Opis:
Aloe vera [Aloe vera (L.) Burm. f.] is considered a valuable medicinal plant worldwide due to its remarkable beneficial effects on human health. However, challenges in A. vera propagation hinder meeting the increasing demand in the health and beauty sectors. As an alternative method, in vitro propagation is crucial for the mass production of Aloe plants, which is a rapid method as well. Therefore, the present study aimed to establish an efficient micropropagation protocol for A. vera by in vitro optimization of the effect of different plant growth regulators (PGRs). For shoot proliferation, sterilized explants were inoculated on the Murashige and Skoog (MS) medium supplemented with 6-benzylaminopurine (BAP) and thidiazuron (0.5, 1.0, 2.0, and 4.0 mg/l) in combination with 0.5 mg/l naphthaleneacetic acid (NAA). Subsequently, indole-3-butyric acid (IBA) (1.0, 2.0, and 3.0 mg/l) was used for root induction. It was found that the explants cultured on the MS medium supplemented with 4.0 mg/l BAP + 0.5 mg/l NAA showed the highest percentage of response (90 ± 1.29) for shoot induction within the minimum number of days (5 ± 0.33). The highest number of shoots (2.7 ± 0.36) and length of shoots (4.7 ± 0.42 cm) per explant were also observed with the same concentration of PGRs. However, the highest number of roots (3.2 ± 0.57), length of roots (5.67 ± 0.21 cm), and root induction (80 ± 1.97 %) were noticed within the minimum number of days (11 ± 0.79) on the MS medium supplemented with 1.0 mg/l IBA. Thus, the proposed method is a quick and effective approach for the mass propagation of A. vera with appropriate dosages of auxins and cytokinins, which may allow meeting the increasing commercial demand.

Coexisting microorganisms are abundant in nature. Plant growth promoting rhizobacteria (PGPR) is a group of beneficial microorganism living around the roots of plants which are able to confer beneficial effects on plant growth. Streptomyces sp. is a gram-positive bacteria as PGPR that can promote plant growth and enhance tolerance in adverse environment. This research was aimed to study the effects of plant growth promotion and stress tolerance of Streptomyces sp. in Arabidopsis and Brassica sp. The amount of indole-acetic acid (IAA) and phosphate solubility were assessed from isolated bacterial. Plant growth promotion was examined in 10-days old seedling with three independent experiments. Our results showed that Streptomyces sp. produced moderate levels of IAA and it was able to solubilize phos- phate. Inoculation of Streptomyces sp. enhanced lateral root number, fresh weight and chlorophyll content in Arabidopsis thaliana. Moreover, the inoculation of Streptomyces sp. significantly increased vegetative growth on Arabidopsis and Brassica sp. by producing higher fresh weight and chlorophyll content. Streptomyces sp. also enhanced tolerance to abiotic stress in Arabidopsis and Brassica sp. by increasing fresh weight under condition of salt and heat stress. Under salt stress, inoculation of Streptomyces sp. in Arabidopsis in- duced activity of catalase enzyme and decreased hydrogen peroxide (H 2 O2 ) and malondial- dehyde (MDA) production. In the molecular levels, Streptomyces sp. induced protein accu- mulations in Arabidopsis including nitrogen assimilation (GS1), carbohydrate metabolism (cFBPase), and the light-harvesting chlorophyll (Lhcb1) protein.

