Opis:
Obesity has become one of the most prevalent chronic diseases worldwide, which affects people's health and daily lives. Therefore, this study aimed to investigate the anti-obesity effects of perilla seed oil (PSO), sunflower oil (SFO), and tea seed oil (TSO) and their potential mechanisms in mice fed a high-fat diet (HFD). Mice were divided into five groups: ND, mice fed a normal diet; HFD, mice fed a high-fat diet; PSO, SFO, and TSO, mice fed a high-fat diet supplemented with PSO, SFO, and TSO at 2 g/kg body weight per day, respectively. Our findings showed that oral supplementation with all three oils for 8 weeks significantly reduced body weight, tissue weight, insulin resistance index, serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), and free fatty acids (FFA), and markedly alleviated hyperglycemia, hyperlipidemia, and hepatic steatosis in obese mice. It also decreased leptin, pro-inflammatory cytokines, such as tumor necrosis factor-alpha (TNF-α), interleukin (IL)-6, and (IL)-1beta (IL-1β), and increased anti-inflammatory adipokine adiponectin at both secretion and mRNA expression levels in the epididymal adipose tissue (EAT). Moreover, PSO, SFO, and TSO administration increased the expression levels of fatty acid β-oxidation-related genes, including peroxisome proliferator-activated receptor-alpha (PPAR-α), carnitine palmitoyltransferase 1a (CPT1a) and CPT1b, and thermogenesis-related genes such as uncoupling protein 1 (UCP1), and decreased the expression levels of lipid synthesis-related genes, including fatty acid synthase (FAS) and PPAR-γ in EAT. In conclusion, PSO, SFO, and TSO supplementation could have potential anti-obesity effects in HFD-fed mice by reducing inflammation and improving lipid metabolism.

Obesity is one of the major causes of metabolic diseases such as diabetes and heart attack, and, hence, can lead to low quality of life. Elaborating adipocyte differentiation is very crucial for formulating the treatment and prevention of obesity. The objective of this study is to investigate the seeding cell number required to obtain optimum lipid accumulation during adipocyte differentiation using the 3T3-L1 cell line. Two sets of 5.48×104 (for Day 0 and Day 8 of differentiation), of 10.96×104 (for Day 8) and of 21.92×104 (for Day 8) of 3T3-L1 cells were seeded in each wells of a 12-well plate. Isobuthylmethylxanthine (IBMX), Dexamethasone, and Insulin-containing differentiation cocktails was added into the medium at Day 0 for 48 hours. The medium was changed every two days. Day 0 and Day 8 samples were then stained using Oil Red O and were examined under the microscope to observe the lipid droplets (red-coloured). The lipid droplets were quantified by measuring the absorbance at wavelength of 550 nm. In the study, seeding the number of 10.96×104 cells produced very significantly higher lipid accumulation, as compared with seeding the number of 5.48×104 cells. However, doubling the seeding number into 21.92×104 cells did not increase the lipid droplets significantly. This study found that the optimum seeding number to obtain the maximum lipid droplets during 3T3-L1 adipocyte differentiation was 10.96×104 cells.

Amphotericin B (AmB) is a well known polyene macrolide antibiotic used to treat systemic fungal infections. Despite its toxicity AmB is still regarded as a life-saving drug. The lack of adequate knowledge of the AmB mechanism of action is a serious obstacle to efficient development of new less toxic derivatives. Complementary to various experimental approaches, computational chemistry methods were used to study AmB mechanism of action. A programme lasting for a decade, that was run by our group covered studies of: i) molecular properties of AmB and its membrane targets, ii) structure and properties of AmB membrane channels, and iii) interaction of AmB with the membrane.

Opis:
Below the melting point temperature of lipids, artificial lipid membranes usually exist in the ordered gel phase. Above these temperatures lipid acyl chains become fluid and disordered (liquid-crystalline phase). Depending on the chemical composition of artificial membranes, phase separation may occur, leading to the formation of transient or stable membrane domains. A similar phase separation of lipids into ordered and disordered domains has been observed in natural membranes at physiological temperature range. Moreover, it has been reported that certain proteins prefer certain organization of lipids, as for example glycosylphosphatidylinositol-anchored proteins or Src family of tyrosine kinases. The aim of present review is to discuss the possibility that some lipid microdomains are induced or stabilized by lipid-binding proteins that under certain conditions, for example due to a rise of cytosolic Ca2+ or pH changes, may attach to the membrane surface, inducing clustering of lipid molecules and creation of ordered lipid microdomains. These domains may than attract other cytosolic proteins, either enzymes or regulatory proteins. It is, therefore, postulated that lipid microdomains play important roles within a cell, in signal transduction and enzymatic catalysis, and also in various pathological states, as Alzheimer's disease, anti-phosphatidylserine syndrome, or development of multidrug resistance of cancer cells.

