Cadmium (Cd) stress significantly affects plant growth and development. Potentilla sericea is typically grown in gardens or as ground cover. In this study, the Cd response of P. sericea was analyzed based on physiological examinations and transcriptome analyses that uncovered the gene expression changes in P. sericea roots induced by a 7-day treatment with 90 μmol/L Cd2+. A total of 53,225 unigenes were identified, including 11,684 differentially expressed genes (DEGs; 8,083 upregulated and 3,601 downregulated). Additionally, 44 gene ontology terms and 127 Kyoto Encyclopedia of Genes and Genomes pathways were significantly enriched among these DEGs. Genes related to glutathione metabolism, plant hormone signal transduction, peroxisome processes, sulfur metabolism, and flavonoid biosynthesis pathways were confirmed as relevant to the Cd response of P. sericea. The molecular biology-related data described here may be useful for the future breeding of transgenic P. sericea plants with increased resistance to heavy metal stresses.

Introduction and objective. Metabolic disorders are associated with a number of negative health consequences. Therefore, the aim of this review was to define the relationships between overweight and obesity, lipid metabolism disorders, hypertension, disorders of carbohydrate metabolism, and cognitive functions. Abbreviated description of the state of knowledge. Metabolic disorders may be linked to the reduction of cognitive functions, such as memory, abstract reasoning, verbal fluency, attention and psychomotor speed. Obesity may lead to changes in the hippocampus, one of the main structures involved in learning and memory. The evidence reviewed suggests that obesity, by inducing inflammation, leads to the dysfunction of brain structures such as the frontal cortex and corpus callosum involved in plasticity processes and cognitive functions. In addition, obesity has been found to be associated with abnormalities in the white matter and glial cells in the brain, which may also cause reduction in processing speed. It is possible that there is a non-linear relationship between lipids and cognitive function moderated by age and the pro- inflammatory process. Disturbances in carbohydrate metabolism can also lead to cognitive impairment, such as: memory, verbal fluency or abstract reasoning. Conclusions. Metabolic disorders are associated with a higher risk of developing cognitive impairment. This association can be moderated by additional factors: lifestyle, oxidative stress, pro-inflammatory processes, genetic factors, age and education.

Sclerostin, which is a glycoprotein produced by osteocytes, reduces the formation of bones by inhibiting the Wnt signal pathway. Thyroid hormones are related with Wnt signal pathway and it has been reported that increased thyroid hormones in hyperthyroidism fasten epiphysis maturation in childhood, and increase the risk of bone fractures by stimulating the bone loss in adults. The aim of this study was to examine the sclerostin serum levels, the relation between sclerostin and thyroid hormones as well as the biochemical markers of the bone metabolism in patients with hyperthyroidism (including multinodular goiter and Graves' disease), whose treatments have not started yet. No difference was found in the serum sclerostin levels between the hyperthyroidism group (n=24) and the control group (n=24) (p=0.452). The serum osteocalcin levels and 24-hour urinary phosphorus excretion were found to be higher in the hyperthyroid group than in the control group (p<0.001, p=0.009). A positive correlation was determined between the sclerostin and bone alkaline phosphatase levels (p<0.001); a negative correlation between the osteocalcin and thyroid stimulating hormone (TSH) (p<0.05); a positive correlation between the osteocalcin and thyroid hormones (FT3,FT4) (p<0.001); and a positive correlation between the deoxypyridinoline and hydroxyproline (p<0.001). No correlation was determined between sclerostin and TSH,FT3,FT4 (p>0.05). Therefore, we consider that a long-term study that covers the pre-post treatment stages of hyperthyroidism, including both the destruction and construction of the skeleton would be more enlightening. Moreover, the assessment of the synthesis of sclerostin in the bone tissue and in the serum level might show differences.

Nematodes were found to synthesize phosphorylcholine-containing molecules not present in higher organisms, i.e. phosphorylcholine-substituted glycosphingolipids and (glyco)proteins. Investigations on the biosynthesis of these structures provided first biochemical evidence for the presence of the Kennedy and Bremer-Greenberg pathways in the model organism Caenorhabditis elegans.

Fatty acids (FAs) are one the most essential substances in intrauterine human growth. They are involved in a number of energetic and metabolic processes, including the growth of cell membranes, the retina and the nervous system. Fatty acid deficiency and disruptions in the maternal-placental fetal metabolism of FAs lead to malnutrition of the fetus, hypotrophy and preterm birth. What is more, metabolic diseases and cardiovascular conditions may appear later in life. Meeting a fetus' need for FAs is dependent on maternal diet and on the efficiency of the placenta in transporting FAs to fetal circulation. "Essential fatty acids" are among the most important FAs during the intrauterine growth period. These are α-linolenic acid, which is a precursor of the n-3 series, linoleic acid, which is a precursor of the n-6 series and their derivatives, represented by docosahexaenoic acid and arachidonic acid. The latest studies have shown that medium-chain fatty acids also play a significant role in maternal-fetal metabolism. These FAs have significant effect on the transformation of the precursors into DHA, which may contribute to a relatively stable supply of DHA - even in pregnant women whose diet is low in FAs. The review discusses the problem of fatty acid metabolism at the intersection between a pregnant woman and her child with reference to physiological pregnancy, giving birth to a healthy child, intrauterine growth restriction, preterm birth and giving birth to a small for gestational age child.

This study compared the ability of daidzein, a soy isoflavone, with that of 17β-estradiol to prevent bone loss in cadmium (Cd)-exposed ovariectomized (OVX) rats during growth. Four week-old female Wistar rats were randomly assigned to five treatment groups of 9 rats each, either (1) sham-operated (SH); (2) OVX and placed on experimental diets (OVX); (3) OVX fed 50 ppm of CdCl2 (OVX-Cd); (4) OVX fed 50 ppm of CdCl2 and 10 µg of daidzein per kg of body mass (OVX-CD-D); or (5) OVX fed 50 ppm of CdCl2 and 10 µg of estrogen per kg of body mass (OVX-CD-E). All rats were given free access to AIN-76 modified diet and drinking water, with or without Cd, for 8 weeks. The OVX groups gained more (P < 0.05) body mass than the SH group. Femoral mass was increased by feeding daidzein and estradiol, whereas femoral length was not (P > 0.05) significantly different among groups. Femoral breaking force was not significantly different among groups, however, femoral BMD was significantly lower in OVX-Cd than in the SH and OVX groups. Morphologically proliferative cartilage and hypertrophic cells in femur showed normal distribution in OVX-Cd-D and OVX-Cd-E groups unlike those in OVX-Cd group. These findings suggest that Cd-OVX-induced osteopenia or osteoporosis probably results from an increase in bone turnover.

