The effects of thiamine (vitamin B1) on the level of spontaneous or radiation-induced genetic changes in human lymphocytes in vitro were studied. Cultured lymphocytes were exposed to increasing concentrations of thiamine (0-500 μg/ml) and irradiated with X-rays. The DNA damage was estimated as the frequency of micronuclei and apoptotic or necrotic morphological changes in fixed cells. The results show that thiamine alone did not induce genetic changes. A significant decrease in the fraction of apoptotic and necrotic cells was observed in lymphocytes irradiated in the presence of vitamin B1 at concentrations between 1-100 μg/ml compared to those irradiated in the absence of thiamine. Vitamin B1 at 1 and 10 μg/ml decreased also the extent of radiation-induced formation of micronuclei. Vitamin B1 had no effect on radiation-induced cytotoxicity as measured by nuclear division index. The results indicate that vitamin B1 protects human cells from radiation-induced genetic changes.

Opis:
Mass casualty scenarios of radiation exposure require high throughput biological dosimetry techniques for population triage, in order to rapidly identify individuals, who require clinical treatment. Accurate dose estimates can be made by biological dosimetry, to predict the acute radiation syndrome (ARS) within days after a radiation accident or a malicious act involving radiation. Timely information on dose is important for the medical management of acutely irradiated persons [1]. The aim of the study was to evaluate the usefulness of the micronuclei (MNi) scoring procedure in an experimental mode, where 500 binucleated cells were analyzed in different exposure dose ranges. Whole-body exposure was simulated in an in vitro experiment by irradiating whole blood collected from one healthy donor with 60 MeV protons and 250 keV X-rays, in the dose range of 0.3–4.0 Gy. For achieving meaningful results, sample scoring was performed by three independent persons, who followed guidelines described in detail by Fenech et al. [2, 3]. Compared results revealed no signifi cant differences between scorers, which has important meaning in reducing the analysis time. Moreover, presented data based on 500 cells distribution, show that there are significant differences between MNi yields after 1.0 Gy exposure of blood for both protons and X-rays, implicating this experimental mode as appropriate for the distinction between high and low dose-exposed individuals, which allows early classification of exposed victims into clinically relevant subgroups.

Opis:
Occupational exposures to environmental toxicants have been associated with the onset of skin lesions-including cancers. Identification and reduction of exposure to such compounds is an important public health goal. We examined the effect of cashew shell oil (CSO), used in skin tattooing for its potential to induce skin transformation in rats. Corn oil and CSO (25, 50, and 100%) were topically applied to depilated sections of Wistar' rat skin (groups: I-IV) for six weeks. Effect of treatments on serum transaminases activity, histological changes in hepatocytes and induction of micronuclei in the bone marrow were examined. In addition, CSO-induced hepatocyte proliferation was also quantified. All animals survived the course of the study. Reduced percentage change in body weight and physical trauma were observed in CSO-treated rat. The effects were more prominent in Group IV (100% CSO). Relative liver weights and number of hepatocytes (cells/mm2) increased significantly in groups II-IV relative to control (p < 0.05). Serum transaminases activities were not significantly (p > 0.05) affected in treated groups. Hepatic histopathology revealed moderate sinusoidal congestion (group II), in addition to portal congestion in (group III), with mononuclear cellular infiltration (group IV) animals. In addition, CSO induced significant micronuclei formation of polychromatic erythrocyte (mPCEs) in the rat bone marrow (p < 0.05) when compared with control. Topical application of CSO disrupted skin cells integrity resulting in physical trauma. In addition, CSO appears to be clastogenic and induces hepatocyte proliferation. Occupational exposure to CSO especially for engraving tattoos in humans should be discouraged and further studies need to be conducted.

The role of glutathione S-transferase Mu 1 (GSTM1) in the biomonitoring of polycyclic aromatic hydrocarbons (PAHs) is not clear. Our purpose has been to evaluate the influence of GSTM1 genotypes on 1-hydroxypyrene (1-OHP), deoxyribonucleic acid (DNA) adducts, and micronucleus frequency in both occupational and non-occupational populations of null and active GSTM1 carriers. We conducted a meta-analysis on 25 articles that met our strict inclusion criteria (11 studies on 1-OHP, 9 on DNA adducts, and 5 on the micronucleus frequency). In the case of occupationally exposed workers, micronucleus frequency was only significantly higher in the null GSTM1 carriers than in the active GSTM1 carriers. In the non-occupationally exposed general population, 1-OHP and micronucleus frequency were significantly higher in the null GSTM1 carriers. The results of Egger’s test and funnel plot analysis indicated no significant publication bias. In conclusion, GSTM1 genotypes may affect the urinary 1-OHP in the non-occupationally exposed general population, and micronucleus frequency in both occupational workers and non-occupational population. Rapid retrospective biological dosimetry allows absorbed dose evaluation post exposure to ionizing radiation. One of the main tools of biodosimetry is based on the analysis of the effects resulting from the impact of ionizing radiation on the cell. Various cytogenetic tests give possibility of the accurate dose estimation. To investigate cell response to radiation one performs the analysis of biomarkers approved by International Atomic Energy Agency e.g. the analysis of dicentric chromosomes or micronuclei frequency. Micronucleus test is relatively a faster and therefore more effective method to study changes in the genetic material, induced by various genotoxic agents. This study confirms that micronulei frequency and nuclear division index analysis allows for appropriate absorbed dose estimation when it comes to ionizing radiation. In order to further optimize and facilitate the micronucleus assay and other cytogenetic tests in rapid retrospective biological dosimetry, the research are still ongoing.

