Opis:
In this research, the response of the plant Gypsophila arrostii Guss. to boron (B) under in vitro conditions was examined. The seeds were cultured on MS medium including 0, 10, 20, 40, 80 mg B l-1. Seedlings obtained from germinated seeds and grown in a culture medium for 8 weeks were analyzed. At the end of this period, stem length (cm), root length (cm), plant weight (g) and elemental content (mg kg-1) of the plants were determined. According to the results, the seeds of G. arrostii Guss. could germinate on media with up to 80 mg B l-1, and the seedlings demonstrated an ability to survive, albeit poorly, a dose of boron as high as 80 mg B l-1. In the experiment, the highest stem length (7.5 cm) was obtained from the 20 mg B l-1 treatment and the highest stem fresh weight (0.9 g) and stem dry weight (0.19 g) were measured in the 10 mg B l-1 variant. No significant statistical difference was determined between the boron treatments in terms of root length, root fresh weight and root dry weight. Our evaluation of the elemental content of plants demonstrated that the amount of boron in the root and stem increased parallel to its increase in the growth media. In the 80 mg B l-1 treatment, 601.9 mg kg-1 boron in root and 1,035.4 mg kg-1 boron in stem were determined. Besides, it was discovered that the contents of K, Mg, Zn, Na in root decreased while the contents of P, B, Mn, Cu in root increased in response to the growing amount of boron in the environment. In response to the increasing boron concentrations, the content of K, P, Mn, Cu, Zn and S increased while the amount of Ca, Mg and Na in the plant stem decreased. Consequently, G. arrostii Guss. was found to be a boron hyperaccumulator, collecting boron in tissues (in the roots and stems), in which it resembled some other types of Gypsophilla.

Opis:
Iris species have great economic importance for their use in ornamental and pharmaceutical industry. Flora of Turkey reports about 43 local taxons of which 16 are endemic. Iris suaveolens Boiss. et Reuter is an endemic species with high seed dormancy and has high tolerance to cold and drought. This study focused on seed dormancy break of I. suaveolens under in vitro conditions. In the first experiment, the seeds were given stratification treatment on MS medium containing different concentrations of BAP with or without NAA. The 2nd experiment reports effects of alternative combinations of cold (at 4°C, 24 h dark) – warm (16 h light photoperiod) conditions on medium containing different concentrations of plant growth regulators on agar solidified MS medium. Both experiments showed about 8.33% seed germination against 64.5% seed viability as confirmed by tetrazolium test. All germinating seeds were abnormal and very weak. In the third experiment, 10 minutes acid scarified seeds cultured at 24°C in 16 h light photo period on MS medium achieved 60.0% germination. Similar treatment under cold + dark condition was inhibitory and failed to show identical results. This underlines the importance of acid scarification, photoperiod and warm treatments of I. suaveolens seeds to break seed dormancy. All germinated seeds showed normal growth and development under field conditions.

Explants of apical buds of sandy everlasting Helichrysum arenarium L. Moench sterilized with calcium hypochlorite – Ca(OCl₂) were placed onto Murashige- -Skoog (MS) growth medium enriched with 1 mg∙dm⁻³ KIN (kinetin) in two consecutive passages. To optimize the procedure of in vitro micropropagation, the axillary shoots of Helichrysum arenarium L. Moench were transferred during the third passage onto 9 different combinations of MS medium without plant growth regulators (PGRs-free medium), with different concentrations of KIN (1.0, 3.0, 4.0, 5.0 mg∙dm⁻³) and KIN (1.0, 3.0, 4.0, 5.0 mg∙dm⁻³) with IAA (indole-3-acetic acid) (0.5 mg dm⁻³). The highest mean number of shoots (24.7) was observed on MS medium with 5 mg∙dm⁻³ KIN and 0.5 mg∙dm⁻³ IAA. The achieved branched shoots were rooted and acclimatized. Rhizogenesis was intensified by the presence of growth hormones: 0.5 mg∙dm⁻³ IAA or 0.5 mg∙dm⁻³ IBA (indole-3-butyric acid). At the stage of acclimation of plantlets the application of a water solution of MS salts (25%) for watering the plants increased the efficacy of plant acclimation from 56 to 75%.
Uzyskane pędy boczne poddano ukorzenianiu i aklimatyzacji. Proces ryzogenezy intensyfikowała zawartość regulatorów wzrostu 0,5 mg∙dm⁻³ IAA i 0,5 mg∙dm⁻³ IBA (kwas indolilo 3-masłowy). Na etapie aklimatyzacji mikrosadzonek zastosowanie do nawadniania 25% roztworu soli MS spowodowało poprawę efektywności aklimatyzacji roślin z 56 do 75%.