Introduction. Positron Emission Tomography (PET) is a non-invasive diagnostic tool that provides tomographic images and measures quantitative parameters of cell viability and metabolic activity of tissues. The most important used tracer in PET is an 18-F-Fluorodeoxy-glucose (FDG), the glucose molecule of which is labeled with a radiotracer and allows measurement and mapping of tissue glucose uptake. There are many studies in PET/CT which rely on some mammalian species, and recently on fish. Objective. The aim of this survey by using FDG-PET/CT are to optimize and determine FDG uptake in fish, in this case, common carp (Cyprinus carpio) using three treatments: Basic fish meal, Vita Pulvis and Probiotics s with two replications were used. Sphere Volume of interest (VOI) were drawn hand for the all organs, and standard uptake value (SUV) means were calculated. Results. The SUV mean for glucose uptake in the liver and gastrointestinal tract of fish were more similar to those of humans than rats or mice. SUV mean in fish fed by Probiotics ss in the major organ were less than those fed by Vita Pulvis and basic fish meal. The results present the opportunity to focus on studies of metabolism and screening for the effects of nutrients on body development.

Insulin resistance (IR) plays a critical role in metabolic syndrome (MS). Previous studies have demonstrated that activated ROCK is increased in MS patients. However, the effect of Rho-kinase (ROCK) on IR has not been definitely determined. Thus, the aims of the present study were to determine whether ROCK activation induces IR or affects myocardial structure and function, as well as the possible mechanisms underlying this process. Wistar rats fed high fat, high glucose and high salt diet sewed as model of MS and we used transmission electron microscopy, echocardiogram technology, and terminal deoxynucleotidyl transferase-mediated DNA nick-end labeling staining to identify any myocardial damage. The protein levels of MYPT-1 (characteristic of ROCK activation), IRS-1 and AKT were analyzed by immunohistochemistry and Western blotting. In hearts from MS rats, we found increased protein levels of phospho-MYPT-1 and phospho-IRS-1 (Ser307) and decreased phospho-AKT compared to levels in normal rats. In conclusion, the results suggest that ROCK-mediated IR is involved in the development of myocardial impairments in MS rats and that this effect is mediated probably via the IRS-1/PI3-kinase/AKT pathway.

Przeszło 30 lat temu zaobserwowano, iż Eskimosi, których dieta opiera się na tłustych rybach morskich, rzadko chorują na choroby układu krążenia takie jak choroba niedokrwienna serca, zawał mięśnia sercowego. Obserwacje te dały początek licznym badaniom epidemiologicznym, klinicznym i doświadczalnym mającym na celu wyjaśnienie tego zjawiska. Obecnie nie ulega wątpliwości, iż ryby będące źródłem niezbędnych nienasyconych kwasów tłuszczowych z grupy n-3 (n-3 PUFA) są ważnym składnikiem pożywienia, gdyż zawarte w nich kwasy tłuszczowe istotnie wpływają na szereg przemian metabolicznych i tym samym na ryzyko rozwoju różnych chorób.
Fatty acids derived from fish and fish oils have a strong beneficial effect on cardiovascular health and neurodevelopment. Dietary omega 3 fatty acids prevent cardiovascular disease development by the influence on eicosanoid synthesis, cytokine and growth factors production, lipid and lipoprotein metabolism, endothelial relaxation. They have a strong antiarrhytmic action on the heart. Enhanced consumption of fish derived fatty acids is related to the low incidence of cardiovascular events and sudden cardiac deaths. High content of mercury in some fishes may attenuate the beneficial effects of omega 3 fatty acids on cardiovascular health and also on neurodevelopment of newborns and children

Nutrition is a factor of major importance for health and also for development of diet related disorders. Nowadays there is no doubts that interaction between genes and nutrition plays a key role in health risk prediction and disease prevention. Individual nutritional status is determined not only by food availability but also by genes that control food intake, nutrient absorption and metabolism. On the other hand nutrients can significantly influence different genes expression and metabolic processes. Knowledge of genetic susceptibility to disease will help identify people at enhance risk and their response to diet. The prevalence of the types of genes of interest may differ between populations. The same genotype does not confer the same risk in all populations because of differences in environmental factors and nutritional habits. Therefore, populations should not simply copy each other’s dietary recommendations. Based on genetic and nutritional studies the best way for expression of healthier phenotypes should be found and used to promote health and prevent different diseases development.

Background. The physical activity of 29 football players using either aerobic or anaerobic energy regimes was tested during official games and in the laboratory. Material and methods. Under conditions of gradually increasing power on a treadmill, maximal running speed (V, km/h), oxygen consumption (VO2, mL/min/kg), heart rate (HR, beats/min), and anaerobic metabolism threshold (AT mL/min/kg) were determined, and blood lactate (BLact, mmol/L) was measured. Results. The correlation between the total length of distance run during the game and treadmill running speed for the outcome measures were as follows: anaerobic metabolism threshold, r=0.54 (p=0.032), BLact, r=0.37 (p=0.034), VO2max, r=0.41 (p=0.027), and HR, r=0.36 (p=0.047). We found aerobic mechanisms were dominant in ensuring the game activity of football players. Specifically, aerobic mechanisms contribute roughly 32.4-43.8% to playing activity, while aerobic-anaerobic energy supply mechanisms provide for only 17%. Lactic and alactic mechanisms provide for 5.6% and 1.6%, respectively. Conclusions. The results provide valuable information on the relationship between football play activity and aerobic and anaerobic energy supply mechanisms. Enhancing the functionality of the anaerobic lactic-alactic energy system can lead to growth and improvements in physical performance in football players.

Resting cyst formation is a crucial process of cryptobiosis in protists. In colpodid ciliates, cyst formation is accompanied by large-scale morphological changes such as changes of cell shape, resorption of cilia, and formation of a cyst wall; additionally, the cell cycle is arrested. These changes provide acquired tolerance against environmental stresses. During cyst formation, mitochondrial membrane potential is reduced and the level of the ATP synthase beta chain is suppressed, strongly indicating that metabolism has ceased. Here, however, we show that ATP levels are elevated during the initial phases of encystment implying that metabolism may not be completely suppressed. This finding suggests another aspect of resting cyst formation that is not applicable to cryptobiosis.

A wide range of disorders, including neuropsychiatric disorders and autism, are associated with increased homocysteine levels in biological fluids. Various B vitamins: B6 (pyridoxine), B12 (cobalamin), and B9 (folic acid) are required as co-factors by the enzymes involved in homocysteine metabolism. Therefore, monitoring of homocysteine levels in body fluids of autistic children can provide information on genetic and physiological diseases, improper lifestyle (including dietary habits), as well as a variety of pathological conditions. This review presents information on homocysteine metabolism, determination of homocysteine in biological fluids, and shows abnormalities in the levels of homocysteine in the body fluids of autistic children.

