The effect of ketotifen on inflammation in the intestine during T. spiralis infection in BALB/c mice was examined. The lack of enteropathy in treated mice was accompanied to affected the size of worms and the number of worms, but did not prevent the retardation of nematodes in the intestine. Fecundity of females in vitro and the number of muscle larvae were not statistically changed after treatment. Ketotifen reduced the number of mast cells and IgA plasma cells in the intestine. As inflammatory influx, epithelial damage and IgA response are related to the action of T cells, ketotifen affected T dependent reactions in the gut of T. spiralis infected mice.

Mast cells play an important role in both, the innate and adaptive immunity, however, clonal proliferation of abnormal mast cells in various organs leads to mastocytosis. A skin variant of the disease, cutaneous mastocytosis (CM) is the most frequent form of mastocytosis in children. HtrA proteases are modulators of important cellular processes, including cell signaling and apoptosis, and are related to development of several pathologies. The above and the observation that mast cells constitutively release the HtrA1 protein, prompted us to investigate a possible involvement of the HtrA proteins in pediatric CM. Levels of the serum autoantibodies (IgG) against the recombinant HtrA proteins (HtrA1-4) in children with CM (n=36) and in healthy controls (n=62) were assayed. Anti-HtrA IgGs were detected using enzyme linked immunosorbent assay (ELISA) and Western-blotting. In the CM sera, levels of the anti-HtrA1 and anti-HtrA3 autoantibodies were significantly increased when compared to the control group, while the HtrA protein levels were comparable. No significant differences in the anti-HtrA2 IgG level were found; for the anti-HtrA4 IgGs lower levels in CM group were revealed. In healthy children, the IgG levels against the HtrA1, -3 and -4 increased significantly with the age of children; no significant changes were observed for the anti-HtrA2 IgG. Our results suggest involvement of the HtrA1 and HtrA3 proteins in pediatric CM; involvement of the HtrA4 protein is possible but needs to be investigated further. In healthy children, the autoantibody levels against HtrA1, -3 and -4, but not against HtrA2, increase with age.

Opis:
Allergic diseases afflict many people worldwide. Allergic inflammation encompassing acute allergic reaction and chronic inflammatory response is the major pathogenesis. To discovery novel therapeutic approach for allergic diseases, we have evaluated the anti-allergic inflammatory effects of irisflorentin using RBL-2H3 cells. The results showed certain concentrations of irisflorentin could suppress histamine release and secretion of TNF-α, IL-1β and IL-6. Further investigations indicated that the effects of irisflorentin were associated with a reduction of intracellular calcium, suppression of NF-κB signaling pathway and inhibition of caspase-1 activity. Therefore, the results of the current study demonstrated that irisflorentin can ameliorate mast cell-mediated allergic inflammation. These results can provide evidences for the discovery of novel therapy for allergic diseases and application of irisflorentin in practice.

Introduction. Risky alcohol consumption is one of the main threats to the health of both individuals and society as a whole. These threats affect, among others, health, safety and financial situation. Their scale and intensity cause the issue of alcohol consumption to become a significant challenge for public health. Objective. The aim of the study was assessment of the occurrence of the problem of alcohol consumption among residents of rural areas in Western Pomerania and the Lubuskie Province of Western Poland. Materials and method. The study was conducted on 350 residents of rural areas – 236 females (66.4%) and 114 males (32.6%), median age – 42 y. (range – 20–76 years). The tools used included MAST and AUDIT questionnaires, Health Behaviour Inventory (HBI), Social Activity Questionnaire (SA), and original questions. Results. In the studied group the problem with proper alcohol consumption occurred. High convergence was observed between results obtained by individual respondents with both tools (r=0.868 with p<0.001). Median on the AUDIT scale indicated the possibility of alcoholism occurrence in the studied group, whereas the MAST test indicated the necessity to conduct an in-depth alcohol diagnosis. Factors that differentiated the points scored by respondents in AUDIT test included gender – more women drank alcohol in a risky way (Z=-2.54, p=0.011), education, health behaviours, professional passivity, and using various forms of support. In the AUDIT test, the alcoholism threat was over 7 times higher in people without permanent employment (vs. employed people), OR=7.41 (95% CI-4.56–12.04), and in MAST test it was over 4 times higher in these individuals: OR =4.45 (95% CI–2.64–7.49). Conclusions. Results indicate the need to intensify the work of regional facilities for solving alcohol problems, as well as educational campaigns in schools and health care facilities.

