Introduction and objective: In June 2009 the World Health Organization announced influenza pandemic caused by A/H1N1/v virus. It became crucial to recognize new cases of A/H1N1/v infection. An effective screening diagnostic procedure was needed for patients suffering from influenza-like symptoms for making an initial diagnosis and analyzing epidemiological pattern of infection. We used a strip test for influenza A/B as a screening diagnostic procedure for patients suffering from influenza-like symptoms for making an initial diagnosis. For comparison, RT PCR for detecting A/H1N1/v was performed. The aim of this study was to assess the efficacy and sensitivity of the strip test and its value for making initial diagnosis of influenza A/H1N1/v. Material and methods: Strip testing for the influenza A/B infection was performed on 1123 patients with influenza-like symptoms in the Admission Unit of the Regional Infectious Diseases Hospital in Warsaw. Strip test results were analyzed according to the age of patients and season of the year. For 97 patients strip test results for detecting A/H1N1 infection were compared with those obtained by RT PCR. Results: There were no statistically significant differences found between the methods and strip testing demonstrated sensitivity of 61% and specificity of 71%. Conclusions: No statistically significant differences were found between the two methods, however, strip test had low sensitivity and specificity.

Influenza is a global health concern. The single most effective way of protecting people against influenza infection and disease is vaccination. However, currently available vaccines against influenza induce only strain-specific immunity, and do not elicit long-lasting serum antibody titers. Therefore, they are ineffective in the case of possible pandemics. There is an urgent need for a new generation vaccine which would induce broad and long-lasting immune protection against antigenically distinct flu viruses. The paper presents recent achievements and the challenges in the field of universal vaccine construction.

A serological surveillance programme for avian influenza A virus (AIV) subtype H5 and H7 in poultry was implemented in Poland in 2008-2013 with two main objectives: i) to detect subclinical infections or previous exposures to AIV H5 and H7 subtypes and ii) to demonstrate the AI- free status of Poland. During this period, over 45 000 serum samples from 2833 holdings were examined using the hemagglutination inhibition test (HI). The presence of HI antibodies was detected in 8 breeder geese holdings (7 positive for H5 and 1 positive for H7 AIV) and in 1 breeder duck holding (H5-positive), which represented 0.32% of all investigated holdings. All seropositive flocks were examined by real time RT-PCR with negative results, which substantiated the AI-free status of Poland. Positive results detected in clinically healthy poultry kept in an open range system indicate prior infections with low pathogenic AIV originating from the wild-bird reservoir.

Dynamically growing number of airline passengers and flights, especially intercontinental ones, is a key factor conducive to the spread of infectious diseases. The threat to public health may be even more noticeable if it is taken into account, that the developing countries record larger increase in air traffic than most European high developed states with good sanitary conditions. The aim of the study is to make a review of the literature concerning risk assessment and ways to prevent transmission of infectious diseases aboard civil aircrafts. Airline-associated spread of infectious diseases can refer to airborne and vector-borne diseases, as well as these transmitted by fecal-oral route. The particular danger of rapid spread of a disease through airline passengers is caused by influenza virus strains with the potential for causing pandemics or coronaviruses that are infectious agents of MERS and SARS. Passengers suffering from active tuberculosis are also a reason of a serious epidemiological concern. Also other airborne diseases like measles can be easily transmitted by air transport when travelling from endemic countries. On the other hand, flight-related food poisonings are less and less frequent – the most common cause are still bacteria of the Salmonella genus. Among vector-borne diseases, particular attention is paid to the problem of possible transfer of mosquitoes infected with West Nile Virus. In the prevention of the spread of communicable diseases via air transport, there are important factors such as: efficient identifying of sick travellers and crew members, use of personal protective equipment (such as masks with HEPA filter) by passengers travelling alongside sick person, efficient ventilation inside the passenger cabin, proper disinfection of aircrafts' interiors, vaccination of cabin crews against influenza and proper preparation and handling of catering provided to aircrafts.

The aim of this study was to investigate the in vitro cytotoxicity of oseltamivir derivatives and determine their activity against A/H1N1/PR/8/34 and A/H3N2/HongKong/8/68 - strains of influenza virus. Antiviral activity of these compounds was determined by using two methods. MTT staining was used to assess the viability of MDCK cells infected with influenza viruses and treated with various concentrations of drugs. In parallel, the effect of drugs on viral replication was assessed using the hemagglutination test. The most toxic compounds were: OS-64, OS-35, OS-29, OS-27 and OS-25, whereas OS-11, OS-20 and OS-23 were the least toxic ones. Statistically significant antiviral effect at a higher virus dose was shown by compounds: OS-11, OS-20, OS-27, OS-35, and OS-64. H3N2 virus was sensitive to 10-times lower concentrations of OS-11 and OS-35 than H1N1. At a lower infection dose, the antiviral activity was observed for OS-11, OS 27, OS-35 and OS-20. OS-64 turned out to be effective only at a high concentration. OS-23 showed no antiviral effect.

