Opis:
There are institutes which research phenomena and processes taking place on islands. Such research is conducted nowadays among others at universities in France, United States, Australia, New Zealand, Great Britain and Scandinavia. Godfrey Baldacchino from the University of Prince Edward Island is a professor of Island Studies (and the editor-in-chief of the "Island Studies Journal"1). One of the actively operating commissions of the International Geographical Union is the IGU Commission on Islands. Therefore, the term "island" is commonly present in the modern reality and so strongly rooted that, as a results, its meaning very rarely raises doubts. Sometimes a group of researchers makes an attempt to reflect upon the definition of the term2. Those attempts are not followed by satisfying results, which does not encourage to further research on islands, be what they may.The following article is another attempt to ponder the nature of islandness and to point those characteristics of an island which differentiate it from other geographical objects. According to the author, the group of geographical objects treated as islands is definitely too wide, and the vast protruding lands, commonly regarded as islands, continue to be islands in the universal awareness only out of habit, or due to the lack of a better, adequate term as a result of existing terminological dichotomy between the terms island/continent.

Opis:
The current study was initiated to isolate and characterize yeasts from wet Arabica coffee processing wastes for bioethanol production. Yeast isolates were retrieved from wet Arabica coffee processing effluent1, effluent2, effluent3, pulp1 and pulp2. The yeast isolates were screened and characterized for ethanol production following standard methods. All the isolates were first tested for carbohydrate fermentation in appropriate medium. Selected ethanol producing isolates were tested for various parameters. Yeast isolates from pulp (ACP12) and effluent (ACE12) showed significantly high counts at 20% glucose concentration with the highest population number of 2.16 ±1.00×108 and 1.21±1.00×108 CFU/ml, respectively. Isolate ACP12 showed higher population number (9.7±1.00×107 CFU/ml) than the standard culture (Saccharomyces cerevisiae) with colonies count of 8.7±1.00×107 CFU/ml at 30 °C. Moreover, yeast isolate (ACP12) showed higher colonies count (9.8 ±1.00×107 CFU/ml) compared to the standard strain (8.7±1.00 ×107 CFU/ml) at pH 5.0. Based on morphological, physiological and biochemical characteristics, the two isolates (ACE12 and ACP12) were tentatively identified as genus Saccharomyces. Total sugar concentration on (90%) was obtained from pulp1. Isolate ACP12 showed the maximum ethanol production (6.2 g/l) from pulp1 compared to the standard isolate (5.49 g/l) and the other test yeasts. From this study, it can be concluded that isolate ACP12 has an inherent potential of ethanol production from coffee pulps compared to the rest yeast isolates and needs further supplementary activities to qualify it for industrial application.

Stem canker and black scurf of potato (Solanum tuberosum L.) caused by Rhizoctonia solani Kühn are important and epidemic diseases in potato-growing regions worldwide, including Iran. In this study, 120 isolates were retrieved from infected stem canker from six potato-growing regions in Iran (Isfahan, Ardebil, Fars, Hamedan, Kurdestan and Kerman). Out of these, 30 isolates were selected as representatives for genetic and virulence analysis. The isolates were analyzed by one sequence analyzes of the ITS-rDNA region, random amplified polymorphic DNA (RAPD) and inter-simple sequence repeat (ISSR), as well as virulence studies. Based on sequence analysis of the ITS-rDNA region, all 30 isolates were assigned to the anastomosis group (AG) and all were assigned to AG-3 PT. Cluster analysis using the unweighted pair group method with the arithmetic averages (UPGMA) method for both RAPD and ISSR markers revealed that they were divided into three main groups, with no correlation to geographical regions of the isolates. Pathogenicity tests showed that all isolates were pathogenic on potato cv. Agria; however, virulence variability was observed among the isolates. The grouping based on RAPD analysis and virulence variability was not correlated.