Objectives To study any possible correlation between arsenic toxicity and the development of oral carcinoma in West Bengal population. Material and Methods Ethical clearance for this study was obtained from the Vivekananda Institute of Medical Sciences. Out of 30 785 patients attending our hospital from November 2012 to July 2015, 107 cases and 50 control individuals were selected. The hair and buccal smear samples were obtained upon their consent for the purpose of the analysis of arsenic count and cytogenetic damage, respectively. Results Ninety-six percent of cases came from the highly arsenic affected districts and 81.3% showed their arsenic count above the safe limit (0.8 μg/g) whereas 96% of the controls’ arsenic count was within the safe limit. The study showed a significant difference of the micronuclei and apoptosis frequency between the cases and controls. Conclusions The difference of micronuclei and apoptosis frequency between cases and controls was significant. The maximum number of cases came from highly arsenic affected areas and a higher percentage of cases showed elevated arsenic count, as compared to controls, which may indicate a possible link between arsenic toxicity and this disease. The function of acetylcholinesterase (AChE) is the rapid hydrolysis of the neurotransmitter acetylcholine (ACh), which is involved in the numerous cholinergic pathways in both the central and the peripheral nervous system. Therefore, AChE measurement is of high value for therapy management, especially during the course of intoxication with different chemicals or drugs that inhibit the enzyme. Pyridinium or bispyridinium aldoximes (oximes) are able to recover the activity of the inhibited enzyme. Since their adverse effects are not well elucidated, in this study the efficiency of HI-6 oxime in protection and/or reactivation of human erythrocyte AChE inhibited by the antineoplastic drug irinotecan as well as its cyto/genotoxicity in vitro were investigated. HI-6 was effective in protection of AChE and increased its activity up to 30%; the residual activity after irinotecan inhibition was 7%. Also, it reactivated the enzyme previously inhibited by 50% irinotecan (4.6 µg/ml) applied at ¼ of the IC50 value. The tested concentrations of HI-6 exhibited acceptable genotoxicity towards white blood cells, as estimated by the alkaline comet assay, DNA diffusion assay and cytogenetic endpoints (structural chromosome aberrations and cytokinesis-block micronucleus assay). The results obtained warrant the further investigation of HI-6 in vivo, as well as its development for possible application in chemotherapy.

Samples of 5 vascular plant species: Achillea millefolium L., Artemisia vulgaris L., Festuca gigantea (L.) VILL., Ranunculus acris L. s. str. and Vicia angustifolia L. were collected near Fuel Storage Station, Chemical Fibre Factory “Wistom” in Tomaszów Mazowiecki and Sewage Treatment Plant in Sulejów. Genotoxicity of environmental pollution was analysed using micronucleus assays in pollen mother and root tip cells in comparison with plant material collected in the Tatra and Babia G óra National Parks. Abnorm alities in morphology of plants growing in polluted areas were also found. The cytogenetical analyses revealed positive correlation between the percentage of micronuclei and the presence of heavy metals in soil. Pollen mother cells (germ line) were found more susceptible to DNA damage as compared with root tip cells (somatic line).

The study of hematological parameters is an important method of studying the health of fish in ecosystems that are exposed to the negative impact of the environment. In this work, we determined the presence of pathological changes in the red blood cells of the crucian carp of the Samara River under conditions of anthropogenic stress. The object of the study were six-year-old individuals of both sexes of silver crucian carp (Carassius gibelio (Bloch, 1782)). Morphological studies of erythrocytes were performed on blood smears, which were stained according to the method of Romanovsky-Gimza. During the analysis of the obtained data, the presence of cells with pathologies and the nature of these pathologies were determined. As a result of the conducted researches pathologies of erythrocytes which arise at toxicological poisoning of fish by heavy metals and organic compounds have been revealed. Were also recorded at all experimental points the pathologies indicating active adaptation of fish to environmental stressors.
