrogen content and a large positive heat of formation are two important properties of these compounds. Two new models are introduced for the reliable prediction of the thermal stability of these compounds through the thermal decomposition temperature (onset). The reported data for 181 compounds have been used to derive and test the new models. For a training set containing 132 heterocyclic aromatic compounds, the values of the average absolute deviation (AAD) and the coefficient of determination (R2) of the improved correlation were 9.72 K and 0.959, respectively. For triazoles, tetrazoles, furazans, triazines, and tetrazines, the predicted results of ADD and R2 for the external test data set for this model containing 41 compounds were 23.03 K and 0.664, respectively, which are closer to experimental data than those obtained by the core correlation, i.e. 26.49 K and 0.653, respectively. The correlation coefficients of cross-validation for leave-one-out (Q2LOO) and 5-fold (Q25CV) of the improved correlation were 0.955 and 0.951, respectively, which confirm that it is not an over-fitted model, robust and well-behaved.
