Purpose: The membrane-anchored MUC1 mucin is typically expressed on normal and cancerous epithelial cells. Non-epithelial localization of this mucin is rare. However, the presence of MUC1 in human skin fibroblasts has been recently unexpectedly revealed. The aim of the study was to prove the expression of MUC1 mucin in human skin fibroblasts and the examine of the influence of luteolin on its expression. Materials and methods: ELISA tests and real-time PCR analysis were used to assess the expression of MUC1 mucin in fibroblast cells cocultured with 30 μM concentration of luteolin. Results: The expression of MUC1 was revealed in human skin fibroblasts. Luteolin decreased the relative level of mucin in cell lysates and media. Statistically significant decreased expression of MUC1 gene after luteolin treatment of fibroblasts cells was also revealed. Conclusion: Our results prove non-epithelial localization of MUC1 mucin. Luteolin inhibits the expression of MUC1 mucin in healthy human skin fibroblasts.

Opis:
The aim of the present study was to determine the effects of Luteolin (LUT) on semen quality, oxidative stress, apoptosis, acrosomal integrity, mitochondrial membrane potential and dead sperm ratio in rabbits. Ejaculates from six New Zealand rabbits were collected, evaluated and pooled. The pooling was divided into five groups as control (no additive) LUT 25 μM, LUT 50 μM, LUT 100 μM and LUT 200 μM and LUT added. It was then filled into a falcon tube with Tris-based extender at a final concentration of approximately 35 x 106 spermatozoa. Diluated rabbit semen samples were drawn into frozen and thawed. Frozen semen straws were thawed at 37°C in 30 seconds. According to our findings, no statistical difference was found between all doses of luteolin and the control group in the CASA (computer assisted sperm analysis) analysis performed at 4°C. However, total motility, progressive motility and rapid sperm percentage were found to be higher in the frozen and thawed rabbit semen at a dose of LUT 50 μM compared to the other groups (p<0.05). While amplitude of lateral head displacement (ALH) and beat cross-frequency (BCF) values were found at the lowest dose of LUT 200 μM, a statistically significant difference was observed between the other groups. When the flow cytometry results were examined, no statistical difference was found between the rate of dead sperm, acrosomal integrity, mitochondrial membrane potential and apoptosis rate. Morever, the H2O2 percentage was found to be lower in all experimental groups compared to the control group (p<0.001). In conclusion, the addition of LUT in long-term storage of rabbit semen provided a protective effect for spermatozoa with its antioxidative properties against damage caused by cryopreservation.

The aim of this work was to study the polyphenolic composition of Deschampsia antarctica È. Desv. plants grown at natural conditions on different locations on the Galindez Island, Argentine Islands, the maritime Antarctic. The plants were collected during the summer season of the 26th Ukrainian Antarctic Expedition (2020–2022). The extracts of 21 plants were obtained and the composition of the extracts was analyzed by means of high-performance liquid chromatography and matrix-assisted laser desorption/ionization mass spectrometry. The antioxidant properties of the extracts were characterized using the DPPH (2,2-diphenyl-1-picrylhydrazyl) test. The extracts were found to contain large amount of polyphenolic compounds, with flavonoids and phenolic acids, as well as their derivatives, being the most common classes of the phenols. Using the HPLC data the content of various phenols in the plants was systematic studied. It has been found that in all plants the most abundant phenols are flavonoids/flavonoid derivatives (on average about 75% of total mass of phenols). Among the flavonoids, luteolin derivatives predominate (86–94% of the total mass of flavonoids), and, among luteolin derivatives, the main compounds are orientin, orientin 2"- O-β-arabinopyranoside and isoswertiajaponin 2"- O-β-arabinopyranoside (67–83% of the total mass of luteolin derivatives). It has been also found that all the extracts possess the high activity in inhibition of DPPH radicals and that the antioxidant activity of the extracts correlates with total content of phenols in the samples. Thus, Deschampsia antarctica É. Desv. plants are a valuable source of natural phenolic antioxidants, and the most common antioxidants in the extracts are orientin, orientin 2"- O-β-arabinopyranoside and isoswertiajaponin 2"- O-β-arabinopyranoside.
