Opis:
Annexin A6 (AnxA6) is a Ca2+-dependent membrane-binding protein involved in vesicular traffic. The likely participation of AnxA6 in the response of lymphocytes to Ca2+ signals has not been investigated yet. The present study focuses on intracellular relocation of AnxA6 in human Jurkat T lymphoblasts upon stimulation followed by transient increase of intracellular [Ca2+] and exocytosis of interleukin-2 (IL-2). Stimulation of the cells under different experimental conditions (by lowering pH and/or by rising extracellular [Ca2+] in the presence of ionomycin) induced time-dependent transients of intracellular [Ca2+] and concomitant changes in AnxA6 intracellular localization and in IL-2 secretion, with only minor effects on cell viability and apoptosis. In resting conditions (in the presence of EGTA or with no ionophore) AnxA6 was localized uniformly in the cytosol, whereas it translocated to vesicular structures beneath the plasma membrane within 5 min following stimulation of Jurkat T cells and rise of intracellular [Ca2+] at pH 7.4. Lowering the extracellular pH value from 7.4 to 6.0 significantly enhanced this process. AnxA6 changed its location from the cytosol to the secretory granules and early endosomes which seem to represent membranous targets for annexin. In conclusion, AnxA6 is sensitive to variations in intracellular [Ca2+] upon stimulation of Jurkat T cells, as manifested by a switch in its intracellular localization from the cytosol to vesicular structures located in close proximity to the plasma membrane, suggestive of participation of AnxA6 in calcium- and proton-dependent secretion of cytokines by lymphocytes.

Gut-derived adenocarcinoma Caco-2 cells were treated with sodium butyrate (NaB) at physiologically relevant concentrations. We characterized its effects on proliferation, differentiation, apoptosis, adhesion to the solid support and interleukin-8 secretion. Differentiation was determined by brush border alkaline phosphatase activity. Apoptosis was assessed by acridine orange and Hoechst stains. Differentiation and apoptosis were analyzed in both adherent and floating cell populations. The transformed Caco-2 cells did not retain their malignant phenotype in the presence of NaB. They appeared to undergo a change in the phenotype induced by NaB, as indicated by reduced proliferation, enhanced differentiation, stimulation of apoptosis leading to decreased viability of cells, and stimulation of interleukin-8 secretion. Considering all the above facts and data, we postulate that Caco-2 cells cultured in NaB supplemented medium could regain the phenotypic characteristics of the phenotype of the parent cell from which originated the Caco-2 line.

We investigated suppression of murine B16(F10) melanoma tumor growth following a therapy which involved concomitant administration of cyclophosphamide and plasmid DNA bearing interleukin-12 gene. Since both therapeutic factors display antiangiogenic capabilities, we assumed that their use in blocking the formation of new blood vessels would result in augmented inhibition of tumor growth. This combined therapy regimen indeed resulted in a considerable suppression of tumor growth. We observed a statistically significant extension of treated animals' lifespan. Interestingly, the therapeutic effect was also obtained using a plasmid without an interleukin gene insert. This observation suggests that plasmid DNA, which has been widely applied for treating neoplastic tumors, contains element(s) that elicit immune response in mice.

Regulatory effect of CD25, an activation antigen the α subunit of interleukin 2 receptor (IL2R) on the activity of natural killer (NK) cells was studied in fifty elderly (57-70 years old) and fifty young people (19-35 years old). Cytotoxic NK activity was assessed by 51Cr release assay, the levels of interleukin 2 (IL2) and tumour necrosis factors α (TNFα) were measured using bioassays and expression of CD16 and CD25 proteins by flow cytometry. Low NK activity in the elderly was associated with decline of full health, lowered serum concentration of IL2 and increased production of TNFα during NK reaction. Inhibition of TNFα activity by anti-TNF monoclonal antibody suppressed exclusively NK activity of low NK responders. Moreover, stimulation in vitro of blood mononuclear cells, with TNFα induced in the elderly low NK responders a significantly higher increase of the CD25 expression on the surface of NK cells as compared with that in the elderly high responders. Since the CD25 molecule constitutes a subunit of the high affinity receptor, binding IL2 to immunocompetent cells, its increased expression on NK cells of low NK responders would enable them to bind even low amounts of the endogenous IL2 available in this group of the elderly. Thus, an overproduction of TNFα seems to be a mechanism compensating, in the non-fully healthy elderly, for the decreased IL2 production, promoting efficient cytotoxic reaction.

Opis:
Studies on serum interleukin (IL)-31 levels in dogs with atopic dermatitis (AD) and their correlation with disease severity are limited. To the author’s knowledge, there are no studies that measured serum IL-31 in dogs treated with lokivetmab injections, a selective inhibitor of this key cytokine in pruritus. The aim of the study was to evaluate serum IL-31 levels in dogs treated with lokivetmab and correlate it with the severity of canine atopic dermatitis using the pruritus visual analog scale (pVAS) and canine atopic dermatitis extent and severity index (CADESI-04). Ten client-owned dogs diagnosed with AD received two injections of lokivetmab four weeks apart. Disease severity was assessed using the pVAS and CADESI-04 scores before and after both injections. In addition, canine serum IL-31 levels were measured at the same moments. Serum IL-31 was detected in all dogs in the study. There was a significant reduction in pVAS scores and serum IL-31 after administrations. However, there was no difference in CADESI-04 scores, and there was no significant correlation between CADESI-04 scores and serum IL-31 in dogs diagnosed with AD. Nonetheless, a significant positive correlation was observed between the pVAS scores and serum IL-31 levels with lokivetmab therapy, which reinforces the role of IL-31 in the pathogenesis of pruritus in dogs with AD. The data presented here provide further evidence that IL-31 is directly involved in pruritus pathogenesis in dogs with AD. In addition, blocking IL-31 has a significant antipruritic effect, but has no influence on skin lesion severity and extension.

