Nontuberculous mycobacteria (NTM) have recently emerged as important bacterial pathogens of animals and humans. Of particular concern is the high level of antimicrobial resistance displayed by these organisms, which complicates treatment and potential successful outcomes. Here, we evaluated the potential of Carlina acaulis L. as a source of novel anti-mycobacterial agents. Our goal was to measure the activity of aqueous, ethanol, and chloroform C. acaulis root extracts against 99 NTM strains. GC-MS spectroscopy analyses were performed to deliver qualitative and quantitative data on the composition of C. acaulis extract. In our study, we have shown for the first time the activity of C. acaulis extracts against NTM. The highest activity was exhibited by the chloroform extract, which inhibited the growth of more than 90% of the strains at the dose of 100 μg/mL (MIC90 = 100 μg/mL). The results of the GC-MS analysis of the C. acaulis chloroform extract contributed to the identification of 37 compounds, with carlina oxide as the most representative compound (69.52%) followed by 3,4-dihydro-2H-phenanthren- -1-one (6.54%) and stigmast-5-en-3-ol (4.14%). Our results indicate that C. acaulis chloroform and ethanol extracts have potential for treatment of NTM infections and that this plant contains anti-mycobacterial compounds.

Nontuberculous mycobacteria (NTM) have recently emerged as important bacterial pathogens of both animals and humans. In this study, we aimed to evaluate the effect of a combination of ten antibiotics with an inhibitor of efflux pumps (EPI), i.e. berberine (BER), against 6 strains of NTM. Our results showed that the BER potentiated the anti-mycobacterial activities of the antibiotics. Overall, our findings show the importance of BER in increasing the efficacy of antibiotics in NTM.

Three Schiff bases derived from thiosemicarbazide and ketones (Vanillin, Benzophenone and Acetophenone) were used to study their antibacterial activities against some pathogenic bacteria by disc diffusion method. Of these, benzophenone thiosemicarbazone showed significant antibacterial activity as compared with that of Kanamycin. All these three compounds were found to possess cytotoxic effect. Minimum inhibitory concentration of these compounds was also determined.

Introduction and objective. Several studies have reported therapy failures in patients with legionnaires’disease; however, antimicrobial resistance of clinical and environmental isolates of Legionella spp. has not yet been documented. Routine susceptibility testing of Legionella spp. is not recommended because of difficulties in determining standard minimal inhibitory concentration values. The purpose of this study was to analyze the antimicrobial susceptibility of Legionella pneumophila. strains isolated from a water supply system. Materials and method. Twenty-eight isolates of L. pneumophila (16 – L. pneumophila SG 1, 12 – L. pneumophila SG 2–14) obtained from water systems in public buildings in Poland were tested. Susceptibility testing was performed using the E-test method. The tested antibiotic were azithromycin, ciprofloxacin, and rifampicin. The medium used for the susceptibility testing was BCYE-, a special medium for Legionella cultivation. Results. Among the tested strains, L. pneumophila was the only one resistant to azithromycin. It was a strain of L. pneumophila SG 2–14 isolated from the water system in a sanitorium. All isolates were found to be sensitive to ciprofloxacin and rifampicin. However, the azithromycin-resistant strain exhibited higher ciprofloxacin and rifampicin MIC (1.5 μg/ml, and 0.19 μg/ml, respectively). The MIC50 for azithromycin, ciprofloxacin, and rifampicin were 0,032, 0,125, and 0,003 μg/ml, respectively. The MIC90 for azithromycin, ciprofloxacin, and rifampicin were 0,032, 0,125, and 0,003 μg/ml, respectively. Conclusions. Azithromycin resistance was found in one strain of L. pneumophila SG 2–14, but the resistance mechanism is unknown and needs further study. It is possible that therapeutic failures in Legionnaires’ disease may be associated with bacterial resistance which should be taken into account. The antibiotic sensitivity testing described in this study could be helpful in detecting the resistance of clinical L. pneumophila isolates. Ciprofloxacin and rifampicin have good in vitro activity against environmental L. pneumophila SG 1 and SG 2–14 in Poland.

Background. With an increase in antibiotic resistance in microorganisms, increasing the effectiveness of antibiotic therapy has become a crucial task for medical and pharmaceutical sciences. One possible way to increase the effectiveness of antibiotic therapy is to monitor biologically active substances like amino acids for their capability of increasing the activity of antibiotics. This research aimed to study the effects of tryptophan and zinc aspartate on the activity of doxycycline in relation to E. coli and P. aeruginosa. Material and methods. The in vitro serial dilution method was used to determine the minimum inhibitory concentration (MIC) of doxycycline independently and in combination with tryptophan and zinc aspartate, for E. coli and P. aeruginosa. Results. Tryptophan and zinc aspartate at a concentration of 2 μg/ml significantly increased the antibiotic activity of doxycycline against E. coli and P. aeruginosa (p<0.0001). Conclusions. Biologically active substances, tryptophan and zinc aspartate have a modulating effect on the antibiotic sensitivity of microorganisms towards antibacterial drugs like doxycycline.

A new series of four transition metal complexes of a Schiff base derived from salicylaldehyde and glycine, viz. [N-salicylidene glycinato diaqua cobalt (II) dimer] (SGCo)2, [N- salicylideneglycinato-di-aqua-nickel(ll)dimer] (SGN)2, [N-salicylideneglycinato-aqua-copper(II)] (SGC) and [N-salicylidene glycinato diaqua zinc(II) dimer] (SGZ)2 have been synthesized and characterized through a rapid, simple, and efficient methodology in excellent yield. These compounds were screened for in vitro antibacterial activities against six pathogenic bacteria, such as Shigella sonnei, Escherichia coli, Bacilus subtilis, Sarcina lutea, Staphylococcus aureus and Pseudomonas arioginosa. The antibacterial activity was determined by the disc diffusion method using DMSO as solvent. The results indicate that (SGC) compound exhibit a significant antibacterial activity, depending on the bacterial strain and (SGCo)2, (SGN)2 and (SGZ)2 compounds show a moderate sensitivity even with higher doses. All these compounds were found to possess cytotoxic effect.

The quaternary derivative of branched polyethylenimine (bPEI-met) was synthesized and its antibacterial activity against Gram-positive bacterium (Staphylococcus aureus) and Gram-negative bacterium (Escherichia coli) was studied. The values of minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of bPEI-met against mentioned bacteria were determined. These indicated that bPEI-met could be considered as an effective alternative to antibiotics for the treatment of selected bacterial strains. The results obtained can be useful in search for drugs allowing treatment of antibiotic resistant bacterial strains.
Zsyntetyzowano czwartorzędową amoniową pochodną rozgałęzionej polietylenoiminy (bPEI-met) i badano jej działanie antybakteryjne przeciwko bakteriom Gram-dodatnim na przykładzie gronkowca złocistego (Staphylococcus aureus) i Gram-ujemnym na przykładzie pałeczki okrężnicy (Escherichia coli). Wyznaczono wartości minimalnego stężenia hamującego (MIC) i minimalnego stężenia bakteriobójczego (MBC) bPEI-met w stosunku do obu wymienionych szczepów bakterii. Wyniki tych badań sugerują, że bPEI-met może być rozważana jako alternatywa dla antybiotyków w zwalczaniu wybranych szczepów bakterii, szczególnie tych, które charakteryzują się antybiotykoopornością.