Purpose. Besides being widely used in cosmetics, retinoids are potent therapeutic agents used topically and systemically as anti-acne agents. The aim of this study was to predict with the use of MetaSite the skin metabolism of selected retinoids employed in treatment of skin disorders and found in cosmeceuticals. The following compounds were studied: retinol, retinaldehyde, retinoic acid, retinyl acetate, retinyl palmitate, acitretin, etretinate, adapalene and bexarotene. Methods. MetaSite, Molecular Discovery Ltd. is a computational model that enables prediction of cytochrome P450-dependant metabolism. This software indicates atoms in the molecule structure that are mostly vulnerable to metabolic changes and predicts the metabolite structures. Results. MetaSite indicated that retinol and retinal metabolites were obtained through hydroxylation of the methyl group located in the position 3 of the aliphatic chain, whereas retinoic acid biotransformation would occur principally in the carbon atom situated in the position 4 in the cyclohexene ring. In acitretin molecule, carbon atom of the methoxy group attached to the benzene ring displayed the highest probability of biotransformation. In etretinate, metabolic reactions would occur principally on the carbon atom of the final ethyl group of the molecule. Conclusions. MetaSite metabolism predictions for retinoic acid, acitretin, etretinate, adapalene and bexarotene were in agreement with experimental findings. In case of compounds being converted by catalysts other than cytochrome P450 enzymes, the primary metabolites predicted by MetaSite differ from those reported previously. In conclusion, MetaSite is a useful tool that can aid identification of the major metabolites of compounds being administered topically.

In the modern security environment, information warfare is an increasingly important threat. The purpose of this article is to determine the conditions for the effective achievement of political objectives through the manipulation of information and disinformation. The conducted research used system analysis, also using case studies, generalizations and synthesis. This allowed to state that the key condition for the effectiveness of conducted information attacks is the mechanism described in the works of Antoni Kępiński as information metabolism. This makes it possible to formulate a postulate on the need to counteract such attacks based on building defensive and offensive capabilities of the state in the sphere of information warfare.

Patients with colorectal carcinoma showed statistically significant lower values of transferrin saturation, total iron binding capacity and serum iron level as compared with control group, while the level of ferritin and the size of labile iron pool in carcinoma patients were higher, although this difference was not statistically significant. Our observations are in favour of the hypothesis which suggests that changes in iron metabolism restrict iron availability for tumour cells and as consequence, slow their growth.

Opis:
Conditions of achieving the maximal accumulation of sulfhydryl metabolites in the leaves of tobacco were explored. Simultaneous production of bacterial O-acetylserine (thiol)-lyase and serine acetyltransferase resulted in the increased thiols contents as compared to single transformants and controls. However, leaf discs feeding experiments differently affected thiols concentration in different plant groups and suggested that the most promising strategy to obtain plants with a high level of non-protein thiol-containing compounds might be sulfate feeding to plants overproducing serine acetyltransferase.Formation of cysteine from sulfide and O-acetyl-L-serine (OAS) is catalyzed by O-acetylserine (thiol)-lyase (OAS-TL) (EC 4.2.99.8) while OAS is synthesized by serine acetyltransferase (SAT) from acetyl-coenzyme A and serine. Molecular interactions between SAT and OAS-TL are involved in the regulation of the enzymatic activities of these proteins in bacteria (Mino et al., 2000) and in plants (Bogdanova & Hell, 1997; Droux et al., 1998). Experiments with Escherichia coli enzymes clearly indicated that OAS-TL activity was reduced to 30% in a complex (Mino et al., 2000). Similarly, a very dramatic decrease of the catalytic activity of OAS-TL bound in a complex has been reported for the plant enzymes (Droux et al., 1998). On the other hand, the bienzyme complex formation stabilizes bacterial SAT (Mino et al., 2000) and increases the apparent affinity for the substrates of plant SAT (Droux et al., 1998). The stability of the complex is negatively affected by OAS and positively by sulfide (Droux et al., 1998). Bacterial SAT is feedback regulated by cysteine, however, no relationship seems to exist between the complex formation and SAT sensitivity to this inhibition (Mino et al., 2000).Both enzymes, SAT and OAS-TL, are located in three compartments of the plant cell: the cytosol, chloroplasts and mitochondria. Different isoforms of these enzymes are in a different way regulated by sulfur nutrition (Nakamura et al., 1999; Takahashi et al., 1997; Warrilow & Hawkesford, 1998). The feedback regulation by L-cysteine of various isoforms of plant SAT has recently been studied (Inoue et al., 1999; Noji et al., 1998). According to the model proposed, the only role of the chloroplastic and mitochondrial isoforms (that are insensitive to the feedback inhibition) would be the production of OAS for cysteine biosynthesis. The cysteine-sensitive cystosolic isoform of SAT would, according to this model, have two roles: (i) OAS production for cysteine biosynthesis in the cystosol and (ii) control of OAS pool for regulatory purposes. The second postulated function is tightly connected with the fact that OAS acts as a positive regulator of genes whose expression is affected by sulfur status (Saito, 2000).Glutathione, the main low-molecular-mass thiol-containing compound in the plant cell, has multiple functions, including involvement in responses to various environmental stresses and maintenance of the redox homeostasis (Foyer & Rennenberg, 2000). Under non-stressing conditions the majority of glutathione is maintained in the reduced form (GSH) and its concentration is determined mainly by the rate of biosynthesis. GSH is produced from cysteine, glutamate and glycine in two steps catalyzed by γ-glutamylcysteinyl synthetase (γ-ECS) and glutathione synthetase (GS), respectively (Noctor et al., 1998). The biosynthesis and accumulation of GSH has been shown to depend on (i) the activity of γ-ECS, (ii) the availability of cysteine, and (iii) the light-dependent formation of glycine through the photorespiratory pathway (Foyer & Rennenberg, 2000).The main aim of this study was to identify the optimal conditions for the maximal accumulation of non-protein sulfhydryl metabolites in plant leaves. The transgenic tobacco plants with cytosolic production of bacterial enzymes of the cysteine biosynthesis pathway, SAT and OAS-TL, were obtained and analyzed for the transgenes expression. Additionally, leaf discs of either single or double transformants, as well as of control plants, were assayed for the thiol contents upon incubation in solutions of various compounds expected to have an influence on sulfur metabolism.

Both urea and urease were subjects of early scientific investigations. Urea was the first organic molecule to be synthesized and jack bean urease was the first enzyme ever to be crystallized. About 50 years later it was shown to be the first nickel metalloenzyme. Since then, nickel-dependent ureases have been isolated from many bacteria, fungi and higher plants. They have similar structures and mechanisms of catalysis. A urease apoenzyme needs to be activated. This process requires participation of several accessory proteins that incorporate nickel into the urease forming catalytic site. In this review, ureases from various organisms are briefly described and the similarities of their structures discussed. Moreover, the significance of urea recycling in plants is explained and recent literature data about the function and activation of plant ureases are presented.

The SNIF-NMR method (site-specific natural isotope fractionation studied by Nuclear Magnetic Resonance) was used to examine the isotopic profile of glucoses derived from plants with different photosynthetic pathways. It is shown that the type of photosynthetic metabolism, either C3 (beet-root, orange, grape), C4 (maize, sugar-cane) or CAM (pineapple), exerts a strong influence on the deuterium distribution in the sugar molecules. The isotopic profile also depends, secondarily, on the physiological status of the precursor plant. Consequently, the isotopic fingerprint of glucose may be a rich source of information in mechanistic comparisons of metabolic pathways. Moreover, from an analytical point of view, it may provide complementary criteria with respect to the ethanol probe for origin inference of sugars.

