The aim of this study was to determine the time-dependent effectiveness of photo-stimulation against bovine sperm cells using a multi-wavelength LED (Light Emitting Diode). Spermatological parameters were evaluated for viability, acrosome structure and motility. In addition, the effect of photo-stimulation on frozen-thawed sperm cells subjected to in vitro capacitation was evaluated in terms of changes in mitochondrial membrane potential. The study consisted of two separate experiments and a total of 32 sperm samples obtained from separate bulls were used. All sperm samples were obtained from Holstein bulls using an artificial vagina. Semen was diluted to a final concentration of 92 x 106 spermatozoa per ml in 0.25 ml straws. The sperm cells were frozen using the conventional method. Straws were kept in a 37°C water bath for 20 seconds and diluted 1:4 in phosphate buffered saline (PBS) to eliminate the potentially deleterious effect of glycerol, the main permeable cryoprotectant in the freezing medium for bull sperm. This dilution also helped in the evaluation of sperm quality parameters. In the first experiment, whereas the 15-10-15 showed no differences with the control, other treatments such as 10-10-10, 5-5-5, and 3-1-3 exhibited significantly higher percentages of viable spermatozoa at 24h. The results obtained for acrosome integrity were pretty much similar to those observed in the sperm viability assessment. In effect, while the treatment consisting of 15-10-15 had no positive effects, shorter treatments exerted a much more positive effect. The percentages of acrosome-intact spermatozoa in 2-1-2 and 1-1-1 were significantly higher than those obtained in the control. The significant differences in mitochondrial membrane potential were observed at 0, 2, 4 and 24h post-photo-stimulation in all treatments, except 15-10-15. The highest increase in the percentage of spermatozoa exhibiting high mitochondrial membrane potential was found in 10-10-10, 5-5-5 and 3-1-3 treatments. With regard to total and progressive motility, whereas 10-10-10 was the best regime, 5-5-5 and 3-1-3 treatments also had a positive effect. However, 15-10-15 appeared to have a stimulating effect upon progressive motility at 2h and 4h but later declined and showed no significant differences with regard to the control at 24h. In the second experiment, not immediately after thawing but after having been kept at room temperature for up to 24h, it was observed that there was no statistical difference in terms of viability, acrosome integrity and total/progressive motility between photostimulation and the control group. This indicates that photo- stimulation is less able to exert a beneficial effect when post-thawed sperm are not immediately stimulated. As a result it was determined that photo-stimulation at a pattern of 10-10-10, 5-5-5, 3-1-3 and, to a lesser extent 2-1-2, increases the resilience of frozen-thawed bull sperm when applied upon thawing.

In this paper, herbal bitters are widely used due to their numerous acclaimed health benefits in many Nigerian homes; however, many have not been subjected to scientific scrutiny. The aim of this study was to determine the phytochemical composition and antioxidant capacity of a non-alcoholic polyherbal formulation, Yoyo bitters, towards validating its broad pharmacological claims. The phytochemical components of Yoyo bitters were ascertained by phytochemical screening assays and gas chromatography-mass spectrometry (GC-MS). The antioxidant activity was investigated in vitro using 2,2-diphenyl-1-picryhydrazyl (DPPH) radical, hydrogen peroxide (H2O2) scavenging activity, total antioxidant capacity (TAC) and ferric reducing antioxidant power (FRAP) assays. Qualitative phytochemical analysis of Yoyo bitters showed the presence of saponins, tannins, flavonoids, terpenoids, cardiac glycosides and anthocyanins. The total phenols, flavonoids, flavanols, tannins and carotenoids content were 14.741 ±0.64 mg GAE/ml, 0.152 ±0.01 mg RE/ml, 0.437 ±0.02 mg RE/ml, 0.368 ±0.04 mg TAE/ml and 0.016 ±0.00 mg CAE/ml respectively. GC-MS chromatogram revealed the presence of forty-three (43) phytochemical compounds with D-allose (41.81%), 1,6-anhydro-beta-D-glucofuranose (24.15%), 5-hydroxymethylfurfural (8.02%) and Z-6-pentadecen-1-ol acetate (3.50%) as the most abundant constituents. Yoyo bitters demonstrated effective antioxidant activity against DPPH and H2O2 with IC50 values of 0.492 mg/ml and 0.629 mg/ml respectively compared to ascorbic acid of 0.161 mg/ml and 0.130 mg/ml respectively. Total antioxidant capacity and ferric reducing antioxidant power of Yoyo bitters were 0.432 mg AAE/ml and 2.236 mg AAE/ml respectively. This study validates the antioxidant capacity of Yoyo bitters and provides chemical basis for its acclaimed pharmacological actions.