Opis:
The in vitro antibacterial activity of pure caffeine powder and its interaction with first line antibiotic against bacterial isolates were investigated with the macrobroth dilution and the checkerboard assay methods. This study showed that caffeine and the antibiotics exhibited various degrees of antibacterial activities. While caffeine had MICs ranging between 67.19 and 268.75 µg/ml, chloramphenicol was characterized by MICs between 0.98 and 31.25 µg/ml, kanamycin - 15.63-62.5 µg/ml, nalidixic acid - 0.49-250 µg/ml, erythromycin - 0.49-62.5 µg/ml, tetracycline - 1.99-62.5 µg/ml and metronidazole - 15.63-31.25 µg/ml. Combining ½ MICs and MICs of caffeine with the antibiotics as well as direct combination of caffeine and the antibiotics resulted in significant reduction of antibiotics' effectiveness. The fractional inhibitory concentration index (FICI) for the combination of ½ MICs of caffeine with different antibiotics showed antagonistic interactions with the antibiotics except kanamycin which had additive and indifferent interactions with caffeine. The FICI of the MICs of caffeine combined with antibiotics showed a reduction in the number of antagonistic interactions as chloramphenicol, nalidixic acid and erythromycin showed some indifferent interactions while kanamycin was the only antibiotic that showed indifferent interaction against all the bacterial isolates. The direct combination of caffeine and the antibiotics resulted in significant antagonistic interactions higher than in the case when caffeine, at the ½ MICs and MICs, was combined with the antibiotics. Although caffeine demonstrated significant antibacterial activity against the selected bacterial isolates, its combination with the selected antibiotics resulted in significant antagonistic interactions. Caffeine should not be combined with antibiotics as this could result in serious therapeutic failure and, possibly, drug toxicity in vivo.

Opis:
Few studies have shown that Cnestis ferruginea possess some therapeutic properties. The present study was aimed screening the extracts of the fruits and leaves of Cnestis ferruginea for phytochemicals using crude screening and gas chromatography-mass spectrophotometer (GC-MS) methods, and also antimicrobial activity against clinical isolates. Preparation of the extracts (aqueous and ethanolic), identification of isolates, and antimicrobial sensitivity were done using previously reported standard methodologies. Replicate readings for the antimicrobial sensitivity were analysed using analysis of variance at 95% level of significance. The result of the crude qualitative screening revealed the presence of phenol, terpenes, alkaloids, saponins, tannin and polyphenol in various amounts in both the leaves and fruits. Quantitative analysis using GC-MS revealed a total of 14 similar phytochemicals each in the leaves and fruits of the study plant in varying amounts. In the fruits, phenol had the highest concentration of 15.01%, followed by terpenes (10.64%), alkaloid (5.43%), and tannins (5.16%). Others were anthocyanins, phytate, phytosterol, steroids, saponins, cardiac glycosides, oxalate, flavonoids, cyanogenic glycosides and coumarin with concentrations that ranged from to 0.01- 4.21%. On the other hand, in the leaves, alkaloids had the highest concentration of 31.62%, followed by phenol (20.59%), phytate (15.18%), and tannin (12.34%). Others include were flavonoid, terpenes, phytosterol, cardiac glycosides, saponins, cyanogenic glycosides, oxalate, anthocyanins, steroids and coumarin with concentrations that ranged from 0.16-8.45%. The isolates were identified as Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa and Proteus spp. The results of the antimicrobial sensitivity revealed varying zones of inhibitions; however, it increased as the concentration of the extract increased. The observed highest zones of inhibitions were 33.86 mm for the fruit ethanolic extract and 30.56 mm for the aqueous extract of the leaves against Proteus and Staphylococcus aureus, respectively.