Influenza virus infection is a major source of morbidity and mortality worldwide. Due to the variable effectiveness of existing vaccines, especially in the early stages of an epidemic, antiviral drugs represent the first line of defense against the virus. Currently, there are two major classes of anti-influenza drugs approved by the FDA for clinical use: M2 protein inhibitors (amantadine and rimantadine) and neuraminidase inhibitors (zanamivir and oseltamivir). However, increasing resistance to these available influenza antivirals among circulating influenza viruses highlights the need to develop alternative approaches for the prevention and/or treatment of influenza. This review presents an overview of currently available drugs for influenza treatment as well as summarizes some new antiviral strategies that are now being tested covering agents targeting both the viral proteins and the host-virus interaction. We discuss their mechanisms of action, resistance and the therapeutic potential as new antiviral drug for use in future influenza pandemics. Additionally, combination therapy based on these drugs is also described.

Poland has experienced four episodes of avian influenza (AI) outbreaks over the past two decades. The first epidemic was caused by a low pathogenicity (LPAIV) H7N7 subtype and occurred in fattening and breeder turkeys in 1995. Two waves of H5N1 high pathogenicity avian influenza (HPAI) took place in 2006 and 2007. In spring 2006, 64 cases of the H5N1 virus were detected, mostly in mute swans. In December 2007, ten outbreaks of H5N1 HPAI were detected in commercial poultry (n=9) and wild birds kept in captivity (n=1). The outbreaks in 2006 and 2007 were caused by genetically similar but clearly distinguishable viruses of the 2.2 clade. In 2013, an H9N2 avian influenza virus was detected in 4 fattening turkey holdings. The virus was low pathogenic and a phylogenetic study has shown a close relatedness to the Eurasian lineage of AIV of the wild bird origin. Neither preventive nor prophylactic vaccinations have ever been used in poultry or other birds. Emergency vaccinations using autogenous vaccine were introduced only to control the H7N7 LPAI outbreaks in 1995. The baseline surveillance for AI in live migratory birds and poultry provides a valuable insight into the ecology of AIV at the wild and domestic bird interface. Passive surveillance is in place of early detection of HPAIV infection in dead or moribund birds.

Opis:
Hemagglutinin (HA), as a major surface antigen of influenza virus, is widely used as a target for production of neutralizing antibodies. Monoclonal antibody, mAb6-9-1, directed against HA of highly pathogenic avian influenza virus A/swan/Poland/305-135V08/2006(H5N1) was purified from mouse hybridoma cells culture and characterized. The antigenic specificity of mAb6-9-1 was verified by testing its cross-reactivity with several variants of HA. The mimotopes recognized by mAb6-9-1 were selected from two types of phage display peptide libraries. The comparative structural model of the HA variant used for antibody generation was developed to further facilitate epitope mapping. Based on the sequences of the affinity- selected polypeptides and the structural model of HA the epitope was located to the region near the receptor binding site (RBS). Such localization of the epitope recognized by mAb6-9-1 is in concordance with its moderate hemagglutination inhibiting activity and its antigenic specificity. Additionally, total RNA isolated from the hybridoma cell line secreting mAb6-9-1 was used for obtaining two variants of cDNA encoding recombinant single-chain variable fragment (scFv) antibody. To ensure high production level and solubility in bacterial expression system, the scFv fragments were produced as chimeric proteins in fusion with thioredoxin or displayed on a phage surface after cloning into the phagemid vector. Specificity and affinity of the recombinant soluble and phage-bound scFv were assayed by suitable variants of ELISA test. The observed differences in specificity were discussed.

The aim of this article was to present selected papers regarding circovirus infections in swine and also swine influenza, raised during the 11th ESPHM in Utrecht. Issues connected with circovirus-related diseases focus on studies of virus genotypes distribution and determination of the dominant PCV2 genotype in different regions of Europe. Also the prevalence of PCV2 and PCV3, the frequency of co-infection with both genotypes and PCV3 infection related gastrointestinal and respiratory symptoms in swine in European countries, were presented and discussed. These works also discussed the possible interaction between PCV2 and porcine parvovirus PPV4, assessed the pressure of PCV2 infection in pigs of different age and the impact of vaccination protocols on virus excretion. There were studies presented on the differences of virus circulation in vaccinated and unvaccinated animals. Also clinical presentation of infections with different PCV2 strains was described. Much attention was paid to assessment of oral fluid examination in diagnostic procedure. Reports regarding swine influenza have focused on the analysis of various diagnostic methods, the assessment of influenza A virus (IAV), its prevalence in piglets in the early and late nursery period and the analysis of IAV occurrence over the seasons. The diversity of influenza A viruses obtained from outbreaks and samples collected during epizootic monitoring in Spain and Portugal was also discussed, as well as the first case of the H1N1 pandemic subtype in a herd of pigs in Belgium.