Menadione (2-methyl-l,4-naphthoquinone) is a prooxidant agent commonly used in laboratory practice. It can induce oxidative damage to many cell biomolecules. The results of earlier studies show that the efficiency of the antioxidant system plays a significant part in the protection of yeast cells from toxic effects induced by menadione. The purpose of this study was to determine the sensitivity of Saccharomyces cerevisiae yeast cells with different types of energy metabolism to menadione. The cells of the wild strain SP4 and its isogenlc respiratory mutant SP4rho~ were used in the experiment. By growing yeast on media containing different carbon sources (fermentable and non-fermentable) cells of different respiratory activity were obtained. The viability of the yeast cells was determined by calculating the colonies grown on solid media in the presence of this prooxidant. Under condition of full respiratory activity of yeast in the presence of the non-fermentable carbon source, ie ethanol, greater sensitivity of yeast cells to menadione was observed than under conditions of repression of the respiratory system (medium containing 2 % or 10 % glucose) in the case of cells of mutants incapable of aerobic respiration and in the presence of the slightly fermentable carbon source, ie raffinose. The results obtained show that the toxicity of menadione is not only connected with the efficiency of cell antioxidant mechanisms but it also depends on the type of energy metabolism of yeast cells.
Menadion (2-metylo-1,4 naftochinon) jest powszechnie stosowanym w praktyce laboratoryjnej czynnikiem prooksydacyjnym. Generuje oksydacyjne uszkodzenia makromolekuł komórkowych. Wyniki wcześniejszych badań wskazują, że sprawność systemu antyoksydacyjnego jest bardzo ważna w ochronie komórek drożdżowych przed toksycznymi efektami indukowanymi menadionem. Celem prezentowanej pracy było określenie wrażliwości komórek drożdży Saccharomyces cerevisiae o różnym metabolizmie energetycznym na menadion. W badaniach wykorzystano komórki szczepu dzikiego SP4 oraz jego izogenicznego mutanta oddechowego SP4 rho-. Hodując drożdże w pożywkach zwierających różne źródła węgla (ulegające fermentacji i nie ulegające fermentacji), uzyskano komórki o różnej aktywności oddechowej. Poziom przeżywalności komórek drożdży oznaczono poprzez liczenie kolonii wyrosłych na pożywkach stałych w obecności tego prooksydantu. W warunkach pełnej aktywności oddechowej drożdży w obecności nie ulegającego fermentacji źródła węgla, jakim jest etanol, zanotowano większą wrażliwość komórek drożdży na menadion niż w warunkach represji układu oddechowego (pożywka zawierająca 2 % lub 10 % glukozę), w przypadku komórek mutanta niezdolnego do oddychania tlenowego oraz w obecności słabo fermentowalnego źródła węgla jakim jest rafinoza. Uzyskane wyniki sugerują, że toksyczność menadionu jest związana nie tylko ze sprawnością komórkowych mechanizmów antyoksydacyjnych, ale także zależy od typu metabolizmu energetycznego komórek drożdży.

The last decades have witnessed tremendous advances in technology. Accelerating rate of technological change has brought about not only humans daily life, but also extraordinary climate change. Almost one-half of the land surface has been transformed by human action, the carbon dioxide concentration in the atmosphere exceeded permissible level, biological diversity is dramatically reduced. This human dominance means that the humanity is responsible for all the havoc wreaked on the earth , atmosphere, stratosphere, and humanity itself. We observed tremendous irreversible havocs and damages wreaked on the planet, and on humanity. The whole planet has endured so big changes that scientists agree to name the current period of the planet’s existence as Anthropocene. E. Fromm alerted a time ago that for the first time in the history the physical survival of the human race depends on a radical change of human heart. It was allegedly A. Einstein, who observed that we cannot solve our problems with the same thinking we used when we created them. It is not difficult to discover what kind of thinking brought humanity on the edge of a precipice. Paradoxically, it is thinking of the highly glorified, the so-called modern science. It was irresponsible transfer of Newtonian and Cartesian thinking from natural sciences into social sciences. As it is well known, the so-called modern science is based on a mechanistic worldview. However, neither ecosystem, nor society is a mega-machine, for which one has a spare parts. People are not equal self-loving atoms separated one from another. Particularly, it is economism, world-wide religion with its basic dogma of consumerism that is responsible for any kind of society’s desertification and biosphere destroying. Market is not an instrument for reaching equality and freedom, ruled by a providential Invisible Hand, but it is a place where mighty exploit the weak (L. Bruni). Human being is a social being, and outside the society can be either beast or god. People are living social atoms, with a Trinitarian nature constituting basic elements of social cells, i.e. family units, which in their turn constitute building material for social tissue. Society is therefore a living organism, having its proper organs. Lotka considered it as the “one huge multiple Siamese twin”. Economy is to be conceptualized as one of various of its exosomatic organs, designated by society and controlled by society. Like biological systems, social systems depend also on continuous throughput of energy and materials in order to maintain their structure and “life”. The concept of metabolism seems to be the unifying concept for any living organism, biological and socio-economical. This paper presents an introduction into economics, or as some prefer, oikonomics, as a science of economy considered as a society’s organ. Society, as a living organism, takes up ( through its organs) resources from the environment, transforms them into useful product and consumes, and excretes needles products of social metabolism again into environment. It is worthy to observe the drastic difference between mainstream economics and economics as a life science. The traditional, mainstreaming approach is based on the principle of using monetary units as the common denominator of all that is important in human life, such an approach was termed by B. Gross as a new philistinism. The alternative approach is based on the metabolic profiles, which are characterized by their material and energy flows. Instead of monetary measurement units, for the description of all society-nature relations there are used physical units. Although the development of a new approach is underway, there are already obtained significant results in measuring the human use of nature, as well as creating material and energy flow accountings.

ObjectivesThe aim of this paper was to investigate the impact of changes in the students’ lifestyle during an examination period on their health condition, as evidenced by changes to the body composition and metabolic rates.Material and MethodsThe study involved 268 Wroclaw Medical University students (second- and third-year students). The study was performed 3 times (at the beginning of a semester, in the middle of a semester, and in the examination period) and included a completion of a questionnaire on the current lifestyle, as well as measurements of the body composition, the basal metabolic rate, and the waist and hip circumferences. The measurements were carried out with the use of a body composition analyzer (Tanita MC-780 MA) and a tape measure with a waist–hip ratio calculator.ResultsIn the examination period, as compared with the rest of the semester, a significant decrease was observed in the metabolic rates, and the body water and muscle mass of the examined students. During that period, there were also more frequent adverse changes in the students’ lifestyle. In the studied periods, a correlation was found between changes in the frequency of snacking, consuming sweets and cola drinks, and the alterations of the subjects’ body fat mass, i.e., r = 0.161, r = 0.135, and r = 0.143, respectively. In turn, differences in the frequency of drinking wine, physical activity, sleep length, and time devoted to learning correlated with changes in the participants’ body water, i.e., r = 0.140, r = 0.152, and r = 0.133 (r = −0.142, r = −0.147), respectively, and muscle mass, i.e., r = 0.141, r = 0.142, and r = 0.126 (r = −0.130, r = −0.142), respectively, and metabolism, i.e., r = 0.127, r = 0.145 and r = 0.135 (r = −0.127, r = −0.135), respectively.ConclusionsThe analysis of the body composition and metabolic rates of the students during an examination period indicates that the changes occurring in various elements of their lifestyle (e.g., dietary habits, physical activity) can result in deteriorating their health condition.

