Opis:
Grapefruit juice (GFJ), a commonly consumed dietary substance, has been shown to alter the disposition of several commonly used drugs. The available data on the effects of GFJ on the paracetamol pharmacokinetics and pharmacodynamics are at variance. The aim of this study was to evaluate the effect of a single or multiple dose of GFJ on some pharmacokinetics and pharmacodynamics aspects of orally given paracetamol (400 mg/Kg). Male mice were randomly divided into three equal groups: mice in the first group were given paracetamol; the second group was given a single oral dose (10 mL/Kg) of GFJ one hour prior to paracetamol administration; the third group was administered multiple oral doses of GFJ (10 mL/Kg) for five consecutive days, and on the last day it was treated with paracetamol. Blood samples were collected 10, 20, 30, and 40 min, and 1, 2, 4, 6 and 8 h after paracetamol administration for subsequent pharmacokinetic analysis. Some mice in the same three groups were also tested for their reactions to thermal (hotplate) and chemical (acetic acid induced – abdominal constriction) nociceptive stimuli. GFJ increased the plasma concentration and area under the plasma concentration curve of paracetamol, to a greater extent after a single dose than multiple doses. It also increased the reaction time in the hotplate test, and reduced abdominal constrictions. GFJ administration increased the plasma concentration and the analgesic effect of paracetamol in mice. The possible implications of these changes in humans and their clinical relevance need to be further investigated.

The present study was aimed to evaluate the activity of Achyranthes aspera against allergy caused by heavy metal potassium dichromate in albino mice. The potassium dichloromate as induced at a concentration of 20 mg/kg. body weight dissolved in distilled water for 10 days continuously. On 11th day, blood was drawn from mice for serum analysis of IgM and IgG to confirm allergy induction in mice. After allergy induction the mice received petroleum ether, chloroform and methanolic extracts of Achyranthes aspera given 3 doses at concentration of – 200 (low), 400 (medium) and 600 (high) mg/kg to mice by oral route for 7 days after allergy induction. On the 8th day, blood was drawn from mice for serum analysis of IgM and IgG in determine the effect of extracts on serum Titre of mice blood.

Diethylnitrosamine (DEN) is proven to be toxic to kidneys and liver and to act as a potent carcinogen mainly in liver. Capsaicin (CAP) is an alkaloid produced by Capsicum genus plants and is considered to be a protective agent against toxicity and carcinogenicity of many substances including DEN. The aim of this study was to assess the toxicity of DEN and CAP in liver and kidneys in mice. The experiment started after two weeks of acclimatisation and was conducted according to the Czech animal welfare protection legal guidelines. At the end of the experiment the mice were sacrificed and the toxicity of DEN and CAP in liver and kidneys were analysed. The histopathological examination of the liver revealed multifocal lymphoplasmacytic reaction in parenchyma in DEN treated group. CAP used as both preventive and therapeutic agent caused reduction in number and extent of lesions. In CAP group mitotic figures were found indicating xenobiotic-induced hepatotoxicity or regenerative changes. In the kidneys DEN revealed also multifocal lymphoplasmacytic reaction that has been mitigated by CAP. Moreover histopathological observation of the kidney in DEN group has revealed granular dystrophy of the renal tubules which has not been presented in CAP treated mice. Levels of ALT, AST activity, total protein and albumin concentration was not statistically different among control and experimental groups. In this study mild protective effect of CAP on DEN-induced hepatotoxicity and nephrotoxicity was shown only in histopathological changes. The toxicity of CAP itself is questionable and further studies should be performed to verify its chemopreventive potential.