Newcastle disease virus (NDV), member of the Paramyxoviridae family and avian influenza virus (AIV), member of the Orthomyxoviridae family, are two main avian pathogens causing serious economic problems in poultry health. Both are enveloped, single-stranded, negative-sense RNA viruses and cause similar symptoms, ranging from sub-clinical infections to severe diseases, including decrease in egg production, acute respiratory syndrome, and high mortality. Similar symptoms hinder the differentiation of infection with the two viruses by standard veterinary procedures like clinical examination or necropsy. To overcome this problem, we have developed a new duplex real-time PCR assay for the detection and differentiation of these two viruses. Eighteen NDV strains, fourteen AIV strains, and twelve other (negative control) strains viruses were isolated from allantoic fluids of specific pathogen-free (SPF), embryonated eggs. Four-weeks-old SPF chickens were co-infected with both viruses (NDV - LaSota and AIV - H7N1). Swabs from cloaca and trachea were collected and examined. The results obtained in this study show that by using duplex real-time PCR, it was possible to detect and distinguish both viruses within less than three hours and with high sensitivity, even in case a bird was co-infected. Additionally, the results show the applicability of the real-time PCR assay in laboratory practice for the identification and differentiation of Newcastle disease and influenza A viruses in birds.

Opis:
Demographic changes and the development of transportation contribute to the rapid spread of influenza. Before an idea of a 'person to person' spread appeared, divergent theories were developed to explain influenza epidemics in the past. Intensified virological and serological tests became possible after isolation of the human influenza virus in 1933. The first influenza virus detection methods were based on its isolation in egg embryos or cell lines and on demonstration of the presence of the viral antigens. Molecular biology techniques associated with amplification of RNA improved the quality of tests as well as sensitivity of influenza virus detection in clinical samples. It became possible to detect mixed infections caused by influenza types A and B and to identify the strain of the virus. Development of reliable diagnostic methods enabled fast diagnosis of influenza which is important for choosing an appropriate medical treatment.

Electrochemical biosensors have emerged as reliable analytical devices suitable for pathogen detection. Low cost, small sample requirement and possibility of miniaturization justifies their increasing development. Thus, we report in this review on the state of the art of avian influenza virus detection with genosensors and immunosensors working by an electrochemical mode. Their working principles focusing on the physical properties of the transducer, the immobilization chemistry, as well as new trends including incorporation of nanoparticles will be presented. Then, we critically review the detection of avian influenza virus in the complex matrices that use electrochemical biosensors and compare them with traditionally applied methods such as ELISA or Western blot.

Analyses and visualizations by the ISSCOR method of influenza virus hemagglutinin genes of different A-subtypes revealed some rather striking temporal relationships between groups of individual gene subsets. Based on these findings we consider application of the ISSCOR-PCA method for analyses of large sets of homologous genes to be a worthwhile addition to a toolbox of genomics - allowing for a rapid diagnostics of trends, and ultimately even aiding an early warning of newly emerging epidemiological threats.

Swine influenza virus (SIV) of H1N1 and H3N2 subtypes are dominated in European pigs population. "Classical swine" H1N1 subtype was replaced by "avian-like" H1N1 subtype. It co-cir- culates with H3N2 reassortant possessing "avian" genes. In the present study, 41 SIV strains isolated from pigs with pneumonia, raised in 20 Polish farms, were identified and characterised. Since it was evidenced that isolates from the same geographic district and the same year of isolation are in 100% similar, 15 strains representing different district and different year of isolation were chosen to construct phylogenetic trees. Two genes, conservative matrix 1 (Ml) and the most variable, haemagglutynin (HA), were sequenced and subjected into phylogenetic analysis. The results of the analysis confirmed that "avian-like" swine H1N1 strains evolved faster than classical SIV strains. HA gene of these isolates have been derived from contemporary strains of "avian-like" SIV. In contrast, the Ml gene segment may have originated from avian influenza viruses. H3N2 strain is located in swine cluster, in the main prevalent European group of H3N2 isolates called A/Port Chalmers/l/73-like Eurasian swine H3N2 lineage, which has evolved separately from the human H3N2 virus lineage around 1973.