Introduction. Cladosporium is one of the most abundant genera of environmental fungi worldwide and a very common respiratory allergen. To-date, many C. cladosporoides infections have been identified. Risk factors for C. cladosporioides infection are primarily injuries, metabolic disorders, organ transplantation and autoimmune diseases, among others. Objective. The aim of the study was to assess the type of pharmacodynamic interactions between fluconazole and some selected essential oils: orange, mandarin, lemon and grapefruit, in an in vitro study against C. cladosporioides. Materials and method. Experiments were carried out using the plate cultivation method. Fluconazole was tested against C. cladosporioides at concentrations ranging from 0.05–3.3 mg / ml, and the activity of essential oils added to PDA medium at concentrations ranging from 1–30%. The dose-effect curves for the collected results were determined with by the log- probit method. Isobolographic analysis of the results allowed determining the type of interactions between fluconazole and the tested essential oils. Results. Lemon essential oil was the most active, and in a concentration of 1% it inhibited the growth of C. cladosporioides by 21%. Isobolographic analysis showed that the combination of fluconazole with orange and grapefruit essential oil had an additive interaction, and with mandarin and lemon – an additive interaction with a tendency to synergy in the plate culture test for C. cladosporioides. Conclusions. The use of isobolographic analysis can contribute to the introduction of natural substances with the desired activities into the pharmacotherapy of many infections and diseases. The use of natural substances can also help to reduce the number of side-effects caused by conventional and standard therapies.

Opis:
Phytochemicals and antioxidant potentials of chloroform extract of leaves of Cyathea latebrosa have been investigated. The metabolites detected are Carbohydrates, starch, glycosides, saponins, alkaloids, tannins, flavonoids, sterols, terpenes, and polyuronides. Quantitative Phytochemical analysis gave alkaloid (0.08×102 ±0.02), saponins (0.16×102 ±0.02), tannin (0.11×102 ±0.01), flavonoid (0.16×102 ±0.01), and phenol (0.18×102 ±0.01). FT-IR (KBr) cm-1 analysis showed -NH-(3433.41 weak), -OH- (3851.01 sharp), -C-F (1408.8 strong) –SO-(1101.39) and Ar (1646.3 weak), GC/MS characterization revealed the extract contained [2-Aziridinylethyl]amine (70.7%), Benzeneethamine, 3-fluoro-β,5-dihydroxy-N-methyl (14.0%), N-Methyltaurine (10.7%) and (R)-(-)-2-Amino-1-propanol (3.15%). The presence of these compounds exposed the leaves as a potential remedy for alleviated cold, cancer, malaria and to regulate immune functions. At 400 µg/ml concentration the extract and ascorbic acid produced 54.34% and 96.02% antioxidant activity respectively. The FRAP values were significantly (p < 0.05) low than the value of ascorbic acid (2µ M). The plant extracts showed low antioxidant potential even on higher concentrations when compared to reference standard ascorbic acid.