Opis:
There are limited published data on in vitro reproduction of Fagus sylvatica L. (European beech). This study was aimed to determine the efficiency of induction of somatic embryogenesis or organogenesis of beech from different types of explants in various culture conditions. Explants derived from immature, fresh seeds (collected in 2011 and 2013) and from mature seeds, stored at –10ºC and some stratified at 3ºC, were placed on induction media with various combinations of plant growth regulators: zeatin, 2,4-dichlorophenoxyacetic acid (2,4-D) and/or benzyladenine (BA). Initial cultures were kept in darkness or weak light (white fluorescent or blue-red LED). Limited success has been achieved in initiation of somatic embryogenesis. We obtained friable, yellow-white callus with characteristic PEM-like structures (cPEM-ls, from embryonic axes or fragments of immature embryos with embryonic axes), which may be an early developmental stage of embryogenic callus of Fagus sylvatica. This type of callus regenerated from explants incubated in darkness, mainly on WPM medium with addition of 6.8 μM zeatin or WPM and MSG media with 9.1 μM 2,4-D and 2.2 μM BA. The highest frequency of regeneration of callus with cPEM-ls was 5%. Instead, we succeeded to induce organogenesis from both immature and mature zygotic embryos and from embryonic axes. The best results were obtained for mature zygotic embryos incubated on ½WPM medium (half-strength Woody Plant Medium) with 9.1 μM 2,4-D and 2.2 μM BA. Adventitious buds were regenerated on up to 15% of the explants. The induced buds developed into shoots, enabling us to establish tissue cultures of beech. Induction of organogenesis from the tested explants was more efficient than induction of somatic embryogenesis.

Opis:
Plant growth throughout the world is often limited by unfavourable environmental conditions. This paper reports results of a study on long- and short-term osmotic stress (−0.5 MPa) followed by a recovery on in vitro translational capacity of polysomes and on the composition of polysome-associated proteins in germinating pea (Pisum sativum L.) seeds. Here we show that, under osmotic stress, cytoskeleton-bound polysomes were charaterized by the highest translation activity, which may be indicative of an important role that this population of polysomes plays in the synthesis of the so-called “stress proteins”. We also find out that in response to osmotic stress, new proteins (22.01, 96.47 and 105.3 kDa), absent in the unstressed sample, associated with the total pool of polysomes, whereas the protein of 22.95 kDa, which was present in the embryonic tissue of seeds germinating under unstressed conditions, disappeared. These changes may have affected both the stability and the translational capacity of polysomes.

Opis:
In this study, an attempt was made to investigate in vitro morphogenetic competence of three shrub species from the Thymelaeaceae family. The studied plant material originated from Russia, Greece and China, and the effectiveness of in vitro shoot formation and rhizogenesis of Daphne caucasica, D. jasminea, and D. tangutica was verified. The multiplication coefficient was compared for different propagation media. Medium composed of WPM mineral salts, MS microelements and a set of vitamins, supplemented with 1.0 mg dm-3 2iP, 0.1 mg dm-3 NAA, and 0.65 g dm-3 calcium gluconate, was appropriate for micropropagation of the tested genotypes. Shoot propagation in medium containing B5 vitamins and microelements was not as effective as on WPM/MS medium. The rooting phase, especially in D. tangutica, needs further optimization in order to reduce the costs associated with acclimatization of microplantlets obtained to in vivo conditions. After stabilization, the plants were successfully cultivated under greenhouse conditions.
Badano kompetencję morfogenetyczną trzech gatunków roślin krzewiastych z rodziny wawrzynkowatych. Materiał roślinny pochodził z Rosji, Grecji i Chin. Regenerowano in vitro pędy i korzenie przybyszowe u wawrzynka kaukaskiego (Daphne caucasica), wawrzynka jaśminowego (D. jasminea) oraz wawrzynka tanguckiego (D. tangutica). Etap ukorzeniania, szczególnie dla wawrzynka tanguckiego, wymaga dalszej optymalizacji w celu zmniejszenia kosztów ponoszonych w trakcie uzyskiwania materiału przydatnego do uprawy. Otrzymane drogą rozmnażania in vitro rośliny przystosowano do uprawy w warunkach szklarniowych.