The plants of the Impatiens L. (Balsaminaceae) have been used for a long time in folk medicine in different painful conditions, and to treat rheumatism, isthmus and crural aches, fractures, superficial infections, fingernail inflammation. This study was undertaken to determine the pharmacological profile of hydroethanolic extracts from Impatiens glandulifera, I. noli-tangere and I. parviflora. A range of behavioral assessments was applied to evaluate the effects of obtained extracts i.e. measurement of body temperature, tests of locomotor activity and motor coordination, nociceptive reaction and anxiety-like behavior. Hydroethanolic extracts were analyzed for total polyphenol (TPC), flavonoid (TFC), flavones/flavonols (TFFC), and flavonones/dihydroflavonols (TFDC) content. Our results show that the extracts from Impatiens species contain high levels of TPC, TFC, TFFC, and TFDC. Oral (i.e., by gavage) administration of Impatiens L. extracts (except for I. noli-tangere) presented an antinociceptive or/and anti-inflammatory activity in the writhing test. The antinociceptive effect of I. parviflora leaves (100 mg/kg) and I. glandulifera flowers (100 mg/kg) was reversed by naloxone. I. glandulifera flowers and roots extracts (100 mg/kg) increased the reaction time to the thermal stimulus in the hot-plate test. All extracts from I. glandulifera (100 mg/kg) showed antianxiety effect in the elevated plus-maze test. It is worth noting that none of the extracts, at the highest used dose – 0.1 ED50 (200 mg/kg), caused coordination impairments or myorelaxation as measured in the rota-rod and chimney tests. These results seem to suggest that the tested extracts are not neurotoxic.

Malaria is an important health and development challenge in Africa, Animal models most particularly mice, have long been employed to study malaria pathogenesis. This paper describes clinical manifestations due to Plasmodium berghei ANKA infection in juvenile mice as a model for understanding the complications of congenital malaria in neonates. Forty-five juvenile mice (5-7 days old) were acquired from University College Hospital, Ibadan and injected with 2×107 (0.2 ml) Plasmodium berghei ANKA parasitized red blood cells (PRBCs). The mice were then transported to the study site, kept in well-ventilated cages and fed daily with a balanced ration. Post-P. berghei infection, the mice were monitored daily for mortality. Clinical manifestations of experimental cerebral malaria (ECM) were assessed and confirmed if at least ruffled fur, hunching, wobbly gait, limb paralysis, convulsions, or coma was observed. Each sign was given a score of 1. Animals with scores ≥4 were considered to have severe ECM. In the experiment, 20 (44%) mice were lost due to natural cause (i.e. stress) at day 2. Between day 4 and 9, 25 (56%) of the study mice presented clinical signs of ECM. This included: ruffled fur – 25 (100%), hunching - 21 (84%), wobbly gait - 17 (68%), limb paralysis - 20 (80%), convulsions - 25 (100%). Survival rate and severity of ECM in the mice differs, 22 (88.0%) had severe ECM and 3 (12.0%) had mild ECM. This study has shown that parasite establishment and malaria complications can manifest as early as 4 days’ post P. berghei infection in 5-7 days old mice.

Essential polyunsaturated fatty acids (PUFAs) in the feed may affect the gastrointestinal microbiota. The present study investigated the effect of 35-day supplementation of mice diet with 5% concentration of high-ω-3 PUFAs in flaxseed with focus on intestinal metabolism of mice. Capillary isotachophoresis method was used for the assessment of the level of organic acids in the gut material and faeces. Supplementation of flaxseed increased the level of organic acids in the caecum (acetic, propionic, butyric and valeric acids) and faeces (lactic, acetic, butyric acids). The most significant effect was observed on day 28 of flaxseed supplementation. The investigated additive had a stimulatory effect on intestinal metabolism and fermentation activity of beneficial bacteria.

