Introduction: Heart failure (HF) is a growing global pandemic that affects millions of people around the world. Despite the progress in medicine, diagnosis and treatment of HF remains problematic. Recently, noncoding micro ribonucleic acids called miRNAs have become significant in the diagnosis and stratification of HF risk. Aim: The aim of this study was the attempt to identify the profile of circulating miRNAs specific for ischemic HF with moderately reduced left ventricular ejection fraction (HFmrEF). Methods and Results: A number of changes in the miRNA profile can characterise patients with ischemic HFmrEF. This is a pilot study before further research on a larger group of patients. Conclusions: Using the quantitative reverse transcription-polymerase chain reaction (qRT-PCR), serum levels of 84 miRNA were measured and compared between a patient with ischemic HFmrEF and a healthy volunteer. Analysis reveals a down-regulation of let-7f-5p and miR-1-3p, as well as up-regulation of miR-100-5p, miR-10b-5p, miR-125a-5p, miR-140-5p, miR-144-3p, miR-149-5p, miR-15b-5p, miR-183-5p, miR-208b-3p, miR-224-5p, miR-26b-5p, miR-27b-3p, miR-302a-3p, miR-320a, miR-7-5p, miR-99a-5p.

Small nucleolar RNAs (snoRNAs) are molecules located in the cell nucleolus and in Cajal bodies. Many scientific reports show that snoRNAs are not only responsible for modifications of other RNAs but also fulfill multiple other functions such as metabolic stress regulation or modulation of alternative splicing. Full-length snoRNAs as well as small RNAs derived from snoRNAs have been implied in human diseases such as cancer or Prader-Willi Syndrome. In this review we describe emerging, non-canonical roles of snoRNAs and their derivatives with the emphasis on their role in human diseases.

Opis:
Wild edible plants are often found to be rich sources of nutrients and medicinally beneficial compounds with pharmacological activities. Gnetum gnemon is a nutritionally important plant and a popular food source in parts of Assam and North-East India. Various microRNAs (miRNAs) have been recently identified in many plants; however, there are no records of identification of miRNAs in any species of Gnetum. The prediction of miRNA-target associations in G. gnemon is an important step to facilitate functional genomics studies in this species. In the present study, all known miRNAs from plants available in public domain were used to search for the conserved G. gnemon miRNA homologues in publicly accessible expressed sequence tags (ESTs) in NCBI database. An aggregate of 20 new potential miRNAs belonging to two diverse miRNA families (miR399 and miR5021) were identified through a homology-based search by following stringent filtering criteria. To investigate the potential cross-kingdom effects of the identified miRNAs, we further identified the putative target genes of G. gnemon miRNAs in human transcriptome and analyzed them against the NCBI non-redundant protein database. The KEGG analysis of the target genes indicated that these genes were involved in different metabolic pathways such as caffeine metabolism, drug metabolism, and nitrotoluene degradation. The target genes of G. gnemon miRNAs in humans were found to be associated with various disorders of both hereditary and non-hereditary origin. These results could help to shed new light on understanding of miRNA-mRNAs functional networks in this species and its potential use as a small RNA-based therapy against some human diseases.

Opis:
The molecular basis for the formation and growth of vestibular schwannomas (VS) has been elucidated in the recent years. The main genetic and epigenetic aberrations, changes in gene expression and specific signaling pathways involved in pathogenesis of sporadic VS and neurofibromatosis type II (NF2) have been defined. These findings facilitated the search for prognostic markers in VS and potential targets for biological therapy. This publication summarizes the main directions of research in the field of molecular diagnostics and pharmacotherapy of VS based on biological agents.

The widespread particularist account of the onset of molecular biology that identifies it with the discovery of the DNA structure in 1953 has been recently contested. The paper contributes to this debate by focusing on a more recent discovery of small noncoding RNAs (microRNAs). First, it outlines a particularist account of the microRNAs discovery and the origins of the particularist predilection of the modern scientometric studies of science dynamics. Next, it discusses its limitations and proposes an alternative, modified processualist account of the discovery. In the final part, the paper applies this approach to unravel network dynamics of the research on the first two microRNAs that were discovered, namely lin-4 and let-7.