The findings of the latter disciplines may and should be useful not only in economics but also in anthropology and metaphysics, which are able to provide answers to the fundamental questions about the roots of human honesty, confidence and willingness to help. Behind this question there is another one: what is the human Self, which is able to trust in and to provide the necessary support to the Other if needed, and how is it formed? The author of the article answers these questions with reference to the ideas of two eminent contemporary thinkers, Emmanuel Levinas (1906–1995) and Antoni Kępiński (1918–1972). Capturing the consistencies, similarities and differences between Kempiński’s description of the situation of a person with schizophrenia and the anthropological vision of Levinas’ philosophy is possible thanks to a scheme composed of three elements: the Self, the border, and the space-time order and hierarchy of values. If the Other is perceived as someone responsible for the Self, then it gives birth to trust of the Self in the Other, i.e. a strong conviction that the Other will not fail and is utterly honest with regard to the Self. Such a conviction is the foundation on which the Self builds a relationship with the Other. In this way, social capital, which is vital for the further development of society, is created. The capital is interfused by trust being the variable without which it is impossible to explain many economic phenomena.

Celem pracy było przedstawienie charakterystyki fenyloketonurii jako rzadkiej choroby metabolicznej. Materiał i metody. Metoda zastosowana w pracy to analiza literatury. Przegląd. Istotą choroby jest brak lub znaczny deficyt aktywności enzymu hydroksylazy fenyloalaninowej (w skrócie PAH), który wytwarzany jest wątrobie. Deficyt prowadzi do zaburzeń przemiany fenyloalaniny (Phe) w tyrozynę. Konsekwencją takiej reakcji jest pojawiająca się w surowicy dziecka nadmierna ilość aminokwasu, co w ostatecznie prowadzi do znacznych uszkodzeń ośrodkowego układu nerwowego. Wnioski. W dzisiejszych czasach osoby chore na Fenyloketonurię mają ogromne szanse na prawidłowy rozwój intelektualny, a sama choroba nie ma wpływu na poziom wykształcenia czy karierę zawodową. Sukces badań przesiewowych jest niewątpliwie namacalny, gdyż poprzez szybką diagnozę daje szansę chorym pacjentom na normalne życie.
Introduction. Phenylketonuria, a metabolic disease, diagnosed late and untreated leads to severe disability. Aim. The aim of the study was to present the characteristics of phenylketonuria as a rare metabolic disease. Material and methods. The method used in the work is the analysis of literature. Overview. The essence of the disease is the lack or a significant deficit in the activity of the enzyme phenylalanine hydroxylase (PAH for short), which is produced by the liver. The deficit leads to disturbances in the transformation of phenylalanine (Phe) into tyrosine. The consequence of such a reaction is an excessive amount of the amino acid appearing in the child's serum, which ultimately leads to significant damage to the central nervous system. Conclusions. Nowadays, people suffering from Phenylketonuria have a great chance for proper intellectual development, and the disease itself does not affect the level of education or professional career. The success of screening is undoubtedly tangible, because through a quick diagnosis it gives patients a chance for a normal life.

In order to understand the roles of abscisis acid in soybean exposed to UV-C radiation, soybean seedlings were exposed to UV-C radiation and ABA was applied. The chlorophyll content, active oxygen metabolism and flavonoids content of soybean seedlings were investigated. The present study revealed that UV-C radiation significantly attenuated the chlorophyll content and suppressed PSII activity. It was observed that the contents of H2O2, malondialdehyde and O2 - were markedly increased, and the activities of antioxidant enzymes such as superoxide dismutase, peroxidase and catalase were enhanced, so were the flavonoids content, whereas the seedling height and biomass were significantly reduced. ABA application improved the activities of superoxide dismutase, peroxidase, catalase and the contents of flavonoids, decreased the contents of H2O2, MDA and O2 -, and also increased the chlorophyll content and enhanced PSII performance, thus enhancing biomass accumulation. The results suggested that ABA alleviated UV-C damage to soybean seedlings by reducing ROS generation and enhancing antioxidase activity. The isoflavones daidzin and genistein were significantly responsive to UV-C.

Yeast metabolism has been a subject of research since the XIX century, when Louis Pasteur had proved that yeast are live organisms and fermentation is one of the processes to produce molecular energy. Yeast ability to ferment is a consequence of alcohol dehydrogenase (ADH) activity, the group of enzymes responsible for sugar conversion in to ethanol as ADH1, and alcohol metabolism as a source of carbohydrates (ADH2). The group of ADH enzymes play a crucial role for yeast adaptation especially in rich sugar environments such as after angiosperm occurrence. Evolution of yeast genes is based on chromosomal multiplication event, previously explained as a Whole Genome Duplication (WGD), presently it has been proven that the yeast common ancestor is a hybrid from different strains. Duplication cause that some multiplaies genes accumulate mutation without cell growth burden and consequently favor formations exploiting new features of proteins. Yeast ability to ferment make them dominate in a sugar environment, however Saccharomyces cerevisiae evolve some quorum-sensing mechanism, which plays crucial role in population messengers as tyrosol which is signaling about cellular density in culture. Moreover there are scientific reports about yeast specific social-wraps which suggest that yeast surviving depends on more complex mechanisms, which pose challenges for researchers. The aim of this article is a short review of yeast metabolic strategy which allow yeast to dominate in high sugar environments and abilities which make yeast so important in nature.