In the present study, the effects of 10, 20, 30 ppm hormone mixtures (indole-3-acetic acid + + gibberellic acid + kinetin) with 0.1, 0.3, 0.5 and 1 ppm zinc (Zn) concentrations alone and their mixtures on the cambial activity of sour cherry (Cerasus vulgaris Miller) cuttings were investigated. Morphological and anatomical developments of the plants were observed. The leaves of the plants treated with zinc were found to be greener than the control. Plants treated with zinc faded earlier than the control. The cambial zone thickness, the cambial zone cell line, the radial and tangential lengths of the cambial zone cells decreased with increasing concentrations of zinc and increased with increasing concentrations of hormones. The radial and tangential wall widths of the cambial zone cells increased with increasing zinc concentrations and decreased with increasing hormone concentrations. As a result, in the 0.1, 0.3, 0.5 and 1 ppm Zn concentrations, the cambial zone thickness decreased by 10, 28, 50 and 65%, respectively, compared to the control. Thirty ppm hormone mixture – H.M. (indole-3-acetic acid + gibberellic acid + kinetin) increased the cambial zone thickness by 65, 15, 5% in 0.1, 0.3 and 0.5 Zn, respectively, compared to the control. It was found that plant hormones importantly improved the harmful effects of zinc on the cambial activity of the plant cuttings.

Opis:
A modular three step synthetic approach of N-methyl indole derivatives has been carried out by the condensation of N-methyl indole with cyanoacetic acid using acetic anhydride as solvent to yield 3-cyanoacetyl N-methyl indole, which further reacts with carbon disulphide and methyl iodide in basic condition to obtain 2-(1-methyl-1H-indole-3-carbonyl)-3,3-bis(methylthio)acrylonitrile as a scaffold. Subsequent, the scaffold when reacts with substituted various amine derivatives via desulfitative displacement forms new derivatives of 3-((substitutedphenyl)amino)-2-(1-methyl-1H-indole-3-carbonyl)-3-(methylthio)acrylonitrile in moderate to good yields. All the novel compounds were evaluated for their antimicrobial activity against Gram+ve and Gram-ve bacteria and different fungal species which demonstrated well to moderate antimicrobial activity.

Arthrospira platensis (Spirulina platensis) is a well-known microalga and has been utilized as a medicinal agent and foodstuff by humans since at least 16th century. The aim of this study was to determine zinc content as well as determine phenolic and indole compounds from commercial preparations containing Arthrospira platensis (lyophilizate, tablets, and capsules) before and after extraction with methanol and incubation with artificial digestive juices. The secondary aim of this study was to evaluate the quality of these preparations. The samples were incubated in artificial stomach juice and in intestinal juice. The samples were mineralized and their zinc(II) ions content was estimated using flame absorption atomic spectroscopy (F-AAS). The maximum zinc(II) ions content released into the digestive juices was found to be up to 1.6 mg 100 g-1 of the preparation. Phenolic compounds identified in the examined extracts are as follows: gallic acid; protocatechuic acid; 3,4-dihydroxyphenylacetic acid; p-hydroxybenzoic acid; syringe acid; cinnamic acid; and quercetin. Furthermore, indole compounds identified were 5-hydroxy-L-tryptophan, 5-methyl-L-tryptophan, L-tryptophan, tryptamine, and 5-methyltryptamine. Consequently, it was also found that the distributed Arthrospira platensis in the form of tablets does not disintegrate in the artificial digestive juices. Among the examined preparations, only hard capsules met the requirements of the European Pharmacopeia 8th ed.

Research of the identification and determination of alkaloid contents in the aboveground parts of Phalaris arundinacea (Poaceae) during various periods of the vegetation season was conducted. Analyses were performed using an apparatus system by Hewlett-Packard (USA), consisting of a gas chromatograph model 589011 and a mass detector model 5971 A. Analyses were conducted at the laboratory of the phytochemistry at the Institute of Bioorganic Chemistry, PAS in Poznań. A total of six alkaloids were identified in the analysed material, i.e. tryptophol and gramine (indole alkaloids) as well as lupanine and 13-OH lupanine and lupanine esters – 13α- isovalericlupanine and 13α-tigloyloxylupanine (quinolysidine alkaloids). Gramine and lupanine are the dominant alkaloids in the growth stage.
Badania dotyczące identyfikacji i określenia zawartości alkaloidów w części nadziemnej rośliny pod względem ilościowym i jakościowym, przeprowadzono u Phalaris arundinacea w różnych okresach sezonu wegetacyjnego na aparacie firmy Hewlett - Packard (USA), w skład którego wchodzi chromatograf gazowy model 589011 oraz detektor masowy model 5971 A, w pracowni Fitochemii Instytutu Chemii Bioorganicznej PAN w Poznaniu. Zidentyfikowano występowanie 6 alkaloidów: tryptofol i graminę (alkaloidy indolowe) oraz lupaninę i 13 -OH lupaninę oraz estry lupaniny: 13α-izowaleroksylupaninę i 13α-tigloyloksylupaninę (alkaloidy chinolizydynowe). Dominującym alkaloidem w fazie wzrostu była gramina, a także lupanina.