Objectives The objective of the present study was to observe the effects of 50 Hz magnetic fields (MFs) on the immune function of splenic lymphocytes in mice. Material and Methods Twenty male Kunming mice (6 weeks old), weighing 18– 25 g, were randomly divided into sham exposure (N = 10) and 500 μT MFs (N = 10) groups. The mice in the MFs group were exposed to 500 μT MFs for 8 h daily (5 days/week) for up to 60 days. In vitro study was carried out to examine the effects of 50 Hz MFs on the expression of inflammatory factor genes and a cluster of differentiation 69 (CD69) in mouse prime splenic lymphocytes activated by para-Methoxyamphetamine (PMA) and ionomycin. In the in vitro experiments, lymphocytes were isolated from the spleen of 10 healthy Kunming mice, the cells were cultured in the Roswell Park Memorial Institute 1640 medium (RPMI-1640) and exposed to 0 μT, 250 μT, 500 μT, or 1 mT MFs in an incubator under 5% carbon dioxide (CO₂) at 37°C for 6 h. The levels of interleukin-2 (IL-2), IL-4, interferon-gamma (IFN-γ), GATA binding protein 3 (GATA-3) and T cell-specific T-box transcription factor (T-bet) were assessed by the real-time quantitative reverse transcription polymerase chain reaction (RT-qPCR), respectively. The expression of CD69 was checked using the flow cytometry. Results Under our experimental conditions, body weight of the mice exposed to occupational, extremely low frequency- electromagnetic fields (ELF-EMFs) significantly decreased on day 20 and day 30. There were no significant changes observed in vivo in spleen weight, splenic coefficient, splenic histology profile and cytokine production in spleen tissues. Our in vitro experiments showed that 50 Hz MFs had no effect on the expression of these genes and CD69 to primary splenic cells. Conclusions In conclusion, under the applied experimental conditions, occupational exposure to 50 Hz magnetic field did not alter responses of inflammatory genes and activation of splenic lymphocytes in mice, except for body weight.

In this study we report the usefulness of nested PCR for screening of the persistent B. microti infections in rodent hosts. Female BALB/c mice were inoculated with 100 μl of donor blood infected with B. microti. Infections were detected using microscopic examination of Giemsa-stained blood smears. To determine whether B. microti DNA was present in blood and/or spleen tissue, nested PCR was performed targeting a specifi c fragment of the gene encoding the 18S rRNA. Blood was sampled every 10 days post-infection (dpi) until day 30, after which mice were sampled every 30 days until the end of experiment at 360 dpi. The most extensive parasitaemia (39% of infected erythrocytes) was observed at 10 dpi. Between 20-60 dpi, less then 1% of infected erythrocytes were detected in blood smears, and from 90 dpi onwards, infected erythrocytes were no longer observed. B. microti DNA was successfully amplifi ed from the blood of mice from 10 dpi until 180 dpi, as well as from spleens of infected mice at 10 and 20 dpi. The presented results show that nested PCR is the method of choice for monitoring infections of B. microti in the blood of rodent hosts, and could therefore be a tool for environmental monitoring of naturally infected rodents which are the predominant source of infection for tick vectors.

Dysfunction of fast axonal transport, vital for motor neurons, may lead to neurodegeneration. Anterograde transport is mediated by N-kinesins (KIFs), while retrograde transport by dynein 1 and, to a minor extent, by C-kinesins. In our earlier studies we observed changes in expression of N- and C-kinesins (KIF5A, 5C, C2) in G93ASOD1-linked mouse model of motor neuron degeneration. In the present work we analyze the profile of expression of the same kinesins in mice with a dynein 1 heavy chain mutation (Dync1h1, called Cra1), presenting similar clinical symptoms, and in Cra1/SOD1 mice with milder disease progression than SOD1 transgenics. We found significantly higher levels of mRNA for KIF5A and KIF5C but not the KIFC2 in the frontal cortex of symptomatic Cra1/+ mice (aged 365 days) compared to the wild-type controls. No changes in kinesin expression were found in the spinal cord of any age group and only mild changes in the hippocampus. The expression of kinesins in the cerebellum of the presymptomatic and symptomatic mice (aged 140 and 365 days, respectively) was much lower than in age-matched controls. In Cra1/SOD1 mice the changes in KIFs expression were similar or more severe than in the Cra1/+ groups, and they also appeared in the spinal cord. Thus, in mice with the Dync1h1 mutation, which impairs dynein 1-dependent retrograde transport, expression of kinesin mRNA is affected in various structures of the CNS and the changes are similar or milder than in mice with double Dync1h1/hSOD1G93A mutations.