The βKlotho gene was identified on 2000, based on sequence similarity with the αKlotho gene. The KLB gene encoded single-pass transmembrane protein, which is expressed predominantly in liver and white adipose tissue. KLB–/–mice, unlike αKL–/– mice, grow and develop quite normally, but they exhibit increased bile acid synthesis and resistance to gallstone formation. Similar phenotype was also observed in mice lacking FGFR4 or FGF15. These observations led to hypothesis, that βKlotho interacts with FGFR4 and functions as a coreceptor for FGF15. Subsequent studies have shown, that βKlotho form complex with FGFR1c and function as coreceptor for FGF21. It was found, that via FGF-FGFR signalling pathway βKlotho can regulate many cellular processes. Functions as coreceptor for FGF19 (the orthologous protein in humans), KLB can regulate inter alia bile acids metabolism and energy homeostasis. Function as coreceptor for FGF21 protein can mediate promotion of lypolisis in white adipose tissue and ketogenesis in liver.
Gen βKlotho (KLB) odkryty został w roku 2000, jako homolog genu αKlotho. Gen KLB koduje monotopowe białko przezbłonowe, ulegające ekspresji głównie w wątrobie i białej tkance tłuszczowej. W odróżnieniu od myszy αKL–/–, myszy KLB–/– rozwijają się prawidłowo, jednakże wykazują one znaczny wzrost biosyntezy kwasów żółciowych oraz oporność na tworzenie się kamieni żółciowych. Podobny fenotyp obserwuje się również u myszy z wyłączonym genem FGFR4 lub FGF15. Dane te były podstawą sugestii, że białko βKlotho oddziałuje z FGFR4 i pełni rolę koreceptora dla FGF15. Dalsze analizy pokazały, że βKlotho tworzy również kompleks z receptorem FGFR1c, funkcjonując jako koreceptor dla czynnika FGF21. Wykazano, że poprzez działanie na szlaku FGF-FGFR, białko βKlotho zaangażowane jest w regulację licznych procesów komórkowych. Funkcjonując jako koreceptor dla czynnika FGF19 (ortolog FGF15 u człowieka), KLB wpływa m.in. na metabolizm kwasów żółciowych oraz homeostazę energetyczną. Natomiast jako koreceptor dla FGF21, białko KLB wpływa na promocję lipolizy w białek tkance tłuszczowej oraz ketogenezy w wątrobie.

Diabetes is one of the major challenges of modern medicine, as it is considered a global epidemic of the XXI century. The disease often leads to the development of serious, health threatening complications. Diabetic foot syndrome is a characteristic set of anatomical and molecular changes. At the macroscopic level, major symptoms are neuropathy, ischemia and chronic ulceration of the lower limb. In every third patient, the neuropathy develops into Charcot neuroarthropathy characterized by bone and joints deformation. Interestingly, all these complications are a result of impaired healing processes and are characteristic for diabetes. The specificity of these symptoms comes from impaired molecular mechanisms observed in type 1 and type 2 diabetes. Decreased wound and fracture healing reflect gene expression, cellular response, cell functioning and general metabolism. Here we present a comprehensive literature update on the molecular factors contributing to diabetic foot syndrome.

Bilirubin is degraded in the human gut by microflora into urobilinoids. In our study we investigated whether the bilirubin-reducing strain of Clostridium perfringens can reduce bilirubin ditaurate (BDT), a bile pigment of some lower vertebrates, without hydrolysis of the taurine moiety. C. perfringes was incubated under anaerobic conditions with BDT; reduction products were quantified by spectrophotometry and separated by TLC. Based on Rf values of BDT reduction products and synthetic urobilinogen ditaurate, three novel taurine-conjugated urobilinoids were identified. It is likely that bilirubin-reducing enzyme(s) serve for the effective disposal of electrons produced by fermentolytic processes in these anaerobic bacteria.

DNA methylation is a reversible epigenetic modification that plays a crucial role in transcriptional gene silencing. Both excessive (hypermethylation) and reduced DNA methylation (hypomethylation) can contribute to the disturbance of the proper course of many important processes in the human body. The aim of the study was to discuss the relationship between methyl nutrients and the DNA methylation process in the course of selected diseases in adults. Methyl nutrients include folates (vitamin B9), riboflavin (vitamin B2), cobalamin (vitamin B12), pyridoxine (vitamin B6) and choline (vitamin B4), as well as methionine and betaine. These substances play the role of both substrates and cofactors in transformations related to one-carbon metabolism. The deficiency of methyl nutrients in the body can lead to disturbances in SAM synthesis, which is the primary donor of methyl groups in the DNA methylation process. However, the mechanism explaining the discussed relationship has not been fully explained so far. Both the concentration in the body and the intake of folate and vitamin B12 in the diet can, to some extent, have an effect on the level of DNA methylation in healthy people. In comparison, data on the effect of excessive intake of vitamin B12 in the diet on the risk of cancer development are inconsistent. An adequate betaine and choline intake in the diet might not only affect the overall improvement of the DNA methylation profile, but, to some extent, also reduce the risk of cancer, the effect of which can depend on the content of folic acid in the body. Research results on the effect of supplementation of methyl nutrients on the DNA methylation process are inconclusive. It is therefore necessary to conduct further research in this area to draw clear conclusions

Type 2 diabetes mellitus (T2D) is a chronic diet-related disease which due to many dangerous complications has become a prominent health problem of the world. The aim of the study was to explore the in vitro activity of Japanese quince (Chaenomeles japonica L., family Rosaceae, JQ) fruit polyphenolic extract as modulator of carbohydrates metabolism. The research was designed to investigate the effect of JQ polyphenolic extract on glucose metabolism in human hepatoma HepG2 cell line cultured under normal non-metabolically changed and hyperglycemic conditions. Pretreatment of the cells with JQ preparation caused decrease of intracellular ROS generation and influenced mitochondrial membrane polarization which seemed to lead to AMPK activation. Further effects observed in HepG2 cells were associated with activation of the enzyme: elevation of glucose uptake and glycogen content, and alleviation of gluconeogenesis through modulation of PEPCK, PTP1B, FOXO1 and GLUT2/4 expression. These findings suggest that JQ polyphenols exhibit hypoglycemic effects via modulation of AMPK signaling in hepatocytes.

α-Amylase (EC 3.2.1.1) was purified from the muscle and intestine of the parasitic helminth of pigs Ascaris suum. The enzymes from the two sources differed in their properties. Isoelectric focusing revealed one form of α-amylase from muscles with pI of 5.0, and two forms of amylase from intestine with pI of 4.7 and 4.5. SDS/PAGE suggested a molecular mass of 83 kDa and 73 kDa for isoenzymes of α-amylases from intestine and 59 kDa for the muscle enzyme. α-Amylase from intestine showed maximum activity at pH 7.4, and the enzyme from muscle at pH 8.2. The muscle enzyme was more thermostabile than the intestinal α-amylase. Both the muscle and intestine amylase lost half of its activity after 15 min at 70°C and 50°C, respectively. The Km values were: for muscle amylase 0.22 μg/ml glycogen and 3.33 μg/ml starch, and for intestine amylase 1.77 μg/ml glycogen and 0.48 μg/ml starch. Both amylases were activated by Ca2+ and inhibited by EDTA, iodoacetic acid, p-chloromercuribenzoate and the inhibitor of α-amylase from wheat. No significant differences were found between the properties of α-amylases from parasites and from their hosts.

Modern metabolic engineering is based on the tools of systems biology and synthetic biology. Mathematical modelling plays an important role in the research related to metabolic network modification, because it enables one to analyze the biological system in a quantitative manner. The flux balance analysis enables one to determine flux values in the network by maximizing the biological objective function. Algorithms of in silico metabolic engineering are based on optimization procedures.