W pracy przedstawiono wyniki obliczeń wytrzymałościowych MES masztu wirnika nośnego wiatrakowca. Analiza została przeprowadzona za pomocą narzędzia obliczeniowego ABAQUS_CAE. Wykonane obliczenia umożliwiły przedstawienie map rozkładu naprężeń, wartości przemieszczenia masztu, kąta skręcenia w trakcie prerotacji naziemnej. Analizom poddano maszt głowicy wiatrakowca I-28 wariant B, wykonywanego w Instytucie Lotnictwa w ramach projektu „Technologia wdrożenia do praktyki gospodarczej nowego typu wiropłatowego statku powietrznego”. W modelu obliczeniowym masztu uwzględniono pracę antywibratora gumowego.
This article is about finite element method strength analysis of a gyroplane rotor head's mast. The studies were made in computational program ABAQUS_CAE. The analysis of numeric research results showed: reduced stress maps, mast's axis displacement values, mast's torsional angle at manoeuvre of ground prerotation. The object of the study is I-28 autogiro's mast, which was made in the Institute of Aviation in the framework of the project "Technologia wdrożenia do praktyki gospodarczej nowego typu wiropłatowego statku powietrznego". Rubber anti-vibrator work was taken into account in the computional model of the mast.

Mast cells, discovered by Paul Burnet over one century ago, have been long recognized only as inductors of IgE-dependent allergic diseases (allergy of type I, Th2 lymphocytes dependent). However, numerous recent studies have indicated that they play an essential role in many other immunological and non-immunological processes. Infection with Toxoplasma gondii elicits the induction of a strong cell-mediated immunity characterized by a highly-polarized Th1 response, which can protect against allergy. Knowledge of the contribution of mast cells to Toxoplasma invasion is still limited, and the present article discusses aspects of the relationship between mast cells and T. gondii.

The instantaneous tangential rigidity (corresponding to the applied horizontal load) of a triguy support with unequal ropes is determined. The differences between the ropes can be due to the different location, both horizontal and vertical, of their anchoring foundations and to the different types of ropes used. The horizontal rigidity of such a support has no general axis of symmetry. Therefore a formula for this rigidity in any direction and a formula for extreme rigidity angles are given in this paper. A notion of the guy’s eigenvalue, standing for the initial tension force above which the guy participates effectively in the support’s rigidity, is introduced. A numerical example is provided.

Próby obciążeniowe zmniejszają ryzyko wprowadzenia do eksploatacji układów nie spełniających zasadniczych wymagań bezpieczeństwa.
Mast and drawworks system for running and pulling drill string, holding it at drilling, casing and tubing running operations influences on mast/derrick load value. Additionally high load of drawworks can be exerted during rig up operation. Load tests of mast and drawworks performed give impact of loading forces on mast framework behaviour, eventuality of design defects and critical deformations. Load tests decrease risk for implementation of systems uncomplying with basic safety requirements

W artykule przedstawiono sposób wzmocnienia masztu oświetleniowego na terenie stacji Dąbrowa Górnicza Towarowa. Dokonano analizy stanu technicznego obiektu, opisu przedstawionych badań i obliczeń sprawdzających wg normy PN-90/B-03200 oraz zakresu wykonanych robót związanych ze wzmocnieniem masztu. Obliczenia wykonano w programie Robot Millenium 17.5. Sformułowano wnioski dotyczące metod monitorowania stanu technicznego masztu oświetleniowego oraz zaleceń dotyczących remontu.
The article presents the method of consolidation the lighting mast at the Dąbrowa Górnicza Towarowa station. The analysis of the technical condition of the facility, a description of the presented tests and verification calculations according to PN–90/B-03200 and the scope of works related to mast consolidation were made. The calculations were made in Robot Millenium 17.5. Conclusions on methods for monitoring the technical condition of the lighting mast and recommendations for renovation were formulated.

W pracy, na przykładzie pewnej realnej konstrukcji masztu o trzonie kratowym, przeprowadzono analizę wpływu osiadania fundamentu centralnego masztu na stan sił i przemieszczeń w maszcie. Obliczenia przeprowadzono dla stanu wstępnego sprężenia i obciążenia konstrukcji wiatrem. Na podstawie uzyskanych wyników obliczeń statycznych sformułowano pewne uwagi i wnioski końcowe, które mogą znaleźć zastosowanie w praktycznym projektowaniu masztów.
The influence of the mast foundation settlement on the static behaviour of the mast structure was analyzed on the basis of a lattice shaft mast. The calculations were made for the pre-stress condition and wind-load condition of the structure. Basing on the results of static calculations some remarks and conclusions were drawn.