The effect of soil drought on leaf water content, proline content, and the activity of guaiacol (GuPX) and ascorbate (APX) peroxidases as well as the level of lipid peroxidation were investigated in leaves of drought resistant red fescue (Festuca rubra) and drought sensitive perennial ryegrass (Lolium perenne). Plants were grown under glasshouse conditions in soil pot culture. 26 day-old grasses were exposed to drought by withholding irrigation for 18 days. Water content in leaves of perennial ryegrass decreased more than in red fescue throughout the experimental period. On the other hand, proline content (PC) was higher in red fescue. The activity of APX and GuPX increased in leaves of red fescue, while it did not change in perennial ryegrass. Our data demonstrate that both red fescue and perennial ryegrass were able to survive applied drought, as shown by a lack of stress-induced lipid peroxidation and hence no evidence of oxidative damage. We speculate, that the observed drought stress tolerance at cellular level was associated with the ability to accumulate proline, and to maintain high activity of APX and GuPX, resulting in protection against oxidative damage and lipid peroxidation. It seems that this mechanism works better in red fescue.

The sources of free radicals and antioxidant defence systems in living organisms are reviewed. Attention is focused on the role of dietary lipid oxidation products and antioxidants to maintain balance between factors promoting free radical mediated oxidation and the antioxidant defence mechanisms. Particular attention is paid to the influence of intake of vitamins with oxidative properties (vitamin C, vitamin E and ß-carotene) and polyphenols compounds in the prevention of coronary heart disease and cancer.

The glycine receptor belongs to the ligand-gated ion channel superfamily. It is a chloride conducting channel composed of four transmembrane domains. It was previously shown that the second transmembrane domain (M2) of the glycine receptor forms an ion conduction pathway throught lipid bilayers. The amino-acid sequence of the transmembrane segment M2 of the glycine receptor has a high homology to all receptors of the ligand-gated ion channel superfamily. In our report, we have used a synthetic M2 peptide. It was incorporated into a planar membrane of known lipid composition and currents induced by M2 were measured by the Black Lipid Membrane technique. When the planar lipid bilayer was composed of 75% phosphatidylethanolamine and 25% phosphatidylserine, the reversal potential measured in a 150/600 mM KCl (cis/trans) gradient was -19 mV suggesting that the examined pore was preferential to anions, PK/PCl = 0.25. In contrast, when 75% phosphatidylserine and 25% phosphatidylethanolamine was used, the reversal potential was +20 mV and the pore was preferential to cations, PK/PCl = 4.36. Single-channel currents were recorded with two predominant amplitudes corresponding to the main-conductance and sub-conductance states. Both conductance states (about 12 pS and 30 pS) were measured in a symmetric solution of 50 mM KCl. The observed single-channel properties suggest that the selectivity and conductance of the pore formed by the M2 peptide of the glycine receptor depend on the lipid composition of the planar bilayer.

Opis:
Paraoxonase 1 (PON1), contained in plasma high-density lipoproteins, plays an important role in the protection of plasma lipoproteins and cell membranes from oxidative damage. Previous studies indicate that human PON1 is stimulated by high NaCl concentrations. The aim of this study was to characterize in more detail the effect of salts on serum PON1. Paraoxon-hydrolyzing activity of human serum was stimulated by 81.6% following the addition of 1 M NaCl. The effect of NaCl was dose-dependent between 0.5 and 2 M. PON1 activity toward phenyl acetate was reduced by 1 M NaCl by 55.2%. Both the paraoxon- and phenyl acetate-hydrolysing activity was slightly lower in heparinized plasma than in serum, but NaCl had similar stimulatory and inhibitory effects on these activities, respectively. In rat, rabbit, and mouse, NaCl reduced PON1 activity. KCl had a similar effect on human PON1 as NaCl. Sodium nitrite also stimulated human PON1 but much less effectively than chloride salts. In contrast, sucrose, sodium acetate and sodium lactate had no significant effect. NaBr was a less effective PON1 activator than NaCl, whereas the effect of NaJ was non-significant. The activity of human PON1 toward homogentisic acid lactone and γ-decanolactone was unaltered by NaCl. These data indicate that: 1) high concentrations of chlorides stimulate human PON1 activity toward paraoxon but not other substrates, 2) PON1 is inhibited by Cl- in other mammalian species, 3) the potency of human PON1 activation by halogene salts increases with decreasing atomic mass of the halide anion.