Studies suggest a variety of biological effects for isoflavones and anthocyanins, but there is little information regarding small intestinal absorption and metabolism. In this article, absorption and metabolism studies of selected phytochemicals, especially isoflavones and anthocyanins are reviewed. To assess small intestinal absorption an isolated preparation of luminally and vascularly perfused rat small intestine was used, with a synthetic perfusate free from blood components as vascular medium. Luminal media consisted of a buffered sodium chloride solution spiked with the phytochemicals. Viability and functional integrity of the organ preparation was maintained during the entire perfusion. Venous and luminal aliquots were analyzed for the phytochemicals with HPLC. Complex foods (tofu) were predigested with an in vitro digestion system using pepsin and pancreatin before perfusion experiments. Vascular uptake of the lipophilic isoflavone genistein was higher than for the more hydrophilic glycosides genistin, daidzin and cyanidin-3-glucoside. The isoflavone glycosides were hydrolyzed to their aglycons, which appeared at the vascular and luminal side. In contrast, anthocyanidin glucosides were not cleaved during perfusion experiments. All of the investigated phytochemicals were partly glucuronidated. These conjugates were vascularly absorbed and luminally secreted. Tofu ingredients influenced isoflavone uptake, extent of glucuronic conjugation and distribution of glucuronides in the small intestine. Ethanol, in contrast, had no significant influence on cyanidin-3-glucoside absorption. The investigated phytochemicals are absorbed at various extent in the small intestine, partly as conjugates. The implication of a simulated digestion and the use of the isolated perfused rat small intestine are suitable tools to investigate and evaluate the influence of food matrix on the intestinal handling of phytochemicals.

Opis:
Both ghrelin and obestatin are peptides derived from the same precursor – preprohormone – encoded on the 3rd chromosome (3p25-26). Both hormones are secreted to the bloodstream. Ghrelin has 28-amino acids with serine at position 3. It is an endogenous ligand of growth hormone receptor (GHS), which has been discovered in hypophysis and hypothalamus. Ghrelin may occur in active and inactive forms. In order to achieve biological activity, ghrelin must contain a N-octa-acetyl group of serine. Total ghrelin is a sum of active and inactive forms. Ghrelin and its receptors GHS-R1a and 1b (growth hormone secretagogue) are widespread in the body, being present mainly in the gastrointestinal tract, central nervous system, reproductive system, heart and kidneys. Obestatin is a recently discovered 23-amino acids long peptide, produced as a result of proteolytic splitting of the preprohormone ghrelin. Systemic distribution of ghrelin is less well known. Both ghrelin and obestatin play a role in energy management (control appetite, body mass, metabolism of fat and glucose, gastrointestinal function), influence cardiovascular, reproductive and immune systems and participate in modulation of central nervous system function. Ghrelin and obestatin regulate processes of cellular proliferation and apoptosis. Variations of ghrelin and obestatin levels, as well as expression of GHS-R in hypophyseal and neuroendocrine tumors, uterine myomas and both benign and malignant ovarian tumors, confirm their role in tumor development and are a promising topic for future studies in oncology.
Ghrelina i obestatyna są peptydami wywodzącymi się z tego samego prekursora – preprohormonu, który kodowany jest na chromosomie 3. (3p25-26); wydzielane są do krwi. Ghrelina to 28-aminokwasowy peptyd, zawierający serynę w pozycji 3. Jest endogennym ligandem receptora wzrostu (GHS), który wykryto w przysadce i podwzgórzu. Występuje ona w formie aktywnej i nieaktywnej. Aby otrzymać aktywność biologiczną, ghrelina musi zawierać N-oktaacetylową grupę seryny. Ghrelina całkowita to suma form aktywnej i nieaktywnej. Ghrelina i jej receptory GHS-R1a i 1b (growth hormone secretagogue) są szeroko rozpowszechnione w organizmie i występują między innymi w przewodzie pokarmowym, ośrodkowym układzie nerwowym, układzie rozrodczym, sercu i nerkach. Obestatyna to ostatnio wykryty 23-aminokwasowy peptyd powstały na skutek proteolitycznego rozszczepienia preprohormonu ghreliny. Dystrybucja obestatyny w organizmie jest mniej znana. Ghrelina i obestatyna biorą udział w kontroli bilansu energii (apetyt, masa ciała, metabolizm tłuszczów i glukozy, funkcje żołądkowo-jelitowe), a także odgrywają rolę w układzie sercowo-naczyniowym, rozrodczym, modulacji immunologicznej i ośrodkowym układzie nerwowym. Regulują procesy proliferacji i apoptozy komórek. Zmiany stężeń ghreliny i obestatyny oraz/lub ekspresja GHS-R w guzach przysadki mózgowej, guzach neuroendokrynnych przewodu pokarmowego (NET), mięśniakach macicy oraz guzach łagodnych i złośliwych jajnika wskazują na ich udział w rozwoju nowotworów i są obiecującym obiektem badań w onkologii.

Opis:
Sulfur is an essential macronutrient for all living organisms. Plants are able to assimilate inorganic sulfur and incorporate it into organic compounds, while animals rely entirely on organic sources of sulfur. In the last decades sulfate availability in soils has become the major limiting factor for plant production in many countries due to significant reduction of anthropogenic sulfur emission forced by introducing stringent environmental legislation. The sulfur flux after transferring plants from optimal conditions to sulfur deficiency is regulated on multiple levels including transcription, translation and activity of enzymes needed for sulfate assimilation and synthesis of sulfur-containing metabolites. Most of these regulatory steps are not yet fully characterized. Plant responses to sulfur limitation are complex and can be divided into phases depending on the degree of sulfur shortage. The initial responses are limited to adaptations within sulfur metabolic pathway, while multiple metabolic pathways and developmental process are affected when sulfur shortage becomes more severe. The major aim of this work is a comprehensive review of recent progress in understanding the regulation of plant adaptations to sulfur deficit.

Opis:
Advances in studies on vitamin D reveal increasingly wider possibilities for its use in the prevention and even treatment of many diseases. Vitamin D is increasingly more frequently called prohormone or cytokine, and nowadays it is not only associated with the regulation of calcium and phosphate metabolism. It is now known that because of the fact that vitamin D has an influence on the production of cytokine, it reduces the incidence of autoimmune illnesses such as diabetes type 1. Vitamin D is also used in the prevention and treatment of cardiovascular diseases, tumors and depression. Changes in metabolism, insufficient acquisition of this compound during skin synthesis, abnormal biochemical changes to the active form, or inadequate dietary intake of this compound may disrupt many important processes for maintaining health.
Postępy w badaniach nad witaminą D ujawniają coraz szersze możliwości jej wykorzystania w zapobieganiu, a nawet leczeniu wielu jednostek chorobowych. Witamina D jest coraz częściej określana również jako prohormon lub cytokina, obecnie nie jest już tylko kojarzona z regulacją gospodarki wapniowo-fosforanowej, wiadomo, że wpływając na produkcję cytokin zmniejsza częstość występowania chorób autoimmunologicznych, np. cukrzycy typu 1. Witamina D ma również zastosowanie w profilaktyce i leczeniu nowotworów, chorób układu krążenia oraz depresji. Zmiany w metabolizmie, zbyt małe pozyskiwanie tego związku w trakcie syntezy skórnej, zaburzenia przemian biochemicznych do formy aktywnej czy niedostateczna podaż w diecie tego związku mogą zaburzyć wiele ważnych dla zachowania zdrowia procesów.

