The glycine receptor belongs to the ligand-gated ion channel superfamily. It is a chloride conducting channel composed of four transmembrane domains. It was previously shown that the second transmembrane domain (M2) of the glycine receptor forms an ion conduction pathway throught lipid bilayers. The amino-acid sequence of the transmembrane segment M2 of the glycine receptor has a high homology to all receptors of the ligand-gated ion channel superfamily. In our report, we have used a synthetic M2 peptide. It was incorporated into a planar membrane of known lipid composition and currents induced by M2 were measured by the Black Lipid Membrane technique. When the planar lipid bilayer was composed of 75% phosphatidylethanolamine and 25% phosphatidylserine, the reversal potential measured in a 150/600 mM KCl (cis/trans) gradient was -19 mV suggesting that the examined pore was preferential to anions, PK/PCl = 0.25. In contrast, when 75% phosphatidylserine and 25% phosphatidylethanolamine was used, the reversal potential was +20 mV and the pore was preferential to cations, PK/PCl = 4.36. Single-channel currents were recorded with two predominant amplitudes corresponding to the main-conductance and sub-conductance states. Both conductance states (about 12 pS and 30 pS) were measured in a symmetric solution of 50 mM KCl. The observed single-channel properties suggest that the selectivity and conductance of the pore formed by the M2 peptide of the glycine receptor depend on the lipid composition of the planar bilayer.

Penetration of the liposome membranes doped with vanadium complex formed in the liquid-crystalline phase from egg yolk lecithin (EYL) by the TEMPO (2,2,6,6-tetramethylpiperidine-1-oxyl) spin probes has been investigated. The penetration process was followed by 360 hours at 24◦C, using the electron spin resonance (EPR) method. The spectroscopic parameter of the partition (F) of this probe indicated that a maximum rigidity of the membrane was at 3% concentration of the vanadium complex. Computer simulations showed that the increase in the rigidity of the membrane corresponds to the closure of gaps in the surface layer of the membrane, and indicates the essential role of the membrane surface in transport processes.

Opis:
Amphotericin B (AmB) is a well known polyene macrolide antibiotic used to treat systemic fungal infections. Despite its toxicity AmB is still regarded as a life-saving drug. The lack of adequate knowledge of the AmB mechanism of action is a serious obstacle to efficient development of new less toxic derivatives. Complementary to various experimental approaches, computational chemistry methods were used to study AmB mechanism of action. A programme lasting for a decade, that was run by our group covered studies of: i) molecular properties of AmB and its membrane targets, ii) structure and properties of AmB membrane channels, and iii) interaction of AmB with the membrane.

Electrochemical impedance spectroscopy was used for the study of two-component lipid membranes. Phosphatidylcholine and ceramide were to be investigated, since they play an important biochemical role in cell membranes. The research on biolipid interaction was focused on quantitative description of processes that take part in a bilayer. Assumed models of interaction between amphiphilic molecules and the equilibria that take place there were described by mathematical equations for the studied system. The possibility of complex formation for two-component system forming bilayers was assumed that could explain the deviation from additivity rule. Equilibria were described by mathematical equations that were further verified experimentally. The determined values of parameters (stability constant, molecular area of complex, capacitance and conductance of the lipid membranes formed from molecules and complexes) were used for calculation of model curves. The comparison of model curves and experimental points verified the assumed model.

Opis:
General anesthesia is defined as impairment of the central nervous system (UON) caused by intravenous or volatile anesthetics. The state of loss of consciousness or even amnesia and the disappearance of perception into external stimuli is achieved by the use of a large group of chemical compounds. The use of nitrous oxide in 1844 revolutionized surgery and medicine at that time. From that moment, anesthesiology develops dynamically, allowing more and more complex procedures. Despite more than 170 years of history of anesthesia, understanding the mechanism of reversible loss of awareness and sensitivity to pain caused by the action of general anesthetics is one of the greatest challenges of modern pharmacology and neuroscience. Incredibly high diversity of anesthetics, including both noble gases and complex steroids, combined with human sensation makes the above problem extremely difficult to solve. The reversibility of the anesthesia phenomenon suggests that the analyzed phenomenon is based on disturbance of weak intermolecular interactions, such as hydrogen bond or van der Walls forces. Anesthetic molecules may bind directly to the hydrophobic region of protein, which causes its conformational changes or disturb ion channel activity by anesthetic-induced perturbations of lipid bilayers. The mechanism of anesthesia is thus very often attributed to both protein and lipid membrane targets. The influence of anesthetic molecules on biomolecular systems can be studied successfully using many different physico-chemical methods, such as, infrared, fluorescence or nuclear magnetic resonance spectroscopy. Vibrational circular dichroism as well as differential scanning calorimetry can also be used.

Investigations were carried out on pollen grains of Scots pine (Pinus sylvestris L.) collected from trees at 1.5, 3, 4 km and control, 20 km from the Luboń factory producing mineral fertilisers. The percentage of germination of pollen formed close to the pollution source was ca 20% lower compared to the control pollen. Lowered vitality of the pollen was effected in changes of the structure of cytoplasmic membranes. Pollen from the polluted area contained ca 15% less total phospholipids, mainly phosphatidylcholine and phosphatytidylinositol and had a lower content of soluble proteins and less of low molecular antioxidants, such as thiols and ascorbic acid. Composition of total fatty acid in phospholipids fractions showed a significant reduction in the degree of unsaturation of fatty acids. Pollen originating from the polluted area and stored at -30°C showed considerably stronger degradation of cytoplasmic membranes than control.

Below the melting point temperature of lipids, artificial lipid membranes usually exist in the ordered gel phase. Above these temperatures lipid acyl chains become fluid and disordered (liquid-crystalline phase). Depending on the chemical composition of artificial membranes, phase separation may occur, leading to the formation of transient or stable membrane domains. A similar phase separation of lipids into ordered and disordered domains has been observed in natural membranes at physiological temperature range. Moreover, it has been reported that certain proteins prefer certain organization of lipids, as for example glycosylphosphatidylinositol-anchored proteins or Src family of tyrosine kinases. The aim of present review is to discuss the possibility that some lipid microdomains are induced or stabilized by lipid-binding proteins that under certain conditions, for example due to a rise of cytosolic Ca2+ or pH changes, may attach to the membrane surface, inducing clustering of lipid molecules and creation of ordered lipid microdomains. These domains may than attract other cytosolic proteins, either enzymes or regulatory proteins. It is, therefore, postulated that lipid microdomains play important roles within a cell, in signal transduction and enzymatic catalysis, and also in various pathological states, as Alzheimer's disease, anti-phosphatidylserine syndrome, or development of multidrug resistance of cancer cells.