Opis:
Urinary tract infection (UTI) is one of the most common diseases encountered worldwide. In this study, urine samples were collected from thirty women of child bearing age (18-25 years) to determine the prevalence of bacteriuria, as well as the presence of pus cells and white blood cells which are signs of infection. The urine microscopy result revealed that 7 (23.3%) of the sample had greater than 10 leucocytes per high power field. Only four of the samples had a significant number of pus cells. The bacteria count recorded ranged from 25 cfu/0.001 ml to 276 cfu/0.001 ml. Of the samples, 13 were positive for bacteriuria having bacterial count greater than 100 cfu/0.001 ml. The bacteria isolates include Klebsiella spp. (3.8%), Streptococcus spp. (2.5%). Escherichia coli (62.5%) and Staphylococcus spp. (21.2%). Of these, Klebsiella spp. was the least prevalent and Escherichia coli had the highest number of occurrence. These organisms were very sensitive to Ciprofloxacin, Pefloxacin, Zinnacef and Gentamycin according to the antimicrobial susceptibility test carried out. Ciprofloxacin was found to be the most effective antibiotics against all the bacterial isolates. Here, zones of inhibition ranged from 15 mm (Streptococcus spp.) to 20 mm (Escherichia coli). All the bacterial isolates were resistant to Amoxicillin, with zones of inhibition between 11 (Klebsiella spp., Escherichia coli, Staphylococcus spp.) and 13 mm (Streptococcus spp.). Since the prevalence of asymptomatic bacteriuria is high, early diagnosis and proper treatment is necessary in order to prevent it from becoming symptomatic.

Opis:
From hydrothermally treated cotyledons of faba bean and pea there were obtained protein isolates characterized by inreased protein content, trace activity of trypsin inhibitors, lowered content of bonded and residual fat, and better taste compared to isolate obtained from non-treated material. This significant improvement of quality was obtained at the cost of 20% yield decrease.
Jakkolwiek wszystkie izolaty zawierały więcej tłuszczu całkowitego aniżeli mąki, to izolaty otrzymane z liścieni parowanych zawierały go o ok. 3% mniej niż izolaty z nasion kontrolnych. Mniejsza była również zawartość tłuszczu związanego. Najbardziej istotną zaletą wstępnej obróbki hydrotermicznej liścieni była zdecydowana poprawa cech organoleptycznych. Izolaty te były pozbawione charakterystycznego, lekko gorzkawego i strączkowego posmaku i zapachu. Ujemną natomiast stroną było, zresztą spodziewane obniżenie o 20% wydajności procesu otrzymania izolatów.

Opis:
Potato virus Y (PVY) causes tobacco veinal necrosis. It be-longs to the group of ten most dangerous plant viruses respon-sible for large economic losses. Within the species Nicotiana tabacum, the resistance to this virus is conditioned by the single recessive va gene resulting from a deletion in the susceptibility Va gene. Many tobacco cultivars have this resistance, including VAM, Wiślica and Virginia SCR (V. SCR). However, their resist-ance is overcome by the virulent PVY isolates, so it is necessary to search for new sources of resistance or combine ones already known. The aim of the study was to identify cultivars with va resist-ance, and to assess its effectiveness depending on the used PVY isolate. Research also included the search for new resistance fac-tors within Nicotiana tabacum cultivars. Twenty five tobacco cultivars were tested for resistance to PVY in biological tests by artificial inoculation under greenhouse conditions. Four virus iso-lates with different levels of virulence were used for the studies. After four weeks, disease symptoms were observed, and DAS-ELISA tests were performed using specific antibodies to confirm the presence of the virus in plants. The tested cultivars showed a different degree of PVY susceptibility depending on the used isolate. The identification of the va gene was carried out by using PCR for amplification of two markers of the length 146 and 402 bp. The presence of amplicons indicated the presence of Va sus-ceptibility gene, while cultivars with the va resistance gene did not amplify the products. The highest resistance was character-istic of cv. VAM which was not infected with isolates IUNG 23 and IUNG 17, defined as weak and medium, although the remain-ing isolates, described as strong (IUNG 22 and IUNG20), caused vein necrosis and the presence of the virus was confirmed in plant tissues by a positive DAS-ELISA test result. Six cultivars that did not show disease symptoms only after applying the weak isolate, had a slightly lower resistance. Other cultivars with va resistance were infected by all used PVY isolates. Another five cultivars, after infection with four isolates, showed symptoms of tolerance, i.e. vein clearing and chlorotic spots of a leaf blade, but they did not have vein necrosis, in spite of that molecular tests confirmed the presence of Va susceptibility gene. Moreover, interesting re-sults were noted for cv. Węgierski Ogrodowy, that, despite the presence of the susceptibility gene, was not infected by weak isolate IUNG23. The last group included susceptible cultivars that reacted with vein necrosis to all used PVY isolates. Since virulent PVY isolates are able to break va resistance, the knowledge of the nature and stability of the resistance of cul-tivars is particularly important, especially within Polish cultivars. In addition, the rising number of necrotic isolates in Poland and in the world, capable of breaking existing sources of resistance, can cause an increased use of tolerant cultivars in tobacco breeding.