Members of Candida species cause significant problems in medicine and in many industrial branches also. In order to prevent from Candida sp. development, essential oils are more and more frequently applied as natural, non-toxic, non-pollutive and biodegradable agents with a broad spectrum of antimicrobial activity. The aim of the research was to determine changes in morphology and metabolic properties of Candida albicans in the presence of thyme and tea tree oils. Changes of enzymatic activity of isolates were observed in the presence of both tested essential oils, and they were primarily associated with loss or decrease of activity of all enzymes detected for control. Furthermore, only for 3 out of 11 isolates additional activity of N-acetyl-β-glucosaminidase, α-mannosidase, α-fucosidase and trypsin was detected. Vivid changes in biochemical profiles were found after treatment with tea tree oil and they were related to loss of ability to assimilate d-xylose, d-sorbitol and d-trehalose. The main differences in morphology of isolates compared to the control strain concerned formation of pseudohyphae structures. Both examined essential oils caused changes in cell and colony morphology, as well as in the metabolism of Candida albicans. However, the extent of differences depends on the type and concentration of an essential oil. The most important finding is the broad spectrum of changes in yeast enzymatic profiles induced by thyme and tea tree oils. It can be supposed that these changes, together with loss of ability to assimilate saccharides could significantly impact Candida albicans pathogenicity.

Tetrahydrobiopterin (BH4) has been recently approved as a treatment of patients with phenylketonuria. However, as a confirmation of BH4-responsiveness, it might require a very expensive trial treatment with BH4 or prolonged BH4-loading procedures. The selection of patients eligible for BH4-therapy by means of genotyping of the PAH gene mutations may be recommended as a complementary approach. A population-wide genotyping study was carried out in 1286 Polish phenyloketonuria-patients. The aim was to estimate the BH4 demand and to cover prospectively the treatment by a National Health Fund. A total of 95 types of mutations were identified. Genetic variants corresponding with probable BH4-responsiveness were found in 28.2% of cases. However, patients with mild or classical phenylketonuria who require continuous treatment accounted for 11.4% of the studied population only. Analysis of the published data shows similar percentage of the "BH4-responsive" variants of a PAH gene in patients from other countries of Eastern Europe. Therefore, it can be concluded, that the proportion of phenylketonuria-patients who could benefit from the use of BH4 reaches approximately 10% in the entire region.

Insufficient treatment adherence after early childhood is frequently observed in patients with phenylketonuria. Assessment of these individuals' long-term metabolic control could enable early detection of the risk of intellectual deterioration resulting from high blood phenylalanine concentration. However, the predictive value of specific parameters related to individual dynamics of hyperphenylalaninemia is not clear. Here, we assessed the impact of blood phenylalanine fluctuations during the first 12 years of life on cognitive outcome in early and continuously treated teenagers with phenylketonuria. We have analyzed a total of 5141 results of blood phenylalanine measurements in 32 patients. The phenylalanine levels of these patients were usually acceptable during their early childhood, but the control of hyperphenylalaninemia worsened and the average treatment adherence dropped to 40% during the late primary school. Our analysis revealed a strong association between the Wechsler intelligence verbal scores and the mean of the yearly means of phenylalanine concentrations (r=-0.62). The correlations of IQ scores with median phenylalanine concentrations and the variability of blood phenylalanine levels gave weaker associations. The Wechsler verbal scores were also strongly correlated with the treatment adherence level during preschool and late primary school (r=0.61 and 0.72). The mean of the yearly means of blood phenylalanine concentrations appears to be a better predictor of cognitive outcome in children with phenylketonuria than other parameters related to phenylalanine fluctuations. The percentage of acceptable phenylalanine levels below 50-60% should be regarded as a "red flag" due to the risk of intellectual deterioration in patients.

Dinitrofenol (DNP) – mieszanina izomerów: 2,3- DNP, 2,4- DNP i 2,6-DNP z przewagą 2,4-DNP, jest stosowany w produkcji barwników, kwasu pikrynowego i pikraminowego, wywoływaczy fotograficznych i materiałów wybuchowych oraz jako pestycyd w rolnictwie i sadownictwie. Dinitrofenol jest trucizną metaboliczną, a mechanizm jego działania toksycznego polega na rozprzęganiu fosforylacji oksydatywnej. Zawodowe narażenie na pary i pyły 2,4-DNP może wywoływać objawy wzmożonego metabolizmu. 2,4-DNP nie jest kancerogenem, nie wykazuje także działania genotoksycznego ani mutagennego. Za podstawę wyliczenia wartości NDS dla 2,4-DNP przyjęto wartość LOAEL dla skutków metabolicznych. Wartość ta u człowieka wynosi 1,2 mg/kg/dzień. W warunkach narażenia drogą oddechową taką dawkę pracownik może wchłonąć, gdy stężenie 2,4-DNP we wdychanym powietrzu wynosi 10,5 mg/m3. Przyjmując współczynnik niepewności równy 16 (2 dla różnic we wrażliwości osobniczej, 2 dla przejścia z wartości LOAEL do wartości NOAEL, 2 dla różnicy w sposobie narażenia i 2 dla ekstrapolacji z narażenia średnioterminowego do przewlekłego), to wyliczona wartość normatywu będzie wynosiła 0,66 mg/m3. W związku z powyższym proponuje się przyjęcie stężenia 0,5 mg/m3 za wartość NDS dla 2,4-DNP. Przyjmując, że 2,4-DNP jest najbardziej toksycznym izomerem dinitrofenolu oraz, że udział tego izomeru w dinitrofenolu – mieszaninie izomerów jest dominujący, proponujemy przyjęcie dla dinitrofenolu – mieszaniny również takiej samej wartości NDS jaką zaproponowano dla 2,4-DNP.
Dinitrophenol (DNP) is a mixture of 2,4-DNP and smaller amounts of 2,3-DNP and 2,6-DNP). It is a yellow, crystalline solid. DNP is used in synthesis of dyes, picric acid, picramic acid, wood preservatives, photographic developers, explosives, and insecticides. In the 1930s, 2,4-DNP was used as a weight-reducing drug. Short-term exposure to DNP may affect metabolism resulting in hyperthermia. High-level exposure may be fatal. The existing data concerning the health effects of 2,4-DNP oral exposure in humans indicate that the characteristic effects of 2,4-DNP for this route are: increased basal metabolic rate and perspiration, weight loss, a sensation of warmth, and – at higher dosage – increased heart and respiratory rate, and increased body temperature. Repeated or prolonged contact with the skin may cause dermatitis. Exposure to DNP may result in changes in the functional state of the peripheral nervous system, cardiovascular system and gastrointestinal system. It may also induce cataracts. Taking into account the results obtained in clinical studies on people ingesting 2,4-DNP (LOAEL for metabolic effects was 1.2 mg/kg/day), a concentration 0.5 mg of dinitrophenol/m3 is proposed as a maximum exposure limit (maximum admissible concentration) with a skin notation. With regard to systemic effects of DNP no STEL value has beeen established.