The main aim of the study was to determine which of the viability and germination capacity of pollen grains estimation methods are optimal and to test if the studied sections and species within the Rosa genus are differentiated in pollen viability andgermination capacity. We analysedandcomparedpollen grains of 14 wild Rosa species using 5 viability tests and13 various liquidandagar media. Viability of pollen grains in the majority of the examinedspecies was greater than their germination capacity. The most viable pollen grains were foundin R. gallica (section Gallicanae), where average of 5 viability tests is 90.69%, andin R. pendulina (section Rosa) – 86.85%. Species from section Caninae have a lower level of viability (from 60.59% in R. rubiginosa to 31.23% in R. inodora). Pollen of species from sections Gallicanae (R. gallica; to 63.4%) and Rosa (R. pendulina; to 47.21%) germinatedmuch better than the pollen of the majority of species from section Caninae (with the exception of R. zalana; to 47.22%). The examinedpollen grains germinatedmost numerously on agar medium with 1.5% agar+15% sucrose+50 ppm boric acidandliquidmed ium with 15% sucrose+ 50 ppm boric acid. Following deep-freeze storage (–25°C) for six months, the pollen grains of nine selected species (with the exception of R. rubiginosa) showed viability higher or similar to that before storage.

Acetylsalicylic acid (ASA) and α-2-pyrazylidene-α-cyano N-butyl acetamide (PD-101), a new antiaggregatory pyrazine derivative were tested for their genotoxicity in human lymphocytes in vitro using the sister chromatid exchange (SCE) technique. Both compounds were found to be inactive in inducing SCE in concentration from 1 µM up to 1000 µM. The agents displayed inhibitory effect on cell kinetics.

The aim of the study was chemical analysis of polysaccharide fractions from sporocarps of Sarcodon imbricatus collected in natural sites and from the mycelium of in vitro cultures. Three polysaccharide fractions (FOI, FOII, FOIII) were isolated from sporocarps and two (FKI, FKII) from in vitro cultures. Qualitative analysis by HPLC method showed that they are composed of galactose and fucose (FOI, FKI) or glucose and fucose (FOII, FKII). FOIII fraction of the sporocarps consisted of glucose only. Molecular weights of isolated fractions ranged from 3.8 to 16.3 kDa for fractions from the sporocarps and from 5.8 to 14.7 kDa for that ones isolated from in vitro culture. The total percentage of sugar content for all fractions ranged from 97.8% to 99.1%. The percentage of uronic acids contents in acidic fractions was 2.6% and 2.7% for the FOI and FKI respectively. The work included also an assessment of cytotoxic activity of polysaccharide fractions in relation to tumor cell lines of human breast cancer MCV-7. FOI polysaccharide fraction of the sporocarps inhibited the growth of cancer cells in 50% compared to the control at a concentration of 0.0125%, while the polysaccharide fraction FKI from in vitro cultures inhibited cell growth in a concentration of 0.016%.
Przedstawiciele gromady Basidiomycota – której reprezentantem jest Sarcodon imbricatus (L.: Fr.) P. Karst., są ważnym źródłem związków aktywnych biologicznie. W ich owocnikach wykryto m.in. związki o działaniu przeciwwirusowym, przeciwbakteryjnym, fungistatycznym, a także przeciwnowotworowym. Najlepiej poznanymi metabolitami grzybowymi są polisacharydy. Coraz częściej podejmowane są badania nad składem chemicznym nie tylko owocników, ale także grzybni pozyskanej z kultur in vitro. W badaniach z tego zakresu dowiedziono istnienia jakościowych i ilościowych różnic w produkcji niektórych grup związków chemicznych przez owocniki i grzybnię z kultur in vitro. Obiektem przeprowadzonych badań były owocniki Sarcodon imbricatus zebrane na terenie lasów świerkowych w roku 2008. Z warstwy hymenialnej owocnika wyprowadzono kultury in vitro, które prowadzono, jako płynne, wytrząsane. W ramach przeprowadzonych badań wyizolowano 3 frakcje polisacharydowe (FOI, FOII, FOIII) z owocników i 2 (FKI, FKII) z mycelium z kultur in vitro. Analiza jakościowa z wykorzystaniem metody HPLC wykazała, że składają się one z galaktozy i fruktozy (FOI, FKI) oraz glukozy i fruktozy (FOII, FKII). Frakcja FOIII z owocników składała się wyłącznie z glukozy. Oznaczono masy cząsteczkowe wyizolowanych frakcji. Wynosiły one od 3.8 do 16.3 kDa dla frakcji z owocników i od 5.8 do 14.7 kDa dla frakcji z kultur in vitro. Całkowita procentowa zawartość cukrów dla wszystkich frakcji mieściła się w przedziale od 97.8% do 99.1%, natomiast procentowa zawartość kwasów uronowych we frakcjach o charakterze kwaśnym wynosiła 2.6% dla frakcji FOI z owocników i 2.7% dla frakcji FKI z kultur in vitro. Druga część pracy obejmowała ocenę aktywności cytotoksycznej wybranych frakcji polisacharydowych w stosunku do linii komórek nowotworowych ludzkich raka sutka MCV-7 w teście in vitro. Frakcje polisacharydowe FOI i FKI wykazywały działanie hamujące na komórki nowotworowe. Frakcja polisacharydowa FOI z owocników hamowała wzrost komórek nowotworowych w 50% w stosunku do próby kontrolnej w stężeniu 0.0125%, natomiast frakcja polisacharydowa FKI z kultur in vitro hamowała wzrost komórek w stężeniu 0.016%.