Opis:
The effects of the phytohormone indole-3-acetic acid and various conditions of illumination on the growth of Parachlorella kessleri were investigated. Two variants of illumination: continuous and photoperiod 16/8 h (light/dark) and two concentrations of the phytohormone – 10-4 M and 10-5 M of indole-3-acetic acid were used in the experiment. The results of this study show that the addition of the higher concentration of indole-3-acetic acid stimulated the growth of P. kessleri more efficiently than the addition of the lower concentration of indole-3-acetic acid. This dependence can be observed in both variants of illumination. Increased biomass productivity was observed in the photo-period conditions. Both the addition of the phytohormone and the conditions of the illumination had an impact on the number of P. kessleri cells. An increased number of cells was observed under the conditions of continuous illumination. This result has shown that the continuous illumination and the higher concentration of the phytohormone stimulated the growth of P. kessleri more effectively than the shorter duration of light (16/8 h (light/dark)).

The research goals of presented work consisted of determination of zinc and indole compounds that are released into artificial digestive juices from Tricholoma equestre species. During the experiment freeze-dried samples of wild growing fruiting bodies and mycelia from in vitro cultures from liquid Oddoux medium were extracted to artificial digestive juices (saliva, gastric juice and intestinal juice). In the next step the determination of examined compounds was done by RP-HPLC for indole compounds and DP ASV for zinc. Furthermore it was decided to check weather this species has any beneficial properties for health. The extraction of researched material in conditions imitating human digestive tract allows determination of true amounts of elements released to artificial digestive juices and their beneficial influence (a specially designed and constructed apparatus Gastroel-2014 was applied). The indole compound with the highest quantity was 5-hydroxy-L-tryptophan, both in fruiting bodies and in biomass from in vitro cultures of T. equestre (up to 352.47 mg/100 g d.w.). Serotonin and L-tryptophan was determined in all analysed samples, but their amounts were significantly lower than the ones found for 5-hydroxy-L-tryptophan. The amounts of zinc on the other hand showed that the biggest concentration of zinc can be obtained for fruiting bodies and biomass from in vitro cultures to both artificial saliva and gastric juice after 120 minutes of digestion (6.83 14.4 mg/100 g d.w. retrospectively) in conditions that imitate human digestive track.

Based on overlapping structural requirements for both efficient aldose reductase inhibitors and PPAR ligands, [5-(benzyloxy)-1H-indol-1-yl]acetic acid (compound 1) was assessed for inhibition of aldose reductase and ability to interfere with PPARγ. Aldose reductase inhibition by 1 was characterized by IC50 in submicromolar and low micromolar range, for rat and human enzyme, respectively. Selectivity in relation to the closely related rat kidney aldehyde reductase was characterized by approx. factor 50. At organ level in isolated rat lenses, compound 1 significantly inhibited accumulation of sorbitol in a concentration-dependent manner. To identify crucial interactions within the enzyme binding site, molecular docking simulations were performed. Based on luciferase reporter assays, compound 1 was found to act as a ligand for PPARγ, yet with rather low activity. On balance, compound 1 is suggested as a promising lead-like scaffold for agents with the potential to interfere with multiple targets in diabetes.