Field evaluation of insect pests and pathogens associated with sesame cultivated in the Niger Delta region of Nigeria was conducted to provide baseline information on their status. The experiment was laid out in Randomized Complete Block Design. Herein, sesame seeds were obtained from the Jos and Maiduguri main markets. These were sown, then data were collected on insect species beginning three weeks after sowing (WAS) for six weeks and identified to species levels. At 10 WAS, plant parasitic nematodes (PPNs) were extracted from both root and soil samples in the rhizosphere of sesame, using the modified Baermann method. Plant parasitic nematodes were identified to generic level. Infected plant parts from field were thoroughly washed, disinfected and plated on PDA, and subsequently incubated at 25 ± 2 °C. Fungal pathogens were isolated, identified and checked for their viability and purity. Data obtained were subjected to both parametric and none parametric analyses. Insect species collected included Leptaulaca fissicolis, Gryllus bimaculatus, Apogonia nitidula, Trilophida conturbata, Elis sp. amongst others. A total of six genera of PPNs, Helicotylenchus, Pratylenchus, Scutellonema, Gracilacus, Meloidogyne and Tylenchus were associated with Sesame. Gracilacus and Pratylenchus had the highest population in soil (37.5%) and roots (37.5%). Three fungal isolates, namely, Fusarium oxysporum, Fusarium solani and Trichoderma sp, were consistently isolated from the plant parts - with Fusarium oxysporum and Fusarium solani being pathogenic. Knowledge of these insect pests and pathogens will aid in their management.

The work presents the results of investigations into the structure and phase composition of newly developed high manganese steels of the X98MnAlSiNbTi24-11 and X105MnAlSi24-11 type. The average density of such steels is 6.67 g/cm3, which is 15% lower as compared to typical structural steels. An analysis of phase composition and structure allows to conclude that the investigated steels feature an austenitic γ-Fe(Mn,Al,C) structure with uniformly distributed and elongated α-Fe(Mn,Al) ferrite grains on the boundaries of austenite grains and carbides. Examinations by diffraction of back scattered electrons (EBSD) allow to conclude that high-angle boundaries dominate in such steels, having significant influence on mechanical properties. Three types of carbides with differentiated chemical composition and size were identified in steel X98MnAlSiNbTi24-11 with scanning and transmission electron microscopy. κ-(Fe,Mn)3AlC carbides, having a regular, face-centered cubic lattice (fcc), were identified in austenite. Transmission electron microscopy examinations have enabled to identify M7C3-type carbide in ferrite. Nb- and Ti-based complex carbides were identified in steel X98MnAlSiNbTi24-11, both, in ferrite and austenite. (NbTi)C2 carbide precipitates were confirmed in an X-ray qualitative phase analysis. The size of the above-mentioned carbides is within several to several dozens of μm. An X-ray qualitative phase analysis has confirmed the precipitates of M7C3 carbides in both steels.

Opis:
Phytophthora cactorum and P. citrophthora were isolated from diseased plants of Sorbus aucuparia. P. cactorum was the dominant species on seedlings and 3−15 years old trees, while P. citrophthora was isolated only from 3−6 years old trees. Isolates of both species obtained from 10 host plants colonized stem fragments and leaf blades of S. aucuparia and S. intermedia. In greenhouse trials only isolates from Pinus sylvestris and Viola wittockiana did not cause root and stem base rot symptoms of rowan seedlings.