Spontaneous parthenogenetic activation of bovine oocytes in an in vitro maturation and fertilization system (IVM/IVF) is described. Altogether, 1403 follicular oocytes, collected by the aspiration method, were matured in vitro and then cultured without insemination in the same conditions as a group of inseminated oocytes. After 48-72 h of additional culture, 141 oocytes (10%) were found to be spontaneously activated. Morphological evaluation revealed that the number of blastomeres within parthenotes ranged from 2 to 16 cells, with a minority (15.7%) comprising of 9-16 blastomeres. According to a cytogenetic analysis, only 1.2% of the analysed parthenotes consisted of more than 9 cells. Parthenotes may not be distinguished from embryos produced in vitro and spontaneous parthenogenetic activation in an IVM/IVF system indicates suboptimal culture conditions. A group of non-inseminated oocytes should be included in each experiment to serve as a control. Spontaneously activated bovine parthenotes only occasionaly developed beyond the 8-blastomere stage in a common IVM/IVF system. The incidence of parthenotes interferes with the efficiency of in vitro embryo production but it is doubtful whether it lowers the pregnancy rate after transfer of IVF embryos.

Five newly synthesised monophosphates of two pyrimidine acyclonucleoside series, namely 1-N-[(2’-hydroxy)ethoxymethyl] and l-N-[(l’,3’-dihydroxy)- 2’-propoxymethyl] derivatives of 5- and 5,6-alkylated uracils were tested in vitro for chromosome aberrations and sister chromatid exchanges (SCE). Metaphase plates were obtained via microculture of human lymphocytes from heparinized peripheral blood. The compounds were tested in doses: 10, 20, 40, 80 and 150 µg per mL of culture. The tested compounds induced mainly chromatid gaps, less frequently chromosome gaps. A low number of mitoses with chromatid and chromosome breaks, acentric fragments, dicentric chromosomes and exchange figures were also observed. The tested compounds in doses: 40, 80 and 150 µg per mL, doubled or tripled the percentage of cells with chromatid gaps and chromosome gaps as compared to the control. The percentage o cells with aberrations (excluding gaps) induced by the tested compounds in all doses did not exceed 2%. The tested compounds induced a higher number of SCE per cell but less than double frequency as compared to the control. SCE frequencies and replication index (RI) values varied depending on the examined compounds. For the highest dose of the tested compounds (150 µg per mL) a significant decrease in RI values was observed for l-N-[(2’-hydroxy)ethoxymethyl]-5,6-tetramethyleneuracil monophosphate and for l-N-[(2’-hydroxy)ethoxymethyl]-5,6-dimethyluracil monophosphate. So far, the results have indicated potential clastogenicity of all the tested compounds except acycloguanosine monophosphate.