Opis:
Gram-positive and nonpathogenic lactic acid bacteria (LAB) are considered to be promising candidates for the development of new, safe systems of heterologous protein expression. Recombinant LAB has been shown to induce specific local and systemic immune response against selected pathogens, and could be a good alternative to classical attenuated carriers. The main goal of our study was to express the avian influenza haemagglutinin (H5) and chicken interleukin 2 (chIL-2) in Lactococcus lactis. Results of this study were anticipated to lead to construction of lactococcal strain(s) with potential vaccine properties against the avian influenza A (H5N1) virus. Expression of the cloned H5 gene, its His-tagged variant and chIL-2 gene, under the control of the ptcB gene promoter was attested by RT-PCR on transcriptional level and Western or dot blot analysis on translational level, demonstrating that system can be an attractive solution for production of heterologous proteins. The results of the preliminary animal trial conducted in mice are a promising step toward development of a vaccine against avian bird flu using Lactococcus lactis cells as antigen carriers.

W artykule zaproponowano graficzną formę prezentacji wyników regresji logistycznej, zastosowanej dla dwustanowej zmiennej zależnej i dwóch numerycznych zmiennych pomiarowych. Przed wyznaczeniem współczynników regresji jedna z tych zmiennych została poddana transformacji z wykorzystaniem logarytmu o podstawie 2. Opisano sposób przeprowadzenia odpowiednich obliczeń i zaprezentowano przykładowe wyniki, bazując na wynikach badania klinicznego (dla N=47 pacjentów) stężeń interleukiny 6 w surowicy dzieci poddawanych zabiegom kardiochirurgicznym w krążeniu pozaustrojowym, narażonych na ryzyko wystąpienia ostrego uszkodzenia nerek (OUSzN). Metoda pozwala na szybką identyfikację ryzyka i podjęcie właściwych działań terapeutycznych, w tym - terapii nerkozastępczej.
In the paper there are proposed nomograms being graphical presentation of logistic regression results. Such graphs depict the probability and odds functions of the considered feature in a readable form. The investigated particular case is suited for the binomial output variable and two numerical input variables. The suggested presentation form has been selected with focus on clinical personnel accustomed to such form of data presentation. After introductory explanation of the logistic regression essentials with emphasis on the Gaussian-based distribution, there are presented assumption and resulting - alternative model formulation supported by the simulation experiment results, basing on the clinically collected data from 47 patients suspected to experience acute kidney injury (AKI) diagnosed accordingly to interleukin 6 serum concentration. The interpretation of unit selection for each input variable is shortly discussed along with the consequences of possible logarithmic transformation. The presented method allows for either quick, graphical, bedside risk identification and mapping of lifethreating condition or early introduction of renal replacement therapy.

Introduction and aim. Reports have shown that there is alteration in haematological and inflammatory processes in patients with cervical cancer. However, there is the dearth of information on the pattern of alteration in Nigerian patients with cervical cancer at various stages of the disease. Therefore, haemocytometric profile and plasma levels of interleukin-6 (IL-6) and IL-12 were determined in Nigerian patients with cervical cancer at various stages of the disease. Material and methods. Eighty-nine adults consisting of 49 patients with cervical cancer and 40 apparently healthy controls were enrolled into this study. Haemocytometric profile was determined using automated haematology analyzer while the plasma levels of interleukin-6 (IL-6) and IL-12 were determined using ELISA. Results. Of the participants with cervical cancer, 6.12%, 24.49%, 53.06% and 16.33% were in stages I, II, III and IV respectively. The mean plasma IL-6 level was significantly higher in patients at stage IV of the cancer compared with those in stages I, II and III. No significant differences were observed in the mean plasma IL-12 level, and the haemocytometric profile when patients in different stages of the cancer were compared with one another. Plasma IL-6 had significant positive correlation with the lymphocytes count and cancer stage but had significant negative correlation with packed cell volume (PCV), haemoglobin and total white blood cells count (WBC) in patients with cervical cancer. Conclusion. Interleukin-6 appears to play an important role in the progression of cervical cancer and could be involved in cervical cancer-associated alteration in haemocytometric profile.

Opis:
The role of interleukin-2 (IL-2) in tumor dynamics is illustrated through mathematical modeling, using delay differential equations with a discrete time delay (a modified version of the Kirshner-Panetta model). Theoretical analysis gives an expression for the discrete time delay and the length of the time delay to preserve stability. Numerical analysis shows that interleukin-2 alone can cause the tumor cell population to regress.
